ABSTRACT

Swine genital leptospirosis is an infectious disease that leads to economic losses due to abortions, stillbirths, and reproductive failures. Considering the scarcity of studies regarding this condition, the objective of the present study was to identify and analyse leptospires infecting the reproductive tract of female pigs slaughtered in the Amazon region. Cervical-vaginal mucus (CVM) from 150 non-pregnant females were collected and submitted to molecular analysis. Initially, polymerase chain reaction (PCR) based on the lipL32 gene was performed. A total of 26.7% (40/150) samples were positive, indicating the presence of Leptospira sp. DNA. Subsequently, positive lipL32-PCR samples were evaluated using secY nested-PCR and sequencing procedures. Eleven amplicons could be sequenced and were identified as Leptospira interrogans (100% identity). Results from phylogenetic analyses led to identification of a putative strain of L. interrogans serogroup Australis, which is indicative of this being a serogroup. In the present study, there was detection of female pigs with leptospires in CVM indicating the possibility of venereal transmission. The large number of genital positive cases could indicate that genital leptospirosis syndrome could also be relevant onto swine production.

ABSTRACT

Doxorubicin (DOX) is known to cause cognitive impairments in patients submitted to long-term chemotherapy (deficits also known as chemobrain). Therefore, there is an urgent need for therapeutic strategies capable of returning cancer survivors back to their previous quality of life. The present study investigated whether resveratrol (RSV) or curcumin (CUR) administration could affect mnemonic function and brain morphological changes following DOX administration in rats. Male Wistar rats were divided into 4 groups: DOX group (2.5 mg/kg/week for 4 weeks, i.p., plus distilled water for 28 days, oral gavage - OG), DOX + RSV group (DOX, 2.5 mg/kg/week for 4 weeks, i.p., plus RSV, 10 mg/kg/day for 28 days, OG), DOX + CUR group (DOX, 2.5 mg/kg/week for 4 weeks, i.p., plus CUR, 100 mg/kg/day for 28 days, OG) and control (CTR) group (0.9% saline solution weekly for 4 weeks, i.p., plus distilled water for 28 days, OG). Behavioral analyses (open field - OF - and the novel object recognition test - NORT) were performed. Brains were collected and analyzed by hematoxylin-eosin and luxol fast blue staining techniques and by immunohistochemistry for GFAP (glial fibrillary acidic protein) expression in astrocytes and Iba1 (ionized calcium-binding adaptor molecule 1) expression in microglia. DOX-injected rats presented short-term and long-term memory impairments as seen in the NORT at 3 and 24 h after habituation and increased GFAP and Iba1 expression, respectively, in astrocytes and microglia of the frontal cortex, hypothalamus and hippocampus. Such cognitive deficits were prevented by CUR at both periods and by RSV at 24 h. DOX-induced astrogliosis and microgliosis were avoided by RSV and CUR. No signs of demyelination or neuronal loss were found in any group. Thus, CUR and RSV prevented memory loss, astrogliosis and microgliosis induced by DOX monotherapy.

ABSTRACT

In this essay, we quantify the concentration of collagen fibers in broiler chickens exposed to increasing concentrations of cupuacu seed by-product. Collection of material was carried out in five chickens per treatment at 70 days old in the groups: control, 5% and 10% inclusion of cupuacu seed by-product. Fragments of Thoracic Pectoralis (PT) and Iliotibial lateralis (ITL) muscles were prepared for light and electronic microscopy. The amount of collagen fibers in the muscle groups was 1.08±0.61% in the PTC group; 6.24±2.58% in PT5% and 7.30±2.75% in PT10%. In the Iliotibial Lateralis groups, the results were 6.96±3.14% in the ITLC; 7.43±4.22% in the ITL5% and 8.66±2.35% in ITL10%. The amount of collagen fibers in the ITL5% and ITL10% groups showed no significant statistical difference. However, when compared to the ILTC group, there was a significant statistical difference. The PT muscle responds to standard nutritional changes, unlike the ILT muscle, which requires a high-nutrient formulation. The use of 5% cupuacu seed by-product has proven to be a viable alternative source of animal feed, as it promotes an increase in the concentrations of collagen fibers in the musculature of broiler chickens and is possibly the determining factor in meat texture.(AU)


