In the article on discusses the problems of origin and duration of chlamydia and herpes-virus infection in children with chronic somatic pathology and different degrees of radiation stress in contaminated territories of Brianskaya region (the level of contamination according to 137Cs = 9.37-19.73 Ci/km2).

ABSTRACT

Presymptomatic DNA diagnosis of Huntington's chorea (HC) was made for two sons of a patient affected with the disease using amplification of the DNA fragment in the area of locus G 8 linked with HC gene. That fragment contains a polymorphous site in the area of restrictase recognition Hind III, being of information value as regards the family under examination. The familial analysis with the use of the DNA diagnosis data makes it possible to exclude the inheritance of HC gene for both the sons of the patient with a probability of 96%.

ABSTRACT

Duchenne muscular dystrophy carrier detection has been performed by using probes XJ1.1 (intragenic probe) and probe 754 for a girl. The carrier probability was estimated by means of a computer program GenRisk combining pedigree and DNA-probe data and turned out to be 95%.

ABSTRACT

Efficiency of transformation by a number of vectors with the different selective markers of a set of cell lines has been studied for three different methods based on using calcium phosphate, polybrene, or electroporation. Electroporation is shown to be the most efficient one. Using this method with the system rat2k-cells-pAGO vector we have obtained the frequencies of transformation up to 2-3.10(-3). We suggest to use this system as a model for investigation of homologous recombination in the framework of the gene therapy project.

ABSTRACT

A total of 1252 workers from 21 enterprises were surveyed by means of questionnaires. The survey uncovered grave shortcomings in the delivery of medical care at all levels. In particular, only 58.2 per cent of the interviewed were satisfied with the work of feldshers providing medical care at health posts. An analysis was also made of workers' suggestions regarding the work of shop physicians, chiefs of health units, administration, public organisations at enterprises and their working collectives.

ABSTRACT

The significance of procedures involved in the technique of differential staining of sister chromatids is estimated by quantitative autoradiography. Photolysis of bromodeoxyuridine-substituted DNA is shown to be the main process determining the quality of differential staining under certain experimental conditions. On the basis of modes provoking DNA degradation and washing DNA fragments off the chromosomes, a simplified method including only irradiation of chromosomes under a mercury lamp and treatment with a sodium citrate solution is proposed.

ABSTRACT

Using polyethylene glycol 6000 (PEG), cell fusion was obtained for the Chinese hamster transplantable culture. A most rational character of cell fusion efficiency was the amount of nuclei found in multinuclear cells (polykaryocytar index) determined within 30-60 minutes after PEG treatment of the culture. After a 1-6 hour treatment, a mass death of polykaryocytes was more than 8 hours; in 3 hours and up to the end of a 24 hour's period of observation an intensive cell division is observed.

ABSTRACT

The duration of the early stages of meiotic prophase was determined in the oogenesis of T. cristatus cristatus by means of autoradiography. The oocytes were being investigated during 39 days from the moment of 3H-thymidine injection. It was shown that preleptotene lasts 1--2, leptotene ca. 4, zygotene 5 and pachytene 26 days. When studying the preparations obtained 1 day after the injection of 3H-thymidine, the silver grains were found to be localized over the nuclei at all stages of meiotic prophase; this suggests the amplification of rDNA which begins in leptotene-zygotene and ends in early diplotene.

ABSTRACT

With the aid of 3-H-uridine radioautography, the effect of gonadotropic hormone choriogonin on the synthesis of RNA in the giant granular loop of XII chromosome of the Triturus cr. cristatus oocytes was studied in normal and after pretreatment of the cells with mutagen. Choriogonin, while obviously stimulating the RNA synthesis, has no effect on the cells pretreated with mutagen.
