ABSTRACT

In Malaysia, rice mutant varieties that are genetically altered to confer resistance against blast disease have been substantially developed through mutational breeding program. However, due to the limited accessible information on the mutant lines, mutant gene variants corresponding to the disease resistance and other useful agronomic traits are yet to be exploited. Here, we conducted whole genome re-sequencing of blast resistance with kernel elongation traits in mutant line, Mahsuri Mutant (87,639,446 bp raw reads), and its parental line, Mahsuri (85,156,783 bp raw reads) using Illumina Novaseq 6000 sequencing platform with 30x sequencing coverage. The generated genome sequences are aimed to facilitate the discovery of causal mutation and single nucleotide polymorphisms (SNPs) related to the intended traits. The identified SNPs can be further employed to develop allele-specific SNP molecular markers to locate the mutant gene regions. The NGS data obtained (FASTQ format) of the parental and mutant lines have been deposited in the National Center for Biotechnology Information (NCBI) database under sequence read archive (SRA) xwith accession numbers SRR24388814 (Mahsuri) and SRR22952097 (Mahsuri Mutant) respectively.

ABSTRACT

We evaluated the effects of a Bacillus-based direct-fed microbial (DFM) on total in vitro gas production, dry matter (DM), neutral detergent fiber (NDF), and starch disappearance of different feedstuffs and total mixed rations (TMR) in three different experiments. In experiment 1, six single fiber-based feedstuffs were evaluated: alfalfa hay, buffalo grass, beet pulp, eragrostis hay, oat hay, and smutsvinger grass. Experimental treatments were control (with no probiotic inoculation; CON) or incubation of a probiotic mixture containing Bacillus licheniformis and B. subtilis (3.2 × 109 CFU/g; DFM). The calculation of DFM dose under in vitro conditions was based on the assumption of a rumen capacity of 70 liter and the dose of 3 g of the DFM mixture/head/d (9.6 × 109 CFU). Total in vitro gas production, DM, and NDF disappearance were evaluated at 24- and 48 h posttreatment incubation. Mean treatment effects were observed at 24- and 48 h gas production (P < 0.0001), as DFM incubation increased in vitro gas production by 5.0% and 6.5%, respectively. For nutrient digestibility, mean DM digestibility was increased at 48 h (P = 0.05), whereas mean NDF digestibility increased at both timepoints by incubating DFM in vitro (P ≤ 0.02). In experiment 2, nine commercial dairy TMR were collected and evaluated for the same variables and treatments described in experiment 1, with the additional analysis of starch digestibility at 7 h post in vitro incubation. The only difference was the concentration of the DFM included, being representative for a dosage of 8.8 × 109 CFU/head/d. In vitro gas production was increased only at 48 h due to DFM incubation (P = 0.05), whereas DM and NDF digestibility were improved at 24 and 48 h (P ≤ 0.02). No treatment effects were observed on in vitro starch digestibility (P = 0.31). In experiment 3, a combined analysis of DM and NDF digestibility was performed by using quality values (NDF and crude protein or CP) of 16 substrates. Regardless of CP and NDF levels of the substrates, DFM improved in vitro 24 and 48 h DM and NDF digestibility (P ≤ 0.03). In summary, incubating a Bacillus-based DFM (B. licheniformis and B. subtilis; BOVACILLUS) improved mean in vitro gas production, DM, and NDF digestibility of single feedstuffs and commercial dairy TMR, highlighting the potential of this combination of Bacillus spp. to improve nutrient utilization, mainly fiber.

ABSTRACT

More than 200 different cultivars of durian exist worldwide but Durio zibethinus or Musang King (MK) is the most premium and prized durian fruit among the recommended varieties. Early identification of this premium variety is critical to protect from non-authentic MK durian cultivars. However, the MK variety's morphological traits are nearly identical to other varieties. Currently, the identification of durian varieties is mostly performed via evaluation of leaf shape, fruit shape, aroma, taste and seed shape and this requires trained personnel for the morphology observation. To enable the rapid identification of the MK variety, PCR amplification of ten durian varieties using six gene candidates from the chloroplast genome was first performed to obtain DNA probes that were specific to the MK durian variety. PCR amplification of ten durian varieties using primers designed confirmed that the nadhA gene sequence showed an obvious difference in the MK variety from other durian varieties. The unique sequence of MK was used as a DNA probe to develop an electrochemical biosensor for the direct identification of the MK durian variety. The electrochemical biosensor was based on the hybridization response of the immobilized DNA probe with the target DNA from the MK variety and was monitored via differential pulse voltammetry technique. Under optimal conditions, the DNA electrochemical biosensor showed a low detection limit at 10% of MK genomic DNA concentration with a wide linear calibration range of 0.05-1.5 µM (R2 = 0.9891) and RSD value of 3.77% (n = 3). The results of the developed DNA biosensor provide high promise for the development of portable sensors employed in the determination of MK variety in the field.

ABSTRACT

Halal authentication has become essential in the food industry to ensure food is free from any prohibited ingredients according to Islamic law. Diversification of food origin and adulteration issues have raised concerns among Muslim consumers. Therefore, verification of food constituents and their quality is paramount. From conventional methods based on physical and chemical properties, various diagnostic methods have emerged relying on protein or DNA measurements. Protein-based methods that have been used in halal detection including electrophoresis, chromatographic-based methods, molecular spectroscopy and immunoassays. Polymerase chain reaction (PCR) and loop-mediated isothermal amplification (LAMP) are DNA-based techniques that possess better accuracy and sensitivity. Biosensors are miniatured devices that operate by converting biochemical signals into a measurable quantity. CRISPR-Cas is one of the latest novel emerging nucleic acid detection tools in halal food analysis as well as quantification of stable isotopes method for identification of animal species. Within this context, this review provides an overview of the various techniques in halal detection along with their advantages and limitations. The future trend and growth of detection technologies are also discussed in this review.

