ABSTRACT

Trimethylsilyl chloride is an efficient activating agent for azines in isocyanide-based reactions, which then proceed through a key insertion of the isocyanide into a N-Si bond. The reaction is initiated by N activation of the azine, followed by nucleophilic attack of an isocyanide in a Reissert-type process. Finally, a second equivalent of the same or a different isocyanide inserts into the N-Si bond leading to the final adduct. The use of distinct nucleophiles leads to a variety of α-substituted dihydroazines after a selective cascade process. Based on computational studies, a mechanistic hypothesis for the course of these reactions was proposed. The resulting products exhibit significant activity against Trypanosoma brucei and T. cruzi, featuring favorable drug-like properties and safety profiles.

ABSTRACT

Las hojas de Galphinia glauca Cav., un arbusto nativo de Centroamérica, proveen efectos duales antialérgicos y sedantes/ansiolíticos por vía oral. El polifenol ácido tetragaloilquínico es su principio activo antiasmático y antialérgico. Su mecanismo de acción está ligado al antagonismo específico sobre los efectos del leucotriene D4 y actúa a dosis de 5mg/kg en modelos in vivo. La actividad ansiolítica reside en su contenido en nor-secitriterpenos llamados galfimias. Las galfimias A y B son los principios activos con mayor actividad ansiolítica en modelos conductuales in vivo. Estos metabolitos pasan la barrera hematoencefálica y actúan en modelos in vivo a dosis de 15 mg/kg. Estudios clínicos han demostrado una eficacia equiparable al lorazepam en el tratamiento de los trastornos de ansiedad generalizada según dos ensayos clínicos a corto y largo plazo. Los tratamientos fueron bien tolerados y no indujo ningún efecto adverso serio. Dichos antecedentes podrían justificar el desarrollo de fitofármacos a partir de fracciones polares a partir de las hojas de G. glauca, estandarizadas en ácido tetragaloilquínico, como agente preventivo de alergias y asma. Por otro lado las fracciones apolares, estandarizadas según el contenido en galfimias A y B, podrían ser de utilidad en el tratamiento del trastorno de ansiedad generalizada (AU)


As folhas de Galphimia glauca Cav., um arbusto originario da América Central, apresentam uma dupla actividade, com efeitos antialérgicos e sedativos/ansiolíticos quando administradas por via oral. O polifenol ácido tetragaloilquinico é o principio como acçao está relacionado com o antagonismo específico sobre os efeitos do Leucotrieno D4 (LTD4), agindo em doses de 5 mg/kg em modelos murinos in vivo. A atividade ansiolítica debe-se á presença de nor-secotriterpenos chamados galfiminas. As galfiminas A e B sao as mais ativas em modelos comportamentais in vivo. Estes compostos atravessam a barreira hemato.encefálica e sao ativos em modelos murinos in vivo em doses de 15 mg/kg. Estudos clínicos demonstraram uma eficacia semelhante ao lorazepem no tratamento da perturbaçao da ansiedade generalizada, tanto a curto como a longo prazo. Os tratamentos foram bem tolerados e nao provocaram efeitos secundários graves. Estas características podem justificar, por um lado, o desenvolvimiento de medicamentos à base de plantas com base em fraçoes polares extraídas a partir das folhas de G. glauca, padronizadas em ácido tetragaloilquinico, para prevençao as fraçoes apolares, padronizadas relativamente ao conteúdo de galfiminas A e B, poderiam ser de utilidade no tratamento da perturbaçao da ansiedade generalizada (AU)


Leaves of Galphimia glauca Cav., a Centroamerican medicinal shrub, are endowed with dual antialergic and anxiolytic effects when administered orally. The polyphenol tetragalloylquinic acid is the antiasthmatic and antiallergic active principle. Its mechanism of action is related to the specific antagonism upon the LTD4 receptors, acting ar doses of 5mg/mL in murine in vivo models. The anxyolitic activity is due to the presence of the norsecotriterpenes ‘galphimines’. Galphimines A and B are the most active in conductual models in vivo. This metabolites are able to cross the blood-brain barrier land are active in murine in vivo models at doses of 15 mg/kg. Clinical studies demonstrated a similar efficacy to lorazepam in the treatment of generalized anxiety disorder both at short and long term. The treatment is well tolerated and no safety concerns were raised. These antecedents may justify on the one hand the development of phytopharmaceutical preparations based on polar fractions from the leaves of G. glauca, standardized in tetragalloylquinic acid, as a preventative agent of allergies and asthma. On the other hand, apolar fractions, enriched and standardized in Galphimines A and B, could be made into medicines for the treatment of generalized anxiety disorder (AU)
