ABSTRACT

The present study evaluated the efficacy of the mineralizing action of Casearia sylvestris ethanolic extract on bovine dentin blocks in its pure form and in dental paste, through scanning electron microscopy. The dentin blocks were immersed in artificial saliva and incubat ed at 37°C for 7 days. Subsequently, six groups were treated with different test substances and analysed qualitatively and quantitatively at 30 and 60 days. The tests used were Kruskal - Wallis and Dunn's. Shapiro - Wilk and ANOVA. The qualitative analysis at 30 days showed a difference between the groups treated with ethanolic extract and toothpaste. Quantitatively, at 30 days, treatment with ethanolic extract of Casearia showed a greater number of open dentinal tubules. At 60 days, the difference persisted on ly for the blocks treated with toothpaste. The results obtained indicated that there is a positive relationship between the use of Casearia sylvestris and obliteration of dentinal tubules

ABSTRACT

OBJECTIVE: Despite commitment by many countries to promote food system transformation, Australia has yet to adopt a national food policy. This study aimed to evaluate Australian Federal Government's (AFG) food policies and policy actions potential to promote healthy and sustainable food systems. DESIGN: This study is a desk-based policy mapping followed by a theoretically guided evaluation of policy actions. This involved three steps: (1) identification of government departments and agencies that could influence Australia's food system; (2) identification of food policies and policy actions within these departments and (3) use of a conceptual framework to evaluate policy actions' potential of changing the food system as adjust (first-order change), reform (second-order change) or transform (third-order change). SETTING: Australia. PARTICIPANTS: None. RESULTS: Twenty-four food policies and sixty-two policy actions were identified across eight AFG departments and the Food Regulation System and evaluated based on the order of change they represented. Most policies were led by individual departments, reflecting the absence of a joined-up approach to food policy in Australia. Most policy actions (n 25/ 56·5 %) were evaluated as having adjust potential, whereas no transformative policy action was identified. CONCLUSIONS: These findings suggest that Australia is likely to proceed incrementally towards achieving food system change through adjustments and reforms but lacking transformative impact. To promote transformative change, all three orders of change must be strategically implemented in a coherent and coordinated matter. A comprehensive national food policy and a national coordinating body are needed to ensure a cohesive approach to policy.

ABSTRACT

This study aimed to evaluate the level of counting by indicator microorganisms, identify the microbial ecology, detect Listeria monocytogenes and Salmonella sp., and determine the presence of virulence genes and biofilm formation. A total of 480 samples were collected from the surfaces of the equipment and utensils using sterile swabs for the detection of L. monocytogenes and Salmonella sp. and counting mesophilic aerobes, Enterobacteriaceae, Escherichia coli, and Pseudomonas sp. The microbial ecology was evaluated by sequencing the 16S rRNA gene. Genes for virulence and biofilm formation were analyzed and adhesion capacity was evaluated for L. monocytogenes and Salmonella sp. The mesophilic aerobe count was the highest in the dairy processing facility, followed by the pork and poultry slaughterhouses. L. monocytogenes was detected in all facilities, with the highest detection in the pork slaughterhouse, followed by the poultry and dairy facilities. Salmonella sp. was only detected in the dairy. Isolates of L. monocytogenes and Salmonella sp. showed poor adhesion to polystyrene surfaces, virulence genes, and biofilm formation. The frequent contaminants in the slaughterhouses were Pseudomonas, Acinetobacter, and Aeromonas in poultry, Acinetobacter, Pseudomonas, and Brevundimonas in pork, and Pseudomonas, Kocuria, and Staphylococcus in dairy. Our results provide useful information to understand the microbiological risks associated with contamination.

ABSTRACT

OBJECTIVE: The choice of terms used to describe 'foods to limit' (FTL) in food-based dietary guidelines (FBDG) can impact public understanding, policy translation and research applicability. The choice of terms in FBDG has been influenced by available science, values, beliefs and historical events. This study aimed to analyse the terms used and definitions given to FTL in FBDG around the world, including changes over time and regional differences. DESIGN: A review of terms used to describe FTL and their definitions in all current and past FBDG for adults was conducted, using a search strategy informed by the FAO FBDG website. Data from 148 guidelines (96 countries) were extracted into a pre-defined table and terms were organised by the categories 'nutrient-based', 'food examples' or 'processing-related'. SETTING: National FBDG from all world regions. PARTICIPANTS: None. RESULTS: Nutrient-based terms (e.g. high-fat foods) were the most frequently used type of term in both current and past dietary guidelines (91 %, 85 %, respectively). However, food examples (e.g. cakes) and processing-related terms (e.g. ultra-processed foods) have increased in use over the past 20 years and are now often used in conjunction with nutrient-based terms. Regional differences were only observed for processing-related terms. CONCLUSION: Diverse, and often poorly defined, terms are used to describe FTL in FBDG. Policymakers should ensure that FTL terms have clear definitions and can be integrated with other disciplines and understood by consumers. This may facilitate the inclusion of the most contemporary and potentially impactful terminology in nutrition research and policies.

