ABSTRACT

Microbial production of carotenoids has gained significant interest for its cost-effectiveness and sustainable nature. This study focuses on 47 red-pigmented yeasts isolated from sediments and plant parts of 13 species of mangrove trees. The relative abundance and distribution of these yeasts varied with plant species and plant parts. The highest number of red yeasts was associated with the mangrove plant Avicennia officinalis (32%). Notably, the leaves harbored the highest percentage (45%) of carotenogenic yeasts, and definite compartmentalization of these yeast species was noticed in mangrove plant parts. All the isolates were molecularly identified and they belonged to the genera of Rhodotorula, Rhodosporidiobolus, and Cryptococcus. The diversity of the pigmented yeasts isolated from A. officinalis was found to be the greatest. Among these strains, Rhodotorula mucilaginosa PV 8 was identified as the most potent producer of carotenoid pigment. Under optimized conditions of physical parameters - 28 °C, pH 5, and 15% salinity led to biomass production of 9.2 ± 0.12 g/L DCW and a pigment yield of 194.78 µg/g. The pigment produced by PV 8 was identified as ß-carotene by thin layer chromatography (TLC) and Fourier transform infrared spectroscopy (FT-IR). This ß-carotene demonstrated strong antioxidant activity. Moreover, the carotenoid displayed promising antibacterial activity against multidrug-resistant organisms, including Aeromonas sp. and Vibrio sp. In vitro studies revealed the probiotic traits of PV 8. The cytotoxicity of R. mucilaginosa PV 8 was assessed in the invertebrate model Artemia salina and the survival rate showed that it was non-toxic. Furthermore, the ß-carotene from PV 8 demonstrated the ability to transfer its vibrant color to various food products, maintaining color stability even under varied conditions. This research underscores the potential of R. mucilaginosa PV 8, as a versatile and valuable resource for the production of carotenoids.

ABSTRACT

Plant Growth-Promoting Yeasts (PGPY) have garnered significant attention in recent years; however, research on PGPY from mangroves remains a largely unexplored frontier. This study, therefore, focused on exploring the multifaceted plant growth-promoting (PGP) capabilities of yeasts isolated from mangroves of Puthuvype and Kumbalam. The present work found that manglicolous yeasts exhibited diverse hydrolytic properties, with the predominance of lipolytic activity, in addition to other traits such as phosphate solubilization, and production of indole acetic acid, siderophore, ammonia, catalase, nitrate, and hydrogen cyanide. After screening for 15 PGP traits, three strains P 9, PV 23, and KV 35 were selected as the most potent ones. These strains also exhibited antagonistic activity against fungal phytopathogens and demonstrated resilience to abiotic stresses, making them not only promising biocontrol agents but also suited for field application. The potent strains P 9, PV 23, and KV 35 were molecularly identified as Candida tropicalis, Debaryomyces hansenii, and Aureobasidium melanogenum, respectively. The potential of these strains in enhancing the growth performance of mangrove seedlings of Rhizophora mucronata, was demonstrated using the pot-experiment. The results suggested that the consortium of three potent strains (P 9, PV 23, and KV 35) was more effective in increasing the number of shoot branches (89.2%), plant weight (87.5%), root length (83.3%), shoot height (57.9%) and total leaf area (35.1%) than the control seedlings. The findings of this study underscore the significant potential of manglicolous yeasts in contributing to mangrove conservation and restoration efforts, offering a comprehensive understanding of their diverse plant growth-promoting mechanisms and highlighting their valuable role in sustainable ecosystem management.

