ABSTRACT

The ability of mononuclear blood cells (PBMC), derived from patients with cancer of the larynx, to produce IFN-gamma in vitro was assessed in this paper. Thirty patients (27 male, 3 female) were qualified to the study. Their mean age was 65 (range: 41 to 78 years), tumour sizes found in the group were from T2 to T4, levels of pathologic malignancy G2 or G3. The percentage rates different blood cells phenotypes (CD3+, CD3+ HLADR+, CD4+, CD8+, CD14+ HLADR+, CD19+, CD56+) were evaluated by means of flow cytometry (Coulter EPICS XL). The control group consisted of 20 healthy blood donors. PBMC were derived by centrifugation of heparinized venous blood on Lymphoprep gradient according to Boyum method. Double cell cultures were performed for 24 hours with antibody anti-CD3 or recombinant interleukins 12 (rhIL-12) or 18 (rhIL-18). The statistic analysis was based on Student t and Mann-Whitney's test with significancy at p<0.05. A significant decrease in the production of IFN-gamma by PBMC in patients with laryngeal cancer after stimulation with antiCD3 (p=0.018), rhIL-12 (p=0.027), rhIL-18 (p=0.016) was found in comparison with the controls. The results suggest a decreased production of IFN-gamma in patients with cancer of the larynx.

ABSTRACT

Voice quality was analysed in 39 patients with the larynx carcinoma after the supraglottis subtotal laryngectomy. Voice pattern was analysed with the use of subjective and objective spectrography before and after the surgery. A deteriorated voice quality was found after the surgery. The spectrographic examination revealed decreased frequency levels of the formants F3 and F4 and the presence of a noise component generated in the glottis area.

ABSTRACT

In our review article we intend to show the importance of IL-4, IL-5, IL-13, IL-10, IFN gamma and IL-12 in pathomechanism of allergic inflammatory process, especially in bronchial asthma. We also want to illustrate the relationships between lymphocytes T, monocytes, macrophages, eosinophils and other inflammatory cells which take part in allergic inflammation.

ABSTRACT

The aim of our investigation was to assess T cell activation markers (CD69, HLA-DR, CD25, CD49a) in idiopathic uveitis. T cells from children aged 9-17 with idiopathic uveitis were studied. Monoclonal antibodies against chosen markers of T cell activation were used. Expression of the CD69, HLA-DR, CD25, CD49a were determined by flow-cytometry. HLA-DR and CD49a--marker of the late activation--were significantly increased.

ABSTRACT

PURPOSE: To evaluate the presence of IL-10 and IL-13 in subretinal fluid of patients with rhegmatogenous retinal detachment. MATERIAL AND METHOD: The studies comprised 14 patients on with retinal detachment operated. The presence of cytokines was evaluated using immunoenzymatic assay. RESULTS: IL-10 and IL-13 were found in all subretinal fluid samples. Our results suggest that the presence of IL-10 and IL-13 has the influence on suppression of inflammatory agent release and activity during retinal detachment.

ABSTRACT

Morphological evaluation was performed with the light and scanning electron microscope of subretinal fluids (SRF) collected during operations from 36 patients with rhegmatogenous retinal detachment. Duration of the disease was 3 weeks, 5-8 weeks, 10 weeks and 6 months. The immunoenzymatic method was used to establish the presence of interleukins IL-1beta, IL-2, IL-4 and IL-6 in SRF. It was revealed that the differentiation of the morphological picture of SRF depended on the duration of detachment. Most cell-rich SRF came from the 3-week detachment. Mononuclear cells were predominant in that period (macrophages, lymphocytes). In the later period (over 8 weeks) a decrease in the number of cells and a change in cellular composition were noted in the fluids examined, while after six months of detachment normal cellular elements were absent. In all time intervals analyzed, no significant increase in IL-1beta, IL-2 and IL-4 was noted in SRF. However, a significant quantity of IL-6 was found, particularly in the 5- to 8-week detachment. The authors assume that the presence of a large number of inflow cells in 'fresh' detachment can trigger a variety of inflammation-based immunological processes which stimulate the development of vitreoretinal proliferation.

ABSTRACT

PURPOSE: The aim of this study was to evaluate the presence of IL-8, TNF-alpha, IFN-gamma in subretinal fluid of patients with rhegmatogenous retinal detachment. MATERIAL AND METHODS: We studied 15 patients operated on retinal detachment. The presence of cytokines was evaluated using immunoenzymatic assay. RESULTS: IL-8, TNF-alpha and IFN-gamma were found in all subretinal fluid samples. The concentration of cytokines in patients with retinal detachment lasting to 2 months was higher than in cases of retinal detachment over 2 months.

