ABSTRACT

This study investigated the levels, mass loadings, removal efficiency, and associated ecotoxicological risks of selected endocrine disrupting chemicals (EDCs), namely, dibutylphthalate (DBP), diethylhexylphthalate (DEHP), dimethylphthalate (DMP), linuron (LNR) and progesterone (PGT) in wastewater, sludge, and untreated dry biosolid (UDBS) samples from twelve wastewater treatment plants (WWTPs) in nine major towns in Kenya. Analysis was done using high-performance liquid chromatography coupled with triple quadrupole mass spectrometry (LC-MS/MS). All the wastewater influents had quantifiable levels of EDCs with DBP being the most abundant (37.49%) with a range of 4.33 ± 0.63 to 19.68 ± 1.24 µg L-1. DEHP was the most abundant in sludge and accounted for 48.2% ranging between 278.67 and 9243.49 ng g-1 dry weight (dw). In the UDBS samples, DEHP was also the most abundant (40%) of the total EDCs detected with levels ranging from 78.77 to 3938.54 ng g-1 dw. The average removal efficiency per pollutant was as follows: DMP (98.7%) > DEHP (91.7%) > PGT (83.4%) > DBP (77.9%) > LNR (72.2%) which can be attributed to sorption onto the biosolid, biological degradation, photolysis, and phytoremediation. The pH was negatively correlated to the EDC concentrations while total dissolved solids (TDS), chemical oxygen demand (COD), biochemical oxygen demand (BOD5), and electrical conductivity (EC) were positively correlated. The mass loadings were as high as 373.33 g day-1 of DBP in the treatment plants located in densely populated cities. DEHP and PGT had their Risk Quotients (RQs) > 1, posing a high risk to biota. DMP, DBP, and LNR posed medium risks as their RQ values were between 0.1 and 1. EDCs are therefore loaded to environmental compartments through either the effluent that loads these pollutants into the receiving aquatic ecosystem or through the UDBS, which are used as fertilizers in agricultural farmlands causing potential toxicological risks to aquatic and terrestrial life.

ABSTRACT

Background: Stress among medical students is related to their academic lifespan; however, information on brain health among medical students from developing countries continues to be scarce. The objective of this study was to establish perceived academic stress levels, assess the ability to cope with stress, and investigate its effects on the visual reaction time (VRT), audio reaction time (ART), and tactile reaction time (TRT) in the somatosensory cortex among medical students of Uganda. Methods: This was a cross-sectional study conducted among preclinical (n = 88) and clinical (n = 96) undergraduate medical students at Kampala International University Western Campus. A standard Perceived Stress Scale (PSS) was used to categorize stress into low, moderate, and severe while the ability to cope with stress was categorized into below average, average, above average, and superior stresscoper (SS). Data on reaction time were acquired through VRT, ART, and TRT using the catch-a-ruler experiment, and this was analyzed using SPSS version 20. Results: This study shows that preclinical students are more stressed than clinical students (PSS prevalence for low stress = preclinical; clinical: 40, 60%). Moderate stress was 48.4 and 51.6% while high perceived stress was 75 and 25% among preclinical and clinical students. Among male and female students in preclinical years, higher TRT and VRT were found in clinical students showing that stress affects the tactile and visual cortical areas in the brain, although the VRT scores were only significantly (P = 0.0123) poor in male students than female students in biomedical sciences. Also, highly stressed individuals had higher TRT and ART and low VRT. SS had high VRT and ART and low TRT in preclinical students, demonstrating the importance of the visual cortex in stress plasticity. Multiple regression showed a close relationship between PSS, ability to cope with stress, age, and educational level (P < 0.05), demonstrating the importance of social and psychological support, especially in the biomedical sciences. Conclusion: Preclinical students suffer more from stress and are poorer SS than clinical students. This strongly impairs their cortical regions in the brain, thus affecting their academic productivity.

