ABSTRACT

OBJECTIVE: Individuals with Down Syndrome (DS) exhibit a higher susceptibility to infections, suggesting potential immunological alterations within this population. Consequently, this study aims to assess the immune response profile in children with DS to identify potential immune dysfunctions associated with recurrent infections. METHODS: The authors conducted a retrospective analysis involving 49 DS patients, examining various epidemiological, clinical, cytogenetic, and laboratory variables. The study's sample comprised patients aged 2-20 years, with a predominance of males. These patients were categorized into two groups based on the presence or absence of recurrent infections, as indicated by the Jeffrey Modell Foundation alert signs. RESULTS: Immunoglobulin (Ig) A, G, and M levels were deemed normal, although individuals with DS experiencing recurrent infections exhibited significantly lower IgA levels. Additionally, CD3, CD4, CD8, and CD19 lymphocyte counts were found to be within normal ranges, with no significant differences between the two groups. While overall data indicated normal seroconversion levels of pneumococcal polysaccharide antibodies, a notable impairment in seroconversion was observed among DS patients with recurrent infections compared to those without such infections. CONCLUSION: The deficiency of anti-polysaccharide antibodies in individuals with DS may constitute an important immunological comorbidity. Therefore, it warrants further investigation, particularly among individuals with recurrent infections.

ABSTRACT

Introdução: A população infantil é frequentemente vítima de maus tratos, violências e negligências por parte dos familiares, o que ameaça sua integridade física e emocional, remanescendo a alternativa de acolhimento em instituições de abrigo infantil com a finalidade de proteção e cuidado. Objetivo: Analisar as condições de saúde e o perfil epidemiológico da população infantil institucionalizada. Métodos: Trata-se de um estudo transversal e retrospectivo, realizado a partir da coleta de dados de prontuários e outros documentos das crianças e adolescentes acolhidos em uma instituição de abrigo infantil do município de Governador Valadares, Brasil, no período entre janeiro de 2016 e dezembro de 2019. Resultados: Durante este período, foram abrigados 105 crianças e adolescentes, sendo a maioria do sexo feminino, com até 4 anos de idade e sem condição de saúde diagnosticada. Para a maioria dos acolhidos não havia informações sobre a história gestacional, dados do nascimento, vacinação e frequência na escola. O uso de álcool e/ou drogas pelos pais, negligência, abandono e violência foram os motivos mais frequentes da institucionalização. Grande parte das crianças e adolescentes permaneceu em acolhimento por até um ano, possuía irmãos também abrigados e foi reintegrada à família ou adotada por familiares. Conclusão: Os resultados do presente estudo poderão contribuir para subsidiar a implantação de políticas públicas que minimizem a vulnerabilidade social, evitando o acolhimento institucional e seus impactos negativos sobre o desenvolvimento infantil.


Introduction: The child population is often the victim of mistreatment, violence, and neglect on the part of family members, which threatens their physical and emotional integrity, remaining the alternative of shelter in child institutions for protection and care. Objective: To analyze the health conditions and the epidemiological profile of the institutionalized child population. Methods: This is a cross-sectional and retrospective study, based on the collection of data from medical records and other documents of children and adolescents admitted to a children's shelter institution in the municipality of Governador Valadares, Brazil, between January 2016 and December 2019. Results: During this period, 105 children and adolescents were sheltered, most of them female, aged up to 4 years old and with no diagnosed health condition. For the majority of sheltered there was no information about the gestacional history, birth data, vaccination e school attendance. The use of alcohol and/or drugs by parents, negligence, abandonment and violence were the most frequent reasons for institutionalization. Most of the children and adolescents remained in children's shelter institution for up to a year, had siblings who were also sheltered and were reintegrated into the family or adopted by relatives. Conclusions: The results of this study may contribute to support the implementation of public policies that minimize social vulnerability, avoiding institutional care and its negative impacts on child development.

