ABSTRACT

This study aimed to identify the toxicity effect of Diuron on Tegillarca granosa by determining accumulation in the soft tissues and alteration on the gill tissue structure. Wild stocks T. granosa were grouped to identify the initial Diuron concentration and 48-h exposure in different concentrations (0, 1, 2, 3, and 4 mg·L-1). The Diuron in tissue was extracted using an established procedure and identified using High-Performance Liquid Chromatography (HPLC). Gills were analysed for histopathological study; tissue structure was classified and scored using a developed scoring system. Diuron concentration in the soft tissue of exposed T. granosa was in the range 1.27-8.09 mg·kg-1, w/w and not proportionately increase along with the exposure concentration. The mean index values of the gill histopathology ranged from 5.25-7.67 and classified as moderate to severe. Gills characteristics and T. granosa protective behaviour potentially limit Diuron accumulation in the soft tissue.

ABSTRACT

Compound 3,4-dichloroaniline (3,4-DCA) is a metabolite of several urea herbicides and intermediate chemical of several industrial products. Moreover, 3,4-DCA has been frequently detected in aquatic ecosystems around the world. This aniline is more toxic than the parent chemicals, and it affects non-target organisms. This study evaluated a 21-day reproductive response of an emerging aquatic vertebrate model, Javanese medaka (Oryzias javanicus), exposed to 3,4-DCA. Fecundity and gonads histopathology were observed. The spawning rate and fertilisation reduced significantly in the highest exposed-group (250 µg/L). Gonadosomatic index (GSI) was significantly low in females exposed to 250 µg/L. No substantial structural alteration of male gonads. However, oocyte development and ovarian cell structure were disrupted in 250 µg/L exposed females. The gonadal developmental was not affected in the males; however, a significant reduction in the developmental of female gonads was observed at 250 µg/L. These results show that 3,4-DCA interfere with the reproduction of Javanese medaka through fecundity and alteration of gonadal tissues.

ABSTRACT

Examination of the impact of Diuron contamination on blood cockles (Tegillarca granosa) was conducted by combining field screening at three sampling events and a toxicity test. Diuron was extracted using the liquid-liquid extraction (LLE) technique and analyzed using HPLC-UV. The median lethal concentration (LC50) of Diuron on T. granosa was tested under a 72-h exposure. Diuron in water samples ranged from not detected (ND) to 3910 ppb, which was the highest concentration detected in samples after the irrigation water was discharged from the paddy plantation. Diuron was not detected in sediment samples. Mortality of T. granosa ranged from 4.74 to 38.33% with the highest percentages recorded after the release of the irrigation water. The LC50 value of Diuron was 1.84 ppm. This study suggests that irrigation water from paddy plantation that drifts to coastal areas containing Diuron harms T. granosa at the study area.

ABSTRACT

Early-life exposure to toxic chemicals causes irreversible morphological and physiological abnormalities that may last for a lifetime. The present study aimed to determine the toxicity effect of 3,4-Dichloroaniline (3,4-DCA) on Javanese medaka (Oryzias javanicus) embryos. Healthy embryos were exposed to various 3,4-DCA concentrations for acute toxicity (5, 10, 25, 50, and 100 mg.L-1) and sublethal toxicity (0.10, 0.50, 1.25, 2.50, and 5.00 mg.L-1) for 96 h and 20 days respectively. Acute toxicity test revealed that the median lethal concentration (96h-LC50) was 32.87 mg.L-1 (95 % CI = 27.90-38.74, R2 = 0.95). Sublethal exposure revealed that 1.25 mg.L-1 at 3 days post-exposure (3 dpe) has a significant lower heartrate (120 ± 12.3 beats/min., p < 0.01), while at 7 dpe those exposed to 5 mg.L-1 (141.8 ± 8.3 beats/min) had significantly (p < 0.01) lower heart rate compared to other treatments. Likewise, at 13 dpe, 5.00 mg.L-1 (110.4 ± 17.3 beats/min) and 2.5 mg.L-1 (130.4 ± 8.3 beats/min) were significantly lower (p < 0.001) compared to control. None of the embryos in 5.00 mg.L-1 and 2.50 mg.L-1 treatment groups survived at the end of the experiment. The results indicated a concentration-dependent response. The lowest observed effect concentration (LOEC) that exerted developmental deformities was 0.5 mg.L-1. Javanese medaka embryo have low sensitivity to acute toxicity of 3,4-DCA, but developmental abnormalities at sublethal concentrations were observed.

ABSTRACT

This study was conducted to investigate the median lethal concentration (LC50) of copper pyrithione (CuPT) at 96-hr exposure on adult Javanese medaka (Oryzias javanicus) in revealing toxicological effects of CuPT contamination in the tropical area. Wild stock fishes were acclimatized for 14-days prior analysis. Triplicate of test tanks for seven test concentrations were placed with ten fishes each, this includes two control tanks. The behaviour of the tested fishes was manually observed through a camera. The LC50 of CuPT at 96-h was found to be 16.58mg/L. Tested fishes swam slowly in vertical movement and swam fast towards food during feeding time as the sign of stress behaviour. Meanwhile, fishes in the two control groups swam actively in a horizontal manner and no excitement during feeding time. No mortality in control groups. Results indicate CuPT to be toxic to Javanese medaka at low concentration and caused behavioural stress.

