ABSTRACT

Drug-drug interactions (DDIs) are responsible for an increase in the incidence of adverse drug reactions. Although CYP is known to be involved in metabolic processes, the DDIs among three or more drugs that involve the same CYP molecular species have not been fully investigated. In this study, we retrospectively examined the relationship between the number of drugs and potential DDIs in metabolic processes involving CYPs in patients who picked up their prescribed drugs at 11 pharmacies in the Kojima Branch of the Okayama Pharmaceutical Association. We found that 66.5% of the 924 patients had potential DDIs; more than half of the patients who took six or more drugs had potential DDIs among three or more drugs. The mean number of CYP3A4-related drugs involved in potential DDIs was 3.52±1.56 in patients who took seven drugs, suggesting the need for careful monitoring of specific symptoms and blood test results for the early detection of adverse drug reactions caused by DDIs among three or more drugs.

ABSTRACT

Lipoxygenase-1 (LOX-1) null 'New Sachiho Golden' is a two-row malting barley (Hordeum vulgare L.) cultivar released in 2015 that was developed at the Tochigi Prefectural Agricultural Experimental Station by backcross breeding using the high-yield leading cultivar 'Sachiho Golden' as a recurrent parent and the LOX-1 null mutant 'Daikei LM1' as a non-recurrent parent. To develop 'New Sachiho Golden' we used a simple LOX activity assay and marker-assisted selection. This is the first LOX-1 null malting barley cultivar in Japan that is resistant to barley yellow mosaic virus (types I-III). Agronomic characteristics and malting qualities of 'New Sachiho Golden' were similar to those of 'Sachiho Golden', except that 'New Sachiho Golden' had no LOX activity in ungerminated grains and had clearly lower LOX activity during malting than 'Sachiho Golden'. The concentrations of a trans-2-nonenal (T2N) precursor in wort and beer made from 'New Sachiho Golden' were significantly lower than in those made from 'Sachiho Golden', both before and after storage.

ABSTRACT

This study examined changes in body mass index (BMI), fasting blood sugar (FBS), total cholesterol (TC) and HDL-cholesterol (HDL-C) levels over a 24-year follow-up period in a pediatric cohort. An appropriate starting age for intervention to prevent cardiovascular diseases is still unclear. The subjects were 655 children, aged 10-12. A follow-up survey was conducted when the subjects reached ages 13-15, 16-18, and 35-45, respectively, and height, weight, and blood tests including FBS, TC and HDL-C were examined. Forty (6%) of these subjects participated. BMI at ages 35-45 were significantly higher than those at ages 10-12 (p < 0.0001), 13-15 (p < 0.001), and 16-18 (p < 0.001). TC levels at ages 35-45 were significantly higher than at ages 10-12 (p < 0.0001), 13-15 (p < 0.0001), and 16-18 (p < 0.0001). BMI at the end of the follow-up (ages 35-45) had a significant correlation with BMI at ages 13-15 (R = 0.38, p = 0.041) and 16-18 (R = 0.41, p = 0.049). TC and HDL-C values at the end of the follow-up had a significant correlation with those at ages 10-12 (R = 0.55, p = 0.0004; R = 0.55, p = 0.016), 13-15 (R = 0.35, p = 0.045; R = 0.42, p = 0.015), and 16-18 (R = 0.47, p = 0.019; R = 0.44, p = 0.028). These results may suggest that intervention for children in Japan with cardiovascular risk factors should be initiated in the early years of life.

ABSTRACT

The methanol extract of the flowers of artichoke (Cynara cardunculus) exhibited remarkable antitumor activity in an in vivo two-stage carcinogenesis test in mice, using 7,12-dimethylbenz[a]anthracene as an initiator and 12-O-tetradecanoylphorbol-13-acetate (TPA) as a promoter. From the active fraction of the methanol extract, four triterpene alcohols and their corresponding acetates were isolated and identified. These compounds were evaluated for their inhibitory effects on TPA-induced inflammation (1 microg/ear) in mice and showed marked anti-inflammatory effects, with a 50% inhibitory dose of 0.50-0.91 micromol/ear.

ABSTRACT

Bioassay-guided fractionation of the cytotoxic MeOH extract from the rhizomes of Alpinia officinarum Hance led to the isolation of two new diarylheptanoids named alpinoid D (1) and E (2), together with fifteen known diarylheptanoids (3 - 17). The structures of compounds 1 and 2 were elucidated on the basis of spectroscopic data and chemical evidence. The cytotoxic activity of the isolated diarylheptanoids was evaluated against the IMR-32 human neuroblastoma cell line. Among the tested compounds, 11, 12 and 14 exhibited the most potent activities with IC (50) values of 0.83, 0.23 and 0.11 microM, respectively.

ABSTRACT

Shorter-wavelength surface-emitting laser sources are important for a variety of fields, including photonics, information processing, and biology. We report on the creation of a current-driven blue-violet photonic-crystal surface-emitting laser. We have developed a fabrication method, named "air holes retained over growth," in order to construct a two-dimensional gallium nitride (GaN)/air photonic-crystal structure. The resulting periodic structure has a photonic-crystal band-edge effect sufficient for the successful operation of a current-injection surface-emitting laser. This represents an important step in the development of laser sources that could be focused to a size much less than the wavelength and be integrated two-dimensionally at such short wavelengths.