ABSTRACT

Botryosphaeran, a (1→3)(1→6)-ß-d-glucan, presents several beneficial activities, such as antiproliferative, hypoglycemic and antitumoural activities. This study evaluated the effects of botryosphaeran on oxidative stress, inflammation and metabolic activities in Walker-256 tumour-bearing non-obese and obese rats. Wistars rats were divided into four groups: control tumour (CT); control tumour + botryosphaeran (CTB); obese tumour (OT), and obese tumour + botryosphaeran (OTB). In ninth week, obese and non-obese rats were inoculated with 1 × 107 Walker-256 tumour cells and treated with botryosphaeran (30 mg/kg/d for 15 days). In 11th week, the following parameters were evaluated glycogen, glucose and lactate levels, pro-oxidant (TBARS) and antioxidant markers (superoxide dismutase [SOD]; catalase [CAT]; glutathione-S-transferase [GST]; reduced glutathione [GSH]; vitamin C) and cytokines. Obesity presented oxidative stress and inflammation, as demonstrated by high levels of TBARS, SOD and TNF-α, and lower levels of CAT, GSH and interleukin-10 (IL-10). Botryosphaeran significantly decreased TBARS and TNF-α and increased GST, GSH, vitamin C and IL-10 in the liver; increased SOD and vitamin C in tumour tissue; decreased TBARS in adipose tissue, and notably decreased the levels of glycogen and lactate in the tumour of CTB rats. Botryosphaeran promoted significant antioxidant, anti-inflammatory, and beneficial metabolic effects in Walker-256 tumour-bearing non-obese and obese rats, which contributed to its antitumour activity.

BACKGROUND: Crohn's disease (CD) and ulcerative colitis (UC), two of the main inflammatory bowel diseases (IBD), have been increasingly diagnosed in South America. Although IBD have been intensively studied in the last years, epidemiologic data in Brazil are scarce. OBJECTIVE: To study the clinical and epidemiologic profile of IBD patients treated in the Clinical Hospital of the Federal University of Uberlândia from 1999 to 2014. METHODS: We performed a retrospective study of the medical records of patients diagnosed with IBD, according to the international classification of diseases (ICD) - ICD K50 for CD and ICD K51 for UC - confirmed by endoscopic examination in the case of both diseases. We analyzed the following variables: age; sex; ethnicity; smoking habit; primary diagnosis; site of disease manifestation; main clinical manifestations; IBD-related complications; extraintestinal manifestations; and established drug and/or surgical treatment. RESULTS: We evaluated 183 IBD cases (91 UC and 92 CD cases). The estimated prevalence rate of UC was 15.06/100.000 inhabitants and of CD was 15.23/100.000. The CU and CD female to male incidence ratios were 1.7 and 1.8, respectively. The average age of patients diagnosed with UC was 39.4 years and of those diagnosed with CD was 31.1 years. White-skinned people were the most affected by UC (66.0%) and CD (69.0%). Few patients were submitted to surgical procedures as treatment alternative. CONCLUSION: The estimated prevalence of IBD in this population was low compared to that of populations of North America, but high compared to that of other regions considered to present low incidence, such as some Asian and Latin American countries.

RESUMO CONTEXTO: A doença de Crohn (DC) e a retocolite ulcerativa (RCU), duas das principais doenças inflamatórias intestinais (DIIs), têm sido cada vez mais diagnosticadas na América do Sul. Embora a DII tenha sido intensamente estudada nos últimos anos, os dados epidemiológicos no Brasil são escassos. OBJETIVO: Estudar o perfil clínico e epidemiológico dos pacientes com DII atendidos no Hospital das Clínicas da Universidade Federal de Uberlândia de 1999 a 2014. MÉTODOS: Foi realizado um estudo retrospectivo dos prontuários de pacientes com diagnóstico de DII, de acordo com a classificação internacional de doenças (CID) - CID K50 para DC e CID K51 para RCU - confirmado por exame endoscópico para ambas as doenças. Analisamos as seguintes variáveis: idade; sexo; etnia; hábito tabágico; diagnóstico primário; local de manifestação da doença; principais manifestações clínicas; complicações relacionadas a DII; manifestações extraintestinais; tratamentos medicamentoso e/ou cirúrgico instituídos. RESULTADOS: Foram avaliados 183 casos de DII (91 casos de RCU e 92 casos de DC). A prevalência estimada de RCU foi de 15,06/100.000 habitantes e de DC foi de 15,23/100.000. As taxas de incidência entre pacientes do sexo feminino e masculino foram de 1,7 para RCU e 1,8 para DC. A idade média dos pacientes com diagnóstico de RCU foi de 39,4 anos e daqueles com DC foi de 31,1 anos. A raça branca foi o grupo étnico mais afetado por RCU (66,0%) e DC (69,0%). Poucos pacientes foram submetidos a procedimentos cirúrgicos como alternativas de tratamento. CONCLUSÃO: A prevalência estimada de DII nesta população foi baixa em comparação com populações da América do Norte, mas elevada em comparação com outras regiões consideradas de baixa incidência, como alguns países da Ásia e da América Latina.

