ABSTRACT

Raising soil contamination with cadmium (Cd2+) and salinization necessitates the development of green approaches using bio-elicitors to ensure sustainable crop production and mitigate the detrimental health impacts. Two field trials were carried out to study the individual and combined effects of foliage spraying of Moringa leaf extract (MLE) and soil application of effective microorganisms (EMs) on the physio-biochemical, osmolytes, antioxidants, and performance of sweet potato grown in Cd2+-contaminated salty soil (Cd2+ = 17.42 mg kg-1 soil and soil salinity ECe = 7.42 dS m-1). Application of MLE, EMs, or MLE plus EMs significantly reduced the accumulation of Cd2+ in roots by 55.6%, 50.0%, or 68.1% and in leaves by 31.4%, 27.6%, or 38.0%, respectively, compared to the control. Co-application of MLE and EMs reduced Na+ concentration while substantially raising N, P, K+, and Ca2+ acquisition in the leaves. MLE and EMs-treated plants exhibited higher concentrations of total soluble sugar by 69.6%, free proline by 47.7%, total free amino acids by 29.0%, and protein by 125.7% compared to the control. The enzymatic (SOD, APX, GR, and CAT) and non-enzymatic (phenolic acids, GSH, and AsA) antioxidants increased in plants treated with MLE and/or EMs application. Applying MLE and/or EMs increased the leaf photosynthetic pigment contents, membrane stability, relative water content, water productivity, growth traits, and tuber yield of Cd2+ and salt-stressed sweet potato. Consequently, the integrative application of MLE and EMs achieved the best results exceeding the single treatments recommended in future application to sweet potato in saline soil contaminated with Cd2+.

ABSTRACT

RATIONALE: Depression is the most prevalent psychiatric disorder worldwide. Although numerous antidepressant treatments are available, there is a serious clinical concern due to their severe side effects and the fact that some depressed patients are resistant to them. Lithium is the drug of choice for bipolar depression and has been used as adjunct therapy with other groups of antidepressants. OBJECTIVES: The present study aims to investigate the effect of lithium augmentation with cerebrolysin on the neurochemical, behavioral and histopathological alterations induced in the reserpine model of depression. METHODS: The animals were divided into control and reserpine-induced model of depression. The model animals were further divided into rat model of depression, rat model treated with lithium, rat model treated with cerebrolysin and rat model treated with a combination of lithium and cerebrolysin. RESULTS: Treatment with lithium, cerebrolysin, or their combination alleviated most of the changes in behavior, oxidative stress parameters, acetylcholinesterase and monoamines in the cortex and hippocampus of the reserpine-induced model of depression. It also improved the alterations in brain-derived neurotrophic factor (BDNF) and histopathology induced by reserpine. CONCLUSIONS: The augmentation of lithium with cerebrolysin showed a clear beneficial effect in the present model of depression suggesting the use of cerebrolysin as an adjuvant in antidepressant treatment.

ABSTRACT

X-chromosomal genetic variants are understudied but can yield valuable insights into sexually dimorphic human traits and diseases. We performed a sex-stratified cross-ancestry X-chromosome-wide association meta-analysis of seven kidney-related traits (n = 908,697), identifying 23 loci genome-wide significantly associated with two of the traits: 7 for uric acid and 16 for estimated glomerular filtration rate (eGFR), including four novel eGFR loci containing the functionally plausible prioritized genes ACSL4, CLDN2, TSPAN6 and the female-specific DRP2. Further, we identified five novel sex-interactions, comprising male-specific effects at FAM9B and AR/EDA2R, and three sex-differential findings with larger genetic effect sizes in males at DCAF12L1 and MST4 and larger effect sizes in females at HPRT1. All prioritized genes in loci showing significant sex-interactions were located next to androgen response elements (ARE). Five ARE genes showed sex-differential expressions. This study contributes new insights into sex-dimorphisms of kidney traits along with new prioritized gene targets for further molecular research.

