ABSTRACT

O objetivo do estudo foi produzir anticorpos policlonais para vírus bovino e testar a reatividade em imunoensaios como imunofluorescência, imunoperoxidase e slot blot. Como imunógeno utilizou-se cepas e/ou isolados dos herpesvírus bovino tipo 1, 2 e 5 (BoHV-1, BoHV-2 e BoHV-5), vírus da diarreia viral bovina (BVDV), vírus respiratório sincicial bovino (BRSV), vírus da língua azul (BTV) e vírus vaccina (VACV) amplificados em cultivo celular. Os coelhos foram imunizados em intervalos regulares e o soro hiperimune produzido foi utilizado como anticorpo primário nos imunoensaios. A diluição de trabalho para cada antissoro foi determinada por diluição seriada limitante e variaram entre 1:800 e 1:51.200. As maiores diluições foram observadas para imunoperoxidase, quando comparadas com a imunofluorescência e slot blot. Diluições menores que 1:800 foram associadas com aumento de reações inespecíficas. Os antissoros anti-BoHV-1, -BoHV-5, -BVDV e -BRSV reagiram frente a isolados heterólogos em ensaios de imunofluorescência e imunoperoxidase. Conclui-se que os antissoros produzidos possuem elevadas concentrações de anticorpos específicos para os vírus e é uma alternativa para a utilização em imunoensaios.(AU)


The objective of the study was to produce polyclonal antibodies to bovine viruses and to test the reactivity in immunoassays such as immunofluorescence, immunoperoxidase and slot blot. Bovine viral diarrhea virus (BVDV), bovine respiratory syncytial virus (BRSV), bovine herpesvirus type 1, 2 and 5 (BoHV-1, BoHV-2 and BoHV-5) blue tongue virus (BTV) and vaccinia virus (VACV) were amplified in cell culture and used to immunize rabbits. Animals were immunized at regular intervals and the hyper immune serum produced was used as primary antibody in the immunoassays. The working dilution for each antiserum was determined by limiting serial dilution and varied from 1: 800 to 1: 51,200. The highest dilutions were observed for immunoperoxidase, when compared with immunofluorescence and slot blot. Dilutions lower than 1:800 were associated with the presence of nonspecific reactions. Anti-BoHV-1, -BoHV-5, -BVDV and -BRSV antisera reacted against heterologous isolates in immunofluorescence and immunoperoxidase assays. It is concluded that the produced polyclonal antibodies have high concentrations of virus-specific antibodies and are an alternative for use in immunoassays.(AU)

ABSTRACT

Background: Uterine tumors are classified according to their origin, epithelial or mesenchymal. These neoplasms are uncommon in most animal species, but are considered as important causes of reproductive disorders. In rabbits, the occurrence of these tumors increase with development of the animals and, although the numbers indicate a high occurrence of this disease in rabbits, reports are scarce. Therefore, this paper has the objective to describe a case of endometrial adenocarcinoma in a rabbit, emphasizing their histological features as well as provide a brief discussion about the epidemiological, clinical, pathological and prevention forms and treatment. Case: A 4-years old New Zealand rabbit breed, from a group of animals maintained only for breeding, had low fertility. The rabbit was twice covered by proven fertile animals, but no pregnancy, becoming unfit reproduction and subjected to euthanasia. At necropsy, tissue samples were collected, subsequently fixed in 10% formalin and then were processed routinely and stained with hematoxylin and eosin for histopathological analysis. Grossly, there was swelling of the uterine body, characterized by two masses that protruded from the endometrium, with rounded and soft look, and the court had cystic cavity filled with whitish gelatinous consistency of content. ABSTRACT

This paper aims to disclose the technique of cystopexy through cystostomy with introduction of foley catheter in the treatment of chronic recurrent perineal hernia with bladder content. Recurrence of perineal hernia is still challenge for surgeons and techniques to prevent or treat this complication should be disseminated. Thus, the realization of cystopexy aims to minimize relapse cases of retroflexion of the organ, considered an emergency situation. In this clinical case, was obtained the cystopexy after conducting cystostomy and introduction of foley catheter through the abdominal wall in a dog with a history of chronic perineal hernia with bladder retroflexion and recurrence after hernia repair with perineal affixing of the pelvic diaphragm muscles. The probe was maintained for eight days to provide adhesion of the urinary bladder with the retroumbilical ventral abdominal wall, allowing satisfactory recovery of the patient. Although some similar techniques have been described, the application of this method was not found in the literature as an adjunct to correction of perineal hernia method.


Este trabalho tem o intuito de divulgar a técnica de cistopexia por meio de cistostomia com introdução de sonda de foley no tratamento de hérnia perineal crônica recidivante com conteúdo vesical. Recidivas da herniação perineal são ainda desafio ao cirurgião e técnicas que previnam ou tratem tal complicação devem ser difundidas. Deste modo, a realização da cistopexia tem por objetivo minimizar os casos de recidiva da retroflexão do órgão, situação considerada de urgência. No presente caso clínico, obteve-se a cistopexia após a realização de cistostomia e introdução de sonda de foley através da parede abdominal em cão com histórico de hérnia perineal crônica com retroflexão de bexiga e recidiva após herniorrafia perinal com aposição dos músculos do diafragma pélvico. A sonda foi mantida por oito dias proporcionando a adesão da vesícula urinária à parede abdominal retro-umbilical ventral, permitindo satisfatória recuperação do paciente. Embora algumas técnicas similares já tenham sido descritas, a aplicação do referido método não foi encontrado na literatura como método adjuvante à correção de hérnia perineal.
