ABSTRACT

Abstract Seven hundred and twenty four fish specimens were captured from March to September 2016. The materials used in the current study were cast nets, hand nets. Eight cyprinid fish species were studied for their length-weight relationships. Parameter b in the LWR was 3.03, 3.06, 3.02, 2.29, 2.82, 3.43, 2.73 and 2.47 for Schizothorax plagiostomus, Schizothorax esocinus, Racoma labiata, Tor putitora, Barilius vagra, Garra gotyla, Puntius ticto and Arassius auratus respectively. Current study is the first attempt on the LWRs of cyprinid species, provide a baseline approach for conservation and /management of local fish fauna of economic importance.


Resumo Um total de 724 espécimes de peixes foi capturado de março a setembro de 2016. Os materiais usados ​​no presente estudo foram redes de lançamento, redes de mão. Oito espécies de peixes ciprinídeos foram estudadas quanto às suas relações peso/comprimento. O parâmetro b no LWR foi 3,03, 3,06, 3,02, 2,29, 2,82, 3,43, 2,73 e 2,47 para Schizothorax plagiostomus, Schizothorax esocinus, Racoma labiata, Tor putitora, Barilius vagra, Garra gotyla, Puntius ticto e Arassius auratus respectivamente. ABSTRACT

Garlic (Allium sativum) is an important cash food crop, and the biotechnology industry has considerable interest in the plant because of its medicinal importance. These medicinal properties are attributed to organosulphur compounds as the accumulation of these compounds varies according to genotype, locality, light quality, and cultivation practices. In this study, we compared a newly developed garlic variety NARC-G1 by National Agricultural Research Centre (NARC), Islamabad, Pakistan with three different garlic cultivars and highlighted the distinctive attributes like phenotypic characteristics, the content of allicin, elemental profile, and gene polymorphism. Phenotypic analysis showed NARC-G1 has significantly higher bulb weight (66.36g ± 18.58), single clove weight (5.87g ± 1.041), and clove width (17.41mm ± 0.95) which directly correlates to the size of the garlic. The analytical analysis showed the highest allicin content (4.82 ± 0.001) in NARC-G1. Genotyping of the alliinase in all four cultivars showed indels in the gene resulting in distinguishable changes in organosulphur compounds' profile. NARC-G1 is unique from other garlic cultivars and could be the best choice for mass production with proper cultivation and irrigation management. Moreover, for Pakistan NARC-G1 could be a potential contender to earn the industrial benefits with inland cultivation instead of importing garlic alleviating the economic burden.

ABSTRACT

Salmo trutta fario is the most important fish species of family Salmonidae inhibited in cool waters all over the world including the nortern parts of Pakistan. This study was aimed to find out the prey selection and feeding habits of the species. One hundred and eighty five Salmo trutta fario were captured from March 2016 to April 2017. Feeding intensity as represented by a fullness index (FI), changing with various length groups and season. Out of 185 Brown trout 24 fish stomaches were found empty. The most important and common food items of brown trout were Brachycentridae, Blepharocera, Hydropsychidae, ephemerella spp. Kruskal Wallis H-test were applied on feeding intensity groups consisting of three month group. The test statistic for K.W-H-test were (H=8.13 with df =3) had a p-value of 0.043 < (alpha=0.05 indicates favor of the alternative hypothesis of at least one difference among the feeding intensity groups. The linear relationship of N with index of relative importance and %IRI denoted by least square regression line (N = 35.2 + 22.1%IRI), shows that for 22 prey there is 1% change in IRI. The relationship between total length (mm) and fish body weight (gm) is expressed by Pearson correlation coefficient (r = 0.976), showed that total length (mm) and fish body weight (gm.) is highly correlated. Descriptive statistics are used for the stomach fullness, which shows that feeding intensity was recorded higher from March to May. A total of 2289 preys was recorded including the most common were; Trichoptera, Hydropsychidae, Brachycentridae, diptera, blepharocera, ephemerala, chironomida, honey bees, grass hopper, locust, trout egg, trochanter, plant tissue, stones were retrieved from the gut contents of brown trout. According to index of relative importance IRI% four preys represents major components of the diet. The highest IRI% was recorded in Brachycentridae (39.38%), followed by blepharocera (13.23%), Hydropsychidae (10.76%) and ephemerella spp (8.28%). The relationship between IRI and FO is (r =0.556) is moderately correlated with coefficient of determination (r2=0.31). This study will help in the development of an artificial diet for the species for better growth performance in captivity.