ABSTRACT

In this context, the objective of the work was to explore the mechanisms of action of antimicrobials, resistance and their importance in public health. Pubmed, Google academic and Scielo databases were used for this research. According to the World Health Organization, resistance to antibiotics is defined when it no longer has an effect. The increasingly frequent insertion of antimicrobials favors resistance, where they put selective pressure on microorganisms, making them resistant to various drugs. The indiscriminate use of antimicrobials is the main factor of microbial resistance, as well as the use of antimicrobials without culture examination and sensitivity test. In this sense, it is concluded that it is extremely important to update protocols that contain the mechanisms of bacterial resistance in order to minimize the indiscriminate use of antimicrobials, as well as to train health professionals for this problem in public health.

ABSTRACT

Background and Aim: Acute kidney injury (AKI) is associated with a grave prognosis. A clinical assessment of kidney function can be performed based on the glomerular filtration rate (GFR). Cystatin C (CysC) can indicate the GFR or kidney function and its measurement is currently performed using immunological methods such as nephelometry, immunoturbidimetry, and enzyme-linked immunosorbent assays in human medicine. However, these techniques are not specific for use in veterinary medicine. This study aimed to validate an immunoturbidimetric assay for serum CysC (sCy) in dogs, determine the sCy reference intervals for healthy dogs, evaluate sCy stability in serum samples, and compare sCy with serum creatinine (sCr) in healthy dogs and dogs with AKI. Materials and Methods: Forty-three dogs were divided into a control group (n = 19) and an AKI group (n = 24). An immunoturbidimetric method including commercially available human CysC calibrated with canine CysC was used to evaluate canine serum samples. Results: An average recovery of 97% was observed for canine serum samples. The reference interval for CysC in healthy dogs was 0.57-1.29 mg/L. The sCy concentration in dogs with AKI was significantly higher (2.82 ± 1.46 mg/L) than in healthy dogs (0.93 ± 0.18 mg/L). Statistical analysis confirmed a strong correlation between sCy and sCr (r = 0.94; p < 0.05) in dogs with AKI. Conclusion: The immunoturbidimetric method of evaluating sCy yielded satisfactory results and can be used for canine samples when a species-specific calibrator is used. Furthermore, sCy is a reliable marker of renal dysfunction in dogs. It is best to store samples for sCy evaluation at temperatures between 4°C and 8°C.

ABSTRACT

Accidents with snakes from the genus Bothrops represent ~90 % of all snakebites in Brazil. Monoclonal antibodies (mAbs) targeting venom components can be important assets for treating envenoming syndromes, for developing diagnostic tests and for research purposes. Therefore, in this study, we aimed to generate murine mAbs against the antigenic mixture of Bothropic venoms traditionally used as immunogen to produce Bothropic antivenoms in Brazil. ELISA showed that one of the produced mAbs recognizes B. alternatus and B. neuwiedi venoms (mAb anti-Ba/Bn) specifically and Western Blot revealed that this mAb binds to a single protein band of molecular mass of ≈50 kDa. MAb anti-Ba/Bn inhibited the coagulant activity but was unable to neutralize hemorrhagic and phospholipase A2 activities caused by the B. neuwiedi venom. MAb anti-Ba/Bn was immobilized to Sepharose beads and used for immunoaffinity chromatography of B. neuwiedi venom. Proteolytic activity assays indicated that the immunoaffinity-purified fraction (BnF-Bothrops neuwiedi fraction) has a serine protease thrombin-like profile, which was supported by coagulability assays in mice. Bottom-up proteomic analysis confirmed the prevalence of serine proteases in BnF using label-free quantification. In conclusion, this work characterized a mAb with neutralizing properties against B. neuwiedi coagulant activity and demonstrates that immunoaffinity chromatography using mAbs can be a useful technique for purification of bioactive toxic proteins from Bothrops spp. snake venoms.

