ABSTRACT

Tannins, which are water-soluble polyhydroxyphenols found in plant structures, offer a sustainable alternative to inorganic products for treating effluents. To overcome their solubility in water, new methods have been developed to immobilize tannin on materials such as nanocellulose, resulting in tannin-based adsorbents. This study investigates the production of different types of tannin-based adsorbents immobilized on nanocellulose nanofibrils using glutaraldehyde as a crosslinking agent. The adsorbents were characterized for their morphology, chemical composition, tannin leaching, thermal stability, and copper adsorption capacity. The main results showed that tannin leaching from the adsorbents in water was greatly reduced with glutaraldehyde. Scanning electron microscopy images showed tannin coating on the surface of nanocellulose nanofibrils due to immobilization. The lyophilized adsorbent was effective in adsorbing Cu (II) within 1 min, with the adsorbed amount increasing gradually until stabilization after 45 min. Overall, this study suggests that tannin immobilized on nanofibrillated cellulose has the potential to offer a sustainable and renewable alternative for effluent treatment.

ABSTRACT

This systematic review presents the potential of using feather waste as a ß-keratin source, including the Brazilian scenario in the generation of this byproduct. The structure and properties of α- and ß-keratin, the methods commonly reported to extract keratin from poultry feathers, and applications of feather keratin-based materials are also covered in this review. The literature search for poultry production data in Brazil was conducted for the last 2 years, for the period 2021-2022. A broad literature search for extraction methods and applications of feather keratin was done for the period 2001-2022. The poultry industry is one of the largest sectors of the food industry, and Brazil was the third-largest world producer of chicken meat with more than six billion chickens slaughtered in 2021. Poultry feathers constitute about 7% weight of broilers; thus, it can be estimated that about one million tons of poultry feathers were generated in Brazil in 2021, and the improper disposal of this byproduct contributes to environmental problems and disease transmission. The most common method of reusing feathers is the production of feather meal. From economic and environmental points of view, it is advantageous to develop processes to add value to this byproduct, including the extraction of keratin. Among natural biodegradable polymers, keratin-based materials have revolutionized the field of biomaterials due to their biocompatibility and biodegradability, allowing their application in biomedical, pharmaceutical, chemical, and engineering areas.

ABSTRACT

Purpose: Retinal microvascular abnormalities measured on retinal images are a potential source of prognostic biomarkers of vascular changes in the neurodegenerating brain. We assessed the presence of these abnormalities in Alzheimer's dementia and mild cognitive impairment (MCI) using ultra-widefield (UWF) retinal imaging. Methods: UWF images from 103 participants (28 with Alzheimer's dementia, 30 with MCI, and 45 with normal cognition) underwent analysis to quantify measures of retinal vascular branching complexity, width, and tortuosity. Results: Participants with Alzheimer's dementia displayed increased vessel branching in the midperipheral retina and increased arteriolar thinning. Participants with MCI displayed increased rates of arteriolar and venular thinning and a trend for decreased vessel branching. Conclusions: Statistically significant differences in the retinal vasculature in peripheral regions of the retina were observed among the distinct cognitive stages. However, larger studies are required to establish the clinical importance of our findings. UWF imaging may be a promising modality to assess a larger view of the retinal vasculature to uncover retinal changes in Alzheimer's disease. Translational Relevance: This pilot work reports an investigation into which retinal vasculature measurements may be useful surrogate measures of cognitive decline, as well as technical developments (e.g., measurement standardization), that are first required to establish their recommended use and translational potential.

ABSTRACT

The aim of this study was to verify the potential of the colorimetric technique in the identification of some species marketed as "tauari" in the Brazilian Amazon. CIE L* a* b* parameters were applied to determine the colour of 35 wood samples from the main wood poles of the State of Para, Brazil, and the scientific collections of the Museu Paraense Emilio Goeldi (Walter A. Egler Collection) and Embrapa Amazônia Oriental. From each sample, data were obtained in the three wood surfaces: transversal, longitudinal tangential and longitudinal radial. The coordinate b*, which showed the yellow pigment, exerted greater weight in the color characterization of the 35 samples marketed as tauari, being more evident in the tangential and radial sections. In PCA, MGW wood samples showed considerably distinct color patterns in relation to PA and IAN samples, and the h and L* parameters provided better informations for distinguishing species from sample sources. The colorimetric technique can be used as an auxiliary tool in the identification of wood. However, the simultaneous use of colorimetry with the anatomical description of wood is suggested, given the complexity of the species-level separation in "tauari" group.

