ABSTRACT

On commercial farms, rabbit does are subjected to a reproductive rhythm that does not account for their welfare or physiology, leading to reduced longevity and consequently high annual replacement. The European Food Safety Authority (EFSA) recommends limited and infrequent use of hormone treatments, and suggests replacement with alternative methods that do not threaten animal welfare when possible. In the present study, we aimed to determine whether the GnRH analogue lecirelin acetate could be administered by inclusion in the seminal dose during insemination. Twenty 9-month-old does (Grigio del Monferrato, autochthonous Italian breed),each having two previous deliveries, were individually housed and divided into two groups at artificial insemination. The control group received 0.2 mL of intramuscular lecirelin (Dalmarelin, Fatro®, Italy) prior to insemination. The intravaginal group was inseminated with a seminal dose that included 0.3 mL Dalmarelin. The experiment was performed for six consecutive reproductive cycles at 42-day intervals, and included a total of 120 inseminations. Prior to each insemination, the heterospermic pooled semen samples were assessed for sperm motility and morphology. Each ejaculate was divided into two samples, with and without lecirelin addition. Compared to the control group, the does with intravaginal Dalmarelin administration showed equal or greater sexual receptivity, which resulted in a higher fertility rate over increasing cycles. The seminal dose volume was very low, possibly explaining the better results in the intravaginal group, which received a similar amount of hormone (0.3 mL/doe) as the control group (0.2 mL/doe). The negative performance of the control group may have also been due to anti-GnRH formation, and the more stressful method of ovulation induction. The number of live-born kits did not significantly differ between groups. Progressive motility was significantly positively correlated with motility characteristics, including VAP, VSL, ALH, BCF, STR, and LIN. Overall, our present findings supported that the incorporation of GnRH in a seminal dose could be used for ovulation induction in rabbit does. Further studies should identify the optimal dose of GnRH for intravaginal administration, taking into account that the intravaginal absorption capacity is about 10 times smaller than the intramuscular absorption capacity.

ABSTRACT

The aim of this study was to analyze the transfer of bioactive compounds from the pasture to the body and meat of organic free-range chickens and to verify the effect of these compounds on the oxidative processes of the meat. Starting at 21 d of age, 100 male naked-neck birds were divided into two homogeneous groups: an indoor group (0.12 m(2)/bird) and an outdoor group (0.12 m(2)/bird indoor and 10 m(2)/bird of forage paddock). At slaughter (81 d of age), blood samples were collected, and the carcasses were stored for 24 h at 4°C (20 birds/group). The grass samples had higher values of carotenoids, tocopherols, and flavonoids respect to standard feed (based on dry matter comparison). The polyunsaturated fatty acid ( PUFA: ) content was also greater in grass, especially the n-3 series (so named because its first double bond occurs after the third carbon atom counting from the methyl at the end of the molecule). The antioxidant profile of the grass improved the antioxidant status of the crop and gizzard contents in the outdoor chickens. The higher antioxidant intake resulted in a higher plasma concentration of antioxidants in outdoor birds; thiobarbituric acid reactive substances ( TBARS: ) and the antioxidant capacity of the plasma were also better in the outdoor than the indoor group. The meat of the outdoor birds had higher levels of antioxidants, mainly due to the higher amount of tocopherols and tocotrienols. Despite the higher antioxidant protection in the drumstick of the outdoor group, the TBARs value was greater, probably due to the kinetic activity of birds, the higher percentage of PUFAs, and the peroxidability index. In conclusion, grazing improved the nutritional value of the meat (PUFA n-3 and the ratio between n-6 and n-3 PUFA) with a minor negative effect on the oxidative stability. Suitable strategies to reduce such negative effects (e.g., reduction of kinetic activity in the last days of rearing) should be studied.