ABSTRACT

Background: Berberine (BER) and mangiferin are known natural dipeptidyl peptidase (DPP-IV) inhibitors. Hence, the study was designed to elucidate the mechanism of action of natural DPP-IV inhibitors (BER and MNG) in experimentally induced diabetes with metabolic syndrome. Aim: The aim of this study was to observe mechanism through which natural DPP-IV inhibitor works in diabetes with metabolic syndrome rat model. Materials and Methods: Wistar rats were fed high-fat diet for 10 weeks and challenged with streptozotocin (STZ) (40 mg/kg) at the 3rd week (high-fat diabetic control [HF-DC] group). After the confirmation of metabolic syndrome in the setting of diabetes, monotherapy (metformin [MET], vildagliptin [VIL], BER, and MNG) and combination (MET + VIL, MET + BER, and MET + MNG) therapy was orally fed to these rats from the 4th to 10th weeks. Results: Insulin resistance (IR) was seen in the HF-DC group as indicated by raised homeostasis model assessment of IR (HOMA-IR) in HF-DC group as compared with normal control (NC) groups. The treatment groups reduced IR as shown by a decrease in HOMA-IR as compared with HF-DC group rats. The marked reduction (P < 0.001) of beta-cell function was observed in the HF-DC group as a reduced level of HOMA for beta-cell function (HOMA-ß) was found as compared with the NC group. Increases in HOMA-ß as compared to the HFDC group were observed in the therapy groups. The treatment group significantly reduced cholesterol and atherogenic index. The treatment group showed significant preservation of beta-cell mass as per immunohistochemistry and significant anti-apoptotic activity as per Terminal Deoxyribonucleotidyl Transferase-Mediated dUTP Nick End Labeling assay report. The treated rats significantly (P < 0.05) reduced high-sensitivity C-reactive protein. Lipid peroxidation (thiobarbituric acid reactive substances) marker (P < 0.001) was significantly reduced in the treatment group. Conclusion: The natural DPP-IV inhibitors BER and MNG treatment showed beneficial effects on various components of metabolic syndrome.

ABSTRACT

An adult Indian buffalo (Bubalus bubalis) presented with corneal opacity, irritation, and excessive lacrimation from the left eye in the Referral Veterinary Polyclinic-Teaching Veterinary Clinical Complex (RVC-TVCC), Indian Veterinary Research Institute, Izatnagar. Clinical examination revealed a whitish thread-like worm in the left eye's anterior chamber. The worm was surgically removed from the eye with supportive nerve blocks. Light microscopy was used for parasite morphological identification, which provided insight into the worm as female Setaria sp. Genomic DNA was isolated, and polymerase chain reaction amplification of 12S rRNA was conducted for molecular confirmation of the parasite. The amplicon was sequenced and analysed by bioinformatics software. Sequence data showed an amplicon size of 243 bp. Phylogenetic analysis with reference data from the NCBI Genbank database revealed the worm was S. digitata, with a similarity of 99.17%. The common predilection site of S. digitata is in the peritoneal cavity of natural hosts like cattle and buffalo and is mostly non-pathogenic. The aberrant migration of the parasite larva to the brain and eye commonly occurs in goats, sheep, and horses, causing clinical conditions like cerebrospinal nematodiasis (lumbar paralysis) and ocular setariasis, respectively. Nevertheless, until now, there have been no reports of ocular setariasis in buffalo. This report is the first unusual occurrence of ocular setariasis in buffalo and its molecular confirmation and phylogenetic analysis using 12S rRNA.

ABSTRACT

Synthetic fertilizers and pesticides are being extensively used in agriculture in order to increase food production to feed the rapidly growing world population. This has negatively affected the soil microbes thereby reducing the agricultural produce. As a safer alternative, microbial fertilizers are now being used as plant growth promoters to improve agricultural yields. A large number of studies are focused on the role of bacteria and multicellular fungi, but plant growth-promoting traits of yeasts, the unicellular fungi are little known. Hence, the present article reviewed the diverse groups of yeasts with the potential to act as plant growth promoters. Plant growth-promoting yeasts (PGPY) have been mainly isolated from the rhizosphere and phyllosphere of major crop plants such as wheat, maize, and rice. Twenty-three genera of yeasts have been reported to have the potential for plant growth promotion (PGP), most of which belong to the phylum Ascomycetes. Dominant PGPY genera include Candida spp., Rhodotorula spp., Cryptococcus spp., and Saccharomyces sp. PGPY are known to exhibit phyto-beneficial attributes viz phytohormone production, phosphate solubilization, siderophore production, improved soil fertility, aid plants to tolerate abiotic stress and also compete effectively against plant pathogens. Over and above these traits, PGPY is Generally Recognized as Safe, making it an ideal candidate to be effectively employed as part of sustainable agricultural practices to ensure food security. The review warrants a need for an in-depth study on the different sources of PGPY other than rhizosphere/phyllosphere and the genes controlling PGP traits.

