ABSTRACT

Nipah virus (NiV), an emerging zoonotic pathogen in Southeast Asia, is transmitted from Pteropus species of fruit bats to a wide range of species, including humans, pigs, horses, dogs, and cats. NiV has killed millions of animals and caused highly fatal human outbreaks since no vaccine is commercially available. This study characterized the immunogenicity and safety of poxvirus-based Nipah vaccines that can be used in humans and species responsible for NiV transmission. Mice were vaccinated with modified vaccinia Ankara (MVA) and raccoon pox (RCN) viral vectors expressing the NiV fusion (F) and glycoprotein (G) proteins subcutaneously (SC) and intranasally (IN). Importantly, both vaccines did not induce significant weight loss or clinical signs of disease while generating high circulating neutralizing antibodies and lung-specific IgG and IgA responses. The MVA vaccine saw high phenotypic expression of effector and tissue resident memory CD8ɑ+ T cells in lungs and splenocytes along with the expression of central memory CD8ɑ+ T cells in lungs. The RCN vaccine generated effector memory (SC) and tissue resident (IN) CD8ɑ+ T cells in splenocytes and tissue resident (IN) CD8ɑ+ T cells in lung cells. These findings support MVA-FG and RCN-FG viral vectors as promising vaccine candidates to protect humans, domestic animals, and wildlife from fatal disease outcomes and to reduce the global threat of NiV.

Introducción: Las geohelmintiasis son las parasitosis intestinales causadas por helmintos que hacen su ciclo de vida en la tierra y representan un problema de salud público a nivel mundial, que afecta en especial aquellos estratos socioeconómicos más bajos. Objetivo: Determinar la prevalencia de geohelmintos en comunidades indígenas del estado Zulia, Venezuela. Métodos: Se analizaron 250 muestras fecales, de individuos de ambos sexos con edades entre 1 a 80 años. Las muestras fueron procesadas con examen directo, concentrado (Ritchie) y recuento de huevos (Kato-Katz). Resultados: El 35,20 por ciento de los individuos presentaron huevos de geohelmintos en sus heces (88 casos). El grupo etario más afectado fue el de escolares (7-12 años: 38,64 por ciento ), seguido de adultos (20 o más años: 26,13 por ciento ). Ascaris lumbricoides con 25,20 por ciento se encontró en primer lugar, seguido de Trichuris trichiura con 14,80 por ciento , y los menos frecuentes Ancilostomideos con 4,40 por ciento . La mayoría de los individuos presentaron infecciones de intensidad leve (A. lumbricoides 74,60 por ciento , T. trichiura 81,08 por ciento y Ancilostomideos 90,91 por ciento ). Conclusiones: Se detecta una moderada prevalencia de geohelmintos, con un mayor porcentaje de las infecciones de intensidad leve. La presencia de los geohelmintos se relaciona con el escaso saneamiento ambiental que poseen estas comunidades indígenas(AU)


Introduction: Geohelminthiases are intestinal parasitic diseases caused by helminths which complete their life cycle in the soil. They are a global public health problem mainly affecting the lowest socioeconomic strata. Objective: Determine the prevalence of geohelminths in native communities from Zulia State, Venezuela. Methods: Analysis was performed of 250 stool samples from individuals of both sexes aged between 1 and 80 years. The samples were processed by direct examination, concentration (Ritchie) and egg count (Kato-Katz). Results: Geohelminth eggs were found in 35.20 percent of the stool samples studied (88 cases). The most affected age group was schoolchildren (7-12 years: 38.64 percent), followed by adults (20 years and over: 26.13 percent). The most common helminth was Ascaris lumbricoides with 25.20 percent, followed by Trichuris trichiura with 14.80 percent. The least common genus was Ancylostoma with 4.40 percent. Infection was mild in most subjects (A. lumbricoides 74.60 percent, T. trichiura 81.08 percent and Ancylostoma 90.91 percent). Conclusions: A moderate prevalence of geohelminth infection was observed, most of which was mild. The presence of geohelminths is related to poor environmental sanitation in these native communities(AU)