Neste estudo, foram quantificadas as concentrações de fibras colágenas de frangos expostos a crescentes concentrações de farinha de cupuaçu. A coleta de material foi realizada em cinco animais por tratamento, aos 70 dias de idade, nos grupos: controle, inclusão de 5% e de 10% de farinha de cupuaçu. Fragmentos dos músculos peitoral torácico (PT) e iliotibial lateral (ITL) foram preparados para microscopia de luz e eletrônica. A quantidade de fibras colágenas nos grupos foi: 1,08±0,61% no grupo PTC; 6,24±2,58% em PT5% e 7,30±2,75% em PT10%. Nos grupos iliotibial lateral, os resultados foram: 6,96±3,14% no ITLC; 7,43±4,22% no ITL5% e 8,66±2,35% em ITL10%. A quantidade de fibras colágenas nos grupos ITL5% e ITL10% não apresentou diferença estatística significativa. No entanto, quando comparados ao grupo ILTC, houve diferença estatística significativa. O músculo PT responde a mudanças nutricionais padrão, ao contrário do músculo ILT, que requer alta formulação nutricional. O uso de 5% de farinha de cupuaçu provou ser uma fonte alternativa viável de alimentação animal, pois promove um aumento nas concentrações de fibras de colágeno na musculatura de frangos de corte e é possivelmente um fator determinante na textura da carne.(AU)

ABSTRACT

The plant diastereoisomeric diterpenes ent-pimara-8(14)-15-dien-19-oic acid, obtained from Viguiera arenaria, and isopimara-8(14)-15-dien-18-oic acid, isolated from Cupressus lusitanica, were distinctly functionalized by the enzymes produced in whole cell cultures of the fungus Preussia minima, isolated from surface sterilized stems of C. lusitanica. The ent-pimaradienoic acid was transformed into the known 7ß-hydroxy-ent-pimara-8(14)-15-dien-19-oic acid, and into the novel diterpenes 7-oxo-8 ß-hydroxy-ent-pimara-8(14)-15-dien-19-oic and 7-oxo-9ß-hydroxy-ent-pimara-8(14)-15-dien-19-oic acids. Isopimara-8(14)-15-dien-18-oic acid was converted into novel diterpenes 11α-hydroxyisopimara-8(14)-15-dien-18-oic acid, 7ß,11α-dihydroxyisopimara-8(14)-15-dien-18-oic acid, and 1ß,11α-dihydroxyisopimara-8(14)-15-dien-18-oic acid, along with the known 7ß-hydroxyisopimara-8(14)-15-dien-18-oic acid. All compounds were isolated and fully characterized by 1D and 2D NMR, especially 13C NMR. The diterpene bioproduct 7-oxo-9ß-hydroxy-ent-pimara-8(14)-15-dien-19-oic acid is an isomer of sphaeropsidin C, a phytotoxin that affects cypress trees produced by Shaeropsis sapinea, one of the main phytopathogen of Cupressus. The differential metabolism of the diterpene isomers used as substrates for biotransformation was interpreted with the help of computational molecular docking calculations, considering as target enzymes those of cytochrome P450 group.

ABSTRACT

Foram estudadas 135 vacas mestiças, provenientes de 10 rebanhos leiteiros no estado do Acre. O objetivo foi identificar espécies de Staphylococcus isoladas dos quartos mamários de vacas com mastite e, posteriormente, avaliar a capacidade de produção de biofilme pela espécie Staphylococcus chromogenes. A caracterização dos isolados presentes nas amostras encontradas, correspondentes a Staphylococcus sp., foi realizada utilizando-se a técnica do MALDI TOF MS (Matrix Associated Laser Desorption-Ionization - Time of Flight - Mass Spectrometry). Foram identificados: S. chromogenes (36), Staphylococcus saprophyticus (5), S. chromogenes ou Staphylococcus hycus (5), Staphylococcus haemolyticus (4), Staphylococcus epidermidis (3), Staphylococcus hycus (3), Staphylococcus aureus (1), Staphylococcus auriculares (1), Staphylococcus kloosii (1) e Staphylococcus xylosus (1). A espécie S. chromogenes correspondeu a 60% dos isolados do gênero (17 isolados coagularam o plasma de coelho no teste da coagulase em tubo), sendo 83,3% dos isolados (30/36) produtores de biofilme, não estando esse fator de virulência associado ao fenótipo de coagulação do plasma. A identificação desses microrganismos é importante para a elucidação da etiologia da mastite bovina. O alto percentual de S. chromogenes, produtores de biofilme, isolados de vacas com mastite é um achado relevante e pode revelar uma mudança de perfil na colonização de agentes etiológicos causadores dessa enfermidade.(AU)


