ABSTRACT

The yolk sac is an extraembryonic membrane, of saccular form, connected to the ventral region of the embryo. It is the main source of nutrition for the embryo during the period when the placenta is not fully formed. The aim of this study was to generate tubular structures using mesenchymal stem cells from the bovine yolk sac (bYS-MSCs) and determine if these structures can be a model for in vitro vasculogenesis. The evaluation of this tissue by histochemistry revealed a strong marking of collagen fibers and PAS technique negativity. In transmission electron microscopy, cytoplasmic organelles with large nuclei were observed. The vessel formation assay on a Matrigel substrate showed that the mesenchymal cells of the yolk sac without growth factors (VEGF) are capable of forming branches, sprouting cells, and tubular structures similar to capillary blood. These tubular structures were xenotransplanted subcutaneously into the mesentery of BALB/c/nude mice; after 45 days, vascularized tissue and extensions of blood vessels around the tubular structures could be observed. Real-time PCR (qPCR) demonstrated an expression of the VEGF gene in different gestational age groups. No difference in distribution or expression was detected among groups. Our results suggest that the spontaneous formation of tubules from the yolk sac can be an experimental model to elucidate initial organogenesis and the possible formation of blood capillaries from in vitro mesenchymal cells and possible route of organoid production.

ABSTRACT

The yolk sac is an extra-embryonic membrane that plays an important role in early embryonic survival. It is the production site for blood cells during embryonic mammalian development and is a likely source of stem cells. The aim of this study was to identify and characterize the putative haematopoietic cells from the yolk sac of bovine embryos at different stages of gestation. The yolk sac regresses according to gestational age and embryos are characterized into groups (I-V) according to the crown-rump measurement. Groups I-III survived in culture longer and exhibited the formation of cell clusters, whereas groups IV and V could not be maintained in culture for an extended period of time. Flow-cytometry analysis revealed that groups I-III had similar characteristics, including high expression levels of the haematopoietic markers CD34, CD90 and CD117. In groups IV and V, decreases were observed in the expression levels of CD117 and CD34. Cells were found to be capable of survival post-cryopreservation and exhibited varying abilities to form colonies in a methylcellulose matrix, depending on gestational age. Cytological analysis revealed the presence of blood cells (lymphocytes and monocytes). Quantitative PCR analysis demonstrated the presence of the haematopoietic progenitor genes GATA3 and LMO2, but not RUNX1. Thus, we have successfully isolated and characterized haematopoietic cells from the bovine embryo yolk sac at varying gestational ages. This study is crucial for the understanding of the development of the haematopoietic system and the embryonic function of this organ. Copyright © 2015 John Wiley & Sons, Ltd.

ABSTRACT

Due to the ecological importance of Procyon cancrivorus, the aim of this study was to characterize morphologically and ultrastructurally the mandibular salivary gland of this specie. We used 10 pairs (right and left) of mandibular salivary glands from five adult animals. The mandibular salivary glands were dissected and measured by precision calipers and later processed by histological routine technique, stained by HE (hematoxylin and eosin) and Picrossirius. Fragments of the glands were processed for analysis by scanning electron microscopy (SEM). The mandibular salivary gland, right and left from P. cancrivorus presents lobulated and oval-shape, and it was positioned between the fossa atlantis and basihyoideum ossis of skull. Microscopically, these glands are lined by a capsule formed by dense irregular connective tissue, which enters into the gland by septa, dividing it into lobules. Into the septa of connective tissue were present blood vessels and nerves, and interlobular excretory ducts. Inside the lobes of the mandibular salivary glands, striatum and intermediate-type ducts are found, and the predominance of mucous acini was observed. The results indicate that the mandibular salivary glands of Procyon cancrivorus, follows the structural pattern described in other mammalian species. However the types of acini may vary among species, and thus further histochemical and cell biology studies are required in order to uncover the significance of this variation for the specie here studied, according with their feeding habit.

ABSTRACT

The yolk sac (YS) is the main source of embryonic nutrition during the period when the placenta has not yet formed. It is also responsible for hematopoiesis because the blood cells develop from it as part of the primitive embryonic circulation. The objective of this study was to characterize the transitional area between the YS and primitive gut using the techniques of light microscopy, transmission electron microscopy, and immunohistochemistry to detect populations of pluripotent cells by labeling with Oct4 antibody. In all investigated embryos, serial sections were made to permit the identification of this small, restricted area. We identified the YS connection with the primitive intestine and found that it is composed of many blood islands, which correspond to the vessels covered by vitelline and mesenchymal cells. We identified large numbers of hemangioblasts inside the vessels. The mesenchymal layer was thin and composed of elongated cells, and the vitelline endodermal membrane was composed of large, mono- or binucleated cells. The epithelium of the primitive intestine comprised stratified columnar cells and undifferentiated mesenchymal cells. The transitional area between the YS and the primitive intestine was very thin and composed of cells with irregular shapes, which formed a delicate lumen containing hemangioblasts. In the mesenchyme of the transitional area, there were a considerable number of small vessels containing hemangioblasts. Using Oct4 as a primary antibody, we identified positive cells in the metanephros, primordial gonad, and hepatic parenchyma as well as in YS cells, suggesting that these regions contain populations of pluripotent cells.