ABSTRACT

Con el objetivo de identificar los factores de riesgo para la infección por leishmaniasis cutánea en individuos del cantón Montecristi, se realizó un estudio durante enero y diciembre del 2019. La muestra estuvo conformada por 297 individuos, con edades comprendidas entre 15 y 50 años, ambos sexos. Se realizó una encuesta para conocer los factores de riesgo y nivel de conocimiento de la enfermedad. Los factores de riesgo identificados donde se mostró diferencias entre los grupos con y sin leishmaniasis fueron: la edad, el sexo, el nivel educativo, ocupación y conocimiento de la enfermedad. Se concluye que los factores de riesgo más relacionados con la enfermedad en la población de Montecristi resultaron ser sexo masculino sobre el femenino y las ocupaciones agricultores-obreros-oficios del hogar, por lo que se deben mantener promoción de salud con la finalidad de dar a conocer las medidas preventivas y así reducir la enfermedad


In order to identify risk factors for cutaneous leishmaniasis infection in individuals from the Montecristi canton, a study was carried out during January and December 2019. The sample consisted of 297 individuals, aged between 15 and 50 years, both sexes. A survey was conducted to find out the risk factors and level of knowledge of the disease. The risk factors identified where differences were found between the groups with and without leishmaniasis were: age, sex, educational level, occupation and knowledge of the disease. It is concluded that the risk factors most related to the disease in the population of Montecristi turned out to be male over female and farmers-workers-housework occupations, so health promotion should be maintained in order to give know the preventive measures and thus reduce the disease

Ante la necesidad de garantizar alimentos inocuos para los consumidores, las agroindustrias han implementado prácticas preventivas y desarrollado investigación que respalde sus procesos. El gremio productor de banano en Guatemala precisó determinar la presencia de Escherichia coli en la cáscara y parte comestible de la fruta, y el crecimiento de esta bacteria durante la postcosecha y almacenamiento. El estudio se realizó en cuatro plantas empacadoras de banano en Guatemala con dos objetivos: el primero determinar si la bacteria E. coli logra infiltrarse hacia la parte comestible del banano durante el lavado postcosecha, y el segundo, determinar si existe presencia de E. coli en la cáscara del banano en estiba y en simulación de almacenamiento a temperatura controlada. Para el primer objetivo se recolectaron al azar dos cajas de banano de una empacadora, que fueron trasladadas al laboratorio donde se replicaron las condiciones de la pileta de lavado y se introdujo E. coli de manera intencional; posteriormente, se utilizó el método de número más probable, para analizar el macerado de la fruta. Para el segundo objetivo, se recolectaron al azar cinco cajas de banano provenientes de tres empacadoras, se realizaron muestreos en la cáscara de dos bananos por caja, tanto en estiba, como durante el almacenamiento a temperaturas entre 17 y 18 °C, los días 4 y 18. Los resultados indicaron ausencia de E. coli en la parte comestible de la fruta y en la cáscara del banano durante el proceso de estiba y almacenamiento en los tiempos evaluados


To ensure innocuous food for consumers, agroindustries have implemented preventive practices and have developed research that supports their processes. e banana producing guild in Guatemala required to determine the presence of Escherichia coli in the peel and edible part of the fruit, and to determine the growth of this bacteria during postharvest and storage processes. e study was carried out in four banana packing plants in Guatemala with two objectives; the first one was to determine if E. coli is able to infiltrate the edible part of the banana during postharvest washing. e second objective was to determine if there is E. coli in the banana peel at stowage and in simulation of controlled temperature storage. For the first objective, two boxes of banana were randomly collected from a packing line, then were transferred to the laboratory where the conditions of the washing tank were replicated and E. coli was introduced intentionally. e most probable number method was used to analyze the maceration of the fruit. For the second objective, five boxes of bananas were randomly collected from three packing plants. Samplings were made to the peel of two bananas per box, at stowage, and then during storage at temperatures between 17 and 18° C, during the 4th and 18th day. e results indicated absence of E. coli in the edible part of the fruit and in the banana peel at stowage area, and at controlled storage temperature during the evaluation time

