ABSTRACT

INTRODUCTION: Trypanosoma cruzi is the agent of Chagas' disease and affects approximately 6-8 million people worldwide. The search for new anti-T. cruzi drugs are relevant because only two drugs exist actually. The objective of this study was to investigate the effect of the extracts from the seeds of Lonchocarpus cultratus on T. cruzi, its cytotoxicity as well as to elucidate its chemical profile. METHODOLOGY: The characterization of the extracts was done using 1H-RMN. T. cruzi forms were treated with increasing concentrations of the extracts and after, the percentage of inhibition and IC50 or LC50 were calculated. Murine peritoneal macrophages were treated with different concentrations of the extracts to evaluate the cellular viability. The hemotoxicity was accessed by verifying the levels of hemolysis caused by the extracts on human red blood cells. RESULTS: Chalcones isocordoin and lonchocarpin were detected in the dichloromethane extract, and chalcone lonchocarpin was detected in the hexane extract. The dichloromethane extract showed higher activity against all the forms of T. cruzi compared to the other two extracts, but the hexane showed the best selectivity index. The cytotoxicity observed in murine macrophages was confirmed in human erythrocytes, with dichloromethane extract having the highest toxicity. The methanolic extract showed the lowest anti-T. cruzi activity but was nontoxic to peritoneal murine macrophages and red blood cells. CONCLUSIONS: L. cultratus extracts have the potential to be explored for the development of new anti-trypanosomal drugs. This study was the first to demonstrate the action of extracts from the genus Lonchocarpus on infecting forms of T. cruzi.

ABSTRACT

This review compiles and discusses the use of genetic markers applied in the study of the fish genus Hypostomus Lacépède, 1803 (Siluriformes: Loricariidae). The database comprises 51 peer-review articles that were published in the last 52 years (1968-2020) and that approach analysis based on different classes of genetic markers. The use of cytogenetic and enzymatic markers was predominantly especially in population studies with the genus Hypostomus, while mitochondrial markers were the majority in phylogenetic studies. Although significant methodological advances have occurred for molecular evaluation, they are still modestly applied to the study of neotropical fish genera, in which Hypostomus is included. New perspectives, especially on integrative approaches, are needed to improve our knowledge of the genetic functionality of fishes.

ABSTRACT

ABSTRACT The goal of this study is to identify the influence of the maturity state in the physical components of the high level youth male soccer players of an elite club in Brazil. Eighty-seven young male soccer players participated in this study, distributed in 34 U-14 players, 23 U-16 players and 30 U-19 players. Peak height velocity (PHV) was estimated: Pre-PHV (<-1.0 years, n=13), mid-PHV (-0.99 to 0.5 years, n=41), and post-PHV (>0.5 years, n=33). Running-based anaerobic sprint test, squat jump, yo-yo and 30m speed test were performed. The results of this study suggest that the maturational development influences vertical jump. VO2max, yo-yo test distance, fatigue index and power. This information is essential for the identification and effective development of talented football players.

ABSTRACT

ABSTRACT Purpose: To present a model of cost allocation for ophthalmic clinics services using cost versus price approach and to evaluate the cost information as a management tool as well as an instrument for cost control and decision-making. Methods: The model was tested applying various costing methodologies: absorption costing, full costing, direct costing and activity based costing. Cost allocation systems were installed in three ophthalmic clinics services with the objective of arriving at the cost of procedures conducted in the period september to november of 2003; data was obtained through financial and management reports and field interviews with staff. Results: The costing system met its objectives: the larger ophthalmic clinic demonstrates a better cost-benefit relationship given its larger volume and variety of procedures and distribution base to spread its fixed costs. With regard to the cost-charge ratio some procedures were found to be profitable and others with low or negative operating results, illustrating the need for cost control to determine profitable services as well as pricing. Conclusion: The proposed model is advantageous for both disseminating and utilizing cost information as well as providing support for management in its decision-making and negotiating activities with potential buyers.

ABSTRACT

BACKGROUND & OBJECTIVE: Sialometric and sialochemical analyses during pregnancy are not consistent, and frequently contradictory in terms of salivary flow rate, pH, and concentration of calcium, phosphorous, sodium, potassium, chloride, glucose and α-amylase. We, therefore, measured the evolution of these parameters throughout pregnancy. METHOD: A cross-sectional study compared sialometric and sialochemical analyses of 30 pregnant women vs. 30 age-matched non-pregnant women, and a longitudinal study evaluated the pregnant women in the first and third trimester of pregnancy. RESULTS: Pregnant women presented acidic non-stimulated saliva, but neutral stimulated saliva pH, and no relevant changes in salivary flow rate. Scialochemical analysis showed decreased calcium levels, increased phosphate levels, and a progressive decrease in glucose levels throughout pregnancy. CONCLUSION: Pregnancy significantly changes the oral biochemical milieu, creating a favorable environment for the development of oral pathology, in particular, dental caries.

