ABSTRACT

Sensing and clearance of dysfunctional lysosomes is critical for cellular homeostasis. Here we show that transcription factor EB (TFEB)-a master transcriptional regulator of lysosomal biogenesis and autophagy-is activated during the lysosomal damage response, and its activation is dependent on the function of the ATG conjugation system, which mediates LC3 lipidation. In addition, lysosomal damage triggers LC3 recruitment on lysosomes, where lipidated LC3 interacts with the lysosomal calcium channel TRPML1, facilitating calcium efflux essential for TFEB activation. Furthermore, we demonstrate the presence and importance of this TFEB activation mechanism in kidneys in a mouse model of oxalate nephropathy accompanying lysosomal damage. A proximal tubule-specific TFEB-knockout mouse exhibited progression of kidney injury induced by oxalate crystals. Together, our results reveal unexpected mechanisms of TFEB activation by LC3 lipidation and their physiological relevance during the lysosomal damage response.

ABSTRACT

A highly spectrophotometric method for the determination of aluminum was developed. This method used the color reaction between m-carboxyphenylfluorone (MCPF) as a novel chemical probe and aluminum in the presence of a surfactant, poly(N-vinylpyrrolidone) (PVP, K-90) (0.03 - 1.40 microg of aluminum in a final volume of 10 ml at 561 nm). The proposed method showed excellent sensitivity (molar absorptibity of 1.70 x 10(5) l mol(-1) cm(-1)) and reproducibility (within-day precision: RSD = 0.35% n = 6, between-day precision: RSD = 0.44% n = 6). Linearity was achieved over the range 3 - 140 microg L(-1) with a correlation coefficient of 0.9999, and the effects of foreign substances were low.

ABSTRACT

Secondary amyloidosis is well recognized as a severe complication in the late stages of rheumatoid arthritis (RA). However, there have been few reported cases of secondary amyloidosis developing early during the course of RA. We here report the case of a 35-year-old woman, in whom RA who had been diagnosed 1 year before, with intractable watery diarrhea as a symptom of RA-induced secondary intestinal amyloidosis. Combination treatment with intravenous hyperalimentation, corticosteroids, and methotrexate (MTX) resulted in a dramatic improvement of her symptoms and objective findings of serological abnormalities. Subsequent administration of corticosteroids and MTX resulted in long-term survival without recurrence. This case indicates that we should be alert for the development of secondary amyloidosis, even in patients with a short history of RA, when the disease is active. Furthermore, combination therapy with intravenous hyperalimentation and strong immunosuppressive agents seems to be very efficacious in the treatment of RA-associated secondary intestinal amyloidosis.