ABSTRACT

Diuron is an alternative biocide suggested to replace organotin in formulating antifouling paints to be applied on water-going vessels hull. However, it is potentially harmful to various non-targeted marine organisms due to its toxic properties. Present study aimed to isolate, screen and identify the potential of Diuron-degrading bacteria collected from the marine sediments of Port Klang, Malaysia. Preliminary screening was conducted by exposing isolated bacteria to 430ng/L (background level), followed by 600ng/L and 1000ng/L of Diuron concentrations. Nine bacteria colonies survived the exposure of the above concentrations. However, only two strains can tolerate to survive up to 1000µg/L, which were then characterised and identified using phenotypic tests and the standard 16S rRNA molecular identification. The strains were identified as Comamonas jiangduensis SZZ 10 and Bacillus aerius SZZ 19 (GenBank accession numbers: KU942479 and KU942480, respectively). Both strains have the potential of Diuron biodegradation for future use.

ABSTRACT

Revealing the potential of seagrass as a bioindicator for metal pollution is important for assessing marine ecosystem health. Trace metal (111Cd, 63Cu, 60Ni, 208Pb, 66Zn) concentrations in the various parts (root, rhizome, and blade) of tape seagrass (Enhalus acoroides) collected from Merambong shoal of Sungai Pulai estuary, Johor Strait, Malaysia were acid-extracted using a microwave digester and analysed via inductively coupled plasma-mass spectrometry (ICP-MS). The ranges of trace metal concentrations (in µgg-1 dry weight) were as follows: Cd (0.05-0.81), Cu (1.62-27.85), Ni (1.89-9.35), Pb (0.69-4.16), and Zn (3.44-35.98). The translocation factor revealed that E. acoroides is a hyperaccumulator plant, as its blades can accumulate high concentrations of Cd, Cu, Ni, and Zn, but not Pb. The plant limits Pb mobility to minimize Pb's toxic impact. Thus, E. acoroides is a potential bioindicator of metal pollution by Cd, Cu, Ni, and Zn in estuarine environments.

ABSTRACT

Passive bioremediation of metal- and sulfate-containing acid mine drainage (AMD) has been investigated in a batch study. Multiple substrates were used in the AMD remediation using spent mushroom compost (SMC), limestone, activated sludge (AS), and woodchips (WC) under anoxic conditions suitable for bacterial sulfate reduction (BSR). Limestones used were of crushed limestone (CLS) and uncrushed limestone, provided at two different ratios in mixed substrates treatment and varied by the proportion of SMC and limestone. The SMC greatly assisted the removals of sulfate and metals and also acted as an essential carbon source for BSR. The mixed substrate composed of 40% CLS, 30% SMC, 20% AS, and 10% WC was found to be effective for metal removal. Mn, Cu, Pb, and Zn were greatly removed (89-100%) in the mixed substrates treatment, while Fe was only removed at 65%. Mn was found to be removed at a greatly higher rate than Fe, suggesting important Mn adsorption onto organic materials, that is, greater sorption affinity to the SMC. Complementary with multiple treatment media was the main mechanism assisting the AMD treatment through microbial metal reduction reactions.

ABSTRACT

Recent bauxite mining activities in the vicinity of Kuantan, Pahang, have been associated with apparent environmental quality degradation and have raised environmental concerns among the public. This study was carried out to evaluate the overall ecological impacts on water and sediment quality from the bauxite mining activities. Water and sediment samples were collected at seven sampling locations within the bauxite mining areas between June and December 2015. The water samples were analyzed for water quality index (WQI) and distribution of major and trace element geochemistry. Sediment samples were evaluated based on geochemical indices, i.e., the enrichment factor (EF) and geoaccumulation index (I geo). Potential ecological risk index was estimated to assess the degree to which sediments of the mine-impacted areas have been contaminated with heavy metals. The results showed that WQIs of some locations were classified as slightly polluted and contained metal contents exceeding the recommended guideline values. The EFs indicated minimal to moderate enrichment of metals (Pb, Cu, Zn, Mn, As, Cd, Cr, Ni, Co, and Sr) in the sediments. I geo showed slightly to partially polluted sediments with respect to As at some locations. The potential ecological risk index (RI) showed that As posed the highest potential ecological risk with RI of 52.35-60.92 at two locations, while other locations indicated low risk. The findings from this study have demonstrated the impact of recent bauxite mining activities, which might be of importance to the local communities and relevant authorities to initiate immediate rehabilitation phase of the impacted area.