ABSTRACT

Purpose:To compare by optical microscopy, tissue reaction caused by the Alloplant ™ to conjunctiva and backs of 24 rabbits. Methods:All rabbits underwent implantation of Alloplant ™ on the inner third of the lower eyelid, replacing the tarsal conjunctiva of one eye. After thirty days six rabbits (Group I) were sacrificed and lower eyelids of both sides and a back fragment (control) routed to optical microscopy. After sixty days six rabbits (group II) were sacrificed and their eyelids and a back fragment (control) sent for microscopic examination. That day were also performed surgical implantation of Alloplant ™ on the back of the twelve remaining rabbits. They were sacrificed ninety days (group III) and one hundred and twenty days (group IV) after the first surgery of the eyelids and microscopic study backs. Results:The “Alloplant™” (aloimplante), caused an intense inflammatory reaction, mixed exudative and proliferative with a predominance of lymphocytes, macrophages and formation of foreign body granulomas in both locations, which evolved with the replacement of the implant by dense fibrous connective tissue. Conclusion:eyelid inflammatory reaction was more intense and more prolonged in the eyelid than in the back. In the back the formation of fibrosis was more intense, what is not suitable for eyelid surgery.

ABSTRACT

Small land mammals possess features that significantly influence the dynamics of ecosystems and participate in various levels of the food web. In the Brazilian Atlantic Forest the richness of these animals is high, which makes them even more ecologically and numerically relevant in this environment. In this context, we investigated the species composition of small mammals in an unexplored area of southern Brazil, and compared the species composition of this area with other Atlantic Forest regions in order to understand how this community is related to others. The study area was located in an interior Atlantic forest formation, at a transition region between deciduous and Araucaria forests. Small mammals were captured at five collection points using pitfall traps. We compared the species composition found in our studied area with the composition of other 11 studies in different regions by a cluster analysis, and we investigated the presence of spatial autocorrelation between communities with a Mantel test. We recorded 779 individuals from 21 species of small rodents (15 species) and marsupials (six species) during the 13 months of the collection period. This richness was high compared to other studies conducted in the Atlantic Forest formations near to coastline and in interior forest formations. This may be a result of the conditions provided by this transition area (deciduous and Araucaria forests), where could be found elements of the both forests formations, which probably allows the establishment of small mammal species from both forest types. Despite differences in sampling effort of the studies, our results suggest that the interior forest formations may harbor a number of species comparable to the formations near the coast. The species composition of this area was similar to those found in other interior forest formations with the same phytophysiognomy characteristics and at nearby regions, and it was less similar to the distant formations located in southeastern and northeastern Brazil and nearby to the coastline. This can be a result of both the spatial autocorrelation (i.e. more nearby communities tend to have more similar species composition) and the differences of forest characteristics among regions.

ABSTRACT

PURPOSE: To compare the change in ocular axial length of the pseudophakic eye versus the fellow eye in children with bilateral cataracts who had surgery in only one eye. METHODS: In this prospective cohort study, 50 eyes of 25 children with bilateral lamellar cataracts were analyzed. A complete ophthalmic examination and evaluation of axial length measurements by contact ultrasound biometry were performed in all eyes undergoing cataract surgery with IOL implantation and in contralateral eyes. The primary outcome measure was the percentage rate of growth between the final and initial measurements, defined as the initial minus the final measurement, with the difference being divided by the initial measurement and the result multiplied by 100. RESULTS: Children aged 4-10 years of age were followed for a mean of 28.5 months. The values for axial length percentage rate of growth were significantly lower in pseudophakic eyes than in the unoperated, contralateral eyes (0.64% vs 2.09%, P < 0.05). Final visual acuity, follow-up time, and initial axial length did not affect the results. Pseudophakic eyes with posterior capsule opacification that underwent neodymium YAG laser showed a significantly higher rate of growth than unoperated eyes. CONCLUSIONS: Axial length in children older than 4 years showed a trend toward stabilization, with lower changes in axial length measurements in pseudophakic eyes and a higher rate of axial growth in contralateral eyes.

