ABSTRACT

Ferulic acid esterases (FAE) were produced by Aspergillus terreus CECT 2808 from vine trimming shoots (VTS) and corn cob. Later, the fungal extracts thus obtained were used to enzymatically release ferulic acid (FA) from both substrates. Our findings showed a higher FAE activity in the enzymatic extracts produced on corn cob (0.070±0.004U/mL). Nevertheless, the enzymatic extracts produced on VTS demonstrated a better performance for FA release from both corn cob (2.05±0.01mg/g) and VTS (0.19±0.003mg/g). This result was probably because of the higher xylanase/FAE ratio determined in VTS extract. Therefore, an additional assay was carried out by supplementing corn cob extract with a commercial xylanase to test the influence of FAE/xylanase ratio in FA release. The results revealed the relevance of the FAE/xylanase ratio for an optimal FA release.

ABSTRACT

Threshold photoelectron spectrum of ozone is presented for the first time at a resolution of 21-38 meV using synchrotron radiation in the energy region of 12-21 eV. The ionization energies of the first ionized states were determined and an interpretation of the O3 spectrum with respect to its first three ionic states, 1 2A1, 1 2B2, and 1 2A2, is presented. Above 16 eV the enhancement of the intensities of the 2 2B1, 3 2A1, and 4 2B2 band systems due to the contribution of indirect processes was observed, not accessible by conventional photoelectron spectroscopy. It was also resolved and assigned the extensive vibrational structures of ozone. Between 15.5 and 18.5 eV the main band contours are similar to those found in conventional photoelectron spectroscopy, except that our threshold photoelectron spectrum reveals extensive additional vibrational structures. The band 2 2B1 was found to present an irregular vibrational spacing DeltaE, with a minimum value of 80 meV at approximately 16.47 eV.

ABSTRACT

Many land plants deviate from the maternal pattern of organelle inheritance. In this study, heterologous mitochondrial and chloroplast probes were used to investigate the inheritance of organelle genomes in the progeny of an intergeneric cross. The seed parent was LB 1-18 (a hybrid of Citrus reticulata Blanco cv. Clementine x C. paradisi Macf. cv. Duncan) and the pollen parent was the cross-compatible species Poncirus trifoliata (L.) Raf. All 26 progeny examined exhibited maternal inheritance of plastid petA and petD loci. However, 17 of the 26 progeny exhibited an apparent biparental inheritance of mitochondrial atpA, cob, coxII, and coxIII restriction fragment length polymorphisms (RFLPs) and maternal inheritance of mitochondrial rrn26 and coxI RFLPs. The remaining nine progeny inherited only maternal mitochondrial DNA (mtDNA) configurations. Investigations of plant mitochondrial genome inheritance are complicated by the multipartite structure of this genome, nuclear gene control over mitochondrial genome organization, and transfer of mitochondrial sequences to the nucleus. In this study, paternal mtDNA configurations were not detected in purified mtDNA of progeny plants, but were present in progeny DNA preparations enriched for nuclear genome sequences. MtDNA sequences in the nuclear genome therefore produced an inheritance pattern that mimics biparental inheritance of mtDNA.