Este estudo teve como objetivo conhecer a seleção de presas e os hábitos alimentares da espécie. Cento e oitenta e cinco Salmo trutta fario foram capturados de março de 2016 a abril de 2017. Intensidade de alimentação representada por um índice de plenitude (FI), mudando com vários grupos de comprimento e estação. De 185 trutas, 24 estômagos de peixes foram encontrados vazios. Os itens alimentares mais importantes e comuns da truta marrom foram brachycentridae, blepharocera, hydropsychidae, ephemerellaspp. O teste H de Kruskal Wallis foi aplicado em grupos de intensidade de alimentação consistindo de um grupo de três meses. A estatística de teste para o teste KW-H foi (H = 8,13 com df = 3) teve um valor de p de 0,043 <(alfa = 0,05 indica favor da hipótese alternativa de pelo menos uma diferença entre os grupos de intensidade de alimentação. relação de N com o índice de importância relativa e% IRI denotado pela linha de regressão do mínimo quadrado (N = 35,2 + 22,1% IRI), mostra que para 22 presas há 1% de mudança no IRI. ABSTRACT

This research was aimed to explore the helminth parasitic diseases in Schizothorax plagiostomus (the snow trout) from river Swat and river Panjkora, Pakistan. Collection of 360 fish specimens have done from the lower, middle and upper reaches of both the rivers through gill nets, cast nets, dragon nets and hooks. All the samples were examined in the University of Malakand, Zoology Department for helminth parasites during the months from January 2015 to December 2016. Of the total examined fish samples 21.9% (n=79) were infected with Rhabdochona spp including 17.7% (n=32/180) in river Swat and 26.6% (n=47/180) in river Panjkora. Highest month-wise prevalence (p=0.09878,p<0.05) was reported in May (30%), then by February and October each (26.6%) while the lowest during August (13.3%). Highest prevalence (p=0.9723, p<0.05) was reported in summer season (26.6%), while lowest in the winter season (20%). Adults were highly infected (p=< 0.0001) in prevalence (63.7%) followed by sub-adults (13.2%) while no infection was found in juvenile specimens. Females fish samples had higher (p=0.0277, p>0.05) prevalence (28.8%) than males (16.6%). Fishes of the lower reaches had highest (p=0.0029, P>0.05) prevalence (31.7%) followed by middle reaches (16.5%) while the lowest prevalence was observed in samples of fish collected from upper reaches (9.87%). Present study address that Rhabdochona spp in the intestine of snow trout has a long term relationship and call as a natural infection in cyprinids and zoonotic risk to human.


Esta pesquisa teve como objetivo explorar as doenças parasitárias por helmintos em Schizothorax plagiostomus (a truta das neves) do rio Swat e do rio Panjkora, Paquistão. A coleta de 360 espécimes de peixes foi feita nos trechos inferior, médio e superior de ambos os rios por meio de redes de emalhar, de lançamento, de dragão e anzóis. Todas as amostras foram examinadas na Universidade de Malakand, Departamento de Zoologia, para helmintos parasitas durante os meses de janeiro de 2015 a dezembro de 2016. Do total de amostras de peixes examinadas, 21,9% (n = 79) estavam infectados com Rhabdochona spp, incluindo 17,7% (n = 32/180) no rio Swat e 26,6% (n = 47/180) no rio Panjkora. A maior prevalência no mês (p = 0,09878, p <0,05) foi relatada em maio (30%), depois em fevereiro e outubro cada (26,6%), enquanto a menor em agosto (13,3%). A maior prevalência (p = 0,9723, p <0,05) foi relatada no verão (26,6%), enquanto a menor no inverno (20%). Os adultos foram altamente infectados (p = <0,0001) na prevalência (63,7%), seguidos por subadultos (13,2%), enquanto nenhuma infecção foi encontrada em espécimes juvenis. As amostras de peixes do sexo feminino tiveram maior (p = 0,0277, p> 0,05) prevalência (28,8%) do que o masculino (16,6%). Os peixes do curso inferior tiveram maior (p = 0,0029, P> 0,05) prevalência (31,7%) seguida do curso médio (16,5%), enquanto a menor prevalência foi observada em amostras de peixes coletados do curso superior (9,87%). O presente estudo aborda que Rhabdochona spp no intestino da truta das neves tem uma relação de longo prazo e pode ser considerada uma infecção natural em ciprinídeos e risco zoonótico para humanos.

