ABSTRACT

Abiotic stress, especially salt stress, is one of the major barriers to crop production worldwide. Crops like onion that belong to the glycophytic group are more sensitive to salinity stress. A huge study regarding the influence of salinity stress on the growth and development of crops has already been done and is still ongoing. One of the major targets of the research is to develop genotypes that have enhanced performance under stress environments. The world needs more of these types of genotypes to combat the ever-growing salt-stressed soils. Therefore, a number of germplasm were studied during the 2019-2020 and 2020-2021 seasons under different salt concentrations to identify tolerant genotypes as well as to study the plants' responses at different growth stages against elevated salinity levels. A 2-year study was conducted where germination potential was evaluated in the first year and carried out in petri dish culture of seeds, followed by plastic pot culture for plant establishment and bulb development evaluation during the second year. Four different saline water solutions having different salt concentrations (0, 8, 10, and 12 dS m-1) were applied to the petri dishes and pots as the source of water for plants in both seasons. Results indicated that a significant reduction in plants' performance occurs under higher salinity levels. Salt concentration had an adverse impact on germination, leaf development and growth, the height of plants, bulb size and shape, and the bulb weight of onion. All the growth phases of onion are sensitive to elevated concentrations. Variable performances were observed in the genotypes under stress conditions, and a few genotypes (Ac Bog 409, Ac Bog 414, Ac Bog 424, Ac Bog 430, Ac Bog 417, Ac Bog 419, Ac Bog 420, Ac Bog 422, and Ac Bog 425) having some sort of tolerance to salt stress were identified, which might be recommended for mass production. Tolerance indices could successfully be applied in selecting the salt-tolerant genotypes. Thus, the present findings and the identified genotypes could be further utilized in salt stress improvement research on onion.

ABSTRACT

INTRODUCTION: Our previous case-control study demonstrated that a high level of intestinal alkaline phosphatase (IAP), an endotoxin-detoxifying anti-inflammatory enzyme secreted by villus-associated enterocytes and excreted with stool, plays a protective role against type 2 diabetes mellitus (T2DM) irrespective of obesity. In the current study, we investigated the long-term effect of IAP deficiency (IAPD) on the pathogenesis of T2DM. RESEARCH DESIGN AND METHODS: A healthy cohort of participants without diabetes (30-60 years old), comprising 188 without IAPD (IAP level: ≥65 U/g stool) and 386 with IAPD (IAP level: <65 U/g stool), were followed up for 5 years. We measured stool IAP (STAP) and fasting plasma glucose, and calculated the risk ratio (RR) using log-binomial regression model. RESULTS: T2DM incidence rates were 8.0%, 11.7%, 25.6%, and 33.3% in participants with 'persistent no IAPD' (IAP level: always ≥65 U/g stool), 'remittent IAPD' (IAP level: increased from <65 U/g stool to ≥65 U/g stool), 'persistent IAPD' (IAP level: always <65 U/g stool), and 'incident IAPD' (IAP level: decreased from ≥65 U/g stool to <65 U/g stool), respectively. Compared with 'persistent no IAPD' the risk of developing T2DM with 'incident IAPD' was 270% higher (RR: 3.69 (95% CI 1.76 to 7.71), χ2 p<0.001). With 'persistent IAPD' the risk was 230% higher (RR: 3.27 (95% CI 1.64 to 6.50), p<0.001). 'Remittent IAPD' showed insignificant risk (RR: 2.24 (95% CI 0.99 to 5.11), p=0.0541). Sensitivity analyses of persistent IAP levels revealed that, compared with participants of the highest persistent IAP pentile (always >115 U/g stool), the rate of increase of fasting glycemia was double and the risk of developing T2DM was 1280% higher (RR: 13.80 (95% CI 1.87 to 101.3), p=0.0099) in participants of the lowest persistent IAP pentile (always <15 U/g stool). A diabetes pathogenesis model is presented. CONCLUSIONS: IAPD increases the risk of developing T2DM, and regular STAP tests would predict individual vulnerability to T2DM. Oral IAP supplementation might prevent T2DM.