ABSTRACT

Studies demonstrate that obesity can increase tumor development. Botryosphaeran, a fungal (1→3)(1→6)-ß-D-glucan, presents antimutagenic, antiproliferative and pro-apoptotic activities. This study evaluated the effects of botryosphaeran on tumor development and metabolic and hematological parameters in tumor-bearing obese and non-obese rats. Obesity was induced by a high-fat and high-sugar diet, while control rats received standard diet and water without sugar for 10 weeks. On 8th-week, Walker-256 tumor cells were inoculated in the rats, and treatment with botryosphaeran (12 mg/Kg b.w.) started. Groups:control tumor-CT, control tumor botryosphaeran-CTB, obese tumor-OT and obese tumor botryosphaeran-OTB. On 10th-week, tumor development, cachexia, metabolic and hematological parameters were analyzed. Tumor development and cachexia were significantly higher in the OT group compared to the CT group, and botryosphaeran attenuated these parameters. OT rats presented accumulation of adipose tissue, reduced muscle mass, glucose intolerance, insulin resistance, hyperglycemia, anemia, leukocytosis, and thrombocytopenia. Botryosphaeran corrected insulin resistance and hyperglycemia, modulated cholesterol levels, and increased leukocyte and lymphocytes in obese rats, which can be attributable to an inflammatory response against the Walker-256 tumor, contributing to a lower tumor development. Our data demonstrated that botryosphaeran was effective in attenuating tumor growth and in improving the metabolic and hematological profiles of the tumor-bearing rats, demonstrating its potential role in the cancer's management.

Obesity is an important risk factor in tumor development. Botryosphaeran, a (1 â†’ 3)(1 â†’ 6)-ß-D-glucan, produced by the fungus Botryosphaeria rhodina (MAMB-05), is a high molecular mass, water-soluble exopolysaccharide. It consists of a main chain of (1 â†’ 3)-linked ß-d-glucose units, with a degree of branching of ~22% at carbon-6 with glucose and gentiobiose residues linked through ß-(1 â†’ 6)-bonds, and presents a triple helix conformation. Botryosphaeran presents anticlastogenic, antiproliferative, pro-apoptotic and anti-obesogenic activities. This study evaluated the effects of botryosphaeran on tumor development in obesity and analyzed its mechanism of action. Obesity was induced in male Wistar rats by a high-fat/high-sugar diet. After 9 weeks, rats were divided into two groups: Obese Tumor (OT) and Obese Tumor Botryosphaeran (OTB), and inoculated with 1 × 107 Walker-256 tumor cells, and treatment with botryosphaeran (30 mg/kg b.w./day via gavage for 15 days) commenced. On the 11th week, biological parameters, tumor development, metabolic profile, erythrogram and protein expression were evaluated. Botryosphaeran significantly reduced tumor growth, body-weight loss and cachexia. Furthermore, botryosphaeran decreased mesenteric fat and insulin resistance, corrected macrocytic anemia, and increased Forkhead transcription factor-3a (FOXO3a) activity. Our study demonstrated the potential role of botryosphaeran in the management of cancer in tumor-bearing obese rats by increasing insulin sensitivity and FOXO3a activity.

ABSTRACT

Copper is an essential micromineral in animal feed; however, when consumed in excess, it can cause liver necrosis, hemolytic crisis, hemoglobinuric nephrosis and death in cattle. Although uncommon in this species, copper poisoning occurs as a result of exacerbated supplementation, deficiency of antagonist microminerals, or previous liver lesions. An outbreak of chronic copper poisoning is reported in semi-confined cattle after supplementation with 50 mg/Kg of dry matter copper. The cattle showed clinical signs characterized by anorexia, motor incoordination, loss of balance, jaundice, brownish or black urine, diarrhea and death, or were found dead, 10 to 302 days after consumption. Of the 35 cattle that died, 20 underwent necropsy, whose frequent findings were jaundice, enlarged liver with evident lobular pattern, black kidneys, and urinary bladder with brownish to blackish content. Microscopically, the liver showed vacuolar degeneration and/or zonal hepatocellular centrilobular or paracentral coagulative necrosis, in addition to cholestasis, mild periacinal fibrosis, apoptotic bodies, and mild to moderate mononuclear inflammation. Degeneration and necrosis of the tubular epithelium and intratubular hemoglobin cylinders were observed in the kidneys. Copper levels in the liver and kidneys ranged from 5,901.24 to 28,373.14 µmol/kg and from 303.72 to 14,021 µmol/kg, respectively. In conclusion, copper poisoning due to excessive nutritional supplementation is an important cause of jaundice, hemoglobinuria, and death in semi-confined cattle.(AU)

