ABSTRACT

In western Colorado, Cytospora leucostoma is ubiquitous in peach orchards and has developed into a major limiting factor of peach production. The pathogen is unable to invade healthy intact phloem tissue of the tree, but instead, it requires a wound as a mode of entry. Bark injuries caused by cold and pruning in commercial orchard systems provide infection courts that, in suitable environment conditions, can lead to many successful fungal infections. Preventive fungicide control is an integral component of management in tree fruit production. Eighteen fungicides were tested at selected label dose rates for C. leucostoma control. All treatments were initially tested in vitro in fungicide-amended media dishes. Successful treatments were then tested under controlled conditions on detached peach branch segments. Effective fungicides identified in the laboratory assays (thiophanate-methyl, captan, lime sulfur, and copper hydroxide) were further tested as spray applications in the field and as wound sealant applications in combination with latex paint and kaolin clay. Of the treatments evaluated, thiophanate-methyl, captan, 50% latex paint, thiophanate-methyl amended in 50% latex paint, captan amended in 50% latex paint, and lime sulfur were most effective in reducing C. leucostoma necrotic area. Copper hydroxide was ineffective in all field trials and in some instances, yielded larger necrotic areas than the nontreated positive control shoots.

ABSTRACT

Changes in quality, phytochemical content and cell wall metabolism of two loquat cultivars (Eriobotrya japonica cvs. 'Morphitiki', 'Karantoki') under different storage regimes were studied. The fruit were harvested at commercial maturity stage and analyzed after 1, 3, 5, 7, and 11 days maintenance at room temperature (RT, ∼ 20°C) or after cold storage (14 days at 4°C) and additional ripening at RT for 1, 3 and 5 days, respectively. Compositional analysis revealed substantial cultivar differences; the 'Morphitiki' fruit was more acidic and showed higher contents of total phenolics, flavonoids and hydroxycinnamic acid-derivatives as well as greater antioxidant potency. Although firmness did not change markedly during storage, the cell wall exhibited extensive remodeling. Greater changes were observed in the pectin backbones than in polyuronide side chains and cross-linking glycans. Polygalacturonase (PG) showed better association with cell wall solubilization at RT than the enzymes involved in arabinan or galactan disassembly. During postharvest ripening after harvest, 'Karantoki' showed more extensive pectin solubilization than 'Morphitiki'. Interestingly, cold storage inhibited the cell wall disassembly in 'Karantoki' but not in 'Morphitiki', suggesting that the cultivars may differ in their susceptibility to chilling-related wall disorders. Low temperature-induced alterations in wall disassembly may impact juice and phytochemical release upon consumption.

ABSTRACT

The incidence of pathogens associated with postharvest fruit rots on the four most extensively cultivated apple cultivars (Red Delicious, Golden Delicious, Granny Smith, and Fuji) in Greece was surveyed during two consecutive storage periods (2008-09 and 2009-10) in five packinghouses located in northern Greece. The fungi isolated were identified based on their morphological characteristics and internal transcribed spacer gene sequencing. In the four cultivars sampled, Penicillium expansum and Botrytis cinerea were the predominant pathogens, accounting for averages of 44.2 and 23.6%, respectively, of the pathogens isolated from the sampled fruit. Two other important rot pathogens were Alternaria tenuissima and Mucor pyriformis, accounting for 16.1 and 6.6%, respectively, of the diseased apple fruit. Other pathogens such as Monilinia laxa, M. fructigena, Botryosphaeria obtusa, Geotrichum candidum, Fusarium avenaceum, and F. proliferatum were isolated at low frequencies and are considered of minor importance. Measurements of the resistance level of the four apple cultivars to fruit rot caused by P. expansum and Botrytis cinerea revealed that Golden Delicious was the most susceptible to blue mold while Fuji was the most susceptible to gray mold infections. Susceptibility to gray mold was negatively correlated with flavonoid and phenol concentration as well to fruit antioxidant activity, while susceptibility to blue mold was negatively correlated with fruit firmness and phenol concentration. Patulin production was significantly higher in Red Delicious and Golden Delicious fruit than in Granny Smith and Fuji fruit and was negatively correlated with the acidity of the fruit. The high incidence of P. expansum and A. tenuissima along with the presence of F. avenaceum and F. proliferatum, all of which are potentially mycotoxin producers, emphasize the risk for mycotoxin contamination of apple fruit juices and by-products. Furthermore, information on the distribution of the pathogens on the main cultivars may be useful for the implementation of strategies to control the diseases and minimize the threat of mycotoxin contamination on each cultivar.