ABSTRACT

BACKGROUND: Markers of cerebrovascular disease are common in dementia, and may be present before dementia onset. However, their clinical relevance in midlife adults at risk of future dementia remains unclear. We investigated whether the Cardiovascular Risk Factors, Ageing and Dementia (CAIDE) risk score was associated with markers of cerebral small vessel disease (SVD), and if it predicted future progression of SVD. We also determined its relationship to systemic inflammation, which has been additionally implicated in dementia and SVD. METHODS: Cognitively healthy midlife participants were assessed at baseline (n=185) and 2-year follow-up (n=158). To assess SVD, we quantified white matter hyperintensities (WMH), enlarged perivascular spaces (EPVS), microbleeds and lacunes. We derived composite scores of SVD burden, and subtypes of hypertensive arteriopathy and cerebral amyloid angiopathy. Inflammation was quantified using serum C-reactive protein (CRP) and fibrinogen. RESULTS: At baseline, higher CAIDE scores were associated with all markers of SVD and inflammation. Longitudinally, CAIDE scores predicted greater total (p<0.001), periventricular (p<0.001) and deep (p=0.012) WMH progression, and increased CRP (p=0.017). Assessment of individual CAIDE components suggested that markers were driven by different risk factors (WMH/EPVS: age/hypertension, lacunes/deep microbleeds: hypertension/obesity). Interaction analyses demonstrated that higher CAIDE scores amplified the effect of age on SVD, and the effect of WMH on poorer memory. CONCLUSION: Higher CAIDE scores, indicating greater risk of dementia, predicts future progression of both WMH and systemic inflammation. Findings highlight the CAIDE score's potential as both a prognostic and predictive marker in the context of cerebrovascular disease, identifying at-risk individuals who might benefit most from managing modifiable risk.

ABSTRACT

PURPOSE: To determine whether a 6-day course of methylprednisolone (MP) improves outcome in patients with severe SARS-CoV­2 (Corona Virus Disease 2019 [COVID-19]). METHODS: The study was a multicentric open-label trial of COVID-19 patients who were aged ≥ 18 years, receiving oxygen without mechanical ventilation, and with evidence of systemic inflammatory response who were assigned to standard of care (SOC) or SOC plus intravenous MP (40 mg bid for 3 days followed by 20 mg bid for 3 days). The primary outcome was a composite of death, admission to the intensive care unit, or requirement for noninvasive ventilation. Both intention-to-treat (ITT) and per protocol (PP) analyses were performed. RESULTS: A total of 91 patients were screened, and 64 were randomized (mean age70 ± 12 years). In the ITT analysis, 14 of 29 patients (48%) in the SOC group and 14 of 35 (40%) in the MP group suffered the composite endpoint (40% versus 20% in patients under 72 years and 67% versus 48% in those over 72 years; p = 0.25). In the PP analysis, patients on MP had a significantly lower risk of experiencing the composite endpoint (age-adjusted risk ratio 0.42; 95% confidence interval, CI 0.20-0.89; p = 0.043). CONCLUSION: The planned sample size was not achieved, and our results should therefore be interpreted with caution. The use of MP had no significant effect on the primary endpoint in ITT analysis; however, the PP analysis showed a beneficial effect due to MP, which consistent with other published trials support the use of glucocorticoids in severe cases of COVID-19.

BACKGROUND: First-degree relatives of people with dementia (FH+) are at increased risk of developing Alzheimer's disease (AD). Here, we investigate "estimated years to onset of dementia" (EYO) as a surrogate marker of preclinical disease progression and assess its associations with multi-modal neuroimaging biomarkers. METHODS: 89 FH+ participants in the PREVENT-Dementia study underwent longitudinal MR imaging over 2 years. EYO was calculated as the difference between the parental age of dementia diagnosis and the current age of the participant (mean EYO = 23.9 years). MPRAGE, ASL and DWI data were processed using Freesurfer, FSL-BASIL and DTI-TK. White matter lesion maps were segmented from FLAIR scans. The SPM Sandwich Estimator Toolbox was used to test for the main effects of EYO and interactions between EYO, Time, and APOE-ε4+. Threshold free cluster enhancement and family wise error rate correction (TFCE FWER) was performed on voxelwise statistical maps. RESULTS: There were no significant effects of EYO on regional grey matter atrophy or white matter hyperintensities. However, a shorter EYO was associated with lower white matter Fractional Anisotropy and elevated Mean/Radial Diffusivity, particularly in the corpus callosum (TFCEFWERp < 0.05). The influence of EYO on white matter deficits were significantly stronger compared to that of normal ageing. APOE-ε4 carriers exhibited hyperperfusion with nearer proximity to estimated onset in temporo-parietal regions. There were no interactions between EYO and time, suggesting that EYO was not associated with accelerated imaging changes in this sample. CONCLUSIONS: Amongst cognitively normal midlife adults with a family history of dementia, a shorter hypothetical proximity to dementia onset may be associated with incipient brain abnormalities, characterised by white matter disruptions and perfusion abnormalities, particularly amongst APOE-ε4 carriers. Our findings also confer biological validity to the construct of EYO as a potential stage marker of preclinical progression in the context of sporadic dementia. Further clinical follow-up of our longitudinal sample would provide critical validation of these findings.

