ABSTRACT

For the functional analysis of mucin related glycan, we synthesized core 3 and 5 structures of mucin type O-glycan and investigated their binding interaction with lectins using sugar chip technology. The construction of Tn antigen moiety containing α-N-acetylgalactosamine residue was achieved by α-selective glycosylation of 2-azido-6-tert-butyldiphenylsilyl-3,4-di-O-chloroacetyl-2-deoxy-galctopyranosyl imidate and glucose moiety, which acts as a hydrophilic spacer when immobilized on a gold-coated sensor chip. Core 3 and core 5 structures were synthesized by the glycosylation of appropriate N-acetylglucosamine and N-galactosamine donors, respectively, and were converted into sugar-chain ligand conjugates according to the method reported. The interaction analysis of lectins with sugar chips coated with ligand conjugates was performed with a surface plasmon resonance (SPR) biosensor. The specific interaction was observed between the core 3 structure and Jacalin (JAC) and kinetic parameters were estimated as ka = 1.5 × 104, kd = 5.8 × 10-3, and KD = 3.8 × 10-7.

ABSTRACT

OBJECTIVE: To investigate the incidence of skeletal-related events (SRE) in urological cancer patients with bone metastases in Japan. METHODS: Five hundred eleven patients with urological cancer and documented bone metastases treated from January 2003 to April 2008 in ten Japanese institutions were included in a retrospective analysis. Type and incidence of SRE (fracture, radiotherapy, spinal cord compression, surgery, hypercalcemia, and bone pain) were determined from patient medical records. RESULTS: The overall incidence of SRE, including 'pain', was 61%. The most common event was radiotherapy for bone metastases, with an incidence of 31%. The overall incidence of events seemed to be similar among Japanese and Western patients with prostate cancer and renal cell carcinoma when comparing data with previously published reports. Nevertheless, a much lower incidence of fracture (19.1%) was observed in Japanese renal cell carcinoma patients. CONCLUSIONS: The overall incidence of SRE in Japanese urological cancer patients with bone metastasis was similar to that in Western patients, but the incidence of fracture was lower in Japanese renal cancer patients.

ABSTRACT

We report a case of primary malignant lymphoma of the prostate. A 76-year-old man was refer to our clinic with a chief complaint of dysuria. Based on a benign prostatic hypertrophy, transurethral resection of the prostate was performed. Histological findings showed diffuse, large cell type malignant lymphoma. His clinical stage was 1 A prostate (+) according to the Ann Abor classification. The combination chemotherapy with THP-COP was performed for 3 courses, followed by irradiation at 30 Gy. His prostate has showed no recurrence for 10 month after treatment. Primary malignant lymphoma of the prostate is rare. About 30 cases have been reported in the japanese literature. Preoperative diagnosis of malignant lymphoma of the prostate is difficult. All cases have been diagnosed after prostatic surgery or biopsy.