ABSTRACT

The double-bond conversion of UV-cured resins prepared from pentaerythritol triacrylate (PETA) was determined by pyrolysis-gas chromatography in the presence of an organic alkali, tetramethylammonium hydroxide (TMAH). The pyrogram of the uncured prepolymer compound, consisting of PETA and a photoinitiator, 2,2-dimethoxy-2-phenylacetophenone, contained specific products reflecting the original acrylate structure, such as methyl acrylate (MA) and methyl ethers of pentaerithritol. Meanwhile, in pyrograms of the UV-cured PETA, the yields of MA considerably decreased. The double-bond conversions of the cured resins, irradiated with various UV dosages, were calculated based on the relative yields of MA among specific products in the pyrograms. The conversions determined by this approach were analyzed by comparing them with those estimated by Fourier-transform infrared spectroscopy.

ABSTRACT

The cross-linking structure of the ultra violet (UV)-cured resin prepared from dipentaerithritol hexacrylate (DPHA) was characterized by matrix-assisted laser desorption/ionization mass spectrometry (MALDI-MS) combined with supercritical methanolysis. The MALDI-mass spectrum of the decomposition products obtained by supercritical methanolysis contained a series of peaks of sodium-cationized methyl acrylate (MA) oligomers up to around m/z = 4000 formed through selective cleavage and methylation occurred at ester linkages in UV-cured DPHA. Furthermore, in order to observe widely distributed sequence lengths in the cross-linking junctions, the decomposed products of the cured resin were then fractionated using size exclusion chromatography followed by the MALDI-MS measurements of the individual fractions. The MALDI-mass spectra of the lower molar mass fractions mainly consisted of a series of peaks of MA oligomers around m/z values of several thousands, whereas those of higher molecular weight showed a broad peak up to m/z ca. 180000. The observed distributions of the supercritical methanolysis products suggested that the network junctions in the given UV-cured resin were composed of up to around 2000 acrylate units.

ABSTRACT

A small amount of an oligomeric hindered amine light stabilizer (HALS) (Adekastab LA-68LD) in polypropylene (PP) materials was directly determined by solid sampling matrix-assisted laser desorption/ionization mass spectrometry (MALDI-MS) using an internal standard method. First the matrix reagent (dithranol), 20 mg, and the empirically selected internal standard, angiotensin I (MW = 1296.5), 5 microg, were premixed in the solid state. The matrix mixture was then co-ground with the PP sample containing the HALS in liquid nitrogen using a freezer mill. The powdered sample mixture was spotted on the sample plate, suspended in ion-exchanged water, dried to adhere on the plate, and subjected to MALDI-MS. Three series of the HALS components accompanied by the oxidized species were clearly observed as their molecular ions (M*+)) along with that of the internal standard in the mass spectra. A fairly good linear relationship (R2 = 0.9991) with a relative standard deviation of ca. 11% was observed between the relative peak intensities of the HALS components and the HALS contents ranging from 0.1-2.5 wt%, which could be used as the calibration line to determine the HALS content in PP composites directly by MALDI-MS. The UV-exposed PP composite samples were evaluated by this method to interpret the photostabilizing action of HALS in the PP materials based on the observed change in the relative abundances of the original and oxidized HALS components as a function of UV-exposure time.

ABSTRACT

The corrosion and tarnish behaviors of three Ag-based alloys (Ag-Pd-Cu-Au alloy, Ag-In alloy, and Ag-Sn-Zn alloy) in polyvinylpyrrolidone iodine (povidone-iodine) solution were examined. The degree of tarnish was evaluated by visible-ray spectrocolorimetry. Corrosion potential measurements and analyses of corrosion products by X-ray diffractometry were carried out to elucidate the corrosion mechanism. The corrosion rate of the three Ag-based alloys in povidone-iodine solution at its practical concentration used as a gargle solution was so fast that the alloys tarnished within 10 seconds of immersion with the formation of AgI. Thermodynamic consideration and the results of surface analysis by X-ray diffractometry revealed that the main anodic and cathodic reactions were Ag + I(-)-->AgI + e- and I2 + 2e(-)-->2I- respectively.

ABSTRACT

A novel method for the direct analysis of small amounts of an oligomeric hindered amine light stabilizer (HALS) occluded in polypropylene (PP) material was developed to study its photostabilizing action on the basis of matrix-assisted laser desorption/ionization mass spectrometry (MALDI-MS) using a solid sampling technique while avoiding troublesome solvent extraction. In this sampling protocol, the powdered mixture of PP composite sample containing trace amounts of an oligomeric HALS, Adekastab LA-68LD (MW = 1900), and the matrix reagent (dithranol) was spotted on the sample plate, then ion exchanged water was deposited onto the mixture to make a suspension, and finally, the dried mixture adhered on the plate was subjected to MALDI-MS measurement. On the mass spectrum thus obtained by the solid sampling MALDI, the molecular ions of the HALS desorbed from the PP composite were clearly observed as three major series of the HALS components in the range up to about m/z 7000 with little interference by the PP substrate and the other additives. Moreover, in the MALDI-MS spectra for the UV-exposed sample, the satellite peaks around the major HALS components proved to enhance significantly, reflecting the oxidized HALS species at the tetramethylpiperidine units to cause the photostabilizing action. In addition, hydrolyzed HALS species were also observed for the irradiated sample. These results suggest that not only the oxidation reaction but also the hydrolysis or decomposition of the oligomeric HALS components competitively proceed in the PP composites during UV exposure.