ABSTRACT

Kraft lignin, so far useful for energy generation, has been gathering considerable attention as an alternative material to replace fossil-based resources mainly due to its high phenolic content. However, the wide molecular weight distribution and chemical composition heterogeneity led to the development of fractionation methods. Herein, to narrow such characteristics we used eucalypt kraft lignin fractionated at pH's 9, 7, 5 and 3 by sequential acid precipitation. These lignin fractions were first characterised by simultaneous pyrolysis and trimethylsilylation (SPyT) with N-Methyl-N-(trimethylsilyl) trifluoroacetamide with posterior tests of antioxidant, antibacterial, and antitumour activities. We observed higher ratio of syringyl/guaiacyl groups and increase in antioxidant activity in those fractions with lower molecular weight (precipitated at lower pH's). Fractions precipitated at pH's 9 and 7 have shown an outstanding antibacterial activity against five bacteria. Moreover, fractions 7 and 5 presented at cytotoxicity tests higher ability to inhibit the growth of U87MG and T98G glioma cells, while only a slight inhibition of adult human fibroblasts (non-tumour cells) was detected.

ABSTRACT

This work describes the production/characterization of low molar mass chitosan nanoparticles derived from waste shrimp shells (SSC), as well as from a commercial chitosan (CC). The production of low molar mass nanochitosan employed thermal shock, alternating between 100 °C and ambient temperature, followed by grinding the dry material (SSC and CC) in a ball mill, producing around 500 g of nanochitosan per batch. A highlight of the methodology employed is that it enables nanochitosan to be obtained even from a low quality commercial raw material. All particles had diameters smaller than 223 nm, with an average diameter below 25 nm (determined by DLS), while reductions of molar mass were between 8.4-fold and 13.5-fold. The depolymerization process resulted in a reduction in crystallinity of 38.1 to 25.4% and 55.6 to 25.9% in the CC and SSC samples, respectively. The production of nanochitosans was also confirmed by TEM through the observation of crystalline domains with diameters between 5 and 10 nm. This work perfectly reproduces the results on bench scale from previous research. The simple and inexpensive processes enable easy scale-up, representing an important advance in the production chain of biopolymers. Graphical abstract.

ABSTRACT

Melanized fungi and black yeasts in the family Herpotrichiellaceae (order Chaetothyriales) are important agents of human and animal infectious diseases such as chromoblastomycosis and phaeohyphomycosis. The oligotrophic nature of these fungi enables them to survive in adverse environments where common saprobes are absent. Due to their slow growth, they lose competition with common saprobes, and therefore isolation studies yielded low frequencies of clinically relevant species in environmental habitats from which humans are thought to be infected. This problem can be solved with metagenomic techniques which allow recognition of microorganisms independent from culture. The present study aimed to identify species of the family Herpotrichiellaceae that are known to occur in Brazil by the use of molecular markers to screen public environmental metagenomic datasets from Brazil available in the Sequence Read Archive (SRA). Species characterization was performed with the BLAST comparison of previously described barcodes and padlock probe sequences. A total of 18,329 sequences was collected comprising the genera Cladophialophora, Exophiala, Fonsecaea, Rhinocladiella and Veronaea, with a focus on species related to the chromoblastomycosis. The data obtained in this study demonstrated presence of these opportunists in the investigated datasets. The used techniques contribute to our understanding of environmental occurrence and epidemiology of black fungi.

