ABSTRACT

ETHNOPHARMACOLOGICAL RELEVANCE: Khaya species, endemic to Africa and Madagascar, continues to be valuable in indigenous traditional medicine. Their bitter tasting barks are decocted to treat fevers, several febrile conditions, microbial infections and worm infestations. In the Budongo rain forest of Western Uganda, non-human primates, especially chimpanzees and baboons, have been observed to eat the bitter non-nutritious bark and occasionally the seed. MATERIALS AND METHODS: Extracts were prepared by sequential fractionation with solvents of increasing polarities and assayed using standard procedures. Bioassay guided purification of the petroleum ether extract by column chromatography yielded three pure limonoids, Grandifolione (1), 7-deacetylkhivorin (2) and 1,3-deacetyldeoxyhavenensin (3). The antitrypanosomal, antileishmanial and antiplasmodial activities of pure compounds (1) and (2) were evaluated in vitro against Plasmodium falciparum K1, Trypanosoma brucei rhodesiense STIB 900, Trypanosoma cruzi trypomastigotes (Tulahuen C4), and axenic Leishmania donovani MHOMET-67/L82 and for cytotoxicity against L6 rat skeletal myoblast cells, in parallel with standard drugs. RESULTS: Of the four extracts tested, the petroleum ether extract showed activity against Plasmodium falciparum (IC50 0.955 µg/ml) and Trypanosoma brucei rhodesiense (IC50 5.72 µg/ml). The pure compounds (1) and (2) demonstrated activity against Plasmodium falciparum (KI strain) and marginal activities against Trypanosoma brucei brucei, Trypanosoma brucei rhodesiense, Trypanosoma cruzi and Leishmania donovani. CONCLUSION: The present study provides evidence justifying the use of Khaya preparations in traditional medicine to treat fevers and microbial infections. The observed antiprotozoal activity of grandifolione and 7-deacetylkhivorin from the seed of Khaya anthotheca further confirms the ethnomedicinal potential of this plant and supports the hypothesis that non-human hominids (chimpanzees and baboons) too, eat the bitter bark and seeds for self-medication and in general, the use of Khaya plant material for medication by humans in disease endemic tropical areas. The antiprotozoal activity of gradifolione, and, the antitrypanosomal and antileishmanial activities of 7-deacetylkhivorin are reported here for the first time.

ABSTRACT

The abundance and species diversity of tabanids were evaluated by trapping of insects using Vavoua traps, during the rainy season, from October 4 to November 30, 2009, in three different habitats: primary forest, secondary forest and village, in the biosphere reserve Ipassa-IRET Makokou in Gabon. Eight species belonging to three genera of tabanids have been identified for a total of 402 specimens caught. The tabanid species numerically the most abundant were: Tabanus secedens Walker, 1854 (55.2%), Tabanus obscurehirtus Ricardo, 1908 (13.9%), Chrysops dimidiatus Wulp, 1885 (11.2%) and Chrysops silaceus Austen, 1907 (10.7%). The less abundant species were Tabanus par Walker, 1854 (3.2%), Tabanus besti arbucklei Austen, 1912 (3%), Tabanus marmorosus congoicola Bequaert, 1930 (1%) and Ancala fasciata fasciata (Fabricius, 1775) (0.5%). Specimens of the genera Tabanus and Chrysops could not be identified, these insects represented respectively 0.7% and 0.5% of the insects trapped. The highest proportion of tabanids was trapped in secondary forest (75.1%) and the lower in primary forest (4.5%).

ABSTRACT

Despite ongoing preventive chemotherapy campaigns, intestinal schistosomiasis is hyper-endemic in shoreline communities living along Lake Albert, Uganda. To provide a deeper insight into the local epidemiology of Schistosoma mansoni, a variety of field-based studies were undertaken focusing upon schistosome-snail interactions and confirmation of transmission foci. Cercarial shedding patterns of field-caught Biomphalaria spp., as identified by morphology, were hourly observed over a ten day period and showed that Biomphalaria stanleyi produced significantly more cercariae than Biomphalaria sudanica. Peak production times in both species were between 12.00 and 14.00h indicating greatest infection risk from lake water exposure is during the early afternoon. Laboratory-bred snails were exposed to locally hatched miracidia and susceptibility of Biomphalaria spp. was confirmed experimentally. Biomphalaria stanleyi was a more permissive host. After ascertaining appropriate conditions for infection of laboratory mice, 28 groups of between 5 and 6 naïve mice were placed in floatation cages at four suspected shoreline transmission sites for a 30 minute period of exposure. Eight weeks later, mice (n=142) were culled and S. mansoni adult worms were retrieved from 10 animals. Taken as a whole, these observations highlight the local importance of B. stanleyi in transmission of intestinal schistosomiasis and clearly demonstrate the risk of infection on the Lake Albert shoreline. To mitigate this risk local environmental modification(s), i.e. improvement in sanitation and hygiene and control of snail populations, is needed to bolster the impact of chemotherapy-based interventions.

ABSTRACT

In Eastern Uganda, paddy-rice growing, which has only become popular in recent years, seems to be associated with the emergence of schistosomiasis mansoni as a new problem in public health. To estimate the magnitude of this problem, a cross-sectional, baseline survey was carried out in six villages of the Kibimba Rice Scheme. The overall prevalence of Schistosoma mansoni infection was found to be 20%. The highest prevalences and intensities of infection were seen in those aged 5-29 years, with more males infected than females. An attempt was then made to identify the important factors in the aetiology of S. mansoni in this area. Odds ratios indicated that working regularly in the rice paddies, fishing with baskets, and being male were statistically associated with an increased risk of S. mansoni infection. It is clear that schistosomiasis mansoni which is emerging as a new health problem in the study area is closely linked to working in the rice paddies. Encouraging the rice farmers to wear knee-high, waterproof boots while in the fields may help control the disease.
