ABSTRACT

In recent years, the presence of Pharmaceutical Active Compounds (PhACs) in ecosystems has become a serious environmental problem due to their capacity to induce harmful effects at extremely low concentrations in both humans and wildlife. Water treatment plants have not been designed to remove these types of compounds efficiently. Thus, the detection of these pollutants is essential to evaluate their negative impacts and is one of the emerging issues in environmental chemistry. The main objective of this study is to determine the bacterial toxicity of two PhACs (both individually and as a mixture) through the quantification of bioluminescence inhibition in the marine bacteria Aliivibrio fischeri, a commonly used method in short-term toxicity tests. In this work, Acetaminophen and Edaravone, two drugs approved by the Food and Drug Administration, have been studied. The acute toxicity of these PhACs has been tested at two exposure times (5 and 15 min) and different concentrations, by estimation of the median effective concentration (EC50) for each individual compound or in combination at different concentrations. Moreover, the EC50 of the binary mixtures Acetaminophen/Edaravone have been forecast using two traditional predictive models, Concentration Addition and Independent Action. The results show that toxicity decreases with exposure time and depends on the concentration tested. Furthermore, a novel semi-empirical Van Laar-based model has been proposed and validated with the experimental data from this study and literature data, obtaining satisfactory estimations of the EC50 for binary mixtures.

ABSTRACT

Microalgae-bacteria biomass cultured in wastewater is an interesting renewable material capable of metabolising nutrients from wastes into carbohydrates, proteins, and lipids through photosynthesis. Despite the interest in the valorisation of this biomass to improve the viability of microalgae-based wastewater treatment processes, very scarce research has been devoted to the fractional recovery of its components. This work evaluates the effect of different pretreatments coupled with enzymatic hydrolysis on the solubilisation of biomass components and on the recovery of fermentable monosaccharides (glucose and xylose) from Scenedesmaceae based biomass grown in a thin layer reactor feed with piggery wastewater. Chemical pretreatments generated high concentrations of byproducts, mainly organic acids. No bacterial DNA was found in these pretreated biomasses. The acid pretreatment provided the highest carbohydrate solubilisation (98%) and monosaccharide recovery (81%). Enzymatic hydrolysis coupled with alkaline NaOH 2 M pretreatment achieved almost complete solubilisation of the biomass components, but high carbohydrate losses. Physical pretreatments remarkably increased the solubilisation of the biomass components during the enzymatic hydrolysis step, especially bead milling, which achieved solubilisation yields of 83% of carbohydrates, 43% of proteins, and 60% of lipids. The presence of viable bacteria in these pretreated biomasses could be related to the high carbohydrate losses and the generation of methanol and ethanol in addition to organic acids as byproducts.

ABSTRACT

This review is focused on the applications of genome cryobanking of aquatic species including freshwater and marine fish, as well as invertebrates. It also reviews the latest advances in cryobanking of model species, widely used by the scientific community worldwide, because of their applications in several fields. The state of the art of cryopreservation of different cellular types (sperm, oocytes, embryos, somatic cells and primordial germ cells or early spermatogonia) is discussed focusing on the advantages and disadvantages of each procedure according to different applications. A special review on the need of standardization of protocols has also been carried out. In summary, this comprehensive review provides information on the practical details of applications of genome cryobanking in a range of aquatic species worldwide, including the cryobanks established in Europe, USA, Brazil, Australia and New Zealand, the species and type of cells that constitute these banks and the utilization of the samples preserved. STATEMENT OF RELEVANCE: This review compiles the last advances on germplasm cryobanking of freshwater and marine fish species and invertebrates, with high value for commercial aquaculture or conservation. It is reviewed the most promising cryopreservation protocols for different cell types, embryos and larvae that could be applied in programs for genetic improvement, broodstock management or conservation of stocks to guarantee culture production.

ABSTRACT

This work evaluated the performance of four open algal-bacterial photobioreactors operated at ≈26days of hydraulic retention time during the treatment of 10 (×10) and 20 (×20) times diluted piggery wastewater (PWW) under indoor (I) and outdoor (O) conditions for four months. The removal efficiencies (REs) of organic matter, nutrients and zinc from PWW, along with the dynamics of biomass concentration and structure of algal-bacterial population were assessed. The highest TOC-RE, TP-RE and Zn-RE (94±1%, 100% and 83±2%, respectively) were achieved indoors in ×10 PWW, while the highest TN-RE (72±8%) was recorded outdoors in ×10 PWW. Chlorella vulgaris was the dominant species regardless of the ambient conditions and PWW dilution. Finally, DGGE-sequencing of the bacterial community revealed the occurrence of four phyla, Proteobacteria being the dominant phylum with 15 out of the 23 most intense bands.

Individual differences in behaviour and physiological responses to stress are associated with evolutionary adaptive variation and thus raw material for evolution. In farmed animals, the interest in consistent trait associations, i.e. coping styles, has increased dramatically over the last years. However, one of limitations of the available knowledge, regarding the temporal consistency, is that it refers always to short-term consistency (usually few weeks). The present study used an escape response during a net restraining test, previously shown to be an indicative of coping styles in seabream, to investigate long-term consistency of coping styles both over time and during different life history stages. Results showed both short-term (14 days) consistency and long-term (8 months) consistency of escape response. However, we did not found consistency in the same behaviour after sexual maturation when the restraining test was repeated 16, 22 and 23 months after the first test was performed. In conclusion, this study showed consistent behaviour traits in seabream when juveniles, and a loss of this behavioural traits when adults. Therefore, these results underline that adding a life story approach to data interpretation as an essential step forward towards coping styles foreground. Furthermore, a fine-tuning of aquaculture rearing strategies to adapt to different coping strategies may need to be adjusted differently at early stages of development and adults to improve the welfare of farmed fish.