ABSTRACT

In the recent decades, there has been a significant uptick on the use of platelet-rich plasma (PRP) as a better alternative for ophthalmologic therapies in pathologies, primarily of the ocular surface. PRP is a class of liquid platelet concentrate containing a supra-physiological concentration of platelets in a relatively small amount of plasma. Its potential to heal various tissues has piqued interest in its therapeutic application as a biomaterial in regenerative medicine. It is currently a popular therapeutic agent in plastic surgery, cardiothoracic surgery, reconstructive surgery, and even oral and maxillofacial surgery. Based on the data from in vitro and in vivo studies, it can be concluded that PRP possesses adequate therapeutic potential in ocular pathologies, especially those involving cornea. In addition, the high concentrations of growth factors (TGF-ß, VEGF, EGF) present in the PRP accelerate the healing of the corneal epithelium. PRP has great therapeutic prospects in veterinary ophthalmology as a regenerative therapeutic modality. However, several variables are yet to be defined and standardized that can directly affect the efficacy of PRP application in different ophthalmic conditions. There is a shortage of research on the use of PRP in ocular surface defects compared to the number of studies and reports on the use of autologous and allogeneic serum eye drops. Therefore, a data-driven approach is required to generate consensus/guidelines for the preparation, characterization, and therapeutic use of PRP in veterinary ophthalmology. This review aims to inform readers of the latest research on PRP, including its preparation methods, physiological and biochemical properties, clinical applications in veterinary ophthalmology, and their safety and efficacy.

For the sustainable farming of disease-free and healthy shrimps, antimicrobial use is frequent nowadays in shrimp-cultured system. Considering the serious impact of global antimicrobial resistance (AMR), the present study was focused to investigate the prevalence of antimicrobial-resistant vibrios among infected shrimps (Penaeus vannamei) from two brackish water-cultured farms. Diverse species of vibrios viz. V. alginolyticus, V. parahaemolyticus, V. cholerae, V. mimicus, and V. fluvialis along with Aeromonas hydrophila, A. salmonicida and Shewanella algae were recovered from the shrimps on TCBS medium. Shannon-Wiener diversity index and H' (loge) were 1.506 and 1.69 for the isolates from farm 1 and farm 2, respectively. V. alginolyticus was found to be the most resistant isolate by showing multiple antibiotic resistance (MAR) index of 0.60 followed by V. mimicus (0.54) and V. parahaemolyticus (0.42). Among the 35 antibiotics of 15 different classes tested, tetracyclines, beta-lactams and cephalosporins were found as the most resistant antibiotic classes. All the isolates possessed a MAR index > 0.2 and the majority exhibited minimum inhibitory concentration (MIC) > 256 mcg/ml, thereby indicating the excess exposure of antibiotics in the systems. An enhanced altered resistance phenotype and a significant shift in the MAR index were noticed after plasmid curing. Public health is further concerning because plasmid-borne AMR is evident among the isolates and the studied shrimp samples are significant in the food industry. This baseline information will help the authorities to curb antimicrobial use and pave the way for establishing new alternative strategies by undertaking a multidimensional "One-Health" approach.