ABSTRACT

Primary health care is integral to diagnosing and managing hypertension. This study aimed to assess the knowledge, attitude, practice, priority, and confidence of primary care physicians in Qatar toward hypertension diagnosis and management, and to measure the determinants of good knowledge, desirable attitudes, and desirable practices. We conducted a cross-sectional web-based survey using a modified version of the World Hypertension League Questionnaire during the period from August 30th to October 23rd, 2020. All primary care physicians working in any of Qatar's 27 publicly run primary healthcare centers were invited to participate in the survey. Out of the 450 primary care physicians working at that time, 197 completed the study questionnaire with a response rate of 43.8%. Most respondents stated that they followed local or international treatment guidelines for hypertension management (96.4%). Primary care physicians were highly confident and prioritized hypertension management. The overall desirable practice score was 73.8%, with the highest score (95.4%) for assessing adherence to antihypertensive medications, whereas the lowest desirable practice score was 33.5% for counseling on home blood pressure monitoring. Fifty-one-point eight percentage and 62.4% correctly identified 140 mm Hg and 90 mm Hg as the systolic and diastolic blood pressure threshold for diagnosing hypertension in most patients. The lowest knowledge scores were for hypertension epidemiology in Qatar and recommended dietary modification for hypertensive patients. Respondents had positive attitudes toward task sharing with nonphysician healthcare workers for most items (58.9%-78.2%), except for drug prescriptions (30.5%). Primary care physicians in Qatar had positive attitudes, with high overall confidence and conviction (priority) scores toward hypertension. However, their knowledge scores and certain practices were suboptimal. Healthcare policymakers in Qatar should raise primary care physicians awareness of the current burden of untreated hypertension in the country by organizing targeted educational programs and emphasizing the importance of following national clinical practice guidelines in the diagnosis and management of hypertension.

ABSTRACT

Objectives: Febrile seizures (FS) are the most common neurological disorder at a young age in humans. Animal models of hyperthermia-induced seizures provide a tool to investigate the underlying mechanisms of FS related to epilepsy development and its co-morbidities. The present study investigates the alterations in monoamine neurotransmitters in two brain areas: the cortex and the hippo-campus in animals subjected to prolonged FS at their immature age. Materials and Methods: Experimental animals were divided into three groups: cage-control group (NHT-NFS), positive hyperthermic control group (HT-NFS), and the hyperthermia-induced febrile seizure group (HT-FS). Each group was further subdivided into young (Y) and adult (A) groups. Results: There were significant changes in the cortical and hippocampal serotonin neurotransmitters that were persistent until adulthood. However, the changes in the two other neurotransmitters, norepinephrine and dopamine, were transient and have been recovered in adulthood. Conclusion: The present study sheds more light on the importance of monoamine neurotransmitters in epileptogenesis following FS.

ABSTRACT

Background: Successful brachial plexus blockade produces sympathetic blockade, resulting in increased skin temperature in the blocked segments. This study aimed to evaluate the accuracy of infrared thermography in predicting failed segmental supraclavicular brachial plexus block. Methods: This prospective observational study included adult patients undergoing upper-limb surgery under supraclavicular brachial plexus block. Sensation was evaluated at the dermatomal distribution of the ulnar, median, and radial nerves. Block failure was defined as absence of complete sensory loss 30 min after block completion. Skin temperature was evaluated by infrared thermography at the dermatomal supply of the ulnar, median, and radial nerves at baseline, 5, 10, 15, and 20 min after block completion. The temperature change from the baseline measurement was calculated for each time point. Outcomes were the ability of temperature change at each site to predict failed block of the corresponding nerve using area under receiver-operating characteristic curve (AUC) analysis. Results: Eighty patients were available for the final analysis. The AUC (95% confidence interval [CI]) for the ability of temperature change at 5 min to predict failed ulnar, median, and radial nerve block was 0.79 (0.68-0.87), 0.77 (0.67-0.86), and 0.79 (0.69-0.88). The AUC (95% CI) increased progressively and reached its maximum values at 15 min (ulnar nerve 0.98 [0.92-1.00], median nerve 0.97 [0.90-0.99], radial nerve 0.96 [0.89-0.99]) with negative predictive value of 100%. Conclusion: Infrared thermography of different skin segments provides an accurate tool for predicting failed supraclavicular brachial plexus block. Increased skin temperature at each segment can exclude block failure in the corresponding nerve with 100% accuracy.

