ABSTRACT

In the south basin of Lake Biwa, Shiga, Japan, overgrown aquatic weeds (submerged macrophytes) impede cruising boats and cause unpleasant odors and undesirable waste when washed ashore. To address this socio-ecological problem, Shiga Prefectural Government implemented a public program to remove overgrown weeds and compost them ashore to conserve the lake environment, while coastal inhabitants and occasional volunteers remove weeds from the beaches to maintain the quality of the living environment. However, these effects are limited because of disjointed social networks. We applied an adaptive and abductive approach to develop community capability to jointly address this problem by sharing academic knowledge with local actors and empowering them. The initial multifaceted reviews, including interviews and postal questionnaire surveys, revealed that the agro-economic value of composted weeds declined in historical and socio-psychological contexts and that most of the unengaged public relied on local governments to address environmental problems. These findings were synthesized and assessed with workshop participants, including local inhabitants, governmental agents, businesspeople, social entrepreneurs, and research experts, to unearth the best solution. The workshops resulted in the development of an e-point system, called Biwa Point, to promote and acknowledge voluntary environmental conservation activities, including beach cleaning. It may contribute to enhancing the socio-ecological capability of communities. Additionally, ethical issues, such as publication of inconvenient truths, undesired interpretation by the researchers, and social constraints in research methods, arose through our research practice. ABSTRACT

INTRODUCTION: Men who require intermittent catheterization may be susceptible to balanoposthitis. Benzalkonium chloride is commonly used to disinfect reusable catheters, but the concentration of this solution is critical. CASE PRESENTATION: A 40-year-old man presented with fever, pus around the glans and foreskin, and difficult catheterization. On physical examination, his urethral meatus was too narrow to insert a catheter. Suprapubic catheterization and 2 weeks of intravenous and topical treatment cured the ulcer of the glans. However, after restarting catheterization, the ulcer of the glans relapsed, and the patient's urethral meatus was completely closed. Circumcision and incision of the meatus were performed, and antibiotics were administered. Unexpectedly, restarting catheterization caused recurrent balanoposthitis. Close inquiry revealed that the patient was using a 10% benzalkonium chloride soaking solution for a reusable catheter, which was a toxic concentration. CONCLUSION: The disinfectant concentration should be confirmed when refractory balanoposthitis is encountered in patients who require catheterization.

ABSTRACT

Claudins (CL) are a family of tetra-transmembrane proteins that are the structural and functional components of tight junctions (TJ). CLs are promising targets for drug development because of their role in mucosal drug absorption and cancer. However, CL-targeted drug development has been delayed because CLs have low antigenicity and preparing CL proteins is difficult. We developed a CL binder by using the C-terminal fragment of Clostridium perfringens enterotoxin (C-CPE) and a baculoviral display system. After screening CL binders from a C-CPE mutant-displaying library by using CL-displaying budded baculovirus (BV) we isolated a C-CPE mutant called m19, which bound to CL1, CL2, CL4 and CL5. A 3-dimensional analysis showed that m19 has a structural backbone similar to C-CPE. The charge density of the CL-binding domains of m19 and C-CPE differed, suggesting that electrostatic interactions may occur between m19 and CLs. Treatment of epithelial cells with m19 decreased the paracellular but not transcellular integrity, and m19 enhanced jejunal absorption. Thus, we successfully created a CL binder with broad specificity. These findings will contribute to future preparation of CL binders for CL-targeted drug development.