ABSTRACT

PURPOSE: The aim of this study was to evaluate the ability of SRF to activate autologous leucocytes derived from the peripheral blood. MATERIAL AND METHOD: We studied 49 patients with idiopathic retinal detachment. Leucocyte migration inhibition, proliferation of T-lymphocytes after PHA stimulation, nonspecific suppressor activity induced by Con A, T-lymphocytes IL-2 production and basophils histamine release were assessed. RESULTS: Lymphocytes obtained from patients with retinal detachment lasting 3-4 weeks exhibited in vitro in the presence of SRF: increased ability to produce IL-2, increased proliferation after PHA stimulation and increased nonspecific T-suppressor activity induced by Con A. SRF enhanced also leucocyte migration inhibition and release of histamine from basophils.

ABSTRACT

AIM: The purpose of the investigation was to evaluate the presence of IL-1 beta, IL-2, IL-6, IL-8, TNF-alpha, IFN-gamma in aqueous humor of patients with cataract. MATERIALS AND METHOD: Aqueous humor was taken from 29 patients during extracapsular cataract extraction. The presence of cytokines was evaluated using immunoenzymatic methods. RESULTS: IL-2, IL-6, IL-8, TNF-alpha and IFN-gamma were found in all aqueous humor samples. There was no evidence for the presence of IL-1 beta. CONCLUSION: Cytokines may play role in the inflammation process after extracapsular cataract extraction.

ABSTRACT

In 16 patients with nonallergic bronchial asthma treated with Broncho-vaxom and 10 healthy persons the mononuclear peripheral blood cells ability for IL-1 beta, IL-2, IL-4 and IL-6 production were studied. Nonallergic asthmatics were characterised by increased levels of IL-1 beta and IL-6 produced by monocytes. After Broncho-vaxom therapy a decreased for IL-1 beta and IL-6, and an increased production of IL-2 were observed. These findings indicate that orally administered Broncho-vaxom affects on biological activity of mononuclear peripheral blood cells.

ABSTRACT

In a group of 14 patients with nonatopic bronchial asthma, with recurrent infections of airways (approximately 5 - 10 episodes/year) and in 9 healthy persons the effect of Ribomunyl on IL-1 beta, IL-2, IL-4 and IL-6 production by mononuclear peripheral blood cells were assessed. After Ribomunyl treatment in asthmatics the significant decrease in IL-1 beta and IL-6 production by monocytes stimulated with lipopolysaccharides (LPS) and significant increase in IL-2 production by lymphocytes were observed.

ABSTRACT

Total lymphocytes number, proportions of T lymphocyte subset, nonspecific suppressor activity induced by Con A and T-lymphocytes ability for IL-2 production were assessed. Group of 46 patients with intrinsic asthma (24 females and 22 males) including 20 asthmatics treated during 5-7 years with glucocorticosteroids were studied. Control group consisted of 12 healthy persons. Statistically significant reduction of total T cells, T-suppressor and T-helper subpopulations, ability for IL-2 production and nonspecific suppressor activity was found.

ABSTRACT

The aim of the investigations was to search the subretinal fluid for the inflammatory interleukins which influence T and B lymphocytes. The levels of IL-1, IL-2, IL-4 and IL-6 were determined. Only IL-6 was found in a significantly high level, especially in cases with retinal detachment lasting 5-8 weeks. The level of other interleukins examined was not significant.

ABSTRACT

Broncho-vaxom was administered to 40 patients with recurrent respiratory infections, allergic-infectious bronchial asthma and chronic bronchitis. Broncho-vaxom was administered in the dose of one capsule for the first month, and 1 capsule for 10 days in the third, fourth, and fifth month. A favourable clinical effect has been achieved in over 50% of the treated patients manifested by the decrease in the number and severity of respiratory infections and reductions in antibiotics use. These results confirm clinical efficiency of this vaccine.

ABSTRACT

Cell-mediated immunity has been measured in vivo in 30 patients with allergic-infectious bronchial asthma with the use of Multitest CMI (Merieux). A decrease in the immunological reactivity has been showed in the asthmatic patients. It was expressed by means of the arithmetic mean value of the positive reactions, and was particularly significant in patients who require long-term corticotherapy. The obtained results suggest that Multitest CMI is a new, valuable device in the diagnosis and monitoring the treatment in asthmatic patients.

ABSTRACT

The T and B lymphocyte subpopulations were evaluated by means of the rosette test and lymphocyte blastogenic index to PHA was determined in 21 asthmatic patients sensitive to house dust mites during specific immunotherapy with Allpyral. After 2-year treatment a marked increase in the capability of T lymphocytes to form active and spontaneous E rosette was noted. The lymphocyte response to PHA was also significantly higher than before hyposensitization. These findings indicate the role of T lymphocytes and the participation of the cell-mediated response in the mechanisms of specific immunotherapy.