O maior IRI% foi registrado em brachycentridae (39,38%), seguido por blepharocera (13,23%), hydropsychidae (10,76%) e ephemerellaspp (8,28%). A relação entre IRI e FO é (r = 0,556) está moderadamente correlacionada com o coeficiente de determinação (r2 = 0,31). Este estudo auxiliará no desenvolvimento de uma dieta artificial para a espécie para melhor desempenho de crescimento em cativeiro.

Highest prevalence (p=0.9723, p 0.05) was reported in summer season (26.6%), while lowest in the winter season (20%). Adults were highly infected (p= 0.0001) in prevalence (63.7%) followed by sub-adults (13.2%) while no infection was found in juvenile specimens. Females fish samples had higher (p=0.0277, p>0.05) prevalence (28.8%) than males (16.6%). Fishes of the lower reaches had highest (p=0.0029, P>0.05) prevalence (31.7%) followed by middle reaches (16.5%) while the lowest prevalence was observed in samples of fish collected from upper reaches (9.87%). Resumo Esta pesquisa teve como objetivo explorar as doenças parasitárias por helmintos em Schizothorax plagiostomus (a truta das neves) do rio Swat e do rio Panjkora, Paquistão. A coleta de 360 espécimes de peixes foi feita nos trechos inferior, médio e superior de ambos os rios por meio de redes de emalhar, de lançamento, de dragão e anzóis. Todas as amostras foram examinadas na Universidade de Malakand, Departamento de Zoologia, para helmintos parasitas durante os meses de janeiro de 2015 a dezembro de 2016. Do total de amostras de peixes examinadas, 21,9% (n = 79) estavam infectados com Rhabdochona spp, incluindo 17,7% (n = 32/180) no rio Swat e 26,6% (n = 47/180) no rio Panjkora. A maior prevalência no mês (p = 0,09878, p 0,05) foi relatada em maio (30%), depois em fevereiro e outubro cada (26,6%), enquanto a menor em agosto (13,3%). A maior prevalência (p = 0,9723, p 0,05) foi relatada no verão (26,6%), enquanto a menor no inverno (20%). Os adultos foram altamente infectados (p = 0,0001) na prevalência (63,7%), seguidos por subadultos (13,2%), enquanto nenhuma infecção foi encontrada em espécimes juvenis. As amostras de peixes do sexo feminino tiveram maior (p = 0,0277, p> 0,05) prevalência (28,8%) do que o masculino (16,6%). Os peixes do curso inferior tiveram maior (p = 0,0029, P> 0,05) prevalência (31,7%) seguida do curso médio (16,5%), enquanto a menor prevalência foi observada em amostras de peixes coletados do curso superior (9,87%). O presente estudo aborda que Rhabdochona spp no intestino da truta das neves tem uma relação de longo prazo e pode ser considerada uma infecção natural em ciprinídeos e risco zoonótico para humanos.

ABSTRACT

Seven hundred and twenty four fish specimens were captured from March to September 2016. The materials used in the current study were cast nets, hand nets. Eight cyprinid fish species were studied for their length-weight relationships. Parameter b in the LWR was 3.03, 3.06, 3.02, 2.29, 2.82, 3.43, 2.73 and 2.47 for Schizothorax plagiostomus, Schizothorax esocinus, Racoma labiata, Tor putitora, Barilius vagra, Garra gotyla, Puntius ticto and Arassius auratus respectively. Current study is the first attempt on the LWRs of cyprinid species, provide a baseline approach for conservation and /management of local fish fauna of economic importance.

