ABSTRACT

We studied the effect of histocompatibility in married couples on the development and severity of gestosis. It was found that gestosis more often develops in women with two common HLA alleles of class II major histocompatibility complex with her husband. The greatest number of coincidences was detected in subgroups with medium and severe gestosis. Perinatal outcomes in women with medium-severe gestosis having HLA-alleles common with husbands are characterized by delivery of babies with lower body weight.

ABSTRACT

Nitric oxide production in organs of pregnant mice (liver, kidney, placenta) was measured using specifical NO traps, Fe-dithiocarbamate complexes (Fe-DETC) forming with NO paramagnetic mononitrosyl iron complexes with DETC (MNIC-DETC), detected by EPR method (-196 degrees C). The amount of NO formed increased in dependence of the pregnancy period (from the first to the third trimesters). Addition of NO-synthase substrate, L-arginine, insignificantly influenced endogenous NO production. However, endogenous NO amount sharply decreased at experimental acute renal failure.

ABSTRACT

Malonic dyaldehyde (MDA) and transferrin (TF) content and total antioxidant activity (AOA) were measured in blood of pregnant women with normal pregnancy (27 women) and preeclamptic pregnancy (46 women) at different pregnancy stages (17-28, 29-34 and 35-40 weeks of pregnancy). It was shown that in normal pregnancy AOA increased from 24 weeks of pregnancy with a maximum at 30 weeks, while MDA level did not change significantly through all time of pregnancy. On the contrary, in preeclamptic pregnancy blood AOA did not change significantly, while MDA content increased at the 26-34 weeks of pregnancy with a maximum at 30-34 weeks of pregnancy. Blood TF content in normal pregnancy increased during pregnancy development with a maximum at 30-34 weeks, while in preeclamptic pregnancy TF changed only a little. These data show the important role of TF in the AOA blood status formation in normal pregnancy, and great pathogenetic role of the absence of such an increase of TF at preeclamptic pregnancy in AOA impairment at this condition. Eiconol (omega-3 polyunsaturated fatty acid concentrate from north fish oil) intake by preeclamptic pregnant women drastically decreased blood MDA level (1,5-2 times compared to the women that did not receive eiconol), especially at 29-34 week of gestation. Blood AOA, that was decreased at preeclampsia increased under influence of eiconol intake at all pregnancy stages studied. Eiconol intake greatly increased also the TF blood content that was decreased at preeclampsia. It is supposed that eiconol ability to increase AOA may be explained by its capacity to increase the level of TF--one of the most important compounds of serum blood antioxidant system.

ABSTRACT

Serum levels/affinities of natural autoantibodies to proteins S100, GFAP and MP65 in patients with physiological pregnancy are usually constant and may vary among individuals within relatively narrow range. In patients with early toxicosis in pregnancy dispersion of serum autoantibodies levels were often found beyond the normal range. This group includes cases with significantly elevated as well as abnormally decreased immunoreactivity parameters. Percent of newborns with different deviations from women with abnormal serum levels/affinities of autoantibodies to proteins S100, GFAP and MP65 was higher 1,5-2 fold compared with population.

ABSTRACT

The levels of progesterone and estrogen receptors, cAMP and prostaglandins E2 and F2 alpha were studied in the decidua of pregnant women, which was obtained during surgical, spontaneous or prostaglandin-induced abortion or by pregnancy termination with vacuum aspiration after RU-486 treatment (before the onset of abortion). Our data have allowed the authors to suggest that (i) for the maintenance of normal activity of decidua during pregnancy, the following points are considerably important: the constant ratio of progesterone and estrogen receptors in favor of progesterone receptors, low PG levels and high cAMP levels; (ii) both spontaneous and induced abortion are characterized by decreases in the levels of progesterone receptor (which in turn changes the ratio of progesterone and estrogen receptors in favor of the latter), by increased PG levels and decreasing cAMP concentrations.

ABSTRACT

Clinical and experimental studies have revealed that salbupart, a beta-mimetic agent, administered in a dose of 0.14 mg/kg (the maximal therapeutic dose) markedly inhibited the spontaneous and oxytocin-induced uterine contractility, caused no embryotoxic or teratogenic effects, and did not influence fetal growth and development. The efficacy of salbupart tocolytic therapy was estimated as 75%. Blood plasma cAMP levels normalized in the pregnant women over the course of therapy. Clinical and experimental results recommend salbupart for pregnancy protecting therapy in cases with threatened spontaneous abortions.