ABSTRACT

The objective of this study was to evaluate the effects of two different probiotic micro-organisms on the performance, egg quality and blood parameters of organically reared hens. A total of 900 16-week-old Hy-Line layer hybrids were randomly assigned to three groups of 300 birds each. The control (CTR) group was fed a corn-soya bean cake-based diet; the L group was fed the same diet supplemented with 0.1% Lactobacillus acidophilus, while the B group was fed the same diet supplemented with 0.05% Bacillus subtilis. Data were recorded at the beginning (weeks 5 and 6: T1) and at the end (weeks 19 and 20: T2) of the experiment, and no differences in hen performance were recorded between dietary groups or sampling times. All of the investigated clinical chemistry parameters, except GGT, were affected by diet (p < 0.05), with the best results recorded for the probiotic-treated groups. The immune-response values showed higher blood bactericidal activity in the B and L groups at T2 (p < 0.05) and a lower lysozime concentration in the B group at T1. Higher antibody production against Newcastle disease virus was observed in the L group compared to the CTR (p = 0.013). No differences in oxidative status were recorded, and no effects of diet on egg quality were observed. Among the physical egg characteristics, only the Roche scale colour was affected by diet (p < 0.05): the egg yolk was paler in the L group. The age of the hen was the most relevant factor affecting physical egg characteristics. The chemical parameters of the egg were almost unaffected by supplementation with probiotics except for the lipid content, which decreased with the L diet (p < 0.05). Both probiotic inclusions had beneficial effects on hen metabolism and welfare, and L. acidophilus induced the best immune response.

ABSTRACT

In order to improve sediment handling following dredging operations, this study aims to statistically derive ranges of distribution for certain microbiological parameters, according to four environmental types inspired by Italian legislation on seaports: ports of international/national importance, ports of regional importance, port channels in brackish environments, and marine coastal areas. A national database was developed using microbiological data from technical reports available at the Italian Ministry of Environment and National Institute of Environmental Protection and Research (ISPRA) for the period 1990-2008. The parameters considered were total coliform bacteria, faecal coliform bacteria, Escherichia coli, enterococci, sulfite-reducing clostridia (SRC), total bacterial counts at 22 °C and at 37 °C, and fungi. The data were statistically analyzed: (1) to verify the correspondence with the identified environmental types and rank them according to the concentration gradient and (2) to describe the data distribution in order to obtain reference ranges typical for each parameter/environmental type. The four environmental types considered were clearly different for enterococci, SRC, and fungi, highlighting a correspondence with Italian legislation. For the remaining parameters, at least two environmental types were merged. In general, the less contaminated environments were small ports and relatively unimpacted coastal areas. The ranges defined for relatively clean coastal areas can be considered a target for other areas both from an environmental point of view and for the sediment management implications. These values could be used as a comparison in environmental surveys addressing marine or brackish sediment handling and may represent a future line of evidence for the assessment of overall sediment quality.

ABSTRACT

The objective of the study was to analyse and increase the quality of rabbit meat by increasing the levels of natural bioactive compounds through providing fresh alfalfa to rabbits as complementary feed. At 50 days of age, forty rabbits were divided into two homogeneous groups and fed pelleted feed (control group) or pelleted feed plus fresh alfalfa (alfalfa group). The lipid content of meat was significantly higher in the control group likely due to the higher feed consumption observed in this group. The same trend was observed in the contents of myristic, palmitoleic, oleic and linoleic acid. On the contrary, the supplementation of fresh alfalfa increased the stearic, linolenic, eicosatrienoic, eicosapentaenoic, docosapentaenoic, docosahexaenoic and total polyunsaturated fatty acid contents; also the total monounsaturated fatty acids and the α- and γ-tocopherol content was significantly lower. Moreover, alfalfa ingestion decreased the n-6 content, n-6/n-3 ratio and thrombogenicity index.