ABSTRACT

The Nucleolar Organizer Regions (NORs - nucleolar organizer regions) are used to describe regions of chromatin stained with silver nitrate and are related to the activity of rRNA synthesis and to the agility and speed of cell proliferation in the tissues studied. The objective of this study was to relate the amount of AgNORs, proliferative activity and stage of pregnancy in horses, using the coloring of Silver Nitrate. The embryonic attachments were collected, fixed in 10% buffered formaldehyde, embedded in paraplast and stained by silver nitrate. The groups were determined according to the gestational age. The amount of the corium NOR found in early pregnancy indicates the onset of cell activity, and in that the pregnancy progresses, the amount of NOR increases, suggesting higher activity and increased synthesis of their importance in maintaining the fetus. Contrary to what occurs in the corium, the quantification of NORs was higher in late pregnancy than in the beginning, suggesting the stabilization of these membranes in late pregnancy. The chorionic girdle and the yolk sac were found in early pregnancy and had lots of NORs, suggesting synthesis function and proliferation in early pregnancy, since their functions is maintenance of the embryo until the complete formation of the true placenta (chorio-allantoic membranes). We conclude that the membranes that develop in a progressive manner in accordance with the growing embryo/fetal (chorion, amnion and allantoic membranes) have an increased number of NORs and the membranes that involute after the formation of the embryo/fetus (yolk sac and chorionic girdle) have a decrease in number, suggesting a reduction in proliferative activity in these membranes.

ABSTRACT

We performed a macroscopic and microscopic study of the tongues of common opossums, Didelphis marsupialis, from South America. We studied two males and two females. We collected morphometric data on the tongue with precision calipers. For the light microscopy and scanning electron microscopy analyses, we fixed tissue fragments in 10% formaldehyde and 2.5% glutaraldehyde, respectively. The opossum tongues averaged 5.87 ± 0.20 cm in length, 3.27 ± 0.15 cm in width at the lingual body, and 3.82 ± 0.15 cm in width at the root. The mean thickness of the lingual body was 1.8 ± 0.1 cm, and the thickness of the root was 3.82 ± 0.15 cm. Sharp filiform papillae were scattered across the entire tongue; conical filiform papillae occurred on the lingual body and tongue tip; fungiform papillae were scattered among the filiform papillae on the lingual body and tongue tip; and there were three vallate papillae at the root of the tongue. We found two strands of papillary projections in the tongue root. Despite the low variability observed in the lingual papillae, the morphological data obtained in this study may be related to the opossum's diverse food habits and the extensive geographic distribution of the species throughout America.

ABSTRACT

For microscopic analysis, tongues were processed routinely through electron microscopy scanning and inclusion in Paraplast; for light microscopy, the fragments were cut by microtome with an average thickness of 5mm and stained with H&E and Picrosirius counterstained with hematoxylin. The macroscopic and microscopic results show that the tongue of coati has filiform, fungiform, vallate and conical papillae distributed in the rostralis, medialis and caudalis regions. Histologically, the tongue is lined by a keratinized stratified squamous epithelium with spinous, granulous and keratin basal layer, with striated longitudinal and transverse skeletal muscles fibers, and several glands. We can conclude that coati´s tongue has macroscopic and microscopic features similar to the ones of Canidae, with differences in the number of vallate papillae and degree of keratinization.

ABSTRACT

A fêmea apresentou dentes menores, caninos arredondados e o macho, caninos maiores. Os animais apresentaram a fórmula dentária 2x(I3/3, C1/1, P4/4, M2/2) (I=incisivos, C=caninos, P=pré-molares e M=molares) = 40.

Nasua nasua, also known as coati, is an omnivorous mammal with wide distribution in South America and found all over Brazil. The evolution adapted the teeth of the multiple mammals for most different diets. Through these variations it is possible to classify the vertebratesï teeth of vertebrates in classes, genera and families. The objective of this paper is to describe and classify morphologically the teeth of Nasua nasua, comparing them with the teeth of Canis familiaris described in the literature. Five adult male and female Nasua nasua were studied. Photographs of the oral cavity of two macerated skulls were taken for documentation of teeth position. From other three animals, teeth were processed for decalcification and embedded in paraffin for histology. It was possible to reveal macroscopically male and female differences in N. nasua, due to a sexual dimorphism. Female have smaller round canine teeth, and male canine teeth are larger than the female ones. The animals presented the dental formula 2x (I3/3, C1/1, P4/4, M2/2) (I=incisive, C=canine, P=premolar, M=molar), with a total of 40 teeth. Microscopically, the teeth were classified as brachyodont with enamel in the most external region, covering the dentine. This type of teeth does not erupt constantly as the hypsodonts, because the enamel stays intact until the time of eruption. Teeth were also classified as anelodont, with a limited period of growth characteristics.

ABSTRACT

Three Procyon cancrivorus females were studied with emphasis for gross and microscopical description of the mammary glands. Samples of the glands were processed with routine techniques for histology. The females studied presented three pairs of mammary glands: one pair of cranial abdominal mammary glands, a second pair of caudal abdominal and a third one, as inguinal mammary glands. Mammary papillae presented a pendulum shape, as in the domestic dogs. Microscopically, the mammary gland consisted from the external to the internal portion (1) of stratified squamous epithelium of the epidermis, (2) dense irregular connective tissue of the derma, and (3) smooth muscle fibers and papillary ducts that flowed with "shower" shape into the lactiferous sinus. The secretory portion consisted of tubule-alveolar glands with cuboidal cells disposed in a simple layer. The results indicate that the set of glandular studies is similar to ones related to dog (Canis familiaris) in such a way in its gross aspect that how much in its microscopically structure. This fact suggests that we can use the raccoon and the dog as similar models of study, for identification of pathologies related to this system.