ABSTRACT

Critically ill hospitalized dogs are subject to certain complications, being acute kidney injury (AKI) a common one. Early diagnosis is crucial, and Cystatin C (CysC) is a reliable and early biomarker. The International Society of Renal Interest (IRIS) states that AKI severity can be assessed by mild changes in creatinine serum levels or reduction of urine output that cannot be considered biomarkers of renal injury but failure or insufficiency. Twenty-eight dogs admitted to the Intensive Care Unit under risk factors for the development of AKI were evaluated. Blood samples were collected for determination of sCr and CysC at admission and after 24, 48, and 72 h. Urine output was measured by daily monitoring, measured by collection in a closed system. The results showed the incidence of AKI was 67.9% based on the IRIS criteria and 78.6% based on cystatin C in critically ill patients' dogs. The measurement of serum cystatin C immediately on admission to the ICU was superior in the early identification of patients with AKI when compared to the IRIS classification and serum creatinine in critically ill dogs.

ABSTRACT

BACKGROUND: Hepatosplenic schistosomiasis mansoni (HS) is associated with thrombocytopenia. Accurate platelet counts are required for identification and management of HS patients. EDTA-dependent pseudothrombocytopenia (EDTA-PTCP) is an in vitro phenomenon of anticoagulant-activated platelet agglutination resulting in low platelet counts by automated methods. The prevalence of EDTA-PCTP in schistosomiasis is unknown and only one case has been described. Our aims were to determine the prevalence of EDTA-PTCP in HS and evaluate alternative methods to overcome this analytical error. METHODS: Blood samples from 56 HS patients and 56 healthy volunteers were collected, and platelet counts were obtained using standard microscopy and automated (electric impedance) methods. Automated platelet counts and the presence of platelet clumps in blood smears were evaluated in samples collected in EDTA or sodium citrate tubes 20 and 180 min after blood collection. RESULTS: EDTA-PTCP was more frequent in HS patients than healthy volunteers (8.92% vs 0.00%, p<0.0285). Platelet clumps and PTCP were also observed in samples collected in sodium citrate tubes, refuting its use as an alternative method. CONCLUSIONS: Automated platelet counts in blood samples from HS patients should be performed right after blood collection in EDTA tubes and verified by manual counts in blood smears.

ABSTRACT

BACKGROUND: The root system plays a major role in plant growth and development and root system architecture is reported to be the main trait related to plant adaptation to drought. However, phenotyping root systems in situ is not suited to high-throughput methods, leading to the development of non-destructive methods for evaluations in more or less controlled root environments. This study used a root phenotyping platform with a panel of 20 japonica rice accessions in order to: (i) assess their genetic diversity for a set of structural and morphological root traits and classify the different types; (ii) analyze the plastic response of their root system to a water deficit at reproductive phase and (iii) explore the ability of the platform for high-throughput phenotyping of root structure and morphology. RESULTS: High variability for the studied root traits was found in the reduced set of accessions. Using eight selected traits under irrigated conditions, five root clusters were found that differed in root thickness, branching index and the pattern of fine and thick root distribution along the profile. When water deficit occurred at reproductive phase, some accessions significantly reduced root growth compared to the irrigated treatment, while others stimulated it. It was found that root cluster, as defined under irrigated conditions, could not predict the plastic response of roots under drought. CONCLUSIONS: This study revealed the possibility of reconstructing the structure of root systems from scanned images. It was thus possible to significantly class root systems according to simple structural traits, opening up the way for using such a platform for medium to high-throughput phenotyping. The study also highlighted the uncoupling between root structures under non-limiting water conditions and their response to drought.

ABSTRACT

Snakebites caused by Crotalus genus are the second most frequent in Brazil. Crotoxin is a beta-neurotoxin responsible for the main envenomation effects of Crotalus biting, while crotamine immobilizes the animal hind limbs, contributing to prey immobilization and to envenoming symptoms. As crotoxin and crotamine represent about 90% of Crotalus venom dry weight, these toxins are of great importance for antivenom therapy. In this sense, knowledge regarding the antigenicity/immunogenicity at the molecular level of these toxins can provide valuable information for the improvement of specific antivenoms. Therefore, the aims of this study are the identification of the B-cell epitopes from crotoxin and crotamine; and the characterization of the neutralizing potency of antibodies directed against the corresponding synthetic epitopes defined in the current study. Linear B-cell epitopes were identified using the Spot Synthesis technique probed with specific anti-C. d. terrificus venom horse IgG. One epitope of crotamine (F12PKEKICLPPSSDFGKMDCRW32) and three of crotoxin (L10LVGVEGHLLQFNKMIKFETR30; Y43CGWGGRGRPKDATDRCCFVH63 and T118YKYGYMFYPDSRCRGPSETC138) were identified. After synthesis in their soluble form, the peptides mixture correspondent to the mapped epitopes was entrapped in liposomes and used as immunogens for antibody production in rabbits. Anti-synthetic peptide antibodies were able to protect mice from the lethal activity of C. d. terrificus venom.