ABSTRACT

The objective of this study was to evaluate the effect of Spirulina and Thyme supplementation on rabbit meat during retail display. At weaning 294 rabbits were allocated to 7 different treatments (42 rabbits/treatment). Rabbits of the control group (C) received a diet without any supplementation throughout the experiment (5-11 weeks of age). The other groups were fed diets containing 5% Spirulina (S), 3% Thyme (T) or both supplements (ST) for the whole trial (5-11 weeks; treatments S, T and ST), or for a part of the growing period (8-11 weeks; treatments C-S, C-T and C-ST). Colour parameters, pH, water holding capacity and drip loss were determined on fresh and stored Longissimus dorsi muscle of 5 rabbits/treatment. Spirulina- and Thyme-supplemented diets had a significant effect on redness and yellowness of Longissimus dorsi. Drip loss was significantly reduced in C-T and T groups that also showed the highest content of α-tocopherol and n-3 fatty acids content and the lower lipid oxidation.

ABSTRACT

According to EC regulation 889/08, different European countries should draw up a list of slow-growing strains adapted to an organic system, and in the meantime, provide this information to operators and the European Union commission. Thus, the aim of the present work was to evaluate the effect of poultry genotype on fatty acid composition and lipid indices of poultry meat. Six poultry genotypes (100 birds each), each with a different growth rate (slow-growing: Leghorn, Ancona, Cornish×Leghorn; medium-growing: Kabir, Naked neck; fast-growing: Ross), were reared under an organic system. Breast meat fatness, fatty acid composition, and indices were largely related to genotype, as slow-growing strains had higher elongase, thioesterase, and Δ5/Δ6 desaturase indices accompanied by a lower Δ9. Differences in the fatty acid profiles were observed by varying contents of total saturated fatty acids, with a higher value seen in Leghorn chickens and a lower value seen in commercial lines. On the contrary, Leghorn and Ancona chickens exhibited higher amounts of stearic acid and total polyunsaturated fatty acids compared with commercial genotypes, both in the total content and in the different fractions (total n-3 and total n-6). Despite the increased consumption of fresh forage, the lower linolenic acid in meat of the slow-growing strain could be explained by the higher conversion of this fatty acid to its long-chain derivatives.

ABSTRACT

Contaminant loadings to the Venice Lagoon peaked from 1950s-1980s and although they have since declined, contaminant concentrations remain elevated in sediment and seafood. In order to identify the relative importance of contaminant sources, inter-media exchange and removal pathways, a modified 10-segment fugacity/aquivalence-based model was developed for octachlorodibenzodioxin/furan (OCDD/F), PCB-180, Pb and Cu in the Venice Lagoon. Results showed that in-place pollution nearby the industrial area, current industrial discharges, and tributary loadings were the main sources of contaminants to the lagoon, with negligible contributions from the atmosphere. The fate of these contaminants was governed by sediment-water exchange with simultaneous advective transport by water circulation. Contaminants circulated amongst the northern and central basins with a small fraction reaching the far southern basin and the Chioggia inlet. As a consequence, we estimated limited contaminant transfer to the Adriatic Sea, trapping the majority of contaminants in the sediment in this "average" circulation scenario which does not account for periodic flooding events.

ABSTRACT

A Monte Carlo analysis is used to quantify environmental parametric uncertainty in a multi-segment, multi-chemical model of the Venice Lagoon. Scientific knowledge, expert judgment and observational data are used to formulate prior probability distributions that characterize the uncertainty pertaining to 43 environmental system parameters. The propagation of this uncertainty through the model is then assessed by a comparative analysis of the moments (central tendency, dispersion) of the model output distributions. We also apply principal component analysis in combination with correlation analysis to identify the most influential parameters, thereby gaining mechanistic insights into the ecosystem functioning. We found that modeled concentrations of Cu, Pb, OCDD/F and PCB-180 varied by up to an order of magnitude, exhibiting both contaminant- and site-specific variability. These distributions generally overlapped with the measured concentration ranges. We also found that the uncertainty of the contaminant concentrations in the Venice Lagoon was characterized by two modes of spatial variability, mainly driven by the local hydrodynamic regime, which separate the northern and central parts of the lagoon and the more isolated southern basin. While spatial contaminant gradients in the lagoon were primarily shaped by hydrology, our analysis also shows that the interplay amongst the in-place historical pollution in the central lagoon, the local suspended sediment concentrations and the sediment burial rates exerts significant control on the variability of the contaminant concentrations. We conclude that the probabilistic analysis presented herein is valuable for quantifying uncertainty and probing its cause in over-parameterized models, while some of our results can be used to dictate where additional data collection efforts should focus on and the directions that future model refinement should follow.