ABSTRACT

Hymenopteran parasitoids are well known for their ubiquitous diversity, important ecological roles and biocontrol potential. We report the first detailed documentation of mite predation by a parasitoid wasp, Bracon predatorius Ranjith & Quicke sp. nov., (Insecta: Hymenoptera), first case of obligate predatory behaviour in the family Braconidae and first case of mite feeding within the superfamily Ichneumonoidea. Larvae of a new wasp species are shown to develop entirely as predators of eriophyid mites that induce leaf galls in a commercially important plant. They display highly modified head capsule morphology that we interpret as being associated with this atypical life style. We propose that the new feeding strategy evolved separately from recently described entomophytophagy in another species of the same genus. The divergent larval morphological adaptations of both species indicate a high degree of evolutionary developmental plasticity in the developmental stage.

BACKGROUND: Home death is one of the key performance indicators of the quality of palliative care service delivery. Such a measure has direct implications on everyone involved at the end of life of a dying patient, including a patient's carers and healthcare professionals. There are no studies that focus on the views of the team of integrated inpatient and community palliative care service staff on the issue of preference of place of death of their patients. This study addresses that gap. METHODS: Thirty-eight participants from five disciplines in two South Australian (SA) public hospitals working within a multidisciplinary inpatient and community integrated specialist palliative care service, participated in audio-recorded focus groups and one-on-one interviews. Data were transcribed and thematically analysed. RESULTS: Two major and five minor themes were identified. The first theme focused on the role of healthcare professionals in decisions regarding place of death, and consisted of two minor themes, that healthcare professionals act to: a) mediate conversations between patient and carer; and b) adjust expectations and facilitate informed choice. The second theme, healthcare professionals' perspectives on the preference of place of death, comprised three minor themes, identifying: a) the characteristics of the preferred place of death; b) home as a romanticised place of death; and c) the implications of idealising home death. CONCLUSION: Healthcare professionals support and actively influence the decision-making of patients and family regarding preference of place of death whilst acting to protect the relationship between the patient and their family/carer. Further, according to healthcare professionals, home is neither always the most preferred nor the ideal place for death. Therefore, branding home death as the ideal and hospital death as a failure sets up families/carers to feel guilty if a home death is not achieved and undermines the need for and appropriateness of death in institutionalised settings.

BACKGROUND AND AIM: The currently available atrophic non-union models rely on wide segmental excision of bone diaphysis to impede the process of healing but lack resemblance to the clinical scenario. The present study focused on developing an in vivo model of atrophic non-union fracture in rabbit radius that can replicate the clinical scenario. MATERIALS AND METHODS: The atrophic non-union fracture model was developed by creating a 10 mm segmental bone defect in the radial diaphysis of five adult New Zealand White rabbits. The periosteum (2 mm) of the cut bone ends was cauterized using electrocautery to induce atrophy. Atrophic non-union was confirmed using radiographic and histologic evaluations on 30th postoperative day. RESULTS: The radiographic signs of healing were completely absent in all the rabbits on 30th postoperative day, indicating inert bone ends. Histological findings further confirmed the presence of inert bone ends, indicating the development of atrophic non-union. CONCLUSION: The combination of the segmental bone defect, electrocautery induced thermal damage of bone end periosteum, and delayed treatment can induce the development of atrophic non-union fracture model in rabbits that can replicate the clinical scenario.

BACKGROUND AND OBJECTIVE: Platelet-rich plasma (PRP) is a category of platelet concentrate that has been widely used as a therapeutic modality in musculoskeletal medicine. The present study was conducted to classify and code the non-activated platelet-rich plasma (nPRP) derived from New Zealand white rabbits for tissue engineering and other regenerative medicine applications. METHODS: PRP was prepared from the whole blood by double centrifugation protocol using a laboratory centrifuge. The prepared nPRP was characterized using the parameters such as platelet dose, the relative composition of platelets, WBC, and RBC. The production protocol was described using the parameters such as platelet enrichment factor, factor increase in WBC concentration, platelet capture efficiency, WBC-reducing efficiency, and RBC-reducing efficiency. The nPRP was also classified and coded using the most recent and universally accepted classification and coding systems. RESULTS: The non-activated leukocyte-poor red cell-rich PRP had an average platelet count of 1875.75 × 109/L, which is higher than the basal platelet concentration in the whole blood. Furthermore, the protocol used in this study has a mean platelet capture efficiency of 47.43 ± 6.42%. CONCLUSION: The protocol described in this study can be used to prepare non-activated leukocyte-poor red cell-rich PRP (Red-PRP IC1) from rabbits that can be coded as 318-00-00.

