ABSTRACT

Cancer is one of the deadliest diseases in the world. Cancer may occur due to gene mutation. Rhaphidophora pinnata is a plant that has many benefits, especially in the leaves which have been used traditionally to treat cancer. The aim of this research is to test the antimutagenic activity of nanoparticles R. pinnata using the micronucleus method. The mice were induced with cyclophosphamide and then followed with the administration of nanoparticles of R. pinnata at the doses of 50, 100, 200 mg/kg for 7 days. The antimutagenic activity was evaluated at the decrease in the number of micronucleus in 200 polychromatic erythrocytes (PCE) cells of mice bone marrow. The result showed that the reduction of amount of micronucleus in PCE of a negative control group, treatment groups, and normal group is 22.65%, 60.3%, 79.6%, 93.8%, and 100%. These results indicate that the antimutagenic activity of nanoparticle of R. pinnata increases proportionally as the doses were increased. It can be concluded that nanoparticles R. pinnata at the doses of 50, 100, and 200 mg/kg have antimutagenic activity.

ABSTRACT

BACKGROUND: Irbesartan and hydrochlorothiazide are a group of anti-hypertensive drugs that are very effective and safe to use to reduce blood pressure and oedema. The combination has a small active ingredient content so that if the treatment didn't meet the requirements for therapeutic doses, it not achieved to the maximum therapy. AIM: The research aims to the simultaneous determination of irbesartan and hydrochlorothiazide in tablets by Ratio subtraction spectrophotometry method. METHODS: The absorption spectra and sample measurement in the Ratio subtraction method performed on Irbesartan at a wavelength of 247.6 nm and 273.6 nm for the Hydrochlorothiazide (HCT) using 0.1 N NaOH as a solution. This method is validated with linearity, accuracy, and precision in intraday and interday, LOD and LOQ and applied in the determination of a mixture of irbesartan and hydrochlorothiazide in the dosage tablet. RESULTS: The validation test for IRB is 101.03 for accuracy, with a precision of 0.57; with precision testing at intraday 0.34 and interday 1.34, and LOD is 0.70 and LOQ is 2.12. Meanwhile, validation for HCT that the accuracy 100.34%; precision 0.89 and precision on intraday 1.20 and interday 1.18, and LOD 0.78 and LOQ 2.37 with IRB levels are 101.03 ± 0.63% and HCT is 100.59 ± 0.91%. CONCLUSION: The ultraviolet spectrophotometric method in subtraction ratio method was validated a method of linearity, accuracy, precision in intraday and interday, LOD, and LOQ and according to ICH guidelines and successfully applied for the determination simultaneous of irbesartan and hydrochlorothiazide in the tablet's dosage form.

ABSTRACT

AIM: This research aimed to determine the characteristics of dried bulbs of Eleutherine palmifolia, the group of active chemical compounds contained in n-hexane extract of Eleutherine palmifolia bulbs and the antifungal activity of n-hexane extract of Eleutherine palmifolia bulbs against Candida albicans and Trichophyton mentagrophytes. METHODS: The Eleutherine palmifolia bulbs were extracted by percolation method using n-hexane solvent. The extract was tested for its antifungal activity against Candida albicans and Trichophyton mentagrophytes by diffusion method using paper discs. RESULTS: The characterisation of dried Eleutherine palmifolia bulbs were obtained water content 9.38%, water-soluble extract content 12.15%, ethanol-soluble extract content 14.48%, total ash content 0.91%, and acid insoluble ash content 0.70%. Determination of Phytochemical content showed alkaloid, flavonoid, tannin, saponin, glycoside, and steroid/triterpenoid compounds. The antifungal activity of Eleutherine palmifolia bulbs n-hexane extract (EPBHE) by concentration 200 mg/ml demonstrated the inhibition diameter of 19.48 and 42.20 mm for Candida albicans and Trichophyton metagrophytes, respectively. CONCLUSION: The antifungal test indicates that n-hexane extract of Eleutherine palmifolia bulbs provides inhibitory power to Candida albicans and Trichophyton mentagrophytes.

ABSTRACT

BACKGROUND: Rhaphidophora pinnata (L.f.) Schott is one of the Indonesian plants, has known as a medicinal plant. This plant is a vine; round stems have sticky roots and hanging roots. The leaves have been used as traditional anticancer in Singapore. Indonesian people have also used R. pinnata plants as a diuretic agent, anticancer and antibacterial. R. pinnata plants contain active substances from alkaloid compounds, flavonoids, saponins, tannins and triterpenoids/steroids. AIM: The purpose of this study was to determine the organic and inorganic content of Rhaphidophora pinnata (L.f.) Schott water extract in the form of fresh leaves, micro simplicia and nano simplicia. METHOD: The collected R. pinnata leaves are drained and grinded to make micro, and nano Simplicia powder of R. pinnata leaves. The size characterisation of R. pinnata leaves was analysing using Particles Size Analyzer. The water extract of R. pinnata leaves, micro simplicia, and nano simplicia R. pinnata leaves was made 10% (w / v) in water. Phytochemical screening of nano simplicia and nano simplicia water extract included an examination of alkaloid compounds, flavonoids, saponins, tannins, glycosides, and steroids/triterpenoids. Thin-layer chromatography analysis of water extract was analysed using TLC scanner. The element that contains in the water extract was analysed using atomic absorption spectrophotometry methods. RESULT: The results of phytochemical screening of nano simplicia powder and nano simplicia water extract showed the presence of flavonoids, alkaloids, saponins, tannins, and glycosides. Eluent, which shows good elution is n-butanol: acetic acid: water (6: 2: 2). This eluent is used to elute polar and semipolar compounds and is very good for separating flavonoids. R. pinnata water extract contains the minerals potassium, sodium, magnesium and calcium. CONCLUSION: R. pinnata water extract contains organic compounds in the form of flavonoids, alkaloids, saponins, tannins, and glycosides. The nano simplicia water extract showed more chemical content than other water extracts on the TLC plate by detection at a wavelength of 250 nm and 300 nm. The most element content in R. pinnata water extract is potassium, followed by magnesium, sodium, and calcium.

ABSTRACT

BACKGROUND: Cancer is one of the causes of morbidity and mortality worldwide. Breast cancer is one of the most common types of cancer in Indonesia. Failures that often occur in the treatment of cancer primarily through chemotherapy, synthetic drugs that have side effects include anemia, alopecia, cardiotoxic and hepatotoxic due to low anti-cancer selectivity and unclear carcinogenesis process. Cyperus rotundus L. rhizome is one of the medicinal plants that potential enough to be developed as an anticancer agent. AIM: The aim of this study was to anticancer activity n-hexane fraction Cyperus rotundus L. rhizomes to breast cancer MCF-7 cell line in vitro. METHODS: Cyperus rutundus L. rhizomes powder was extracted ethanol by percolation then fractionated with n-hexane. Phytochemical screening was then carried out. The cytotoxic activity of the n-hexane fraction was determined by observing this extract on MCF-7 cells using the (3- (4,5-dimethylimidazole-2-il) -2,5-diphenyl tetrazolium bromide) (MTT). Selectivity index (IS) of normal cells (Vero cells). Cell cycle and apoptosis induction were analyzed by flow cytometry. RESULTS: The result showed that the fraction n-hexane Cyperus rutundus L. rhizome has anticancer activity against breast cancer MCF-7 cells with accumulation cell cycle in the G0-G1 phase and through induction of apoptosis. CONCLUSION: The n-hexane fraction Cyperus rotundus L. rhizome has potent anticancer activity.