ABSTRACT

Strains of Lactobacillus acidophilus WFA1 (KU877440), WFA2 (KU877441), and WFA3 (KU877442) were isolated from indigenous Dahi (yogurt), screened, and selected based on acid and bile tolerance along with the antimicrobial activity. These selected strains were further assessed for their probiotic and functional attributes. Results for simulated gastric and intestinal tolerance/ resistance revealed that all three strains can resist and survive under the following mentioned conditions. To access cell surface hydrophobicity, bacterial adhesion to hydrocarbons (BATH), cellular auto-aggregation, and salt aggregation were performed. In BATH, adhesion of strains against three hydrocarbons namely xylene, dichloromethane, and hexadecane was conducted. The results show that strains showed the least adhesion to xylene (54.25%) as compared to dichloromethane (55.25%) and hexadecane (56.65%). WFA1 showed maximum adherence percentage (55.48%) followed WFA2 (55.48%) and WFA3 (51.38%). Cellular auto-aggregation varied from 21.72% to 30.73% for WFA3 and WFA1, respectively. In the salt aggregation test (SAT), WFA1, WFA2, and WFA3 aggregated at 0.6, 1.0, and 2.0 molar concentrations of ammonium sulfate, respectively. PCR amplification of bile salt hydrolase gene (bsh) was performed and sequences were submitted to the public database of NCBI and Gene bank under accession numbers, KY689139, KY689140, and KY689141. Additionally, a cholesterol-lowering assay was conducted and up to 26% reduction in cholesterol was observed by the strains. Regarding functional properties, exopolysaccharide (EPS) production, and antioxidant potential, strain WFA1 showed promising results EPS (1.027mg/ml), DPPH (80.66%), ABTS (81.97%), and reducing power (1.787). It can be concluded from the present study that the mentioned strains of L. acidophilus (WFA1, WFA2, and WFA3) are strongly hydrophobic; thus having an ability to survive and colonize under the gastrointestinal tract which confirms their probiotic nature. Regarding their functional properties, L. acidophilus WFA1 (KU877440) showed excellent properties of antioxidants and EPS production.

ABSTRACT

A prolific three-breed (Malpura, Patanwadi, and Garole) cross Avishaan sheep has been developed in the semi-arid zone to improve farmer's income. Nutritional scarcity is a major limitation in animal husbandry during the dearth period of semi-arid tropics. Therefore, before the inaugural launch of the breed into the field, a study was designed to evaluate the effect of nutritional stress on physiological parameters and seminal attributes of native-crossbred rams in semi-arid tropics. Thus, 16 native adapted (Malpura) and 16 native-crossbred rams were equally distributed into four groups, namely, native control (G1), native nutritional stress (G2), native-crossbred control (G3), and native-crossbred nutritional stress (G4). Both the control groups (G1 and G3) were kept on their maintenance requirement as per their body weight, whereas the nutritional stress groups (G2 and G4) were provided 30% less than their maintenance requirement. The body weight of G4 decline (P<0.05) as compared to their initial weight. The plasma glucose level of G2 and G4 reduced (P<0.05) in comparison with G1 and G3, respectively. The total motile sperm percentage, rapid motile sperm percentage, and sperm viability decrease significantly (P<0.05) within the acceptable limit in native-crossbred rams (G4) under nutritional scarcity. However, the similar blood biochemical along with acceptable seminal attributes of all the rams reflected that native-crossbred rams can cope with the nutritional scarcity in semi-arid tropics and have the potential to contribute to the sustainable small ruminant production system for livelihood security in this region.