ABSTRACT

ABSTRACT: The objective of this research was to determine the optimal plot size and the number of replications to evaluate the fresh matter of ryegrass sown to haul. Twenty uniformity trials were conducted, each trial with 16 basic experimental units (BEU) of 0.5 m2. At 117, 118 and 119 days after sowing, the fresh matter of ryegrass in the BEUs of 5, 10 and 5 uniformity trials, respectively, were determined. The optimal plot size was determined by the maximum curvature method of the variation coefficient model. Next, the replications number was determined in scenarios formed by combinations of i treatments (i = 3, 4, ... 50) and d minimum differences between means of treatments to be detected as significant at 5% of probability by the Tukey test, expressed in experimental mean percentage (d = 10, 11, ... 20%). The optimal plot size to determine the fresh matter of ryegrass seeded at the haul is 2.19 m2, with a variation coefficient of 9.79%. To identify as significant at 5% probability, by the Tukey test, differences between treatment means of 20%, are required five, six, seven and eight replications, respectively, in ryegrass experiments with up to 5, 10, 20 and 50 treatments.

Abstract Objective To analyse the effects of warm shower, perineal exercises with a Swiss ball or both during the labour in maternal and perinatal parameters. Methods Randomised controlled trial with 101 low-risk birthing women admitted in two public midwife-led birth centres, between June, 2013 and February, 2014, with minimal age 18 years, full-term gestation, single live foetus in cephalic presentation, cervical dilation 3-8 cm, pain score ≥5, without clinical or obstetric pathologies or mental illness, non-users of psychoactive drugs or synthetic or natural corticosteroids and who had not used tobacco, caffeine and analgesics in the previous two, four and six hours before inclusion in the study, respectively. The non-pharmacological interventions were for 30 minutes performed. Maternal and perinatal parameters were assessed before and 30 minutes after the interventions, including: maternal blood pressure, heart rate, respiratory rate, uterine contractions, cervical dilation, foetal heart rate, baseline, variability, accelerations and decelerations using cardiotocography and Apgar score (at the 1stand 5thminutes after birth); The participants were randomly assigned in group A warm shower (33), B Swiss ball (35) and C combined interventions (33). Results Concerning maternal parameters, systolic blood pressure was kept above 100 mmHg, with a little increase in the group B. Diastolic blood pressure decreased in all the groups, however was maintained above 70 mmHg. The heart rate decreased in the group B and C and was above 80 bpm. The respiratory rate was above 20 rpm in all groups after the interventions, while the cervical dilation before the interventions were in average 5.0 cm and increased 1.3 cm after the interventions in all groups. Concerning the foetal parameters, foetal heart rate was normal in more than 90% in all groups at both evaluation times, transient acceleration was present in more than 80% in all groups at both evaluation times and no decelerations were found before the intervention in approximately 58.4% of the cases. Decelerations were observed in 52.5% of the cases, mainly in the groups A and B. Variability was normal in more than 80% of the cases, and the Apgar score ˂7 at the first minute after birth was observed in 14 cases only. No significant differences were found in maternal blood pressure, pulse rate, foetal heart rate including the occurrence of transient accelerations, variability or decelerations and Apgar at the inter and intragroup analysis or by evaluation time. By comparing maternal parameters before and 30 minutes after the interventions, increased maternal respiratory rate (p=0.037) and cervical dilation (p<0.001) were found for the all intervention groups. At the intergroup analysis, group A (p=0.041) and group C (p=0.021) stimulated labour progression regarding the uterine contractions increased in comparison to the group B. Conclusion The interventions alone or in combination are a safe way for childbirth assistance as they do not result in negative effects on maternal and perinatal parameters.