Las parasitosis intestinales representan uno de los problemas más importantes de salud pública que afecta principalmente a países en vías de desarrollo. El objetivo de este trabajo fue determinar la prevalencia de parasitosis intestinales en escolares del cantón Paján, Ecuador. Se analizaron 351 muestras de heces con solución salina fisiológica y lugol de niños en edades comprendidas entre 5 y 9 años de uno u otro sexo. Se determinó una prevalencia general de parasitados de 45,30% (159/351) prevaleciendo los monoparasitados sobre los poliparasitados (91,82% / 8,18%). Las principales especies encontradas fueron los protozoarios Complejo Entamoeba 26,50% (93 casos) E. coli (6,55%) y G. lamblia (6,27%) y de los helmintos A. lumbricoides (1,14%) y E. vermicularis (0,57%). Se concluye que los niños en edad escolar del cantón Paján tienen una mediana prevalencia de parasitados por lo que existen condiciones permisivas para la trasmisión de enteroparásitos; lo que es necesario hacer seguimiento a las medidas preventivas y de tratamiento de las parasitosis.


Intestinal parasites are one of the most important health problems that are mainly used in developing countries. The objective of this work was to determine the prevalence of intestinal parasitosis in the schools of the Paján canton, Ecuador. We analyzed 351 stool samples with physiological saline solution and lugol from children between the ages of 5 and 9 years of either sex. A general prevalence of parasites of 45.30% (159/351) was determined, prevailing monoparasites over polyparasites (91.82% / 8.18%). The main characteristics of the species were the protozoa Entamoeba Complex 26.50% (93 cases) E. coli (6.55%) and G. lamblia (6.27%) and the helminths A. lumbricoides (1.14%) and E. vermicularis (0.57%). It is concluded that children in the school age of Paján canton have a prevalence of parasites for permissive conditions for the transmission of enteroparasites; that is necessary to follow up on preventive measures and treatment of parasitosis.

Con la finalidad de determinar la prevalencia de enteroparásitos en niños de una comunidad indígena del municipio Machiques de Perijá del estado Zulia, fueron analizadas 139 muestras fecales provenientes de niños en edades comprendidas entre 0 a 12 años de edad. A cada espécimen fecal se le realizó un examen coproparasitológico al fresco con solución salina fisiológica al 0,85% y coloraciones temporales NAIR y lugol, así como la técnica de concentración de Ritchie. Se observó una prevalencia general de niños parasitados de 87,7 % No hubo diferencia significativa en cuanto al estudio de las variables parasitosis y sexo. Se observó un predomino del poliparasitismo (68, 35%) sobre el monoparasitismo (20,15%). En cuanto al grupo de edades, el estrato de 2 a 6 años resultó el mayormente parasitado (43,92 %). El χ2 reveló que si existe relación significativa entre las variables parasitismo y edad. Las especies parasitarias que obtuvieron una mayor prevalencia fueron: Entamoeba coli (46,06%), Blastocystis sp. (43,16%), Trichuris trichiura (30,94%) y Ascaris lumbricoides (21,58%), en todo el grupo estudiado. Estos resultados son similares a los obtenidos en investigaciones previas. La alta prevalencia de protozoarios sugieren un patrón de transmisión directa entre la población estudiada o a través del consumo de agua y/o alimentos contaminados fecalmente.


