ABSTRACT

T-cell intracellular antigen 1 (TIA1) and TIA1-like/related protein (TIAL1/TIAR) are two members of the classical family of RNA binding proteins. Through their selective interactions with distinct RNAs and proteins, these multifunctional regulators are involved in chromatin remodeling, RNA splicing and processing and translation regulation, linking them to a wide range of diseases including neuronal disorders, cancer and other pathologies. From their discovery to the present day, many studies have focused on the behavior of these proteins in order to understand their impact on molecular and cellular processes and to understand their relationship to human pathologies. The volume of research on these proteins in various fields, including molecular biology, biochemistry, cell biology, immunology and cancer, has steadily increased, indicating a growing interest in these gene expression regulators among researchers. This information can be used to know the most productive institutions working in the field, understand the focus of research, identify key areas of involvement, delve deeper into their relationship and impact on different diseases, and to establish the level of study associated with them.

ABSTRACT

Deregulated intracellular Ca2+ and protein homeostasis underlie synaptic dysfunction and are common features in neurodegenerative diseases. DREAM, also known as calsenilin or KChIP-3, is a multifunctional Ca2+ binding protein of the neuronal calcium sensor superfamily with specific functions through protein-DNA and protein-protein interactions. Small-molecules able to bind DREAM, like the anti-diabetic drug repaglinide, disrupt some of the interactions with other proteins and modulate DREAM activity on Kv4 channels or on the processing of activating transcription factor 6 (ATF6). Here, we show the interaction of endogenous DREAM and presenilin-2 (PS2) in mouse brain and, using DREAM deficient mice or transgenic mice overexpressing a dominant active DREAM (daDREAM) mutant in the brain, we provide genetic evidence of the role of DREAM in the endoproteolysis of endogenous PS2. We show that repaglinide disrupts the interaction between DREAM and the C-terminal PS2 fragment (Ct-PS2) by coimmunoprecipitation assays. Exposure to sub-micromolar concentrations of repaglinide reduces the levels of Ct-PS2 fragment in N2a neuroblastoma cells. These results suggest that the interaction between DREAM and PS2 may represent a new target for modulation of PS2 processing, which could have therapeutic potential in Alzheimer's disease (AD) treatment.

ABSTRACT

Introducción. El mantenimiento de la intimidad dentro del Hospital es a veces olvidada a favor de otros aspectos más técnicos. Desde el punto de vista jurídico se considera desde tres niveles distintos: una esfera pública, una esfera privada y una estrictamente íntima. Algunos autores enfermeros han identificado varias facetas dentro de ella: la intimidad física, la psíquica, la socio-cultural y la relacionada con la información y la confidencialidad. Metodología. Estudio cualitativo de tipo descriptivo. Los métodos para recoger los datos han sido entrevistas en profundidad y observación participante. Resultados. Se establecen cuatro grandes categorías centrales: significado de la intimidad, la relación con los convivientes en el Hospital, factores extrínsecos que determinan la vivencia de la intimidad y factores intrínsecos. Discusión. Pocos estudios originales con enfoque fenomenológico que traten el tema. Sí aparecen estudios que abordan temas relacionados con la intimidad en el Hospital como las relaciones humanas, el espacio físico, o la implicación de la familia en el cuidado (AU)


Introduction. The maintenance of the intimacy inside the Hospital is sometimes forgotten for other more technical aspects. From the juridical point of view it is considered from three different levels: a public sphere, a private sphere and the strictly intimate sphere. Some authors nurses have identified several facets inside it: the physical intimacy, the psychic one, the sociocultural one and the related one to the information and the confidentiality. Methodology. Qualitative study of descriptive type. The methods to gather the information have been depth interviews and participant observation. Results. Four big central categories are established: meaning of the intimacy, the relation with the cohabitants in the Hospital, extrinsic factors that determine the experience of the intimacy and intrinsic factors. Discussion. Few original studies with phenomenological approach that treat the topic. There are studies that approach topics related to the intimacy in the Hospital as the human relations, the physical space, or the familys implication in the care (AU)