AIM: The purpose of this study was to determine the possible effects of genetic and environmental factors on dentofacial complex using monozygotic twins. MATERIALS AND METHODS: The study sample was made of 21 pairs of monozygotic twins (14 female pairs and seven male pairs) between 10 and 25 years. Pretreatment lateral cephalo-grams were used which were traced and digitized, and various landmarks to determine the anteroposterior and vertical proportions were marked. Samples were divided into two groups. The correlation between groups was found by calculating Pearson's product moment correlation coefficients. RESULTS: The range of the correlation coefficient was from 0.705 to 0.952. Gonial angle showed the highest correlation coefficient (0.952), while saddle angle showed the lowest correlation coefficient (0.705). CONCLUSION: The growth and development of craniofacial complex is under mutifactorial control. However, genetic influences do tend to play a dominant role. CLINICAL SIGNIFICANCE: By studying identical twins, we can study about the interaction of the environment with the genes and how it affects the growth and development of the body in general and dentofacial complex in particular. By utilizing twin studies, we can identify whether a particular trait, disease, or disorder is influenced more strongly by genetics or by the environment. Success of orthodontic treatment depends on a proper diagnosis of the problem including its etiological factors. Genetic studies let the orthodontists to understand the effects of genetic and environmental factors in the growth and development of dentofacial complex better and allows to prevent or treat malocclusions and skeletal anomalies in better ways.

AIM: The aim of this study was to test the effect of varying alcohol concentrations of various commercially available mouth-washes on force decay of elastomeric chains in prestretched and unstretched condition. MATERIALS AND METHODS: A total of five specimen groups were tested with a total sample size of 180 specimens. A specimen is described as a three-link, short module, clear elastomeric chain (3M Unitek, Ortho Plus, Ortho Organizer). The control and test groups were independently submerged in separate 37°C artificial saliva to simulate the oral conditions. Effects of different solution groups and time (initial, 1, 7, 14, 21, and 28 days) on three different elastomeric chains and force (cN) are analyzed with two-way analysis of variance; pair-wise comparisons are done by t-test. RESULTS: Higher mean force was recorded initially followed by 24 hours >7 days >14 days >21 days and 28 days respectively. The difference in mean force decay among the different time intervals was found to be statistically significant (p < 0.001). Among the different solutions, higher mean force was recorded in 21.6% alcohol mixture > Listerine 8.38% > Wokadine > alcohol mixture 8.38% > artificial saliva. CONCLUSION: In conclusion, alcohol-containing mouth rinses cause an increase in force decay of elastomeric chain over time. CLINICAL SIGNIFICANCE: These mouth rinses are commonly recommended during orthodontic treatment. The orthodontist can know better that the alcohol content of the mouth rinses and the structural and molecular modification results in the decay of elastomeric chain.