Extensive experimental data were used to develop a comprehensive kinetic model for the methanol to propylene (MTP) process over a ZSM-5 catalyst. Preliminary experiments were performed to determine the reaction conditions that would ensure the absence of external (film) and internal mass transfer resistances. The kinetic experiments were subsequently carried out at 420-500 °C under conditions where mass transfer limitations were absent. A detailed reaction network was proposed for the MTP process based on the experimental product distribution and various reported kinetic models in the literature. According to the first series of experiments (without C4 and C5/C6 recycle streams) conducted at various temperatures, the best yield for propylene production was achieved at 480 °C with a water to methanol ratio of 0.7. Subsequently, kinetic experiments were performed at 480 °C and a water to methanol ratio of 0.7 using feeds with different amounts of C4 and C5/C6 hydrocarbons as recycle streams. Species material balances for the integral tubular reactor along with power-law rate functions and the Arrhenius equation for rate constants were employed in an optimization algorithm to obtain the kinetic parameters. The predictive ability of the model was checked against experimental data, and the kinetic parameters were validated by additional experiments.

Parkinson's disease (PD) is the second most prevalent neurodegenerative disease worldwide and represents a challenge for clinicians. The present study aims to investigate the effects of cerebrolysin and/or lithium on the behavioral, neurochemical and histopathological alterations induced by reserpine as a model of PD. The rats were divided into control and reserpine-induced PD model groups. The model animals were further divided into four subgroups: rat PD model, rat PD model treated with cerebrolysin, rat PD model treated with lithium and rat PD model treated with a combination of cerebrolysin and lithium. Treatment with cerebrolysin and/or lithium ameliorated most of the alterations in oxidative stress parameters, acetylcholinesterase and monoamines in the striatum and midbrain of reserpine-induced PD model. It also ameliorated the changes in nuclear factor-kappa and improved the histopathological picture induced by reserpine. It could be suggested that cerebrolysin and/or lithium showed promising therapeutic potential against the variations induced in the reserpine model of PD. However, the ameliorating effects of lithium on the neurochemical, histopathological and behavioral alterations induced by reserpine were more prominent than those of cerebrolysin alone or combined with lithium. It can be concluded that the antioxidant and anti-inflammatory effects of both drugs played a significant role in their therapeutic potency.

PURPOSE: This study investigated the correlation between the diameter of the bony cochlear nerve canal (BCNC), as determined by Temporal bone CT, and MRI findings of cochlear nerves (CN) in children with sensorineural hearing loss (SNHL). MATERIALS AND METHODS: A prospective study design was followed. Radiological data (Temporal bone CT and MRI) of fifty children with sensorineural hearing loss (age <18 y) were included in the study. All patients (100 ears) underwent routine MRI protocol in addition to 3D CISS (3-D constructive interference in steady state). RESULTS: Based on CT findings, the BCNC was classified according to its diameter into three groups; group 1 (<1.4 mm), group 2 (1.4-2.0 mm), and group 3 (>2.0 mm). A significant difference between the three groups at degrees of SNHL (p < 0.001) was observed. Significant difference (p < 0.001) was also observed in the mean level of pure tone audiometry (PTA) average in group 1 compared to group 2. The CN was absent in 20 ears of group 1 CT results (29%), CN hypoplasia was noticed in 40 ears of group 1 CT (58%). However, CN was present in 9 ears of group 1 CT (13%), while in group 2 and 3, CN was present in 100% of the cases (27, and 4 ears, respectively, p < 0.001). CONCLUSIONS: MRI and CT imaging are valuable in the diagnosis of SNHL in children. Moreover, with BCNC stenosis, there was a high probability of CN aplasia or hypoplasia.