Objective: To test the hypothesis that among cognitively healthy individuals, distinct groups exist in terms of amyloid and phosphorylated-tau accumulation rates; that if rapid accumulator groups exist, their membership can be predicted by Alzheimer's disease (AD) risk factors, and that time points of significant increase in AD protein accumulation will be evident. Methods: The analysis reports data from 263 individuals from the BIOCARD and 184 individuals from the Baltimore Longitudinal Study of Aging with repeated cerebrospinal fluid (CSF) and positron emission tomography (PET) sampling, respectively. We used latent class mixed-effect models to identify distinct classes of amyloid (CSF and PET) and p-Tau (CSF) accumulation rates and generalized additive modeling to investigate non-linear changes to AD biomarkers. Results: For both amyloid and p-Tau latent class models we confirmed the existence of two separate classes: accumulators and non-accumulators. The accumulator and non-accumulator groups differed significantly in terms of baseline AD protein levels and slope of change. APOE ε4 carrier status and episodic memory predicted amyloid class membership. Non-linear models revealed time points of significant increase in the rate of amyloid and p-Tau accumulation whereby APOE ε4 carrier status associated with earlier age at onset of rapid accumulation. Conclusions: The current analysis demonstrates the existence of distinct classes of amyloid and p-Tau accumulators. Predictors of class membership were identified but the overall accuracy of the models was modest, highlighting the need for additional biomarkers that are sensitive to early disease phenotypes.

In this study, a strategy was adopted to enhance the use of ramie fibers as raw material for isolation of cellulose nanofibers (CNFs). Ramie pulp was produced by alkaline organosolv followed by bleaching. CNFs were produced by mechanical defibrillation, and films were fabricated via casting. Effects of number of passes in the mechanical grinding on physical and mechanical properties of CNF films were comprehensively studied. Potential of ramie fibers was proved by fabricating homogeneous nanofibers with average thickness of 8.72 nm, which led to CNF films with dense and non-porous networks, and crystallinity index of 76-78%. Tensile strength (42-82 MPa) and dynamic mechanical (9-11 GPa) performance were good only for less severe mechanical defibrillation. Lower solubility (1.85-2.43%). and activity (0.69) in water, and outstanding barrier properties against water vapor and oxygen make ramie suitable for more sustainable extraction of cellulose nanofibers and production of CNF films for diverse applications.

ABSTRACT

The large amount of residues generated by the peach palm agroindustry and its cellulose content (34 g 100g-1) motivated the present investigation, where outer sheaths were used for nanocellulose production through ultrafine grinding. Protocols combining different chemical delignification with defibrillation intensity (10 and 20 cycles) were applied and their influence over some properties and characteristics of nanofibers evaluated. At all protocols applied it was possible produce cellulose at nanometric level (widths ˜100 nm), with high crystallinity index (49.8-54.5%) and great thermal stability. The delignification influenced the defibrillation, where lignin removal resulted in well dispersed bundles of fibrils with lower widths. Lower lignin contents combined with higher shear forces released more nanofibrils, with more adsorbed water with defibrillation extension. These behavior influenced the suspension stability, that was higher at larger number of cycles (20). The results evidenced that was possible produced FNC with appropriate technical characteristics from pupunha discards.

Se efectuó un estudio descriptivo y transversal de 40 pacientes con VIH pertenecientes al Policlínico Docente Frank País García de Santiago de Cuba, con vistas a identificar las reacciones adversas a medicamentos antirretrovirales durante el primer semestre de 2018. En la casuística, la mayoría de las notificaciones las realizaron los especialistas en Medicina General Integral (61,4 por ciento), seguidos por los de Medicina Interna y los licenciados en Enfermería (8,8 y 17,6 por ciento, respectivamente); asimismo, entre los efectos adversos predominantes figuraron las náuseas, la cefalea y los mareos, fundamentalmente con los esquemas terapéuticos constituidos por lamivudina-zidovudina-nevirapina y zidovudina-nevirapina-efavirenz. Se concluye que durante el tratamiento antirretroviral la mayoría de dichas reacciones fueron clasificadas como definitivas, independientemente del esquema terapéutico prescrito y las enfermedades concomitantes.


A descriptive and cross sectional study of 40 patients with HIV belonging to the Teaching Polyclinic Frank País García in Santiago de Cuba was carried out with the aim of identifying the adverse reactions to antirretroviral drugs during the first semester of 2018. Most of the notifications were carried out by specialists in Comprehensive General Medicine (61,4 percent) in the case material, followed by those of Internal Medicine and Licentiate in Nursing (8,8 and 17,6 percent, respectively); also, among the predominant adverse effects there were nauseas, migraine and sickness, fundamentally with the therapeutic schemes constituted by lamivudina-zidovudina-nevirapina and zidovudina-nevirapina-efavirenz. It is concluded that during antirretroviral treatment most of these reactions were classified as definitive, independently of the prescribed therapeutic scheme and the concomitant diseases.