In order to determine the prevalence of intestinal parasites in children from an indigenous community in the municipality Machiques de Perija of Zulia state, they were analyzed fecal samples from 139 children aged between 0-12 years old. Each stool specimen was conducted a coproparasitological fresh examination with physiological saline 0.85% and temporary colorations NAIR and lugol, as well, Ritchie´s concentration technique. An overall prevalence of children parasitized 87.7% was observed, no significant differences in the studies of the variables parasitosis and sex. It was observed a predomine of polyparasitism (68,35%) on monoparasitism (20,15%). As for the age group, stratum 2 to 6 years was the mostly parasitized (43.92%). The X² revealed that if there is significant relationship between variables and age parasitism. Parasitic species obtained a higher prevalence were: Entamoeba coli (46.06%), Blastocystis sp. (43.16%), Trichuris trichiura (30.94%) and Ascaris lumbricoides (21.58%) throughout the study group. These results are similar to those obtained in previous research. The high prevalence of protozoan suggest a pattern of direct transmission between the study population or through consumption of water and/or fecally contaminated food.

ABSTRACT

The purpose of the present study is to evaluate an individualized intervention based on Coach Effectiveness Training (CET) principles, using a case study. Two basketball coaches selected 3 target behaviors to improve. Behavioral assessment revealed that Coach 1 achieved positive changes in all his 3 target behaviors. In turn, Coach 2 improved on 2 of his 3 target behaviors. Changes in coaches’ behaviors were mostly perceived by players in the evaluation stage. Specifically, players’ perceptions of Coach 1 showed an increase of General Encouragement and Mistake-Contingent Encouragement, and players of Coach 2 perceived a clear increase of General Encouragement, Reinforcement and Mistake-Contingent Technical Instruction. Results are discussed in line with CET principles and potential applications of our program are presented


El objetivo del presente estudio es evaluar una intervención individualizada basada en los principios del Coach Effectiveness Training (CET) mediante un enfoque de estudio de caso. Dos entrenadores de baloncesto seleccionaron 3 conductas objetivo a mejorar. La evaluación conductual reveló que el Entrenador 1 consiguió cambios positivos en sus 3 conductas objetivo. Por su parte, el Entrenador 2 mejoró en 2 de sus 3 conductas objetivo. Estos cambios en la conducta de los entrenadores fueron mayoritariamente percibidos por sus jugadores en la fase de evaluación. Concretamente, las percepciones de los jugadores del Entrenador 1 mostraron un claro aumento de Ánimo General y Ánimo Contingente al Error, y las de los jugadores del Entrenador 2 una clara mejoría de Ánimo General, Refuerzo e Instrucción Técnica al Error. Los resultados se discuten en base a los principios del CET y se proponen potenciales aplicaciones de nuestro programa


O objetivo do presente estudo é avaliar uma intervenção baseada nos princípios do Coach Effectiveness Training(CET) através de doisestudos de caso. Dois treinadores de basquetebol selecionaram 3 comportamentos - objetivo a melhorar. A avaliação comportamental revelou que o Trei-nador 1 conseguiu mudanças positivas nos 3 comportamentos - objetivo. O Treinador 2 melhorou em dois dos 3 comportamentos que definiu como ob-jetivo. Estas mudanças nos comportamentos dos treinadores foram, na sua maioria, percebidos pelos seus jogadores na fase de avaliação. Concretamente,as perceções dos jogadores do Treinador 1 indicam um aumento evidente de Animo Geral e Animo após o Erro, e as perceções dos jogadores do Treinador2 indicam uma clara melhoria de Animo Geral, Reforço e Instrução Técnica ao Erro. Os resultados são discutidos com base nos princípios do CET e sãoapresentadas propostas de potenciais aplicações do nosso programa de intervenção