ABSTRACT

ABSTRACT: The objective of this study was to characterize the production of biquinho pepper through the interpretation of parameter estimates from the logistic model and its critical points obtained by the partial derivatives of the function, and to indicate the best cultivar and growing season for subtropical climate sites. For this, a 2x3 factorial experiment was conducted with two cultivars of biquinho pepper (BRS Moema and Airetama biquinho) in three growing seasons (E1: October 2015, E2: November 2015, E3: January 2016). The logistic non-linear model for fruit mass was specified as a function of the accumulated thermal sum, and the critical points were calculated through the partial derivatives of the model, in order to characterize the productive performance of the crop by the biological interpretation of the estimates of the three set parameters. In E3, temperatures close to 0 ºC during the experiment were lethal to the plants, and a linear regression model was used in this case. The production of the cultivars in E1 and E2 were well characterized by the estimated logistic models, and the most productive cultivar was Airetama biquinho in all evaluated seasons. This cultivar also presented higher concentration of production. The two cultivars did not differ significantly with regards to productive precocity. For E3, it was not possible to interpret the parameters in the same way as for E1 and E2, since the use of the linear model did not allow the same interpretations performed for the nonlinear model, reaffirming its applicability horticultural crops of multiple harvests.

ABSTRACT

BACKGROUND: Acute kidney injury (AKI) is a common complication in patients undergoing liver transplant (LT) and is associated with high morbidity and mortality. We aim to evaluate the pattern of urine and plasma neutrophil gelatinase-associated lipocalin (NGAL) elevation during the perioperative period of LT and to assess it as a prognostic marker for AKI progression, need for dialysis and mortality. METHODS: We assessed NGAL levels before induction of anesthesia, after portal reperfusion and at 6, 18, 24, and 48 h after surgery. Patients were monitored daily during the first week after LT. RESULTS: Of 100 enrolled patients undergoing liver transplant, 59 developed severe AKI based on the KDIGO serum creatinine (sCr) criterion; 34 were dialysed, and 21 died within 60 days after LT. Applying a cut-off value of 136 ng/ml, UNGAL values 6 h after surgery was a good predictor of AKI development within 7 days after surgery, having a positive predictive value (PPV) of 80% with an AUC of 0.76 (95% CI 0.67-0.86). PNGAL at 18 h after LT was also a good predictor of AKI in the first week, having a PPV of 81% and AUC of 0.74 (95% CI 0.60-0.88). Based on PNGAL and UNGAL cut-off criteria levels, time to AKI diagnosis was 28 and 23 h earlier than by sCr, respectively. The best times to assess the need for dialysis were 18 h after LT by PNGAL and 06 h after LT by UNGAL. CONCLUSION: In conclusion, the plasma and urine NGAL elevation pattern in the perioperative period of the liver transplant can predict AKI diagnosis earlier. UNGAL was an early independent predictor of AKI development and need for dialysis. Further studies are needed to assess whether the clinical use of biomarkers can improve patient outcomes. ABSTRACT

ABSTRACT: The objectives of this work were estimate the leaf area of squash 'Brasileirinha' by linear dimensions of the leaves and check models available in the literature. An experiment was conducted in the 2015/16 sowing season. Were collected 500 leaves and in each one, were measured the length (L), width (W) and length×width product (LW) and determined the real leaf area (LA). Then, 400 leaves were separated to generate models of the leaf area (LA) as a function of linear dimension (L, W or LW) of squash. The remaining 100 leaves were used for the validation of models. A second experiment was conducted in the 2016/17 sowing season. Were collected 250 leaves, used only for the validation of the models of the first experiment. There is collinearity between L and W and, therefore, models using the LW product are not recommended. The model LA=0.5482W2 + 0.0680W (R²=0.9867) is adequate for leaf area estimation of squash 'Brasileirinha'.

ABSTRACT

ABSTRACT Objective The perception of marijuana among mental health professionals is unknown in Brazil. The aim of this study was to compare mental health professionals' perception and knowledge standards about marijuana in three institutions with different profiles. Methods Seventy professionals (psychiatrists, psychologists, social workers, occupational therapists and nurses) from three mental health services - Nossa Senhora de Fátima Hospital (philanthropic hospital), CAPS-AD (public specialized service on treating drugs and alcohol dependents), and the Institute of Psychiatry at the Clinics Hospital, Medical School, University of São Paulo (academic and assistance service) were evaluated through a 31 affirmatives questionnaire created by researchers. Results Psychiatrists disagree more often than other health professionals on the need to treat marijuana users and on the idea that it causes cognitive damages. The professionals from Nossa Senhora de Fátima Hospital are more in favor of the prohibition of marijuana use and consider more the possible damages associated with its use, when compared to professionals from the other two institutions. Conclusions The perception of marijuana among mental health providers may vary according to their profession and work place.
