ABSTRACT

The southern root-knot nematode, Meloidogyne incognita, is the most damaging pathogen of cotton in the United States, and both resistance and tolerance to M. incognita could be valuable management approaches. Our objectives were to evaluate advanced cotton breeding lines for resistance and tolerance to M. incognita and to determine if a relationship between resistance and tolerance exists. Reproduction of M. incognita was evaluated on 17 breeding lines, a susceptible control (Delta and Pine Land DP5415), and a resistant control (M-120) in two greenhouse trials with six replications in a randomized complete block design. Two-week-old seedlings were inoculated with 8,000 M. incognita eggs and assessed for egg production 8 weeks later. Reproduction on the resistant control was only 10% of that on the susceptible control. Eight breeding lines supported 45% to 57% less (P <= 0.05) nematode reproduction than the susceptible control, and none of them were as resistant as M-120. Yield was determined in 2001 and 2002 in fumigated (1,3-dichloropropene at 56 liters/ha) and nonfumigated plots in a strip-plot design with three replications in a field naturally infested with M. incognita. Yield suppression caused by nematode infection differed among genotypes (P /= 0.10). However, only three genotypes had no significant yield suppression in both years, of which two also were resistant to M. incognita. Regression analysis indicated that yield suppression decreased linearly as nematode resistance increased.

ABSTRACT

The green stink bug, Acrosternum hilare (Say), the southern green stink bug, Nezara viridula (L), and the brown stink bug, Euschistus servus (Say), were predominant phytophagous Pentatomidae detected during 1995-1997 in cotton in South Carolina. These species occurred in similar numbers in conventional and transgenic cotton 'NuCOTN33B', containing the gene for expression of CryIA(c) delta-endotoxin of Bacillus thuringiensis Berliner variety kurstaki. Adult stink bugs moved into cotton from wild and cultivated alternate hosts during July, and reproducing populations usually were detected in cotton from late July into September. Applications of either methyl parathion (0.56 kg [AI]/ha) directed for stink bugs or lambda-cyhalothrin (0.037 kg [AI]/ha) or cyfluthrin (0.056 kg [AI]/ha) for control of cotton bollworm, Helicoverpa zea (Boddie), provided effective control of pentatomids in NuCOTN33B or conventional 'DP5415' and increased yields compared with untreated plots. Fiber quality did not differ among treated or untreated plots of NuCOTN33B. The ground-cloth technique was used to estimate populations of stink bugs, and data indicated that treatment at one bug per 2 m of row adequately protected cotton from yield loss due to stink bug damage. Observations on boll damage indicated that treatment might be necessary if >20-25% reveal internal symptoms of feeding injury during mid- to late season. More detailed damage thresholds should be developed to complement an approach based on population monitoring. This study validated current recommendations for management of pentatomids in cotton, demonstrated the necessity of threshold use for stink bugs in transgenic cultivars expressing endotoxin from B. thuringiensis, and provided insight into further development of management options for pentatomids in the crop.

ABSTRACT

Fasting produces multiple cardiovascular, metabolic, and behavioral responses. To examine the interrelationship between these responses, male spontaneously hypertensive rats (SHR; n = 8) implanted with cardiovascular telemetry devices were housed in metabolic chambers at 23 degrees C for 22-h daily measurements of physiological variables. The experimental apparatus was designed so that ingestive behavior was detected by photobeams and locomotion was detected by a load sensor. Cardiovascular and metabolic status were determined as both a function of the circadian cycle (12-h dark and 10-h light), as well as during periods of inactivity (no ingestion and minimal locomotion) within the dark and light phases. Data were obtained during baseline, 48-h of caloric deprivation, and 6 days of refeeding. Fasting produced significant reductions in mean arterial pressure (dark: -9.2+/-1.3 from 143.7+/-3.7 mm Hg; light: -8.6+/-1.8 from 140.1+/-3.7 mm Hg), heart rate (dark: -43.4+/-5.2 from 330.0+/-5.2 beats/min; light: -27.4+/-5.2 from 294.0+/-5.2 beats/min), and oxygen consumption (dark: -5.0+/-0.6 from 20.6+/-0.3 ml x min(-1) x kg (0.75); light: -2.7+/-0.2 from 14.9 +/-0.2 ml x min(-1) x kg(0.75)). Analysis of inactive periods during both light and dark phases revealed that these reductions were not dependent on behavioral effects. We conclude that fasting produces concurrent and interrelated reductions in cardiovascular and metabolic function in the SHR. The merging of cardiovascular telemetry, indirect calorimetry, and behavioral monitoring provides a powerful approach for investigation of the integrative physiological responses to energetic challenges.