BACKGROUND: Chemosensory dysfunction (CD) has been reported as a common symptom of SARS-CoV-2 infection, but it is not well understood whether and for how long changes of smell, taste and chemesthesis persist in infected individuals. METHODOLOGY: Unselected adult residents of the German federal state of Schleswig-Holstein with Polymerase Chain Reaction (PCR)-test-confirmed SARS-CoV-2 infection were invited to participate in this large cross-sectional study. Data on the medical history and subjective chemosensory function of participants were obtained through questionnaires and visual analogue scales (VAS). Olfactory function (OF) was objectified with the Sniffin Sticks test (SST), including threshold (T), discrimination (D) and identification (I) test as well as summarized TDI score, and compared to that in healthy controls. Gustatory function (GF) was evaluated with the suprathreshold taste strips (TS) test, and trigeminal function was tested with an ampoule containing ammonia. RESULTS: Between November 2020 and June 2021, 667 infected individuals (mean age: 48.2 years) were examined 9.1 months, on average, after positive PCR testing. Of these, 45.6% had persisting subjective olfactory dysfunction (OD), 36.2% had subjective gustatory dysfunction (GD). Tested OD, tested GD and impaired trigeminal function were observed in 34.6%, 7.3% and 1.8% of participants, respectively. The mean TDI score of participants was significantly lower compared to healthy subjects. Significant associations were observed between subjective OD and GD, and between tested OD and GD. CONCLUSION: Nine months after SARS-CoV-2 infection, OD prevalence is significantly increased among infected members of the general population. Therefore, OD should be included in the list of symptoms collectively defining Long-COVID.

ABSTRACT

Context: Chronic kidney disease (CKD) is a public health burden worldwide. Epidemiological studies observed an association between sex hormones, including estradiol, and kidney function. Objective: We conducted a Mendelian randomization (MR) study to assess a possible causal effect of estradiol levels on kidney function in males and females. Design: We performed a bidirectional two-sample MR using published genetic associations of serum levels of estradiol in men (n = 206,927) and women (n = 229,966), and of kidney traits represented by estimated glomerular filtration rate (eGFR, n = 567,460), urine albumin-to-creatinine ratio (UACR, n = 547,361), and CKD (n = 41,395 cases and n = 439,303 controls) using data obtained from the CKDGen Consortium. Additionally, we conducted a genome-wide association study using UK Biobank cohort study data (n = 11,798 men and n = 6,835 women) to identify novel genetic associations with levels of estradiol, and then used these variants as instruments in a one-sample MR. Results: The two-sample MR indicated that genetically predicted estradiol levels are significantly associated with eGFR in men (beta = 0.077; p = 5.2E-05). We identified a single locus at chromosome 14 associated with estradiol levels in men being significant in the one-sample MR on eGFR (beta = 0.199; p = 0.017). We revealed significant results with eGFR in postmenopausal women and with UACR in premenopausal women, which did not reach statistical significance in the sensitivity MR analyses. No causal effect of eGFR or UACR on estradiol levels was found. Conclusions: We conclude that serum estradiol levels may have a causal effect on kidney function. Our MR results provide starting points for studies to develop therapeutic strategies to reduce kidney disease.

Background and Aims: Infants exposed to major surgery are at risk of injuries to the immature brain because of reduced arterial oxygen saturation. This study compared the effect of volume-controlled ventilation (VCV) versus pressure-controlled ventilation (PCV) on cerebral oxygenation in neonates subjected to repair of tracheoesophageal fistula (TEF) under video-assisted thoracoscopic surgery (VATS). Methods: This randomised controlled study included 30 full-term neonates scheduled for VATS for managing TEF under general anaesthesia. They were randomised to either VC group (n = 15), who received VCV, or PC group (n = 15), who received PCV. Cerebral oxygenation (rScO2) was monitored throughout the surgery with documentation of episodes of cerebral desaturation. Peripheral oxygen saturation, partial pressure of carbon dioxide (PaCO2), and end-tidal carbon dioxide were recorded at baseline, after induction of anaesthesia, and every 30 min till the end of the surgery. Results: rScO2 was significantly higher in the PC group than the VC group at baseline and was significantly higher in the VC group after 15 min (P = 0.041). Later, it was comparable in both the groups up to 60 min after starting the surgery. Cerebral desaturation was significantly more common in the PC group (80%) compared to VC group (33.3%) (P = 0.010). PC group required higher fraction of inspired oxygen and positive end-expiratory pressure to prevent cerebral desaturation. PaCO2 was significantly higher in the PC group than the VC group at 30 and 60 min (P = 0.005 and 0.029). Conclusion: VCV is safer than PCV for cerebral oxygenation during VATS in neonates.