BACKGROUND: Mangiferin (MNG) is known to possess antidiabetic and antioxidant activity. However, there is no experimental evidence presently available in the literature with regard to its ameliorating effects on diabetes mellitus coexisting with metabolic syndrome. OBJECTIVE: The present study was designed to evaluate the protective effects of MNG on various components of metabolic syndrome with diabetes as an essential component. MATERIAL AND METHODS: Adult Wistar rats were fed high-fat diets for 10 weeks and challenged with streptozotocin (40 mg/kg) at week three (high-fat diabetic control group). After the confirmation of metabolic syndrome in the setting of diabetes, MNG 40 mg/kg was orally fed to these rats from the fourth to tenth week. RESULTS: The treatment with MNG showed beneficial effects on various components of metabolic syndrome, such as reduced dyslipidemia (decreased triglyceride, total cholesterol, low-density lipoprotein cholesterol, and increased high-density lipoprotein cholesterol) and diabetes mellitus (reduced blood glucose and glycosylated hemoglobin). In addition, an increase in serum insulin, C-peptide, and homeostasis model assessment-ß and a reduction in homeostasis model assessment of insulin resistance-IR were observed in MNG-treated group compared with high-fat diabetic control group. MNG was also found to be cardioprotective (reduction in creatine phosphokinase-MB levels, atherogenic index, high-sensitivity C-reactive protein). Reduction in serum dipeptidyl peptidase-IV levels in the MNG-treated group correlated with improvement in insulin resistance and enhanced ß-cell function. CONCLUSION: The present study has demonstrated antidiabetic, hypolipidemic, and cardioprotective effects of MNG in the setting of diabetes with metabolic syndrome. Thus, MNG has the potential to be developed as a natural alternative to synthetic dipeptidyl peptidase-IV inhibitors beneficial in this comorbid condition.

INTRODUCTION: Berberine, an isoquinoline alkaloid isolated from the Berberis aristata, has been shown to display a wide array of pharmacological activities (hypoglycaemic and hypolipidemic). AIM: The present study was designed to investigate whether these pharmacological properties translate into the cardioprotective effects of Berberine in the setting of diabetes mellitus. MATERIALS AND METHODS: Necessary approval from the Institutional Animal Ethics Committee was taken for the study. Experimental diabetes was produced with single dose of Streptozotocin (STZ): 45mg/kg ip and myocardial infarction was induced by administering Isoproterenol (ISP): 85mg/kg, sc to rats on 35(th) & 36(th) day. After the confirmation of diabetes on 7(th) day (>200mg/dl), Berberine (100 mg/kg) was administered orally to experimental rats from day 8 and continued for 30 days thereafter. Various anti-diabetic (Glucose, HbA1c), cardioprotective (CPK-MB), metabolic (lipid profile), safety {liver function (SGPT, kidney function (Creatinine)} and histopathological indices of injury were evaluated in Healthy Control, Diabetic Control and Berberine treated groups. RESULTS: Administration of STZ-ISP resulted in a significant decrease in body weight (p<0.001), diabetic changes (increase in blood glucose, HbA1c), cardiac injury (leakage of myocardial CPK-MB), altered lipid profile, SGPT, creatinine levels (p<0.001) in the diabetic control group rats as compared to healthy control. Berberine treatment demonstrated significant antidiabetic as well as myocardial salvaging effects as indicated by restoration of blood glucose, HbA1c and CPK-MB levels (p<0.001) compared to diabetic control group. In addition, Berberine favourably modulated the lipid parameters (total cholesterol, triglycerides, HDL, LDL). Subsequent to ISP challenge, histopathological assessment of heart, pancreas and biochemical indices of injury confirmed the cardioprotective effects of Berberine in setting of diabetes. In addition, Berberine was found to be safe to the liver and kidney. CONCLUSION: Berberine treatment produced myocardial salvaging effects in the setting of diabetes challenged with ISP induced myocardial necrosis. Cardioprotection may be attributed to anti-diabetic and hypolipidemic activities.