Con la finalidad de comparar la prevalencia de Microsporidiosis intestinal en niños con desnutrición severa y niños eutróficos de la Unidad de Recuperación Nutricional del Hospital Chiquinquirá de Maracaibo, Estado Zulia, se realizó la técnica de reacción en cadena de la polimerasa (PCR) para la identificación de Encephalitozoon intestinalis y Enterocytozoon bieneusi en las muestras de heces de 50 niños desnutridos graves y 50 niños eutróficos, cuyas edades fueron estratificadas de la siguiente manera: lactantes menores (0-11 meses), lactantes mayores (12-23 meses), preescolares (2-6 años), y escolares (7 a 12 años). De las especies de microsporidios investigadas, se evidenció Enterocytozoon bieneusi en el 14% de los niños desnutridos graves y 8% en los niños eutróficos, no se obtuvieron amplificaciones para Encephalitozoon intestinalis en los grupos estudiados; al aplicar la prueba estadística X², resultó no significativa para las variables microsporidiosis intestinal y desnutrición. Se concluye que la medida en la que se relacionan la desnutrición, y la microsporidiosis es difícil de esclarecer, ya que la desnutrición es una condición multifactorial y aún no está bien establecido cuál de estos dos factores es la causa y cual la consecuencia.


In order to compare the prevalence of intestinal microsporidiosis in children with severe malnutrition and eutrophic children at the Nutritional Recovery Unit, Chiquinquirá Hospital in Maracaibo, State of Zulia, a chain reaction technique was performed on the polymerase (PCR) to identify Encephalitozoon intestinalis and Enterocytozoon bieneusi in stool samples from 50 healthy and 50 severely malnourished children, whose ages were stratified as follows: younger infants (0-11 months), older infants (12-23 months), preschool (2-6 years) and school children (7-12 years). Of the microsporidia species investigated, Enterocytozoon bieneusi was evident in 14% of severely malnourished children and 8% of the eutrophic children. No amplifications for Encephalitozoon intestinalis were obtained in the groups studied. On applying the chi-square statistical test, the result was not significant for the variables intestinal microsporidiosis and malnutrition. Conclusions are that the extent to which malnutrition and microsporidiosis relate is difficult to clarify, because malnutrition is a multifactorial condition, and it is not yet well established which of these two factors is the cause and which is the consequence.

Para determinar la prevalencia de Entamoeba histolytica como productora de diarrea, se realizó un estudio en niños menores de cinco años con diarrea que asistieron a diversas consultas del Servicio Autónomo Hospital Universitario de Maracaibo, Venezuela. A las muestras de heces obtenidas, se les realizó examen macroscópico, microscópico con solución salina fisiológica 0,85% y lugol, así como coloración de Kinyoun. El resto de la muestra se congeló hasta la extracción del ADN y luego se amplificaron mediante PCR separadas para E. histolytica y E. dispar. De las 50 muestras analizadas mediante examen microscópico, ninguna presentó trofozoítos y/o quistes de Entamoeba histolytica/dispar/moshkovskii, ni coccidios intestinales. Los parásitos detectados fueron Giardia lamblia (6%), Blastocystis sp. (6%), Pentatrichomonas hominis (2%), Ascaris lumbricoides (2%) y Trichuris trichiura (2%). Mediante PCR, 6 muestras (12%) presentaron ADN de E. dispar y 2 (4%) ADN de E. histolytica. Ningún niño presentó asociación de ambas amibas. Los dos niños que presentaron E. histolytica tenían 1 año de edad. E. dispar si fue detectada en niños de menor edad. Se sugiere que la prevalencia de E. histolytica en niños menores de cinco años con diarrea es realmente baja.


To determine the prevalence of Entamoeba histolytica as a producer of diarrhea, a study was conducted in children, less than five years of age, with diarrhea who attended several out patient clinics of the Servicio Autónomo Hospital Universitario, Maracaibo, Venezuela. A macroscopic and microscopic examination with physiological saline, lugol and Kinyoun staining were performed to the stool samples obtained. The remainder of the sample was frozen until DNA extraction, and PCR amplification was performed separately for E. histolytica and E. dispar. Microscopic examination showed no trophozoites and/or cysts of Entamoeba histolytica/dispar/moshkovskii, or intestinal coccidians in any of the 50 samples analyzed. Parasites detected were Giardia lamblia (6%), Blastocystis sp. (4%), Pentatrichomonas hominis (2%), Ascaris lumbricoides (2%) and Trichuris trichiura (2%). By PCR, six samples (12%) had DNA of E. dispar and two (4%) had DNA from E. histolytica; no child showed association of both amoebae. The two children who had E. histolytica were one-year-old. E. dispar was detected in younger children. We suggest that the prevalence of E. histolytica in children under five years is really low.