Drought stress destructively affects the growth and productivity of sorghum crop, especially under saline soils. Therefore, Field trials were performed to determine the influence of water stress on water productivity (water productivity for grain, (G-WP) and water productivity for forage, (F-WP), yield of sorghum and soil properties in salt-affected soil (8.20 dS m-1) under different sowing dates and irrigation regimes. The summer sowing (SS) was performed on 1 April while fall sowing (FS) was established on 2 August. The irrigation regimes were; 100, 90, 80, and 70% of crop evapotranspiration (ETc). The findings displayed that the fodder and grain yields were increased by 23% and 26% under SS compared to FS over the two seasons 2017 and 2018, respectively. Among irrigation levels, the maximum values of grain and fodder yield were given by 100% of ETc, while a non-significant difference was observed between 100% and 90% of ETc. Moreover, the maximum values of G-WP (1.31%) and F-WP (9.00%) were recorded for 90% of ETc. Interestingly, the soil salinity was decreased in 0-0.6 m depth, and more decline was noted in 0-0.2 m depth using 90% of ETc. The highest salt accumulation withinside the soil profile was recorded under 70% of ETc in comparison to 100% of ETc. Thereupon, under water scarcity, application of 90% of ETc is recommended with SS to save 10% of the applied irrigation water without a significant decrease in grain yield (GY).

Inoculating rice plants by plant growth promoting rhizobacteria (PGPR) may be used as a practical and eco-friendly approach to sustain the growth and yield of drought stressed rice plants. The effect of rice inoculation using plant growth hormones was investigated under drip full irrigation (FI; 100% of evapotranspiration (ETc), and deficit irrigation (DI; 80% of ETc) on growth, physiological responses, yields and water productivities under saline soil (ECe = 6.87 dS m-1) for 2017 and 2018 seasons. Growth (i.e. shoot length and shoot dry weight), leaf photosynthetic pigments (chlorophyll 'a' and chlorophyll 'b' content), air-canopy temperature (Tc-Ta), membrane stability index (MSI%), and relative water content, (RWC%) chlorophyll fluorescence (Fv/Fm) stomatal conductance (gs), total phenols, peroxidase (PO), polyphenol oxidase (PPO), nitrogen contents and water productivities (grain water productivity; G-WP and straw water productivity; S-WP) were positively affected and significantly (p < 0.05) differed in two seasons in response to the applied PGPR treatments. The highest yields (3.35 and 6.7 t ha-1 for grain and straw yields) as the average for both years were recorded under full irrigation and plants inoculated by PGPR. The results indicated that under water scarcity, application of (I80 + PGPR) treatment was found to be favorable to save 20% of the applied irrigation water, to produce not only the same yields, approximately, but also to save more water as compared to I100%.

Two successive field trials were carried out at the experimental farm of the Agriculture Department of Fayoum University, Fayoum, Egypt, to investigate the sole or dual interaction effect of applying a foliar spray of Aloe saponaria extract (Ae) or potassium silicate (KSi) on reducing the stressful salinity impacts on the development, yield, and features of roselle (Hibiscus sabdariffa L.) plants. Both Ae or KSi were used at three rates: 0% (0 cm3 L-1), 0.5% (5 cm3 L-1), and 1% (10 cm3 L-1) and 0, 30, and 60 g L-1, respectively. Three rates of salinity, measured by the electrical conductivity of a saturated soil extract (ECe), were also used: normal soil (ECe < 4 dS/m) (S1); moderately-saline soil (ECe: 4-8 dS/m) (S2); and highly-saline soil (ECe: 8-16 dS/m) (S3). The lowest level of salinity yielded the highest levels of all traits except for pH, chloride, and sodium. Ae at 0.5% increased the values of total soluble sugars, total free amino acids, potassium, anthocyanin, a single-photon avalanche diode, stem diameter, fruit number, and fresh weight, whereas 1% of Ae resulted in the highest plant height, chlorophyll fluorescence (Fv/Fm), performance index, relative water content, membrane stability index, proline, total soluble sugars, and acidity. KSi either at 30 or 60 g L-1 greatly increased these abovementioned attributes. Fruit number and fruit fresh weight per plant also increased significantly with the combination of Ae at 1% and KSi at 30 g L-1 under normal soil conditions.

