ABSTRACT

Lectins are ubiquitous proteins involved in the immune defenses of different organisms and mainly responsible for non-self-recognition and agglutination reactions. This work describes molecular and biological characterization of a rhamnose-binding lectin (RBL) from Rhodnius prolixus, which possesses a 21 amino acid signal peptide and a mature protein of 34.6 kDa. The in-silico analysis of the primary and secondary structures of RpLec revealed a lectin domain fully conserved among previous insects studied. The three-dimensional homology model of RpLec was similar to other RBL-lectins. Docking predictions with the monosaccharides showed rhamnose and galactose-binding sites comparable to Latrophilin-1 and N-Acetylgalactosamine-binding in a different site. The effects of RpLec gene silencing on levels of infecting Trypanosoma cruzi Dm 28c and intestinal bacterial populations in the R. prolixus midgut were studied by injecting RpLec dsRNA into the R. prolixus hemocoel. Whereas T. cruzi numbers remained unchanged compared with the controls, numbers of bacteria increased significantly. The silencing also induced the up regulation of the R. prolixus defC (defensin) expression gene. These results with RpLec reveal the potential importance of this little studied molecule in the insect vector immune response and homeostasis of the gut bacterial microbiota.

ABSTRACT

BACKGROUND: The association of behavioural phenotype assessment with cytogenomic characterisation may provide a better comprehension of genotype-phenotype correlations in syndromes caused by chromosomal abnormalities, such as 18p deletion syndrome. METHOD: We report on four Brazilian patients with 18p deletion syndrome characterised by cytogenomic techniques and detailed neuropsychological evaluation. Intellectual, adaptive and behavioural characteristics were assessed through the Wechsler's Scales, the Vineland-II Scale and the Child Behaviour Checklist, respectively. Socio-economic measures including main caretaker educational level and family income as defined by Brazilian criteria for social class classification were also collected to evaluate a possible contribution of environmental factors in neurocognitive variability. RESULTS: Two out of four patients showed intellectual disability (IQ < 70). Wechsler's scale results suggest that in our sample, interpretation of social situations based on observation of non-verbal behaviour constitute a cognitive strength while judgement of social rules and language skills associated with word knowledge and verbal fluency may be a cognitive weakness. Concerning adaptive behaviour, motor and socialisation domains showed to better develop than communication and daily living skills on the Vineland-II Scale. Only one patient presented internalising behavioural problems based on the Child Behaviour Checklist. Our results also suggested that socio-economic status may contribute to overall patient development. CONCLUSION: Our results suggest that some 18p deletion syndrome patients may present average intellectual performance and that the segment deletion size and some families' socio-economic conditions may influence cognitive development.

Abstract Characterising the external morphology of mosquito eggs is important, since it facilitates the identification of material from breeding sites and contributes to the understanding of species biology and group systematics. Therefore, was to compare eggs from different Psorophora ferox populations using scanning electron microscopy (SEM). Eggs were obtained from adult female of Ps. ferox collected in the Poços das Antas Biological Reserve (Reserva Biológica de Poços das Antas, Rio de Janeiro, Brazil). From each female, one portion of eggs (n = 5) was reared for identification purposes, and the second portion (n = 10) was examined using SEM for morphometric analysis. The egg morphology was then compared to that of eggs from Ps. ferox populations in Florida (USA) and Arena (Trinidad). The exochorion ornamentation of the populations differs considerably in the morphology of the tubercles of the chorionic cells, external chorionic reticulum, micropylar collar, and micropyle.


Resumo Caracterização da morfologia externa dos ovos do mosquito é importante, uma vez que facilita a identificação de materiais a partir de locais de reprodução e contribui para a compreensão da biologia das espécies e sistemática do grupo. O objetivo do presente estudo foi comparar os ovos de diferentes populações Psorophora ferox usando MEV. Os ovos foram obtidos a partir de fêmeas de Ps. ferox que foram coletadas na Reserva Biológica de Poço das Antas (Reserva Biológica de Poço das Antas, Rio de Janeiro, Brasil). A partir de cada fêmea, uma parte dos ovos (n = 5) foi criado para fins de identificação, e uma segunda parte (n = 10) foi examinado usando MEV para análise morfométrica. Foi então comparada a morfologia dos ovos de populações de Ps.ferox da Flórida (EUA) e Arena (Trinidad). A ornamentação do exocório das populações difere consideravelmente na morfologia dos tubérculos das células coriônicas, retículo coriônico externo, colar micropilar e micrópila.