ABSTRACT The present work provides a comparative account of the morpho-anatomy of six species of Eucalyptus, namely E. badjensis Beuzev. & Welch, E. benthamii Maiden & Cambage, E. dunnii Maiden, E. grandis W.Hill, E. globulus Labill. and E. saligna Sm., Myrtaceae. Leaf samples of these six species were investigated by light and scanning electron microscopy. The observed microscopic features that can be useful in the identification and quality control of the studied species include the morphology of epicuticular waxes, presence of prismatic crystals on the leaf surface, leaf midrib shape and arrangement of its vascular system, and the presence or absence of the sclerenchymatous fiber caps in the vascular bundle.

Analyzes of samples of stem and fiber Boehmeria nivea var. Murakami were performed to increase information on the plant and disseminate its multiple possibilities of use. The histochemical analyzes with dyes and reagents showed the presence of lignified, suberous and cutinized cell walls with Safranin in the xylem region. The presence of pectin-cellulose with Astra Blue was confirmed in the regions of fiber bundles (cortex). In all regions of the stem were identified substances such as fat and mucilage with Methylene Blue evidencing high concentrations in the regions of fiber bundles and bark (violet blue). By means of Lugol (dark brown) starches were identified in several parts of the stem with concentrations in the xylem and epidermal regions. Through analyzes by EDS it was possible to identify F, Mg, Al, Si, K and Ca with predominance of K. In the chemical analyzes of the fibers were found 71.75 of cellulose, 12.11 of hemicelluloses, 1.06 of lignin and 1.70% of ashes. The percentages of extractives soluble in hot and cold water were 5.28 and 3.12% respectively, for Ethanol-Toluene was 8.55 and NaOH (1%) 27.27%. Histochemical analyzes revealed important characteristics of the fiber and stem contributing with better knowledge of the species.


Análises de amostras de caule e de fibras Boehmeria nivea var. ‘Murakami’ foram realizadas para aumentar o conhecimento da planta e difundir suas múltiplas possibilidades de uso. As análises histoquímicas com corantes e reagentes mostraram paredes celulares lignificadas, suberosas e cutinizadas com Safranina na região do xilema. Confirmou-se a presença de pectocelulose com Azul de Astra nas regiões de feixes de fibras (córtex). Em todas as regiões do caule, foram identificadas substâncias como gordura e mucilagem com Azul de Metileno, evidenciando elevadas concentrações nas regiões dos feixes de fibras e da casca (azul violeta). Por meio de Lugol (marrom escuro), foram identificados amidos em várias partes do caule com concentrações nas regiões do xilema e da epiderme. Por meio de análises de EDS, foram identificados F, Mg, Al, Si, K e Ca, com predominância do K. Nas análises químicas das fibras, foram encontrados 71,09 de celulose, 12,11 de hemiceluloses, 1,06 de lignina e 1,70% de cinzas. As porcentagens de extrativos solúveis em água quente e fria foram de 5,28 e 3,12%, respectivamente. Para o Etanol-Tolueno, foi 8,55 e para o NaOH (1%), 27,27%. ABSTRACT

Intensive aquaculture needs to adopt techniques that are able to contribute towards sustainability. Closed systems that employ water recirculation can combine intensive production with environmental sustainability, since there is no exchange of water or discharge of effluents into the environment. In order to achieve this, effective filtration systems are required to ensure that the water quality is satisfactory for the cultivation of aquatic organisms. Chitosan, an industrial waste material derived from crustacean farming, is a renewable natural material that is biodegradable and possesses adsorbent characteristics. In this work, chitosan foam was incorporated in filters and was evaluated as an adsorbent of aquaculture pollutants, adding value to the material and at the same time providing a use for industrial waste. The foam was characterized by scanning electron microscopy and energy dispersive spectroscopy, apparent density, and water absorption capacity. It was used to remove ammonia, nitrite, orthophosphate, and turbidity from aquaculture effluents. The foam consisted of a bilayer with smooth and porous sides, which presented low density, flexibility, and high water absorption capacity. The best proportion of the foam, in terms of the mass of foam per volume of solution (% m v-1), was 0.10, which resulted in removal of 32.8, 57.2, 89.5, and 99.9% of ammonia, nitrite, orthophosphate, and turbidity, respectively. This biopolymer produced is biodegradable, and when saturated with organic compounds from aquaculture, and no longer suitable for reuse as a filter material, it can be employed as a fertilizer, hence closing the sustainability cycle of the aquaculture production chain.