Objetivo: Determinar la prevalencia de enteroparásitos en niños con desnutrición moderada atendidos en dos centros hospitalarios de la ciudad de Maracaibo, Estado Zulia. Materiales y método: Se analizaron 84 muestras fecales de niños de ambos sexos con desnutrición moderada hospitalizados en la Unidad De Recuperación Nutricional del hospital Chiquinquirá y General del Sur Dr. Pedro Iturbe, en edades comprendidas entre los 8 meses a 12 años de edad. A cada espécimen fecal se le realizó un examen coproparasitológico directo al fresco con solución salina fisiológica al 0,85% y coloración temporal de lugol, así como la técnica de concentración de Ritchie. Resultados: Se observó una prevalencia general de niños parasitados de un 58% donde el género femenino obtuvo un 34,52% mientras que el masculino 23,81%. Hubo predomino del monoparasitismo en un 57,14%. En cuanto al grupo de edades, el estrato de 3 a 6 años resultó el mayormente parasitado (30,95% %). El ² reveló que si existe relación significativa entre las variables Parasitismo y edad. Las especies de enteroparásitos encontradas fueron: Blastocystis sp (49,39%), Giardia intestinalis (21,52%) quienes ocuparon los primeros lugares, seguidos de Complejo Entamoeba histolytica/Entamoeba dispar/ moshkovskii (11,39%), Entamoeba coli (6,33%) y Trichuris trichiura (6,33%), en todo el grupo estudiado. Conclusiones: Los resultados son similares a los obtenidos en investigaciones previas. La alta prevalencia de protozoarios sugieren un patrón de transmisión directa entre la población estudiada o a través del consumo de agua y/o alimentos contaminados fecalmente. Se plantea la necesidad de evaluar el comportamiento en la higiene alimentaria y abastecimiento de agua en la población estudiada.


Objective: To determine the prevalence of intestinal parasites in moderately malnourished children treated at two hospitals in the city of Maracaibo, State of Zulia. Materials and Methods: Eighty-four fecal samples from moderately malnourished children of both sexes between 8 months and 12 years of age, hospitalized in the Nutrition Rehabilitation Unit of the Chiquinquirá Hospital and South General Hospital, Dr. Pedro Iturbe, were analyzed. Each fecal specimen underwent a fresh test with saline solution 0.85%, Lugol staining and Ritchie concentrate. Results: An overall prevalence of parasitized children (58%) was observed; 34.52% were female, 23.81% were male. There was a predominance of monoparasitism 57.14%. Regarding the age group, the 3 to 6 stratum was most heavily parasitized (30. 95%). The c² revealed a significant relationship between parasitism and age. Species found in the group under study were: Blastocystis spp. (49. 39%), Giardia intestinalis (21.52%), Entamoeba histolytica / dispar / moshkovskii (11.39%), Entamoeba coli (6.33%) and Trichuris trichiura (6.33%). Conclusions: The results are similar to those obtained in previous research. The high prevalence of protozoa suggests a pattern of direct transmission among the studied population or through consumption of water and /or food. The need to evaluate behaviour related to food hygiene and water supply forth estudied population is proposed.