As intoxicações, apesar de incomuns nessa espécie, ocorrem devido a suplementação exacerbada de cobre, pela deficiência de microminerais antagonistas ou secundária a lesão hepática prévia. Relata-se um surto de intoxicação crônica por cobre em bovinos semiconfinados após suplementação com 50mg/kg de cobre em matéria seca. Os bovinos manifestaram sinais clínicos caracterizados por anorexia, incoordenação motora, perda de equilíbrio, icterícia, urina acastanhada ou negra, diarreia e morte ou foram encontrados mortos, após 10 a 302 dias do início de consumo. De 35 bovinos que morreram 20 foram submetidos à necropsia sendo achada frequente icterícia, fígado aumentado e com padrão lobular evidente, rins pretos e bexiga urinária repleta de conteúdo acastanhado a enegrecida. Microscopicamente, no fígado havia degeneração vacuolar e ou necrose coagulativa hepatocelular zonal, centrolobular ou paracentral, além de degeneração vacuolar com corpúsculos de Councilman, colestase, fibrose periascinar leve, e inflamação de discreta a moderada. Nos rins havia degeneração e necrose do epitélio tubular assim como cilindros de hemoglobina intratubulares. Os níveis de cobre no fígado e rim foram de 5.901,24 a 28.373,14µmol/kg e 303,72 a 14.021µmol/kg respectivamente. A suplementação nutricional excessiva com cobre pode causar doença hemolítica com icterícia, hemoglobinúria e morte de bovinos mantidos em sistema de semiconfinamento.(AU)

ABSTRACT

AIMS: Cancer is a multifactorial disease characterized by an uncontrolled growth of cells that can lead to cachexia-anorexia syndrome. Botryosphaeran, a fungal (1 â†’ 3)(1 â†’ 6)-ß-D-glucan produced by Botryosphaeria rhodina MAMB-05, has presented antimutagenic, antiproliferative, pro-apoptotic, hypoglycemic and hypocholesterolemic effects. This study evaluated the effects of botryosphaeran (30 mg/kg b.w./day) on tumor development and cachexia syndrome in Walker-256 tumor-bearing rats, and also the metabolic and hematological profiles of these animals. MATERIALS AND METHODS: Male Wistar rats were divided into 3 groups: control (C), control tumor (CT) and control tumor botryosphaeran (CTB). On the first day, 1 × 107 Walker-256 tumor cells were inoculated subcutaneously into the right flank of the CT and CTB rats, and concomitantly treatment with botryosphaeran (30 mg/kg b.w./day) started. After the 15th day of treatment, biological parameters, tumor development, cachexia, glucose and lipid profiles, hemogram and protein expression were analyzed. KEY FINDINGS: Botryosphaeran significantly reduced tumor development (p = 0.0024) and cancer cachexia, modulated the levels of glucose, triglycerides and HDL-cholesterol, and corrected macrocytic anemia. Botryosphaeran also increased significantly the bax expression in the tumor tissue (p = 0.038) demonstrating that this (1 â†’ 3)(1 â†’ 6)-ß-D-glucan is increasing the apoptosis of tumor cells. p53, p27, bcl-2, caspase-3 and Forkhead transcription factor 3a (FOXO3a) protein expression were similar among the groups. SIGNIFICANCE: This study demonstrated that botryosphaeran was effective in decreasing tumor development and cachexia by direct and indirect mechanisms increasing apoptosis and improving the metabolic and hematological profiles.

ABSTRACT

Background: Actinobacillosis in adult horses is rare, and is often associated with infection due to Actinobacillus equulisubsp. equuli, which is a commensal organism inhabiting the oral cavity of healthy animals. It is an opportunistic pathogen,and the infection usually occurs secondary to another primary disease or due to predisposing factors such as pre-existinglesions, mainly in the oral cavity. Infection may be associated with peritonitis, bacterial endocarditis, pneumonia, enteritis,infectious periorchitis, abortion, and septicemia. This paper aims to describe the clinical, microbiological, and pathologicalaspects of actinobacillosis in the oral cavity of a horse.Case: A case of actinobacillosis has been reported in a 22-year-old gelding Quarter Horse. According to the owner, theanimal presented with progressive weight loss during the previous three months, and leakage of food from the oral cavity.Examination of the oral cavity revealed sharp enamel overgrowths, which had been corrected during a previous dentalprocedure. However, five days after the procedure, the animal developed halitosis. Ceftiofur sodium (4.4 mg/kg intramuscularly, every 24 h) was prescribed; two days after the commencement of antimicrobial therapy, the animal presented withsialorrhea, bilateral submandibular lymphadenomegaly, protrusion of the tongue, edema of the base of the tongue, cheeks,and gums, in addition to black-colored sores and scales in the oral mucosa, particularly affecting the base of the tongue andgums. Samples from ulcerative lesions found at the base of the tongue and gums were collected using sterile swabs, andculture and antibiogram were requested. Microbiological culture led to the growth of small colonies measuring less than5 mm in diameter, that were non-hemolytic, slightly whitish in color but non-opaque, smooth and bright in appearance.These were identified as gram-negative rods on microscopy, and further characterized...