ABSTRACT

Maternal breast milk, which is a complete food for the infant's growth, development, and health, contains fats and lipids making it susceptible to accumulation of lipophilic compounds like polycyclic aromatic hydrocarbons (PAHs). This study aimed at analyzing correlates of measured levels of PAHs in breast milk of nursing mothers to frequently used household fuels and cooking methods in Uganda, and estimate the potential health risks of PAHs to infants through breastfeeding. Sixty breast milk samples were collected from healthy and non-smoking mothers who had lived in Kampala capital city (urban area) and Nakaseke district (rural area) for at least five years. Sample extracts were analyzed for PAHs using a gas chromatograph coupled with a triple quadrupole mass spectrometer. ∑13PAHs in samples from Kampala ranged from 3.44 to 696 ng/g lw while those from Nakaseke ranged from 0.84 to 87.9 ng/g lw. PAHs with 2-3 rings were more abundant in the samples than PAHs with 4-6 rings. At least 33 % of the variance in the levels of ∑13PAHs in the breast milk samples was attributable to the fuel type and cooking methods used. Nursing mothers who used charcoal for cooking accumulated higher levels of ∑13PAHs in their breast milk samples compared to those who used firewood. Levels of ∑13PAHs in breast milk of mothers increased depending on the cooking methods used in the order; boiling< grilling< deep-frying. In all samples, hazard quotients for PAHs were <1 and estimated incremental cancer risks were all between 10-6 and 10-4, indicating that the health risks to infants due to the ingestion of PAHs in breast milk was tolerable. Further studies with large datasets on PAHs and their derivatives and, larger samples sizes are needed to confirm these findings.

ABSTRACT

In this study, we initiated an effort to generate information about beef safety in Uganda. Our entry point was to assess by atomic absorption spectrophotometry the levels of essential elements copper (Cu), cobalt (Co), iron (Fe) and zinc (Zn), and non-essential elements lead (Pb), chromium (Cr), nickel (Ni), and cadmium (Cd) in 40 beef samples collected from within and around Soroti (Uganda). The information was used to evaluate the safety of consuming such beef against the World Health Organization (WHO) limits. The latter was accomplished by (i) estimating the daily intake (EDI) of each metal in the study area, (ii) modeling the non-cancer health risk using the target hazard quotient (THQ) and (iii) modeling the cancer risk using the incremental lifetime cancer risk (ILCR). The study finds that the mean concentrations (±95% CI) and EDI were in the order of Fe > Zn > Cr > Ni > Pb > Co > Cu > Cd. Cancer risk was found to be due to Ni > Cr > Cd > Pb and significantly higher in children than adults. The latter particularly demonstrates the importance of Ni poisoning in the study area. Overall, while essential elements in our beef samples were below WHO limits (hence no health risks), non-essential elements had high health and cancer risks due to higher levels of Cr and Ni.

ABSTRACT

The impact of oil exploration and production activities on the environment of sub-saharan African countries is not well studied. This study aimed at determining concentrations, sources, and bioaccumulation of 13 polycyclic aromatic hydrocarbons (PAHs) in sediments and fish from the White Nile near Melut oil fields, South Sudan. The study also assessed the ecological and human health risk associated with PAHs in this aquatic system. Total (∑13) PAH concentrations ranged from 566 to 674 ng g-1dry weight (dw) in sediments, while those in fish were 191-1143 ng g-1 wet weight (ww). ∑13PAH concentrations were significantly higher in C. gariepinus than in other fish species. Low molecular weight PAHs (LPAHs) dominated the profile of PAHs in sediments (constituted 95% of ∑13PAHs) and fish (97% of ∑13PAHs). Compared to Sediment Quality Guidelines of the United States Oceanic and Atmospheric Administration, the levels of LPAHs in this study were all above the threshold effect limits, but below the probable effect level, while those of high molecular weight PAHs (HPAHs) were all below the lowest effect levels. The carcinogenic potency equivalent concentrations of PAHs in L. niloticus and C. gariepinus were above the US EPA screening level; suggesting consumption of these species could adversely affect human health. Biota-sediment accumulation factor values (range: 0.006-3.816 g OC g-1 lipid) for PAHs showed high bioaccumulation of LPAHs in fish muscle, and that bioaccumulation decreased with increase in hydrophobicity of the compounds. This is possibly because LPAHs have higher aqueous solubilities which increases their bioavailability through water-gill transfers compared to HPAHs. Profiles of PAHs in the White Nile environment indicate predominant contribution from petrogenic sources, which could be attributed to presence of crude oil reservoirs and oil production operations. More research into the levels of other environmental pollutants in the oil-rich area is recommended.