ABSTRACT

Measurements of prostaglandins E2 and F2 alpha in the endometrium of 35 women of a reproductive age, suffering from habitual abortions in late pregnancy terms, have shown elevation of these prostaglandin levels in the secretory phase as against the reference group and their normal values during the proliferative phase. It is possible, that disordered prostaglandin synthesis in the endometrium, manifesting in pregnancy, is one of the causes of habitual abortions. cAMP levels were somewhat decreased in the secretory phase in this patient population; this permits a hypothesis on disorders in the adenylate cyclase system.

ABSTRACT

Estrogen and progesterone reception was examined in the endometrial cytoplasmic and nuclear fraction from apparently healthy subjects and patients with habitual late incompetent pregnancy in the early proliferative and late secretory periods of a menstrual cycle. The reception of the two reproductive hormones in the nuclei and the binding of estrogens in the endometrial cytosol were demonstrated to be significantly higher in the female patients than in healthy subjects. The cytosol estrogen to progesterone reception ratio was increased in the patients in the secretory phase of a menstrual cycle than in normal females. There was a complete normalization of endometrial reproductive hormone receptor levels in the patients undergoing hyperbaric oxygenation.

ABSTRACT

A study was made of estrogen and progesterone reception of the endometrial cytoplasmatic and nuclear fractions of healthy women and women suffering from late habitual abortion in the early proliferative and late secretory phases of the menstrual cycle. Reception of both sex hormones in the nuclei and estrogen binding in the endometrial cytosol of women with late habitual abortion was significantly higher than that in healthy women. The estrogen receptors/progesterone receptors ratio in the cytosol in the pathological endometrium at the secretory phase of the menstrual cycle was higher than in the normal one. Hyperbaric therapy given to such patients resulted in complete normalization of endometrial sex hormones reception.

ABSTRACT

Comparative trials of two tocolytic preparations, salbupart (Polfa) and partusisten, showed their high efficacy (75 and 80 per cent, respectively). The success of management depends on the outcome of the preceding pregnancies, the course of current pregnancy in the 1st and 2nd trimesters, the cervical status, and extent of therapy. Normalization of cAMP levels concurrent with reduced force of uterine contractions and elimination of clinical signs of threatening pregnancy termination are also indicative of benefits of tocolytic therapy.

ABSTRACT

The content of prostaglandins and cAMP was determined in the endometrium of healthy women with a regular menstrual cycle in its different phases. It was shown that the PGE2 level in the proliferative (the 7th-10th day of the cycle) and secretory (the 24th-26th day of the cycle) phases was the same. At the same time the level of PGE2 in the secretory phase of the cycle was 3 times higher than in the proliferative phase. The level of cAMP in the secretory phase of the cycle was 2 times higher than in the proliferative phase.

ABSTRACT

Plasma estrogen, progesterone, cortisol and prolactin concentrations were determined by RIA in 20 healthy women on the 7th-10th and 24th-26th days of the menstrual cycle. The content of estrogen and progesterone receptors in the cytosol and nuclei of the endometrium was studied by a method of adsorption on dextran-covered carbon; pathomorphological investigation of the endometrium was performed. Sex hormone concentration was shown to be significantly lower in the middle (proliferative) phase as compared to the late one (secretory). Cortisol and prolactin concentrations in both periods did not change. The content of both sex hormone receptors was significantly higher in the middle (proliferative) phase. On the basis of the content and ratio of plasma sex hormones and receptor content in the endometrium a conclusion was made that the main factor in the regulation of receptor levels by sex hormones was the ratio of their concentrations in the plasma.

ABSTRACT

PIP: The effects of methyl ester of 15-methyl-prostaglandin F2alpha (15-Me-PGFalpha) on the content in plasma of female peripheral blood of 15-keto-13, 14-dihydro-PGF2alpha--the basic metabolite of PGF2alpha--was investigated. The studies were performed in 46 women who received vaginal suppositories of 1 mg 15-Me-PGF2alpha or suppositories with placebo in order to dilate the uterine cervix before abortion in the 1st trimester of pregnancy. It was found that 6-12 hours after administration of suppositories with 15-Me-PGF2alpha, the PGF2alpha metabolite level in the blood increases 1 1/2-2 fold. The concentration of PGF2alpha metabolite in the blood is neither altered in the earlier periods or after administration of suppositories with placebo. (author's)^ieng
