ABSTRACT

Microarray data analysis is a very critical step and it is currently facing a major challenge to dig out research-related information and provide professional interpretation. Gene function enrichment analysis appears to address the bottleneck issue with reasonable option. This article focuses on the principle, the relevant analytical tools and applications of gene function enrichment analysis, thus further laying the foundation for a wide range of applications.

ABSTRACT

Data analysis and mining is a key issue to microarray technology and is usually implemented through software development. This paper summarizes the state-of-art software development in cDNA microarray data analysis and mining. The updated software developments are discussed in three stages: data inquisition from cDNA microarray tests, statistical treatment of cDNA data and data mining from gene network.

ABSTRACT

Objective To discover new method studying renal deficiency and syndrome of blood stasis by gemellary research method. Methods Through clinical investigation of 300 instances of twins,we choice three twins, include mistress Yang from Qionglai city who is being later stage of older;mistress Yu from Chengdu city who is being presenium;mistress Lu from Beijing who is being Middle-age using five species of ovi-type appraisement methods, and syndrome-penthemeron investigation by blood stasis, stagnation of blood measuring scale, renal deficiency measuring scale, and eight principles measuring scale,in this text.Results(1)Get three family constellation graphs, which provide inspiration for syndrome-penthemeron studing.(2)Though investigating to three twins,we discover that this relative value of eight principles measuring scale has augmented tendency with time-lapse.3.There haven’t regularity from the stagnation of blood measuring scale and renal deficiency measuring scale. Conclusion Using gemellary research method to study Diabetes Mellitus is perfect. In spite of there haven’t ideal result, but it provid prospective thinking and suggestion for us.

ABSTRACT

BACKGROUND: Jinguishenqi pills (JP) can ameliorate the symptoms of kidney deficiency, but the molecular mechanism of the effect is unclear.OBJECTIVE: To observe the differentially expressed genes of panic-induced kidney deficiency model mice treated with JP using suppression subtractive hybridization (SSH).DESIGN: Randomized control animal experiment.SETTING: Institute of Traditional Chinese Medicine Genetics, Chengdu University of Traditional Chinese Medicine; Sichuan Key Laboratory of Molecular Biology and Biotechnology, College of Life Science, Sichuan University.MATERIALS: This experiment was carried out at the Experimental Animal Center of Chengdu University of Traditional Chinese Medicine. Totally 30 BALB/c male adult mice were used in the experiment, and they were randomly divided into model group, JP-treated group and control group, with 10 in each.METHODS: Mice models of kidney deficiency were created in the mice of model group and JP-treated group with the methods of "terrifying mouse with cat" and "suspending mouse over water". Mice of the JP-treated group were intragastrically administrated of JP (produced in Tongrentang Pharmacy Co., Beijing, Z11020147, composed of Guizhi, Fuzi, Shudi, Shanyao,Shanzhuyu, Zexie, Fuling, Danpi, etc) 20-fold as human taken according to the bodyweight once per day. Modeling and administration were conducted in 14 days. Mice of normal control group were fed normally without any extra stimulation. Twenty-four hours after the last administration, 100 μL blood was collected respectively from mice of each group. Switch Mechanism at 5' end of RNA Template (SMART) technique was used in reverse transcription and amplification to obtain the whole length of total cDNA of mice in each group; Forward and reverse SSHs were used to acquire 6 groups of differential expression cDNA segments which were under 3 states(at kidney deficiency, after treatment and at normal physiological status).MAIN OUTCOME MEASURES: Effect of JP on gene expression of kidney deficiency mice RESULTS: ① Differentially expressed cDNA segments were found in each group; The differentially expressed cDNA profile of model mice had restored to that of the normal physiological state by JP treatment. cDNA of the same kind was used as the tester and driver of SSH, and almost all the homologous fragments were subtracted completely, which showed that the subtraction efficiency was very high. ② 6 cDNA libraries were acquired. CONCLUSION: Panic-induced kidney deficiency is related to differential expression of some genes. The gene expression of the kindey deficiency model mice could be affected by JP treatment, making it trend to normal physiological status.

ABSTRACT

Syndromes-genome research performed eight years ago, has made a series of achievements. At the same time, there are still many problems which have to be solved. Based on the research experience, six major problems were summed up, such as difficult to refine into a sample, difficult to sample, difficult to analysis mass data, difficult to combine TCM with western medicine disease, difficult to dock and function between TCM theory and genes, difficult to apply the result of Syndrome- genomics. In this paper, the author also put forward possible new strategies, such as establishing the characteristics of the gene database, combining with the known gene functions likely to explain the evidence of the mechanism for the realization of the modernization of TCM theory, speeding up the unknown gene research, occupying gene annotation territories.

ABSTRACT

Objective:To identify the deficient cold syndrome relevant genes and functional modules, based on gene expression profiles and gene functional knowledge to study the molecular mechanism of deficient cold syndrome. Methods:Screening the differential expression genes with Array Tools, and testing the classificational results to the samples. The Gene Ontology Tool was used for annotating characteristics functional gene, selecting functional modules enriched with differentially expressed genes;and then identifing the feature modules and analyzing their association with the deficient cold syndrome. Results:The accuracy rate of classification samples was 93% with 25 different gene, the accuracy rate was 87%-100% with 11 functional modules. Conclusion:These features function modules had some relevance to the occurrence of deficient cold syndrome. It was more favorable interpretation of the disease by studying functional modules based on gene expression profiles.

ABSTRACT

In modern information age,the Chinese medicine scientific research is developing gradually to the direction of mathematization.Mathematics will become the middle axle, key and tool of chinese medicine multi-disciplinary research. To revive the classical medicine thoughts including "Do anything abiding by the theory of yin and yang and exercising shu shu/healthy techniques"(Suwen?Shanggu Tianzhen Lun).It is a urgent and forward-looking strategy to integrate the mathematics and the traditional Chinese medicine in the 21st century.

ABSTRACT

Objective To study the importance of back pain-lassitude as the key symptom in the differentiation of kidney-deficiency syndrome in diabetes patients. Methods Totally 460 diabetics were divided into the pain group(154 cases)and non-pain group(306 cases). The 39 symptoms, signs and behavior were abstracted and each patient was scored according to the details of kidney deficiency scale to analyze the constitution and distribution of kidney-deficiency syndrome. Results There was a significant difference in the total symptom score of the two groups(P