ABSTRACT

This study aimed to explore attentional patterns among children with inattentive attention-deficit/hyperactivity disorder (ADHD-I) and children with typical development (TD), using a latent class analysis (LCA). Patterns of brain connectivity were also explored. The sample comprised 29 ADHD-I and 29 TD matched children. An LCA was conducted to reclassify subjects according to their attentional performance, considering cognitive measures of attention and behavioral symptoms, regardless of group of origin. The new clusters were then compared in respect to brain white matter measurements (extracted from diffusion tensor imaging). Participants were rearranged in 2 new latent classes, according to their performance in an attention task and the results of behavioral scales, resulting in groups with more homogeneous attentional profiles. A comparison of the 2 new classes using the white matter measurements revealed increased fractional anisotropy in the left inferior fronto-occipital fasciculus and left inferior longitudinal fasciculus for the class composed by participants with a higher risk of attentional problems. The findings indicated that it was possible to observe variability regarding neuropsychological profile, accompanied by underpinning neurobiological differences, even among individuals with the same disorder subtype - inattentive ADHD. This specific data-driven clustering analysis may help to enhance understanding of the pathophysiology of the disorder's phenotypes.

ABSTRACT

In this study, we investigated the chemical composition, and antioxidant and antibacterial properties of ethanolic extracts of propolis (EEP) from Melipona quadrifasciata quadrifasciata and Tetragonisca angustula. Chemical composition of EEP was determined by colorimetry and chromatographic (HPLC-DAD and UPLC-Q/TOF-MS/MS) analysis. Antimicrobial activity of EEP was evaluated against gram-positive (S. aureus, methicillin-resistant S. aureus, E. faecalis) and gram-negative (E. coli and K. pneumoniae) bacteria by the minimal inhibitory concentration (MIC) test using the microdilution method. Furthermore, the growth curve and integrity of cell membrane of S. aureus and E. coli were investigated using standard microbiological methods. HPLC-DAD analysis showed that the EEP of M. quadrifasciata quadrifasciata has a more complex chemical composition than the EEP of T. angustula. Moreover, UPLC-MS analyses of M. quadrifasciata quadrifascita indicated flavonoids and terpenes as major constituents. The bactericidal activity of both EEPs was higher against gram-positive bacteria than for gram-negative bacteria. The EEP from M. quadrifasciata quadrifasciata presented MIC values lower than the EEP from T. angustula for all tested bacteria. The EEP from M. quadrifasciata quadrifasciata caused lysis of the bacterial wall and release of intracellular components from both E. coli and S. aureus. Our findings indicate that the chemical composition of propolis from stingless bees is complex and depends on the species. The extract from M. quadrifasciata quadrifascita was more effective against gram-positive than gram-negative strains, especially against S. aureus and methicillin-resistant S. aureus compared to T. angustula extract, by a mechanism that involves disturbance of the bacterial cell membrane integrity.

BACKGROUND: Cri du Chat syndrome (CdCS) is a genetic syndrome caused by deletions in the short arm of chromosome 5. Although the main clinical features of CdCS are well known, the neurocognitive and behavioural characteristics of the phenotype are rarely described in detail in the literature. In this study, we analysed the main phenotypic features of CdCS from a parental perspective. METHOD: A questionnaire was sent to 700 Brazilian families that were registered in the Brazilian Association of CdCS. The questions involved specific domains of CdCS, such as pregnancy and birth conditions, recurrence of the disease in the family, current major health problems, and aspects of cognitive development. RESULTS: In total, 73 questionnaires were completed: 44 females and 29 males, ranging from 9.5 months old to 40 years old (mean = 13.8 years; median = 12 years). Most of the parents noticed the typical cat-like cry at birth (94.4%). The age at diagnosis of CdCS ranged from the time of birth to 180 months (mean = 14 months; median = 6 months), while one case was diagnosed during pregnancy. In all of the cases, the diagnosis of CdCS was made by G-banding karyotype analysis. In 66.2% of the cases, the parents underwent cytogenetic investigation. A total of 52.1% of the parents answered that they did not remember what the recurrence risk of CdCS was in their family. The main health problems that were reported were as follows: swallowing problems (80.3%), feeding problems (80.3%), congenital heart disease (31.5%), spine abnormalities (28.8%), and neurological symptoms (20.5%), including seizures (11%). The behavioural problems that were reported were as follows: aggressive behaviour, stereotypies, anxiety, phobias, and genital manipulation/masturbation. Neurodevelopmental delay was reported in all of the cases. Independent walking was achieved in 72.2% of the patients. Approximately 50% of the patients never presented expressive language, and most of the patients are dependent on others for their daily activities. CONCLUSIONS: The questionnaire was a pioneer initiative in the CdCS support group, and the answers used in this study can improve the health care assistance to these patients because they focus attention on the demands from a parental perspective. In addition, nearly half of the families stated that they did not remember information regarding recurrence risk, which reinforces the importance of genetic counselling follow-up and the need for the expansion of genetic services in Brazil.