ABSTRACT

This study proposes test methods for assessing the printability of concrete materials for Additive Manufacturing. The printability of concrete is divided into three main aspects: flowability, setting time, and buildability. These properties are considered to monitor the critical quality of 3DCP and to ensure a successful print. Flowability is evaluated through a rheometer test, where the evolution of shear yield strength is monitored at a constant rate (rpm), similar to the printer setup. Flowability limits were set based on the user-defined maximum thickness of a printed layer and the onset of gaps/cracks during printing. Setting time is evaluated through an ultrasonic wave pulse velocity test (UPV), where the first inflection point of the evolution of the UPV graph corresponds to the setting time of the concrete specimen. The results from this continuous non-destructive test were found to correlate with the results from the discrete destructive ASTM C-191 test for measuring setting time with a maximum difference of 5% between both sets of values. Lastly, buildability was evaluated through the measurement of the early-age compressive strength of concrete, and a correlation with the UPV results obtained a predictive model that can be used in real-time to non-destructively assess the material buildability. This predictive model had a maximum percentage difference of 13% with the measured values. The outcome of this study is a set of tests to evaluate the properties of 3D printable concrete (3DP) material and provide a basis for a framework to benchmark and design materials for additive manufacturing.

ABSTRACT

Introduction and importance: Echinococcus infection affects the liver and lungs. However, in unusual cases it may take location in the heart. Furthermore, the settling of the cysts in the interventricular septum are especially rare. Case presentation: We report a case of a 17-years old male presented with dyspnea and productive cough. Ultrasound showed an incidental finding of a cyst in the distal part of the interventricular septum and the apex of left ventricle. He was treated with a course of antibiotics with albendazole 800 mg then was referred to the cardiac surgery department. The cyst with all its layers was resected. Clinical discussion: Cardiac hydatid cyst is extremely rare. Especially the involvement of the interventricular septum as it is responsible for only 4% of cardiac cases. Its symptoms maybe nonspecific and the diagnoses is based on echography and computed tomography. The surgical treatment under cardiopulmonary by-bass with complementary course of albendazole 800 mg seems to have a good prognostic outcome. Conclusion: Cardiac echinococcus should be kept in mind in endemic regions. The diagnosis should be made in the early and uncomplicated stages since it may be fatal. Echocardiography is sensitive and useful for the diagnosis.

ABSTRACT

Identification of plant variety and cultivar is pivotal in the agricultural sector due to the abundance of plant varieties and cultivars developed in many crop species. However, plant variety and cultivar identification via basic morphological features is problematic and challenging when differentiating closely related species not only due to their limited differences but also due to technical limitations of the process being time-consuming, labour-intensive and costly, and statistically imprecise information being available due to phenotypic plasticity. Therefore, it is imperative to have rapid and highly efficient techniques to mitigate these limitations. This review provides an overview and summarization of the development and application of molecular markers such as Random Amplified Polymorphic DNA (RAPD), Restriction Fragment Length Polymorphism (RFLP), Simple Sequence Repeats (SSR), Inter-simple sequence repeats (ISSR), Amplified Fragment Length Polymorphism (AFLP), Single nucleotide polymorphism (SNP) and DNA barcoding, High-resolution melting (HRM) and biosensor technology as potential tools in the identification of plant variety and cultivar.

ABSTRACT

The aim of this experimental study is to develop high strength and lightweight concrete mixture suitable for structural applications. This work investigates the effect of replacing normal aggregate either partially or totally with expanded perlite aggregate. This material allows for better thermal insulation properties, thus decreasing the energy usage within the life cycle of the concrete structure. Expanded perlite aggregate was used in concrete by 20%, 40%, 60%, 80%, and 100% in replacement of the natural aggregate. Material characterization tests of compressive strength, flexural strength, and thermal conductivity were carried out for six concrete mixtures. In addition, microstructure analysis was performed with the aid of a micro-computed tomography system to investigate the effects and relation of microstructure quantities on material properties. The proposed concrete mixture, which has 100% of expanded perlite aggregate, has a unit weight of 1703 kg/m3 and achieved reduction percentage of thermal conductivity around 62% (1.81 to 0.69 W·m-1·K-1) and a compressive strength of 42 MPa at 28 days; and thus is ideal for structural applications with enhanced properties.

ABSTRACT

Out of hospital cardiac arrest (OHCA) is a leading cause of death worldwide. Early cardiopulmonary resuscitation (CPR) and early defibrillation are key to improving outcomes of patients with OHCA including return of spontaneous circulation (ROSC) and survival to hospital discharge with good neurologic outcomes. Lebanon like other developing countries, suffers from absence of organized prehospital cardiac arrest care bundle and from absence of a legal framework for community involvement in cardiac arrest care. Scientific societies, involved non-governmental organizations (NGOs) and local governmental stakeholders organized a national meeting to launch a strategy aiming at improving OHCA outcomes in Lebanon. This article represents a position statement of the Lebanese Society of Cardiology and the Lebanese Society of Emergency Medicine summarizing the strategy to improve out-of-hospital CPR. Participating stakeholders developed and submitted a law proposal of a "Good Samaritan Law" to the Lebanese parliament. Several of activities were also launched aiming at establishing public access defibrillation programs and at training bystanders in different areas in Lebanon to perform bystander CPR and use automated external defibrillators (AEDs). Additional recommendations were proposed to local emergency medical system (EMS) agencies to improve prehospital care and introduce medical direction to prehospital activities.
