ABSTRACT

The number of patients diagnosed with Alzheimer's disease (AD) increases each year, and there are currently few treatment strategies to decrease the symptoms of AD; furthermore, these strategies are not sufficient to reduce memory loss in AD patients. In this work, in vitro and in silico studies were performed to evaluate the effects of fucosterol, which was extracted from an algal source and characterized by liquid chromatography-mass spectra (LC-MS), as an inhibitor of Aß1-42 aggregation. Experimental studies, including protein gel electrophoresis, atomic force microscopy and fluorescence studies with thioflavin T (ThT), highlighted that fucosterol can decrease oligomer formation more than galantamine, which was used as a positive control. Docking and molecular dynamics simulations coupled with an MMGBSA approach showed that fucosterol is capable of recognizing the hydrophobic regions of monomeric Aß1-42, suggesting that fucosterol could affect amyloid-beta (Aß1-42) aggregation by preventing the formation of oligomers, preventing the development of AD.Communicated by Ramaswamy H. ABSTRACT

Enterotoxigenic Escherichia coli (ETEC) is an important diarrhea-causing pathogen for humans. Heat-stable enterotoxin (ST) plays a crucial role in triggering diarrhea and ETEC pathogenesis. However, ST is a small peptide that lacks immunogenic activity itself but becomes immunogenic when it is coupled to a carrier molecule. In this study, the ß-glucans (BG) from yeasts have been used to test their immunomodulatory activity and adjuvant effect on the properties of ST. This study aimed to synthesize and characterize a conjugate of yeast-derived ß-glucan with the ST enterotoxin (BG-ST) and evaluate the antigenic and antioxidant activities in mouse splenocytes. Fourier transform infrared spectroscopy and scanning electron microscopy analysis showed new bands and changes in morphology, respectively, confirming ST was successfully coupled to beta glucan. Additionally, according to the enzyme-linked immunosorbent assay (ELISA), conjugation efficiency was almost 90%. Cellular viability, phagocytic cell proportion, and respiratory burst enhanced splenocytes stimulated by BG-ST. In addition, nitric oxide production and antioxidant enzymes increased in cells stimulated with BG-ST, BG and ST. In conclusion, the results revealed the successful conjugation of ß-glucan with ST peptide enhancing immune and antioxidant parameters to a greater extent than their individual components.

ABSTRACT

The Baja California Peninsula in México has about 670 species of macroalgae along its coast. Species richness increases the probability of finding native macroalgae with potential as sources of bioactive compounds suitable for health, pharmacological, and cosmetic ingredients. To understand the biotechnological value of macroalgae from the peninsula, ethanol extracts from 17 macroalgae (four Chlorophyta, six Rhodophyta, seven Ochrophyta) were screened for antioxidant potential. To determine the antioxidant capacity of macroalgal extracts, 2,2-diphenyl-1-picrylhydrazyl (DPPH), ferric reducing antioxidant power, and nitric oxide radical scavenging as well as total phenolic content (TPC) were measured. Extracts of the brown macroalgae were most active. Among these, Eisenia arborea, Padina concrecens, and Cystoseira osmundacea had the highest TPC and exhibited the strongest radical scavenging activities. Correlations were found between TPC macroalgal and scavenging capacity, indicating an important role of polyphenols as antioxidants. This suggests that some brown macroalgae from Baja California Peninsula may be a good source of natural bioactive compounds.