Para determinar la seroprevalencia de anticuerpos contra Entamoeba histolytica y la prevalencia de los enteroparásitos en indígenas de la comunidad de Toromo, estado Zulia, Venezuela, se estudiaron 69 individuos de ambos sexos, de 1 a 60 años de edad, en el año 2012. Se recolectaron muestras de heces y suero; a todas las muestras fecales se les realizó examen microscópico con solución fisiológica y lugol, concentración con formol-éter y coloración de Kinyoun; para la detección de anticuerpos IgG anti-Entamoeba histolytica en suero, se empleó una técnica de ELISA comercial. El 98,6% de los individuos estudiados estaban parasitados, predominando el poliparasitismo (88,2%), detectándose asociaciones de hasta 10 especies parasitarias. Entre las especies encontradas están, Ascaris lumbricoides el principal helminto (65%), y Blastocystis sp. (59%) el protozoario más frecuente. Entamoeba histolytica/dispar/moshkovskii se detectó por microscopia en 25 individuos (36% de prevalencia), donde se incluyó un caso de infección por Entamoeba histolytica, por haberse encontrado trofozoítos hematófagos. Se observó una seroprevalencia de anticuerpos contra E. histolytica, del 83%. De los resultados obtenidos, se concluye que existe una elevada seroprevalencia de anticuerpos contra E. histolytica, así como una alta prevalencia de enteroparásitos en la población indígena estudiada.


In order to determine the seroprevalence of antibodies against Entamoeba histolytica and intestinal parasites in the indigenous community of Toromo, Zulia state, Venezuela, 69 individuals of both sexes, from 1 to 60 years of age were studied during the year 2012. Stool and serum samples were collected. Microscopic study of fecal specimens was carried out by saline solution dilution, Lugol staining, formol-ether concentration and Kinyoun’s stain. For determination of anti-Entamoeba histolytica IgG antibodies, a commercial ELISA technique in serum was used; 98.6% of the population were parasitized, predominating the polyparasitism (88.24%) and finding up to 10 different parasite species were associated. The main parasitic species found were: Ascaris lumbricoides (65%), followed by Blastocystis sp. (59%). Entamoeba histolytica/dispar/moshkovskii was detected by microscopy in 25 individuals (36% prevalence), which included a case of infection by Entamoeba histolytica as hematophagous trophozoites were observed. A seroprevalence of 83% for IgG anti-Entamoeba histolytica antibodies in the individuals studied was found. According to the results it is concluded, that there is a high seroprevalence of antibodies against E. histolytica, as well as a high prevalence of intestinal parasites in the indigenous population studied.

Microsporidioses are considered emerging and opportunistic infections in immunocompromised individuals worldwide. The purpose of this study was to identify the species of intestinal microsporidia in patients with HIV-AIDS from the Servicio Autónomo Hospital Universitario de Maracaibo, Venezuela (SAHUM). Fecal samples were collected from 50 patients with confirmed diagnosis of HIV, during the years 2007 and 2008; the CD4 values were obtained from 42 patients. The samples were analyzed by separate PCRs to identify Encephalitozoon intestinalis and Enterocytozoon bieneusi. Microsporidia species showed a 36% prevalence: ten patients had Encephalitozoon intestinalis, four Enterocytozoon bieneusi and four both species. An inverse and statistically significant relationship between the CD4 count and the presence of microsporidia in the fecal sample was also found. It is remarkable the high prevalence of microsporidia species observed in the HIV patients studied, with a predominance of E. intestinalis.

Con la finalidad de determinar la prevalencia de Blastocystis sp. y especies de protozoarios comensales intestinales del hombre según diferentes aspectos como la edad y el sexo, se evaluaron muestras de heces de 111 individuos varones y mujeres de todas las edades, pobladores de Santa Rosa de Agua, Maracaibo, estado Zulia. Cada muestra fecal se analizó a través de un examen al fresco, tinción de lugol y técnica de concentración de Ritchie. Se utilizó la tinción de hematoxilina férrica para identificar trofozoitos de Dientamoeba fragilis. Las especies del complejo Entamoeba histolytica/Entamoeba dispar se diferenciaron mediante técnicas de PCR. Blastocystis sp. fue la especie predominante (45,6%), seguida por Entamoeba coli (17,9%), Endolimax nana (14,8%), Chilomastix mesnili (6,2%), Entamoeba dispar (5,6%), Dientamoeba fragilis, Pentatrichomonas hominis, Iodamoeba butschlii (3,1%) respectivamente y Entamoeba hartmanni (0,6%). Hubo afinidad parasitaria entre las especies Entamoeba coli y Endolimax nana. No se demostró asociación entre presencia de infección y sexo. Los grupos etarios preescolares y escolares demostraron asociación significativa con la infección parasitaria. Se determinó una elevada prevalencia de Blastocystis sp. y especies comensales intestinales, todas indicadoras de contaminación fecal, donde probablemente factores ambientales y socioculturales promovieron su transmisión.