BACKGROUND: Gestational diabetes is a risk factor for future development of type 2 diabetes. The primary aim of this study was to estimate the prevalence of postpartum glucose tolerance status evaluation in pregnancies complicated by gestational diabetes 6-12 weeks after delivery. The secondary one was to identify the factors that are implicated with postpartum glucose retesting. METHODS: This was a retrospective study performed with a cohort of women with gestational diabetes, with prenatal care and delivery at a tertiary care center, from January 2013 to April 2017. The diagnosis of gestational diabetes was based on IADPSG criteria (Fasting ≥ 92 mg/dl, 1 h ≥ 180 mg/dl and/or 2 h ≥ 153 mg/dl, respectively) and the diagnosis of type 2 diabetes and prediabetes were made using the 2016 ADA's criteria (fasting and 2 h after glucose load ≥ 126 mg/dl and/or ≥ 200 and 100-125 mg/dl and/or 140 and 199 mg/dl, respectively). All women had an appointment scheduled 6-12 weeks postpartum with the results of a 75-g oral glucose tolerance test (OGTT). RESULTS: Of the 152 evaluated women, 21 (13.8%) returned with the postpartum OGTT results. Of these, 9 (45.0%) had a diagnosis of prediabetes. The use of insulin during gestation was the only factor implicated in a higher adherence rate to postpartum testing OR 6.33 (p 0.002). No significance was found for other demographic and clinical variables (age, family income, years of study, parity, gestational age at first visit, smoking, family history of type 2 diabetes, diagnosis of gestational diabetes before the third trimester, pregestational body mass index, previous history of gestational diabetes and ethnicity). CONCLUSION: The majority of patients with gestational diabetes did not return postpartum to perform OGTT and in our study the only factor implicated in a higher postpartum return was the use of insulin during pregnancy. Considering that 45.0% were diagnosed with prediabetes, diabetes care teams should initially identify non-adherent patients.

The aim of this systematic review and meta-analysis was to compare the survival rate of the implants and the peri-implant tissue changes associated with implants inserted in fresh extraction sockets and those inserted in healed sockets. This review has been registered at PROSPERO under the number CRD42016043309. A systematic search was conducted by two reviewers independently in the databases PubMed/MEDLINE, Embase, and the Cochrane Library using different search terms; articles published until November 2016 were searched for. The searches identified 30 eligible studies. A total of 3,049 implants were installed in a total of 1,435 patients with a mean age of 46.68 years and a minimum of 6 months of follow-up. The survival rate of delayed implants (98.38%) was significantly greater than immediate implants (95.21%) (p=.001). For the marginal bone loss (p=.32), implant stability quotients values (p=.44), and pocket probing depth (p=.94) there was no significant difference between the analysed groups. The immediate implants placed in fresh sockets should be performed with caution because of the significantly lower survival rates than delayed implants inserted in healed sockets.

ABSTRACT

Culex (Melanoconion) pedroiSirivanakarn & Belkin 1980 and Culex (Melanoconion) ribeirensisForattini & Sallum 1985 are two morphologically very similar species of the Pedroi subgroup of mosquitoes in the Spissipes section of the subgenus Melanoconion of the genus Culex L. 1758. We carried out an analysis of the mitochondrial cytochrome c oxidase I (COI) DNA variation between these two species. The recent observation of sympatric coexistence in a forested area of Rio de Janeiro State (Brazil) triggered the need to validate these two species previously identified based on morphology. We concluded that the COI is a useful tool for identification of Cx. pedroi and Cx. ribeirensis.

ABSTRACT

O plasmocitoma ósseo solitário é uma neoplasia incomum, não secretória que pode acometer tanto o esqueleto apendicular como o axial. Os sinais clínicos dos plasmocitomas ósseos incluem dor, claudicação e alterações neurológicas relacionadas à compressão medular. A suspeita diagnóstica é baseada na avaliação radiográfica caracterizada por lesões ósseas líticas e a confirmação diagnóstica é realizada por meio do exame histopatológico, sendo o mieloma múltiplo um dos principais diagnósticos diferenciais. O tratamento para pacientes portadores de plasmocitoma ósseo solitário incluem cirurgias, radioterapia ou ainda tratamento paliativo direcionado para o controle da dor óssea quando a cirurgia não for uma opção viável. O objetivo do presente relato é descrever três casos de plasmocitomas ósseos solitários em cães sendo dois em vértebras lombares, que foram submetidos à remoção cirúrgica e que tiveram boa resposta ao tratamento empregado. Apesar de pouco descrito na literatura, o plasmocitoma ósseo solitário é um importante diferencial para lesões líticas em cães e pode apresentar tempo livre de progressão de doença e sobrevidas superiores a outras neoplasias ósseas como osteossarcoma quando submetidos ao tratamento adequado.