ABSTRACT

The effect of the organic production system and genotype on chicken behavior and muscle fiber characteristics was assessed. Three hundred day-old male chicks from slow-growing (Leghorn), medium-growing (Kabir), and fast-growing (Ross 208) genotypes were assigned to 2 different production systems: conventional, housing in an indoor pen (0.12 m(2)/bird); and organic, housing in an indoor pen (0.12 m(2)/bird) with access to a grass paddock (4 m(2)/bird). Behavioral observations were recorded from 73 to 80 d of age in the morning and afternoon. At 81 d of age, blood samples were collected to measure lactate dehydrogenase and creatine kinase, and 20 birds per strain and rearing system were slaughtered. Samples of pectoralis major, ileotibialis lateralis, and semimembranosus muscles were obtained for histological evaluations. Behavioral observations showed that genetic selection of animals for a better growth rate modified their behavior, reducing kinetic activity. Indeed, Leghorn birds were characterized by moving activities, whereas Kabir and Ross strains were discriminated on the basis of their lying, standing, and eating activities, and these activities were strongly associated with energy conservation, growth, and muscle fiber characteristics. Fiber characteristics and muscle enzyme functions were affected by rearing system only in animals adapted to the organic system. Interesting results relative to Leghorn chickens are the presence of alpha-Red fiber in breast muscle and the increased cross-sectional area of the ileotibialis lateralis muscle, which together with behavioral data could affirm that this genotype is the most adapted to the organic rearing system.

ABSTRACT

The synergistic effect of dietary linolenic acid and vitamin E on the oxidative stability and nutritional and eating characteristics of fresh and stored rabbit meat was studied. One-hundred hybrid male rabbits were divided into two homogenous groups and fed ad libitum two diets differing in the amount of sunflower and flaxseed and in the level of α-tocopherol, as follows: control diet: 0.08 kg kg(-1) sunflower, 50 mg kg(-1) α-tocopheryl-acetate and LNA-VE diet: 0.08 kg kg(-1) flaxseed, 200 mg kg(-1) α-tocopheryl-acetate. At 85 days, 20 rabbits per group were slaughtered and the thiobarbituric-acid reactive substances (TBA-RS), chemical composition, fatty acid profile and sensory quality were assessed on the longissimus dorsi muscles (fresh and stored for 8 days at 4 °C). The proximate composition of the fresh muscle was not significantly affected by the dietary treatment. Rabbits fed the LNA-VE diet showed a good capability to elongate and desaturate linolenic acid and this diet enriched the n-3 PUFA content of the meat without affecting its peroxidative stability. The sensory quality of the fresh and stored muscle was slightly affected by the dietary treatment, even though final tenderness (fresh meat) and overall acceptability (stored meat) of the LNA-VE rabbits showed significantly higher scores.

ABSTRACT

The effect of organic production on broiler carcass and meat quality was assessed. Two hundred and fifty Ross male chickens were assigned to two different systems of production: conventional, housing in an indoor pen (0.12 m(2)/bird); organic, housing in an indoor pen (0.12 m(2)/bird) with access to a grass paddock (4 m(2)/bird). At 56 and 81 days of age, 20 chickens per group were slaughtered to evaluate carcass traits and the characteristics of breast and drumstick muscles (m. pectoralis major and m. peroneus longus). The organic chickens had carcasses with a higher breast and drumstick percentages and lower levels of abdominal fat. The muscles had lower pHu and water holding capacity. Instead cooking loss, lightness values, shear values, Fe, polyunsaturated fatty acids of n-3 series and TBA-RS were higher. The sensory quality of the breast muscle was better. Organic production system seems to be a good alternative method, due to better welfare conditions and good quality of the carcass and meat. A negative aspect was the higher level of TBA-RS in the muscles, probably due to greater physical activity.