ABSTRACT

Honey has multifaceted nutritional and medicinal values; however, its quality is hinged on the floral origin of the nectar. Taking advantage of the large areas that they cover; honeybees are often used as bioindicators of environmental contamination. The focus of the present paper was to examine the quality of honey from within the vicinity of an abandoned pesticide store in Masindi District in western Uganda. Surficial soils (<20 cm depths) and honey samples were collected from within the vicinity of the abandoned pesticide store and analysed for organochlorine pesticide (OCP) residues using gas chromatograph coupled to an electron capture detector (GC-ECD). The mean level of ∑DDTs in all the soil samples was 503.6 µg/kg dry weight (d.w). ∑DDTs contributed 92.2% to the ∑OCPs contamination loads in the soil samples, and others (lindane, aldrin, dieldrin, and endosulfans) contributed only 7.8%. Ratio (p, p'-DDE+p, p'-DDD)/p, p'-DDT of 1.54 suggested historical DDT input in the area. In all the honey samples, the mean level of ∑DDTs was 20.9 µg/kg. ∑DDTs contributed 43.3% to ∑OCPs contamination loads in the honey samples, followed by lindane (29.8%), endosulfans (23.6%) and dieldrin (3.2%), with corresponding mean levels of 14.4, 11.4 and 1.55 µg/kg, respectively. Reproductive risk assessment was done based on the hazard quotient (HQ) and hazard index (HI) procedure. In our study, the calculated HIs for adults (102.38), and children (90.33) suggested high potential health risks to the honey consumers. Lindane, endosulfan and p, p'-DDD detected in the honey samples at levels exceeding the acute reference dose (ARfD) are known risk factors for spontaneous abortion, reduced implantation, menstrual cycle shortening, impaired semen quality, and prostate cancer in exposed individuals and experimental animal models.

ABSTRACT

Persistent organic pollutants (POPs) are ubiquitous contaminants with adverse health effects in the ecosystem. One of such effects is endocrine disruption in humans and wildlife even at background exposure concentrations. This study assessed maternal breastmilk concentrations of POPs; brominated flame retardants (BFRs), polychlorinated biphenyls (PCBs) and polychlorinated dibenzo-p-dioxins/furans (PCDD/Fs), and the potential health risks posed to the nursing infants. We also evaluated the association of these POPs with total 3,3',5-triiodo-L-thyronine (T3), L-thyroxine (T4), and 3,3',5'-triiodo-L-thyronine (rT3) levels measured in human breast milk. Thirty breastmilk samples were collected from Kampala, Uganda between August and December 2018. Hexabromobenzene was not detected while the maximum level of 2,2',4,4',5,5'-hexabrombiphenyl was 64.7 pg/g lw. The median levels of total indicator PCBs, PBDEs, dioxin-like PCBs, and PCDD/Fs in the samples were 159 pg/g lw, 511 pg/g lw, 1.16 pg TEQ/g lw, and 0.4 pg TEQ/g lw, respectively. These levels were lower than those reported in other countries. Owing to their bio accumulative nature, PCBs -81, -169, and ∑PCDD/Fs increased with increase in maternal age. Estimated dietary intakes for dioxin-like PCBs and PCDD/Fs were lower than those reported elsewhere but were higher than the WHO tolerable daily intakes suggesting potential health risks to nursing infants. In adjusted single pollutant models, PCB-126, PCB-169, and ∑PCBTEQ were negatively associated with T3, while 1,2,3,4,5,7,8-HpCDF was positively associated with rT3. Although these associations did not persist in multipollutant models, our findings suggest potential thyroid hormone disruption by POPs in mothers. This may reduce the levels of thyroid hormones transferred from the mother to the neonates and, hence, adversely influence infant growth. A temporal study with a bigger sample size is required to corroborate these findings.