With the purpose of determining Blastocytis sp. and other commensal intestinal protozoa species prevalence according to different aspects such as age and sex, we evaluated 111 feces samples from male and female individuals of all ages, living at Santa Rosa del Agua, Maracaibo, Zulia State, Venezuela. Each fecal sample was analyzed fresh, stained with lugol, and by Ritchie’s concentration test. We used ferric hematoxilin stain to identify Dientamoeba fragilis trophozoites. Species belonging to the Entamoeba histolytica/Entamoeba dispar complex were differentiated through PCR techniques. Blastocytis sp. was the predominant species (45.6%), followed by Entamoeba coli (17.9%), Endolimax nana (14.8%), Chilomastix mesnili (6.2%), Entamoeba dispar (5.6%), Dientamoeba fragilis, Pentatrichomonas hominis, Iodamoeba butschlii (3.1%, respectively), and Entamoeba hartmanni (0.6%). There was parasitic affinity between the Entamoeba coli and Endolimax nana species. There was no association between presence of infection and sex. Pre-school and school age groups showed a significant association with parasite infection. A high prevalence of Blastocystis sp. and intestinal commensal species was determined, all indicative of fecal contamination, where environmental and sociocultural factors probably promote transmission.

Los microsporidios pueden provocar infecciones emergentes y oportunistas en individuos inmunocomprometidos de todo el mundo. Se realizó éste estudio para identificar las especies de microsporidios intestinales presentes en pacientes con VIH-SIDA del Servicio Autónomo Hospital Universitario de Maracaibo (SAHUM). Se recolectaron 50 muestras fecales de individuos con diagnóstico confirmado de VIH durante los años 2007-2008; se obtuvieron las cifras de CD4 de solo 42 pacientes. Las muestras se analizaron mediante PCR separadas para la identificación de Encephalitozoon intestinalis y Enterocytozoon bieneusi. Las especies de microsporidios presentaron un 36% de prevalencia, 10 pacientes presentaron Encephalitozoon intestinalis, 4 Enterocytozoon bieneusi y 4 ambas especies. Se determinó una relación inversamente proporcional y estadísticamente significativa entre el contaje de CD4 y la presencia de microsporidios en la muestra fecal. Es destacable la elevada prevalencia de especies de microsporidios observada en los pacientes VIH estudiados, donde predominó E. intestinalis.

ABSTRACT

To determine the prevalence of Entamoeba histolytica as a producer of diarrhea, a study was conducted in children, less than five years of age, with diarrhea who attended several out patient clinics of the Servicio Aut6nomo Hospital Universitario, Maracaibo, Venezuela. A macroscopic and microscopic examination with physiological saline, lugol and Kinyoun staining were performed to the stool samples obtained. The remainder of the sample was frozen until DNA extraction, and PCR amplification was performed separately for E. histolytica and E. dispar. Microscopic examination showed no trophozoites and/or cysts of Entamoeba histolytica/dispar/moshkovskii, or intestinal coccidians in any of the 50 samples analyzed. Parasites detected were Giardia lamblia (6%), Blastocystis sp. (4%), Pentatrichomonas hominis (2%), Ascaris lumbricoides (2%) and Trichuris trichiura (2%). By PCR, six samples (12%) had DNA of E. dispar and two (4%) had DNA from E. histolytica; no child showed association of both amoebae. The two children who had E. histolytica were one-year-old. E. dispar was detected in younger children. We suggest that the prevalence of E. histolytica in children under five years is really low.