ABSTRACT

The environmental impacts with regard to agro-based biofuel production have been associated with the impact of greenhouse gas (GHG) emissions. In this study, field GHG emissions during plantation stage of palm oil-based biofuel production associated with land use changes for oil palm plantation development have been evaluated. Three different sites of different land use changes prior to oil palm plantation were chosen; converted land-use (large and small-scales) and logged-over forest. Field sampling for determination of soil N-mineralisation and soil organic carbon (SOC) was undertaken at the sites according to the age of palm, i.e. <5 years (immature), 5-20 and >21 years (mature oil palms). The field data were incorporated into the estimation of nitrous oxide (N2O) and the resulting CO2-eq emissions as well as for estimation of carbon stock changes. Irrespective of the land conversion scenarios, the nitrous oxide emissions were found in the range of 6.47-7.78 kg N2O-N/ha resulting in 498-590 kg CO2-eq/ha. On the other hand, the conversion of tropical forest into oil palm plantation has resulted in relatively higher GHG emissions (i.e. four times higher and carbon stock reduction by >50%) compared to converted land use (converted rubber plantation) for oil palm development. The conversion from previously rubber plantation into oil palm plantation would increase the carbon savings (20% in increase) thus sustaining the environmental benefits from the palm oil-based biofuel production.

ABSTRACT

A possible tributyltin (TBT)-degrading bacterium isolated from contaminated surface sediment was successfully identified as Klebsiella sp. FIRD 2. It was found to be the best isolate capable of resisting TBT at a concentration of 1000 µg L(-1). This was a concentration above the reported contaminated level at the sampling station, 790 µg L(-1). Further studies revealed that the isolate was Gram negative and resisted TBT concentrations of up to 1500 µg L(-1) in a Minimal Salt Broth without the addition of any carbon source within the first 48 h of incubation. It is expected that additional work could be conducted to check the degradation activity of this new isolate and possibly improve the degradation capacity in order to contribute to finding a safe and sustainable remediation solution of TBT contamination.

ABSTRACT

The morphological expressions and histopathological analysis of the gonads of a tropical marine neogastropod species (Thais sp.) from East Malaysia revealed new evidence of mechanical sterility in the imposex affected females. The gradual development of imposex was classified into five stages (Stage 0 to Stage 4) with three types of sterility conditions; Type A caused prohibition of copulation and capsule formation; Type B prohibits the releasing process of eggs; and gonads in Type C are infertile. Further analysis is needed to confirm, if the gonad malformation in imposex affected snails is generated specifically by tributyltin (TBT) or by other possible factors. The levels of imposex incidence (stages and percentages) were greater in a marina and decreased with increasing distance from the marina. Organotin tissue burden across the sexes showed that dibutyltin (DBT) as well as TBT might be the elements inducing imposex in Thais sp. from Miri in East Malaysia.

ABSTRACT

Present study was conducted to evaluate current status of trace elements contamination in the surface sediments of the Johor Strait. Iron (2.54 +/- 1.24%) was found as the highest occurring element, followed by those of zinc (210.45 +/- 115.4 microg/g), copper (57.84 +/- 45.54 microg/g), chromium (55.50 +/- 31.24 microg/g), lead (52.52 +/- 28.41 microg/g), vanadium (47.76 +/- 25.76 microg/g), arsenic (27.30 +/- 17.11 microg/g), nickel (18.31 +/- 11.77 microg/g), cobalt (5.13 +/- 3.12 microg/g), uranium (4.72 +/- 2.52 microg/g), and cadmium (0.30 +/- 0.30 microg/g), respectively. Bioavailability of cobalt, nickel, copper, zinc, arsenic and cadmium were higher than 50% of total concentration. Vanadium, copper, zinc, arsenic and cadmium were found significantly different between the eastern and western part of the strait (p < 0.05). Combining with other factors, Johor Strait is suitable as a hotspot for trace elements contamination related studies.

ABSTRACT

Concentrations of 11 trace elements (V, Cr, Co, Ni, Cu, Zn, As, Ag, Cd, Pb, and U) were determined in the intertidal surface sediments of Peninsular Malaysia. The average trace element concentrations are ranked as follows: Zn>V>As>Cr>Pb>Cu>Ni>Co>U>g>Cd. Interim Sediment Quality Guidelines (ISQGs) employed in present study are the Australia and New Zealand joint guideline (ANZECC/ARMCANZ), and the Hong Kong authorities. From the pooled data, none of these trace elements have the average concentration above the ISQG-high values. However, As and Ag average concentrations were over the ISQG-low values. Some elements were found to have the average concentration above the ISQG-high and/or ISQG-low in certain locations, including Kampung Pasir Putih (JPP), Lumut Port (ALP), Kuala Perai (PKP), Port Dickson (NPD), and others. The lowest and highest concentrations in a specific sampling location and maritime area varied among the elements, variations that were greatly affected by natural and anthropogenic activities in a given area. For each trace element, there were various levels of concentration among the sampling locations and maritime areas. These patterns indicated pollutant sources of an element for each area perhaps derived from nearby areas and did not widely distributed to other locations. It is necessary for Malaysia to develop an ISQG for effective quick screening and evaluation of the coastal environment of Peninsular Malaysia.