ABSTRACT

Abstract Purpose: To verify the influence of two different intensities and volumes of resistance training on intraocular pressur. Method: Nineteen athletes boxers (13 male and 6 female; mean age of 22 ± 3 years) were randomly assigned to two different resistance exercise sessions: muscular endurance with 3 sets of 15 repetition maximum and muscular hypertrophy with 4 sets of 8 RM. The intraocular pressure was measured with a Perkins tonometer before, during and after the resistance exercise sessions. Data were expressed as mean and standard deviation. A repeated-measures analysis of variance (ANOVA) was used and the Tukey post hoc test for multiple comparisons were applied. Results: A reduction in intraocular pressure during the resistance exercise session was observed. However, the muscular endurance exercise produced a significant reduction in intraocular pressure when compared with muscular hypertrophy. At the end of session, intraocular pressure returned to pre-exercise levels in muscular endurance session and above to pre-exercise levels in hypertrophy session. Conclusion: Resistance exercises influence intraocular pressure values, thus 3 sets of 15 repetitions with 60%1RM promote better intraocular pressure responses than 3 sets of 8 repetitions with 80% 1RM. This finding of the present research can contribute in prescription of resistance exercise to people with glaucoma risk factors. .

ABSTRACT

Species identification through DNA barcoding is a tool to be added to taxonomic procedures, once it has been validated. Applying barcoding techniques in public health would aid in the identification and correct delimitation of the distribution of rodents from the subfamily Sigmodontinae. These rodents are reservoirs of etiological agents of zoonoses including arenaviruses, hantaviruses, Chagas disease and leishmaniasis. In this study we compared distance-based and probabilistic phylogenetic inference methods to evaluate the performance of cytochrome c oxidase subunit I (COI) in sigmodontine identification. A total of 130 sequences from 21 field-trapped species (13 genera), mainly from southern Brazil, were generated and analyzed, together with 58 GenBank sequences (24 species; 10 genera). Preliminary analysis revealed a 9.5% rate of misidentifications in the field, mainly of juveniles, which were reclassified after examination of external morphological characters and chromosome numbers. Distance and model-based methods of tree reconstruction retrieved similar topologies and monophyly for most species. Kernel density estimation of the distance distribution showed a clear barcoding gap with overlapping of intraspecific and interspecific densities < 1% and 21 species with mean intraspecific distance < 2%. Five species that are reservoirs of hantaviruses could be identified through DNA barcodes. Additionally, we provide information for the description of a putative new species, as well as the first COI sequence of the recently described genus Drymoreomys. The data also indicated an expansion of the distribution of Calomys tener. We emphasize that DNA barcoding should be used in combination with other taxonomic and systematic procedures in an integrative framework and based on properly identified museum collections, to improve identification procedures, especially in epidemiological surveillance and ecological assessments.

ABSTRACT

PURPOSE: To determine the incidence of congenital nasolacrimal duct obstruction (CNLDO) in premature and full-term children. METHODS: Medical records of 200 premature and 200 full-term children from July 2004 to November 2009 at the Peri-Peri Health Center were retrospectively reviewed. Fifty-three nasolacrimal ducts in 32 premature children (21 patients were bilateral) and 9 nasolacrimal ducts in 7 full-term children (2 patients were bilateral) diagnosed as having CNLDO were retrospectively examined. RESULTS: Of the 200 premature children, 32 (16%) had CNLDO compared with 7 (3.5%) of the 200 full-term infants (P < .0001). Thirty premature infants and five full-term children underwent conservative treatment. Bilateral obstruction was found in all 3 extremely premature children (gestational age < 28 weeks), in 9 (69.23%) moderately premature children (gestational age of 28 to 31 weeks), in 9 (56.25%) mildly premature children (gestational age of 32 to 36 weeks), and in 2 (28.57%) full-term children (gestational age > 37 weeks). CONCLUSIONS: This study reported a higher incidence of CNLDO in preterm infants when compared to full-term infants. This might be expected in light of the normal development of the nasolacrimal duct system, the patency of which normally occurs after the preterm infant is born.