Estrogen receptor-α (ESR1) single nucleotide polymorphisms (SNPs) have been related to breast cancer (BC) susceptibility. In this retrospective study we investigated ESR1 SNPs in association with survival and treatment response in BC patients. Seven ESR1 SNPs were genotyped using TaqMan probe assay in 100 formalin-fixed paraffin embedded blocks of Egyptian ER+BC patients. Log-binomial regression was used to assess the association of 5 ESR1 SNPs with relative risk of non-response to adjuvant-hormonal treatment. We compared the performance of five machine learning classification models for prediction of treatment response. Predictive models were developed using rs1801132, rs2228480, and rs9322354 that were significantly associated with increased risk for non-response along with the relevant clinical features. Survival analysis was performed to detect prognostic significance of ESR1 SNPs in ESR+BC patients. rs1801132 (C), rs2228480 (A), and rs9322354 (G) minor alleles significantly increased the risk of non-response to tamoxifen by more than 81, 84, and 117%, respectively, in ER+BC patients on anthracycline/anthracycline-taxanes-based chemotherapy. Multivariate Cox regression survival analysis revealed that rs1801132 (C) and large tumor size were independent predictors for poor survival outcome in ER+BC. The best response predictive model was a combination random forest, K-nearest neighbor, and decision tree having an area under the curve of 0.94 and an accuracy of 90.8%. Our proposed predictive model based on ESR1 rs1801132, rs2228480, and rs9322354 SNPs represents a promising genetic risk stratification for selection patients who could benefit from tamoxifen therapy in such a way that might facilitate personalized medicine required to improve ER+BC patients' outcome.

ABSTRACT

Phosphorus (P) is an essential macronutrient necessary for plant growth, development, and reproduction. Two field experiments were carried out in 2018/2019 and 2019/2020 on P-deficient soil to evaluate the impact of foliar fertilization with nanophosphorus (nP) on growth, yield, and physio-biochemical indices, as well as trigonelline content of fenugreek plants under deficient irrigation (dI) stress (a deficit of 20 and 40% of crop evapotranspiration; dI-20 and dI-40). The growth and yield traits, leaf integrity (relative water content and membrane stability index), photosynthetic pigment contents, leaf and seed P contents, and stem and leaf anatomical features significantly decreased under dI-20, with greater reductions recorded under dI-40. In contrast, water-use efficiency, osmoprotective compounds, including free amino acids, soluble sugars, proline, and trigonelline, along with antioxidant contents (ascorbate, glutathione, phenolics, and flavonoids) and their activity increased significantly under both dI-20 and dI-40. However, foliar feeding with nano-P considerably increased plant growth and yield traits, leaf integrity, photosynthetic pigments contents, leaf and seed P contents, and anatomical features. Besides, water-use efficiency, osmoprotectant contents, and antioxidant content and activity were further increased under both dI-20 and dI-40. The positive effects were more pronounced with the smaller nP (25 nm) than the larger nP (50 nm). The results of this study backed up the idea of using foliar nourishment with nP, which can be effective in modulating fenugreek plant growth and seed production.