Solitary osseous plasmocytoma (SOP) is an uncommon, non-secretory neoplasm that can affect both the appendicular and axial skeletons. Clinical signs of bone plasmocytomas include pain, lameness and neurological changes related to spinal cord compression. The diagnosis is based on radiographic evaluation characterized by lytic bone lesions and confirmation is through histopathological examination, with multiple myeloma being one of the main differential diagnoses. Treatment of patients with SOP includes surgery, radiation therapy or palliative treatment directed at bone pain control when surgery is not a viable option. The objective of the present report is to describe three cases of SOP, two in lumbar vertebrae that were submitted to surgical removal with good response to the treatment. Although less described in the literature, SOP is an important differential for lytic lesions in dogs and may present disease free interval and survival time superior to other bone neoplasms such as osteosarcoma when submitted to adequate treatment.

Eggs were obtained from adult female of Ps. ferox collected in the Poços das Antas Biological Reserve (Reserva Biológica de Poços das Antas, Rio de Janeiro, Brazil). From each female, one portion of eggs (n = 5) was reared for identification purposes, and the second portion (n = 10) was examined using SEM for morphometric analysis. The egg morphology was then compared to that of eggs from Ps. ferox populations in Florida (USA) and Arena (Trinidad). The exochorion ornamentation of the populations differs considerably in the morphology of the tubercles of the chorionic cells, external chorionic reticulum, micropylar collar, and micropyle.


Resumo Caracterização da morfologia externa dos ovos do mosquito é importante, uma vez que facilita a identificação de materiais a partir de locais de reprodução e contribui para a compreensão da biologia das espécies e sistemática do grupo. O objetivo do presente estudo foi comparar os ovos de diferentes populações Psorophora ferox usando MEV. Os ovos foram obtidos a partir de fêmeas de Ps. ferox que foram coletadas na Reserva Biológica de Poço das Antas (Reserva Biológica de Poço das Antas, Rio de Janeiro, Brasil). A partir de cada fêmea, uma parte dos ovos (n = 5) foi criado para fins de identificação, e uma segunda parte (n = 10) foi examinado usando MEV para análise morfométrica. Foi então comparada a morfologia dos ovos de populações de Ps.ferox da Flórida (EUA) e Arena (Trinidad). A ornamentação do exocório das populações difere consideravelmente na morfologia dos tubérculos das células coriônicas, retículo coriônico externo, colar micropilar e micrópila.

O plasmocitoma ósseo solitário é uma neoplasia incomum, não secretória que pode acometer tanto o esqueleto apendicular como o axial. Os sinais clínicos dos plasmocitomas ósseos incluem dor, claudicação e alterações neurológicas relacionadas à compressão medular. A suspeita diagnóstica é baseada na avaliação radiográfica caracterizada por lesões ósseas líticas e a confirmação diagnóstica é realizada por meio do exame histopatológico, sendo o mieloma múltiplo um dos principais diagnósticos diferenciais. O tratamento para pacientes portadores de plasmocitoma ósseo solitário incluem cirurgias, radioterapia ou ainda tratamento paliativo direcionado para o controle da dor óssea quando a cirurgia não for uma opção viável. O objetivo do presente relato é descrever três casos de plasmocitomas ósseos solitários em cães sendo dois em vértebras lombares, que foram submetidos à remoção cirúrgica e que tiveram boa resposta ao tratamento empregado. Apesar de pouco descrito na literatura, o plasmocitoma ósseo solitário é um importante diferencial para lesões líticas em cães e pode apresentar tempo livre de progressão de doença e sobrevidas superiores a outras neoplasias ósseas como osteossarcoma quando submetidos ao tratamento adequado.

Solitary osseous plasmocytoma (SOP) is an uncommon, non-secretory neoplasm that can affect both the appendicular and axial skeletons. Clinical signs of bone plasmocytomas include pain, lameness and neurological changes related to spinal cord compression. The diagnosis is based on radiographic evaluation characterized by lytic bone lesions and confirmation is through histopathological examination, with multiple myeloma being one of the main differential diagnoses. Treatment of patients with SOP includes surgery, radiation therapy or palliative treatment directed at bone pain control when surgery is not a viable option. The objective of the present report is to describe three cases of SOP, two in lumbar vertebrae that were submitted to surgical removal with good response to the treatment. Although less described in the literature, SOP is an important differential for lytic lesions in dogs and may present disease free interval and survival time superior to other bone neoplasms such as osteosarcoma when submitted to adequate treatment.(AU)