ABSTRACT

A quantitative structure-activity relationship (QSAR) study of twenty flavonoid derivatives with antimutagenic activity against 3-nitrofluoranthene (3-NFA) was performed by Partial Least Squares (PLS), using Ordered Predictors Selection (OPS) algorithm for variable selection. Four descriptors (PJI2, Mor27m, G1e and R4u+) were selected and a good model (n = 19; R(2) = 0.747; SEC = 0.332; PRESS(cal) = 1.768; F((2,27)) = 23.585; Q(LOO)(2) = 0.590; SEV = 0.388; PRESS(val) = 2.858; R(pred)(2) = 0.591; SEP = 0.394; ARE(pred) = 5.230%; k = 1.005; k' = 0.990; |R(02) - R'(02)| = 0.109) was built with two latent variables describing 83.410% of the original information. Leave-N-out cross validation (LNO) and y-randomization were performed in order to confirm the robustness of the model. The topological descriptors selected indicate that the antimutagenic activity against 3-NFA depends on molecular size, shape and Sanderson electronegativity of flavonoids. The proposed model may provide a better understanding of the antimutagenic activity of flavonoids and can be used as a guidance for proposition of new chemopreventive agents.

ABSTRACT

PURPOSE: To verify the association between two different types of strangling with intraocular pressure variation in jiu-jitsu athletes. METHODS: An observational study was performed on 9 athletes of jiu-jitsu, with at least 6 month of training, male, aged 20 to 30 years, without any physical and eyeball lesions. Associations between intraocular pressure and Cross Choke from the guard strangling (E1), and E2 - Cross Choke from mount strangling were gotten. Intraocular pressure was determined by using Perkins tonometer, at first in the absence of physical exercise over the last 24 hours and after each strangling. Then it was carried out the intraocular pressure measure at each 3 minutes, during 12 minutes of exercise recovery (R1, R2, R3, and R4) keeping the athletes lied down. Statistical analysis was done using ANOVA test and Bonferroni post-test. RESULTS: Meaningful reduction of both eyes intraocular pressure occurred at the E2 situation comparing to the E1 situation at all exercise recovery measures: R1 (OD: 8.22 +/- 1.39 vs.11.33 +/- 2.00 / OE: 8.55 +/- 1.23 vs. 11.88 +/- 1.90), R2 (OD: 8.44 +/- 1.87 vs.10.22 +/- 2.53 / OE: 9.00 +/- 1.80 vs. 10.44 +/- 2.35), R3 (OD: 8.44 +/- 1.74 vs.9.78 +/- 2.54 / OE: 8.55 +/- 1.42 vs. 10.33 +/- 1.93) all with p<0,01 e R4 (OD: 8.88 +/- 2.08 vs.9.55 +/- 2.87 / OE: 9.11 +/- 1.53 vs. 10.44 +/- 2.18) with p<0.05. Meaningful IOP reduction (p<0.05) was observed at the R1 moment of E2 strangling (OD: 10.77 +/- 1.92 vs.8.22 +/- 1.39 / OE: 11.44 +/- 1.94 vs. 8.55 +/- 1.23). CONCLUSION: There is association between intraocular pressure and jiu-jitsu strangling exercises, with intraocular pressure reduction.

ABSTRACT

PURPOSE: To verify the intraocular pressure (IOP) after sub-maximal strength test. METHODS: 145 Physical Education freshmen (22.04 +/- 4.17 years old; female and male) from Superior Physical Education School of Jundiaí (ESEFJ) were evaluated in an observacional study. The exclusion criteria were: media opacity and eyeball absence or changes. All subjects agreed to take part in this research and signed up the Informed Consent. IOP was measured by Perkins tonometer: i) pretest: just before the submaximal strength test performance and ii) post-test: immediately after the strength test. The strength test consisted in the one-repetition-maximum-assessment through repetition until fatigue. Resistance training exercises such as bench press, pulley dorsal high, shoulder press, arm curl and leg press 45 masculine were performed. Statistical analyses were obtained through paired Student's 't' test. RESULTS: Decrease of IOP was observed after the strength test: 13.48 +/- 3.32 vs.10.20 +/- 3.72 (p<0.001) in the right eye and 13.13 +/- 3.96 vs.9.74 +/- 3.33 (p<0.001) in the left eye. CONCLUSION: IOP showed relevant decrease after submaximal strength test performance in college students.

ABSTRACT

ABSTRACT

ABSTRACT

The population genetic structure of two sympatric species of sigmodontine rodents (Oligoryzomys nigripes and Euryoryzomys russatus) was examined for mitochondrial DNA (mtDNA) sequence haplotypes of the control region. Samples were taken from three localities in the Atlantic Rain Forest in southern Brazil, along an altitudinal gradient with different types of habitat. In both species there was no genetic structure throughout their distribution, although levels of genetic variability and gene flow were high.