ABSTRACT

Shilajit is used commonly as Ayurvedic medicine worldwide which is Rasayana herbo-mineral substance and consumed to restore the energetic balance and to prevent diseases like cognitive disorders and Alzheimer. Locally, Shilajit is applied for patients diagnosed with bone fractures. For safety of the patients, the elemental analysis of Shilajit is imperative to evaluate its nutritional quality as well as contamination from heavy metals. The elemental composition of Shilajit was conducted using three advanced analytical techniques (LIBS, ICP, and EDX). For the comparative studies, the two Shilajit kinds mostly sold globally produced in India and Pakistan were collected. Our main focus is to highlight nutritional eminence and contamination of heavy metals to hinge on Shilajit therapeutic potential. In this work, laser-induced breakdown spectroscopy (LIBS) was applied for qualitative and quantitative analysis of the Shilajit. Our LIBS analysis revealed that Shilajit samples composed of several elements like Ca, S, K, Mg, Al, Na, Sr, Fe, P, Si, Mn, Ba, Zn, Ni, B, Cr, Co, Pb, Cu, As, Hg, Se, and Ti. Indian and Pakistani Shilajits were highly enriched with Ca, S, and K nutrients and contained Al, Sr, Mn, Ba, Zn, Ni, B, Cr, Pb, As, and Hg toxins in amounts that exceeded the standard permissible limit. Even though the content of most elements was comparable among both Shilajits, nutrients, and toxins, in general, were accentuated more in Indian Shilajit with the sole detection of Hg and Ti. The elemental quantification was done using self-developed calibration-free laser-induced breakdown spectroscopy (CF-LIBS) method, and LIBS results are in well agreement with the concentrations determined by standard ICP-OES/MS method. To verify our results by LIBS and ICP-OES/MS techniques, EDX spectroscopy was also conducted which confirmed the presence above mentioned elements. This work is highly significant for creating awareness among people suffering due to overdose of this product and save many human lives.

BACKGROUND: Weight loss in patients with metabolic syndrome has positive effects on cardiovascular and type 2 diabetes risks, but its effects on peripheral cytokines and lipid profiles in patients are still unclear. AIM: To determine the effects of diet-induced weight loss on metabolic parameters, lipids and cytokine profiles. METHODS: Eighteen adult males with metabolic syndrome (defined according to IDF 2009) and Body Mass Index (BMI) between 25 and 35 kg/m2 were subjected to a balanced hypocaloric diet for 6 months to reach at least a 5% body weight loss. RESULTS: After weight loss, a significant improvement in BMI, waist circumference, insulin, fasting blood glucose and HOMA-IR (homeostasis model assessment of insulin resistance) was observed. The analysis of LDL (low-density lipoprotein cholesterol) and HDL (high-density lipoprotein cholesterol) lipoproteins showed a change in their composition with a massive transfer of triacylglycerols from HDL to LDL. This was associated with a significant reduction in peripheral pro-inflammatory cytokines such as IL-6, TNF-α, IL-8 and MIP-1ß, leading to an overall decreased inflammatory score. An interesting positive correlation was also observed among peripheral cytokines levels after diet and peripheral levels of CETP (cholesteryl ester transfer protein), an enzyme with a key role in lipid change. CONCLUSION: Weight loss through caloric restriction is associated with an improvement in peripheral lipid and cytokine profiles that may play a major role in improving cardiovascular risk.

ABSTRACT

Two chemometric assisted spectrophotometric models were applied for the quantitative analysis of velpatasvir and sofosbuvir in their newly FDA approved pharmaceutical dosage form. The UV absorption spectra of velpatasvir and sofosbuvir showed certain degree of overlap which exhibited degree of difficulty for the choice of certain method provides simultaneous quantitative analysis of the cited drugs. Artificial neural networks and genetic algorithm artificial neural networks were the suitable model for the quantitative analysis of velpatasvir and sofosbuvir in their binary mixture. Experimental design and building the calibration set for the binary mixture were achieved to implement the described models. The proposed models were optimized with the aid of five-levels, two factors experimental design. Spectral region of 380-400 nm was rejected which resulted in 181 variables. GA reduced absorbance matrix to 72 and 36 variables for velpatasvir and sofosbuvir respectively. The models succeeded to estimate the studied drugs with acceptable values of root mean square error of calibration and root mean square error of prediction. The developed models were successfully applied to the quantitative analysis of the two drugs in Epclusa® tablets. The results were statistically compared with another published quantitative analytical method with no significant difference by applying Student t-test and variance ratio F-test.