ABSTRACT

(1) Background: In this study, the effects of different pH values ​​(2.4, 3.2, 4.4 and 5.0), temperatures (30, 35, 40, 45 and 50°C) and agitation (100 rpm) on the enzymatic decolourisation of twenty-two dyes belonging to the chromophore groups anthraquinone, azo and triphenylmethane were assessed. (2) Methods: In all conditions, it was used a crude enzyme broth containing 30 U mL-1 laccases produced by Pleurotus sajor-caju PS-2001 in submerged process. (3) Results: Regarding the effects of pH values, the best results were obtained at pH 3.2 and 30°C, in which bleaching was observed for all dyes evaluated. In assays conducted at different temperatures, highest levels of decolourisation were observed at 35°C and pH 3.2 for nineteen of the dyes assessed. Thirteen dyes presented colour reduction exceeding 50% after the enzymatic treatment, including all acid and all disperse dyes evaluated. The reciprocal agitation of 100 rpm promoted negative effect on decolourisation. (4) Conclusion: From the results achieved, one can conclude that the laccase-containing preparation of P. sajor-caju PS-2001 has potential for the decolourisation of some dyes widely used in different industrial sectors, especially in the textile industry, and therefore could be used in future strategies for the biotreatment of coloured wastes.

ABSTRACT

Bócio é o aumento não inflamatório e não neoplásico da glândula tireoide em animais adultos e recém-nascidos. Uma das principais causas envolvidas é a deficiência nutricional de iodo. Relata-se neste trabalho, a ocorrência de três surtos de bócio em bovinos. Na primeira propriedade (Propriedade A), 60 bezerros foram afetados, sendo que 20 morreram logo após o nascimento, 30 recuperaram-se e 10 permaneceram doentes e tiveram remissão dos sinais apenas após tratamento parenteral com iodo. Na segunda propriedade (Propriedade B) uma vaca e seu feto foram acometidos e na terceira (Propriedade C) dois bezerros foram afetados. Os principais sinais clínicos observados nos bezerros foram aumento de volume bilateral na região cervical ventral, emagrecimento, dificuldade respiratória, hipotricose e desenvolvimento corpóreo retardado. Na propriedade A foi relatada ainda a ocorrência de abortamentos. Macroscopicamente, os bezerros e o feto apresentavam tireoide aumentada, vermelho-escura, com vascularização evidente e edema subcutâneo cervical. No surto dois também foi observado, à necropsia, aumento de volume da glândula tireoide da vaca. Microscopicamente, a tiroide dos bezerros e do feto apresentava folículos tireoidianos hiperplásicos, heterogêneos, destituídos de coloide e com interstício acentuadamente vascularizado. A tireoide da vaca era semelhante à dos bezerros, no entanto, possuía quantidade maior de coloide. Em todos os casos a suplementação mineral era realizada por meio da mistura de sal mineral com sal branco. Na Propriedade A o sal branco era não iodado e misturado em partes iguais com o sal mineral. Na Propriedade B o sal também era misturado a um sal branco não iodado na proporção e 1:2, respectivamente. Na Propriedade C o sal mineral e o sal branco não iodado eram ofertados em cochos separados no campo e, segundo relato do proprietário, os animais priorizavam o consumo do sal branco. Em todas as propriedades acompanhadas foi recomendada a interrupção da adição de sal branco na mistura mineral e a administração de iodo aos bezerros acometidos. Após essas medidas não foram observados novos casos nas propriedades. Apesar de ser uma enfermidade bem conhecida, ainda são poucos os relatados de casos de bócios em bovinos no Brasil. Além disso, pouco se sabe sobre as reais deficiências minerais de cada região, e que simples orientações de manejo nutricional ainda são necessárias.(AU)