BACKGROUND: Bangladesh is a densely populated country with a high demand for non-vegetable protein. Poultry meat is an important source of protein due to its affordability. Trace metals are persistent in the environment and are subject to bioaccumulation in the food chain. Contaminated poultry feed can pose a risk to human health as it biomagnifies and bioconcentrates toxic metals up the food chain, which can enter the human body and cause adverse and toxic health effects. OBJECTIVES: The aim of the current study was to assess the concentration and distribution of metals such as arsenic (As), cadmium (Cd), cobalt (Co), chromium (Cr), copper (Cu) and nickel (Ni) in different types and parts of poultry in markets in Dhaka, Bangladesh. METHODS: A total of 15 chickens were collected from five commercial markets of Dhaka city. Three types of poultry were selected, broiler, layer and cock. Altogether, 45 chicken samples were analyzed for toxic metals concentrations using inductively coupled plasma optical emission spectroscopy. RESULTS: The concentrations of As, Cd, Co, Cr, Cu and Ni obtained in the broiler, layer and cock chickens were 0.728, 0.232, 0.392; 0.595, 0.245, 0.271; 0.058, 0.016, 0.096; 5.275, 1.562, 22.180; 3.571, 2.269, 4.241, and 0.332, 0.211, 0.433 mg/kg (wet weight), respectively. The results indicate that the concentrations of As, Cr, and Cu in the analyzed samples exceeded the maximum permissible levels, whereas the concentrations of Cd and Ni were within acceptable limits. Metal concentrations decreased in the order of Cr > Cu > As > Cd > Ni > Co. The target hazard quotient and cancer risk values were estimated for each metal due to consumption of the different types of chicken. CONCLUSIONS: The estimated metal concentrations of As, Cr and Cu were higher than the permissible levels of international standards, indicating a health risk. Groundwater in many parts of the country is contaminated with As and is a probable source of As contamination in poultry. High concentrations of Cr and Cu in chicken may be caused by contaminated poultry feed. In the present study, layer chickens were comparatively less contaminated with metals than other types of chicken. The estimated target hazard quotient and cancer risk values of the analyzed chicken samples were lower than acceptable limits for all individual trace metals in the three types of chicken, indicating no non-carcinogenic and cancer health risk from ingestion of a single trace metal through consumption of these chickens. ABSTRACT

BACKGROUND: Here, we report the phase distribution of chronic myeloid leukemia (CML), defined based on the World Health Organization criteria, among 63 patients in Bangladesh. All patients were diagnosed based on complete blood count, bone marrow examination including bone marrow aspiration and reverse-transcriptase polymerase chain reaction (RT-PCR). Out of 63 patients, 42 were male and 21 were female. The mean age of the subjects was 37.4 years, with an age range of 17-60 years. The majority of patients (86%) were classified in the chronic phase (CP), 7 (11%) in the accelerated phase (AP) and two (3%) in blast crisis (BC). The most frequent patient age ranges were 21-30 years for CP, 41-50 years for AP and 41-50 years for BC. RESULTS: The Philadelphia chromosome was detected in 48 patients by RT-PCR. The mean total leukocyte counts, platelet counts, hemoglobin levels and marrow blast frequencies were 101 × 10(9)/L, 409 × 10(9)/L, 12.2 g/dl and 2.8% for CP; 121 × 10(9)/L, 418 × 10(9)/L, 8.7 g/dl and 15% for AP and 311 × 10(9)/L, 396 × 10(9)/L, 9.2 g/dl and 26% for BC, respectively. CONCLUSION: This study concluded that most CML patients in Bangladesh are from a younger age group (31-40 years). In addition, males were more commonly affected, although females were afflicted with this disease at a younger age.

ABSTRACT

BACKGROUND: Withania somnifera, also known as ashwagandha, is an important herb in ayurvedic and indigenous medical systems. The present study was designed to evaluate the antioxidant and antibacterial activities of an 80% aqueous methanolic extract of W. somnifera roots (WSREt), fruits (WSFEt) and leaves (WSLEt). METHODS: Several assays were performed to determine the antioxidant properties of this herb including 1,1-diphenyl-2-picrylhydrazyl radical (DPPH) scavenging activity, ferric reducing antioxidant power (FRAP), ferrous chelation and inhibition of ß-carotene bleaching. RESULTS: The values for DPPH, FRAP, ferrous chelation and inhibition of ß carotene bleaching for the three types of extracts ranged from 101.73-801.93 µg/ml, 2.26-3.29 mM Fe/kg, 0.22-0.65 mg/ml and 69.87-79.67%, respectively, indicating that W. somnifera, particularly the leaves, possesses significant antioxidant properties. The mean ascorbic acid content was 20.60-62.60 mg/100 g, and the mean anthocyanin content was 2.86-12.50 mg/100 g. Antibacterial activities were measured using the agar well diffusion method and five pathogenic Gram-negative bacteria: Escherichia coli, Salmonella typhi, Citrobacter freundii, Pseudomonas aeruginosa and Klebsiella pneumoniae. The leaf extracts displayed the highest activity against S. typhi (32.00 ± 0.75 mm zone of inhibition), whereas the lowest activity was against K. pneumoniae (19.00 ± 1.48 mm zone of inhibition). The lowest minimum inhibitory concentration value was 6.25 mg/ml, which was against S. typhi, followed by 12.5 mg/ml against E. coli. CONCLUSION: In addition to its antioxidant properties, W. somnifera exhibited significant antibacterial activities against Gram-negative bacteria, particularly S. typhi.