ABSTRACT

Bwindi Impenetrable National Park situated southwest of Uganda is a biodiversity hotspot that is home to about half of the world's endangered mountain gorilla (Gorilla beringei). Given its ecological significance and mounting pressures from agricultural activities such as tea growing, continuous monitoring of the levels of chemical toxins like pesticides in the park and surrounding areas is needed for effective conservation strategies. Furthermore, persistent organochlorine pesticides (OCPs) like DDT were used in agricultural gardens and indoor spraying in Kanungu district between the 1950s and 80s. The focus of this study was to explore the possible exposure of mountain gorillas to OCPs and cypermethrin used by the farmers in the areas near the park. Data from our interviews revealed that glyphosate is the most widely used pesticide by the farmers in areas surrounding the park, followed by cypermethrin, and mancozeb. Samples of leaves from plants consumed by mountain gorillas along the forest edges of the park and surficial soils (15-20 cm depths) were collected from three sites (Ruhija, Nkuringo and Buhoma) and analysed for the presence of cypermethrin and OCPs residues. Concentrations of total (∑) DDTs and ∑endosulfans were up to 0.34 and 9.89 mg/kg dry weight (d.w), respectively in soil samples. Concentrations of ∑DDTs and ∑endosulfans in samples of leaves ranged from 0.67 to 1.38 mg/kg d.w (mean = 1.07 mg/kg d.w) and 0.9 to 2.71 mg/kg d.w (mean = 1.68 mg/kg d.w), respectively. Mean concentration of ∑DDTs in leaves exceeded the European pharmacopeia and United States pharmacopeia recommended maximum residue limit values for DDTs in medicinal plants (1.0 mg/kg). In addition, calculated hazard indices for silverbacks (36.35), females (57.54) and juveniles (77.04) suggested potential health risks to the mountain gorillas. o,p'-DDT/p,p'-DDT ratios (0.5-0.63) in samples of leaves confirmed recent input of dicofol-DDT type in Bwindi rainforest.

ABSTRACT

Per- and polyfluoroalkyl substances (PFASs) are known organic pollutants with adverse health effects on humans and the ecosystem. This paper synthesises literature about the status of the pollutants and their precursors, identifies knowledge gaps and discusses future perspectives on the study of PFASs in Africa. Limited data on PFASs prevalence in Africa is available because there is limited capacity to monitor PFASs in African laboratories. The levels of PFASs in Africa are higher in samples from urban and industrialized areas compared to rural areas. Perfluorooctane sulfonic acid (PFOS) and perfluorooctanoic acid (PFOA) are the dominant PFASs in human samples from Africa. Levels of PFOS and PFOA in these samples are lower than or comparable to those from industrialized countries. PFOA and PFOS levels in drinking water in Africa are, in some cases, higher than the EPA drinking water guidelines suggesting potential risk to humans. The levels of PFASs in birds' eggs from South Africa are higher, while those in other environmental media from Africa are lower or comparable to those from industrialized countries. Diet influences the pollutant levels in fish, while size and sex affect their accumulation in crocodiles. No bioaccumulation of PFASs in aquatic systems in Africa could be confirmed due to small sample sizes. Reported sources of PFASs in Africa include municipal landfills, inefficient wastewater treatment plants, consumer products containing PFASs, industrial wastewater and urban runoff. Relevant stakeholders need to take serious action to identify and deal with the salient sources of PFASs on the African continent.