ABSTRACT

Background. The incidence of metabolic syndrome co-existing with diabetes mellitus is on the rise globally. Objective. The present study was designed to develop a unique animal model that will mimic the pathological features seen in individuals with diabetes and metabolic syndrome, suitable for pharmacological screening of drugs. Materials and Methods. A combination of High-Fat Diet (HFD) and low dose of streptozotocin (STZ) at 30, 35, and 40 mg/kg was used to induce metabolic syndrome in the setting of diabetes mellitus in Wistar rats. Results. The 40 mg/kg STZ produced sustained hyperglycemia and the dose was thus selected for the study to induce diabetes mellitus. Various components of metabolic syndrome such as dyslipidemia {(increased triglyceride, total cholesterol, LDL cholesterol, and decreased HDL cholesterol)}, diabetes mellitus (blood glucose, HbA1c, serum insulin, and C-peptide), and hypertension {systolic blood pressure} were mimicked in the developed model of metabolic syndrome co-existing with diabetes mellitus. In addition to significant cardiac injury, atherogenic index, inflammation (hs-CRP), decline in hepatic and renal function were observed in the HF-DC group when compared to NC group rats. The histopathological assessment confirmed presence of edema, necrosis, and inflammation in heart, pancreas, liver, and kidney of HF-DC group as compared to NC. Conclusion. The present study has developed a unique rodent model of metabolic syndrome, with diabetes as an essential component.

ABSTRACT

BACKGROUND: Assess prevalence and severity of malocclus ion in 12 - 16 year old population of Travancore using IOTN index. Tertiary care setting, with the study being conducted in the Dept of Orthodontics at PMS Dental College & the samples being collected from four different dental health care centres. Study design is descriptive study. MATERIALS AND METHODS: 120 pre- treatment study models of patients aged 12-16 yrs were collected, 30 each from four dental health centres spread across the state. These casts were subjected to three stages of screening based on IOTN index to arrive at the prevalence and severity of malocclusion in the targeted group. Kappa statistics and stratified kappa statistics. RESULTS: Results of the study showed that the prevalence of malocclusion and treatment need was 53.3%. CONCLUSIONS: The prevalence of orthodontic treatment need in South Kerala was found to be 53.3%.

Vibrio sp. V26 isolated from mangrove sediment showed 98 % similarity to 16S rRNA gene of Vibrio cholerae, V. mimicus, V. albensis and uncultured clones of Vibrio. Phenotypically also it resembled both V. cholerae and V. mimicus. Serogrouping, virulence associated gene profiling, hydrophobicity, and adherence pattern clearly pointed towards the non-toxigenic nature of Vibrio sp. V26. Purification and characterization of the enzyme revealed that it was moderately thermoactive, nonhemagglutinating alkaline metalloprotease with a molecular mass of 32 kDa. The application of alkaline protease from Vibrio sp. V26 (APV26) in sub culturing cell lines (HEp-2, HeLa and RTG-2) and dissociation of animal tissue (chick embryo) for primary cell culture were investigated. The time required for dissociation of cells as well as the viable cell yield obtained by while administering APV26 and trypsin were compared. Investigations revealed that the alkaline protease of Vibrio sp. V26 has the potential to be used in animal cell culture for subculturing cell lines and dissociation of animal tissue for the development of primary cell cultures, which has not been reported earlier among metalloproteases of Vibrios.

ABSTRACT

Four long-chain aliphatic organic diluents, triethylene glycol dimethacrylate (TEGDMA), ethylene glycol dimethacrylate (EDMA), triethyleneglycol diacrylate (TEGDA) and tetraethylene glycol diacrylate (TTEGDA) were used as diluents for preparing a visible-light-cured dental composite. Cured samples of pastes prepared using the above diluents were stored separately for a period of 60 days in distilled water at 37 degrees C and tested for changes in compressive strength, diametral tensile strength and microhardness at specific intervals of 1, 7, 14, 21, 28 and 60 days. Samples prepared using TEGDMA showed better strength values initially compared with the composite specimens prepared using the rest of the diluents although they are found to deteriorate slowly upon storage in distilled water up to 60 days. Samples prepared using EDMA showed retention in properties whereas samples prepared using TEGDA and TTEGDA showed large deviations in strength values during the first 28 days, did not deteriorate further and were found either to show improvement upon ageing or to remain constant. Sorption values seem to reach a maximum for all composites and then to decrease further after 14 days whereas solubility is found to increase with time.