A total of 135 crossbred cows was studied, from ten dairy herds on the state of Acre. The purpose was to identify species of Staphylococcus isolated from the mammary quarters of cows with mastitis. Additionally, the capacity of biofilm production by the species Staphylococcus chromogenes was evaluated. The sample characterization of the isolates was performed using the MALDI TOF MS (Matrix Associated Laser Desorption-Ionization - Time of Flight - Mass Spectrometry). The following species were identified by MALDI TOFF MS: S. chromogenes (36), Staphylococcus saprophyticus (5), S. chromogenes or Staphylococcus hycus (5), Staphylococcus haemolyticus (4), Staphylococcus epidermidis (3), Staphylococcus hycus (3), Staphylococcus aureus (1), Staphylococcus auriculares (1), Staphylococcus kloosii (1) and Staphylococcus xylosus (1). S. chromogenes corresponded to 60% of the Staphylococcus isolates (17 isolates positive on tube coagulase test). From those, 83.33% (30/36) of them were biofilm producers. This virulence factor had no association with the plasma coagulation phenotype. The identification of these microorganisms is important for the elucidation of the bovine mastitis etiology. The high percentage of S. chromogenes, biofilm producers, isolated from cows with mastitis is an important finding. This may reveal a profile change on the colonization of etiologic agents that cause this disease.(AU)

ABSTRACT

Criadores de animais silvestres sempre buscam métodos eficientes de manter sua criação. Parasitos intestinais afetam os animais em cativeiro, diminuindo sua sobrevivência e reprodução. Este estudo teve como objetivo avaliar a eficácia do uso de cal virgem na desinfecção do solo e na prevenção da reinfestação de endoparasitos em pacas (Cuniculus paca) em cativeiro. Amostras de solo das baias de criação foram analisadas usando-se o método de Rugai modificado. Duas etapas foram avaliadas: a primeira aos cinco, 45 e 105 minutos após os tratamentos, e a segunda aos sete dias e aos 14 dias após os tratamentos. Os valores de pH das amostras foram verificados após a aplicação de quatro tratamentos. Foram aplicados 400g/m2 de cal virgem a lanço ou 500mL/m2 de três soluções de cal virgem (10%, 20% ou 40% p/v). Na primeira etapa, a cal a lanço foi ineficaz na modificação do pH do solo e no controle de parasitos. A solução a 10% proporcionou o melhor controle dos parasitos. Na segunda etapa, as soluções foram eficazes na elevação do pH, porém não houve diferença significativa nos achados larvais. Concluiu-se que, nas condições do estudo, a solução a 10% aumentou o pH do solo e teve boa capacidade de inativação dos ovos de Strongyloides sp., o que economicamente é interessante. Porém, para o controle eficiente de Strongyloides sp. em cativeiros de pacas, deve-se considerar a frequência da aplicação de soluções com até 20% de cal, juntamente com a adoção de um programa de vermifugação.(AU)


Wild animal breeders are always looking for effective ways to care for their livestock. Intestinal parasites affect animals in captivity, reducing their survival and reproduction. This study aimed to evaluate the effectiveness of quicklime for use in soil disinfection and prevention of reinfection of endoparasites in paca (Cuniculus paca) kept in captivity. Soil samples of pacas' stalls were analyzed using the modified Rugai method. Two experimental stages were evaluated; the first at 5, 45 and 105 minutes after treatment and the second at 7 days and 14 days after treatment. The pH values of the samples were checked after the application of four treatments. An application of 400g/m2 of quicklime spread out on ground or 500mL/m2 of three lime solutions (10%, 20% or 40%, w / v) applied on ground. In the first stage, the lime spread out on soil was ineffective in parasites control, while the 10% solution provided the best control of parasites. In the second stage, the solutions were effective in raising the pH, but there was no significant difference in larval findings. In conclusion, under the conditions of the study, 10% solution increased the soil pH and had good ability to inactivate the eggs of Strongyloides sp.; that is economically interesting. However, for effective control of Strongyloides sp. in pacas kept captive, both the frequency of application of solutions up to 20% of lime and the adoption of a good worming schedule should be taken into consideration.(AU)

O Amblyomma geayi é um carrapato da família Ixodidae, encontrado principalmente em mamíferos e aves silvestres de regiões tropicais da América do Sul. Descreve-se a ocorrência de um carrapato ixodídeo da espécie A. geayi, encontrado em uma preguiça (Bradypus variegatus) proveniente do Parque Zoobotânico da Universidade Federal do Acre, Rio Branco, Acre, Amazônia Ocidental.(AU)

This case report the occurrence of an ixodid tick species A. geayi found in a brown-throated sloth (Bradypus variegatus) from the botanical zoo Park of the Federal University of Acre, Rio Branco, Acre, Western Amazon.(AU)