ABSTRACT

Germin and germin-like proteins (GLPs) are a broad family of extracellular glycoproteins ubiquitously distributed in plants. Overexpression of Oryza sativa root germin like protein 1 (OsRGLP1) enhances superoxide dismutase (SOD) activity in transgenic plants. Here, we report bioinformatic analysis and heterologous expression of OsRGLP1 to study the role of glycosylation on OsRGLP1 protein stability and activity. Sequence analysis of OsRGLP1 homologs identified diverse N-glycosylation sequons, one of which was highly conserved. We therefore expressed OsRGLP1 in glycosylation-competent Saccharomyces cerevisiae as a Maltose Binding Protein (MBP) fusion. Mass spectrometry analysis of purified OsRGLP1 showed it was expressed by S. cerevisiae in both N-glycosylated and unmodified forms. Glycoprotein thermal profiling showed little difference in the thermal stability of the glycosylated and unmodified protein forms. Circular Dichroism spectroscopy of MBP-OsRGLP1 and a N-Q glycosylation-deficient variant showed that both glycosylated and unmodified MBP-OsRGLP1 had similar secondary structure, and both forms had equivalent SOD activity. Together, we concluded that glycosylation was not critical for OsRGLP1 protein stability or activity, and it could therefore likely be produced in Escherichia coli without glycosylation. Indeed, we found that OsRGLP1 could be efficiently expressed and purified from K12 shuffle E. coli with a specific activity of 1251 ± 70 Units/mg. In conclusion, we find that some highly conserved N-glycosylation sites are not necessarily required for protein stability or activity, and describe a suitable method for production of OsRGLP1 which paves the way for further characterization and use of this protein.

ABSTRACT

Salmo trutta fario is the most important fish species of family Salmonidae inhibited in cool waters all over the world including the nortern parts of Pakistan. This study was aimed to find out the prey selection and feeding habits of the species. One hundred and eighty five Salmo trutta fario were captured from March 2016 to April 2017. Feeding intensity as represented by a fullness index (FI), changing with various length groups and season. Out of 185 Brown trout 24 fish stomaches were found empty. The most important and common food items of brown trout were Brachycentridae, Blepharocera, Hydropsychidae, ephemerella spp. Kruskal Wallis H-test were applied on feeding intensity groups consisting of three month group. The test statistic for K.W-H-test were (H=8.13 with df =3) had a p-value of 0.043 < (alpha=0.05 indicates favor of the alternative hypothesis of at least one difference among the feeding intensity groups. The linear relationship of N with index of relative importance and %IRI denoted by least square regression line (N = 35.2 + 22.1%IRI), shows that for 22 prey there is 1% change in IRI. The relationship between total length (mm) and fish body weight (gm) is expressed by Pearson correlation coefficient (r = 0.976), showed that total length (mm) and fish body weight (gm.) is highly correlated. Descriptive statistics are used for the stomach fullness, which shows that feeding intensity was recorded higher from March to May. A total of 2289 preys was recorded including the most common were; Trichoptera, Hydropsychidae, Brachycentridae, diptera, blepharocera, ephemerala, chironomida, honey bees, grass hopper, locust, trout egg, trochanter, plant tissue, stones were retrieved from the gut contents of brown trout. According to index of relative importance IRI% four preys represents major components of the diet. The highest IRI% was recorded in Brachycentridae (39.38%), followed by blepharocera (13.23%), Hydropsychidae (10.76%) and ephemerella spp (8.28%). The relationship between IRI and FO is (r =0.556) is moderately correlated with coefficient of determination (r2=0.31). This study will help in the development of an artificial diet for the species for better growth performance in captivity.

ABSTRACT

Mechanobiology has underpinned many scientific advances in understanding how biophysical and biomechanical cues regulate cell behavior by identifying mechanosensitive proteins and specific signaling pathways within the cell that govern the production of proteins necessary for cell-based tissue regeneration. It is now evident that biophysical and biomechanical stimuli are as crucial for regulating stem cell behavior as biochemical stimuli. Despite this, the influence of the biophysical and biomechanical environment presented by biomaterials is less widely accounted for in stem cell-based tissue regeneration studies. This Review focuses on key studies in the field of stem cell mechanobiology, which have uncovered how matrix properties of biomaterial substrates and 3D scaffolds regulate stem cell migration, self-renewal, proliferation and differentiation, and activation of specific biological responses. First, we provide a primer of stem cell biology and mechanobiology in isolation. This is followed by a critical review of key experimental and computational studies, which have unveiled critical information regarding the importance of the biophysical and biomechanical cues for stem cell biology. This review aims to provide an informed understanding of the intrinsic role that physical and mechanical stimulation play in regulating stem cell behavior so that researchers may design strategies that recapitulate the critical cues and develop effective regenerative medicine approaches.