Polybrominated diphenyl ethers (PBDEs) are ubiquitous environmental pollutants with adverse effects on the foetus and infants. This study aimed at assessing in utero exposure levels and transplacental transfer (TPT) characteristics of BDE congeners in primiparous mothers from Kampala, the capital city of Uganda. Paired human samples (30 placenta and 30 cord blood samples) were collected between April and June 2018; and analysed for a suite of 24 tri-to deca-BDE congeners. Extraction was carried out using liquid-liquid extraction and sonication for cord blood and placenta samples, respectively. Clean-up was done on a solid phase (SPE) column and analysis was performed using gas chromatography/mass spectrometry (GC/MS). Total (∑) PBDEs were 0.25-30.9 ng/g lipid weight (lw) (median; 7.11 ng/g lw) in placental tissues and 1.65-34.5 ng/g lw (median; 11.9 ng/g lw) in cord blood serum, with a mean difference of 1.26 ng/g lw between the compartments. Statistical analysis showed no significant difference between the levels of PBDEs in cord blood and placenta samples (Wilcoxon signed rank test, p = 0.665), possibly because foetus and neonates have poorly developed systems to metabolise the pollutants from the mothers. BDE-209 was the dominant congener in both matrices (contributed 40.5% and 51.2% to ∑PBDEs in placenta and cord blood, respectively), suggesting recent and on-going maternal exposure to deca-BDE formulation. Non-significant associations were observed between ∑PBDEs in maternal placenta and maternal age, household income, pre-pregnancy body mass index (BMI), and beef/fish consumption. This suggested on-going exposure to PBDEs through multiple sources such as dust from indoor/outdoor environments and, ingestion of other foods. Based on absolute concentrations, the extent of transplacental transport was greater for higher congeners (BDE-209, -206 and -207) than for lower ones (such as BDE-47), suggesting alternative TPT mechanisms besides passive diffusion. More studies with bigger sample sizes are required to confirm these findings.

ABSTRACT

The aim of this study was to investigate levels of polybrominated diphenyl ethers (PBDEs) in breast milk samples from healthy primiparous mothers who had lived in Kampala capital city (urban area) and Nakaseke district (a rural area) for the last five years. Fifty samples were collected between March and June 2018 and were extracted by dispersive solid-phase extraction (SPE). Clean-up was performed on an SPE column and analysis was done using gas chromatography-mass spectrometry. Total (∑) PBDEs (BDE 28, 47, 49, 66, 77, 99, 100,138,153, 154, 183 and 209) ranged from 0.59 to 8.11 ng/g lipid weight (lw). The levels of PBDEs in samples from Kampala capital city were significantly higher than those from Nakaseke (p < 0.01, Mann-Whitney U test). The most dominant congeners were BDE-209 and -47 (contributed 37.1% and 20.2%, respectively to ∑PBDEs), suggesting recent exposure of mothers to deca-and penta-BDE formulations. Fish and egg consumption, plastics/e-waste recycling and paint fumes were associated with higher levels of BDE-47, -153 and -99, respectively, implying that diet and occupation were possible sources of the pollutants. Estimated dietary intakes (ng kg-1 body weight day-1) for BDE-47, -99 and -153 were below the US EPA reference doses for neurodevelopmental toxicity, suggesting minimal health risks to nursing infants who feed on the milk. Generally, the risk quotients for BDE-47, -99 and -153 were <1 in majority (96%) samples, indicating that the breast milk of mothers in Uganda was fit for human consumption.

ABSTRACT

This paper reviews literature for the last two decades with emphasis on levels, toxic equivalencies and sources of polychlorinated dibenzo-p-dioxins (PCDDs), dibenzofurans (PCDD/Fs) and dioxin-like polychlorinated biphenyls (dl-PCBs) in Africa. Further, we comprehensively analysed data, interpret differences and identify existing gaps with those from other continents. We observed that high levels of PCDD/Fs and dl-PCBs were reported in environmental and biological samples near densely populated urban and industrialised areas compared to those in rural settings. In general, the concentrations of PCDD/Fs and dl-PCBs in the blood samples from Africa were in the same range as those from Asia but lower than those from Europe. The concentrations of dioxins and dioxin-like compounds in the atmosphere in Africa were comparable to and/or higher than those in developed countries. The reported sources of PCDD/Fs and dl-PCBs in Africa were industrial emissions, obsolete pesticide stockpiles, household heating, recycling of electronic waste, and incineration and combustion of domestic waste. Regional and intercontinental transport of dioxins could not be confirmed because of the lack of sufficient literature on them. Further data about the levels and sources of PCDD/Fs and dl-PCBs in Africa need to be generated to complete the chemical inventories for the continent and to facilitate the implementation of the Stockholm Convention on persistent organic pollutants. The reviewed literature shows that most analyses have been carried out in laboratories outside Africa because of the limited institutional capacity in Africa. More support needs to be given to laboratories in Africa to develop the capacity to accurately quantify dioxins on routine basis.