ABSTRACT

Surface functionalization of nanoparticles (NPs) for therapeutic siRNA delivery into cancer cells has gained interest. The present study was designed for surface functionalization of gold nanoparticles (AuNPs) for efficient siRNA delivery and knockdown in cancer cells. In order to achieve this objective, AuNPs were coated with HER2-siRNA in the presence of 11-mercaptoundecanoic acid (11-MUA), calcium chloride (CaCl2) and polyethyleneimine (PEI) in alternate charge bearing successive layers. MCF-7 cells were cultured and transfected with fabricated assembly of AuNPs. Cytotoxicity analysis revealed that the half inhibitory concentration (IC50) for the formulation was 45.35 nM  . Total RNA was isolated from transfected cells, reverse transcribed into complementary DNA (cDNA) and real-time polymerase chain reaction (RT-PCR) was performed. The RT-PCR based delta-delta Ct analysis in treated cells revealed a significant 18.94 times decrease (p<0.001) in the expression of HER2 gene standardized with ACTB housekeeping gene as compared to untreated cells, which makes this formulation a potent approach for siRNA delivery and  gene knockout.

Medicinal plants are long been used for pharmaceutical and cosmetic industry. Among medicinal plants, Polygonum amplexicaule of family polygonaceae has traditional use in medicines and skin care. P. amplexicaule belongs to genus Polygonum that contains several important phytochemicals and considered as a rich source of antioxidants. The present study was designed to formulate herbal gel containing P. amplexicaule extract and evaluate its different physical properties as well as antioxidants and antityrosinase activities. Chitosan gel base was used as gelling agent and different gel formulations were prepared by different concentrations of extracts and polymers. Physical properties like pH, colour, odour, appearance and homogeneity, spreadability, extrudability and stability were optimized and analysed. A stable gel formulation containing 1% chitosan gel base and 5% plant extract was prepared that showed good appearance and homogeneity, easily spread ability and excellent extrudability. This gel formulation was tested for antioxidant and skin whitening properties by DPPH free radical scavenging assay and tyrosinase inhibition assay respectively and ascorbic acid was used as reference standard. DPPH scavenging activity with an IC50 value of 0.446 mg/mL and tyrosinase inhibition activity with an IC50 value of 0.805 mg/mL was observed and results indicated that this herbal gel formulation has a good potential for cosmetic use.

Sheep reared in hot semi-arid environments are generally exposed to heat and nutritional stress in some seasons of the year, which affects both production and reproduction. To assess the effect of high ambient temperature and feed scarcity on superovulation, 16 adult Malpura ewes were randomly divided into two groups of 8 animals each. G1 (control) was kept under a shed and offered a maintenance diet, and G2 (combined stress) was subjected to both nutritional (30% less of maintenance diet) and heat (38-44 °C for 6 h/day) stress. Ewes were superovulated without estrus synchronization by a combination of single injection of 200 IU eCG and 8 injections of FSH (Folltropin-V) at 12-h intervals in tapering doses of 5 mg/kg body weight, starting from the day 7 of natural estrus. eCG was given with the first injection and PGF2α (10 mg) was given with the second last FSH injection. G2 increased respiration rate and rectal temperature (P < 0.01), and blood urea level (P < 0.05), whereas it decreased average daily gain, plasma T4 concentration (P < 0.01) and body weight (P < 0.05). Plasma estradiol level was lower (P < 0.05) in G2 ewes as compared to control (G1) ewes. However, the number of ewes showed a superovulatory response (88 vs 66% ewes ≥ 3 corpus luteum), ovulation rate (8.75 vs 5.88) and embryo production (5.5 vs 3.9) decreased, and the number of large follicles (anovulation) increased (1.0 vs 2.14) in G2 ewes. G2 had a comparable effect on the superovulatory response compared to control ewes although physiological changes occurred as an adaptive mechanism to stress. Therefore, the well-adapted cyclic sheep of the semi-arid region may be used for superovulation despite the stressful condition of heat exposure and nutritional insufficiency.
