ABSTRACT

While circadian rhythms are entrained to the once daily light-dark cycle of the sun, many marine organisms exhibit ~12h ultradian rhythms corresponding to the twice daily movement of the tides. Although human ancestors emerged from circatidal environment millions of years ago, direct evidence of ~12h ultradian rhythms in humans is lacking. Here, we performed prospective, temporal transcriptome profiling of peripheral white blood cells and identified robust ~12h transcriptional rhythms from three healthy participants. Pathway analysis implicated ~12h rhythms in RNA and protein metabolism, with strong homology to the circatidal gene programs previously identified in Cnidarian marine species. We further observed ~12h rhythms of intron retention events of genes involved in MHC class I antigen presentation, synchronized to expression of mRNA splicing genes in all three participants. Gene regulatory network inference revealed XBP1, and GABP and KLF transcription factor family members as potential transcriptional regulators of human ~12h rhythms. These results suggest that human ~12h biological rhythms have a primordial evolutionary origin with important implications for human health and disease.

ABSTRACT

The central dogma of molecular biology is responsible for the crucial flow of genetic information from DNA to protein through the transcription and translation process. Although the sequence of DNA is constant in all organs, the difference in protein and variation in the phenotype is mainly due to the quality and quantity of tissue-specific gene expression and methylation pattern. The term methylation has been defined and redefined by various scientists in the last fifty years. There is always huge excitement around this field because the inheritance of something is beyond its DNA sequence. Advanced gene methylation studies have redefined molecular genetics and these tools are considered de novo in alleviating challenges of animal disease and production. Recent emerging evidence has shown that the impact of DNA, RNA, and protein methylation is crucial for embryonic development, cell proliferation, cell differentiation, and phenotype production. Currently, many researchers are focusing their work on methylation to understand its significant role in expression, disease-resistant traits, productivity, and longevity. The main aim of the present review is to provide an overview of DNA, RNA, and protein methylation, current research output from different sources, methodologies, factors responsible for methylation of genes, and future prospects in animal genetics.

ABSTRACT

Advancements in the Nuclear Magnetic Resonance (NMR) and Mass Spectrometry (MS) along with recent developments in omics sciences have resulted in a better understanding of molecular mechanisms and pathways associated with the physio-pathological state of the animal. Metabolomics is a post-genomics tool that deals with small molecular metabolites in a given set of time which provides clear information about the status of an organism. Recently many researchers mainly focus their research on metabolomics studies due to its valuable information in the various fields of livestock management and precision dairying. The main aim of the present review is to provide an insight into the current research output from different sources and application of metabolomics in various areas of livestock including nutri-metabolomics, disease diagnosis advancements, reproductive disorders, pharmaco-metabolomics, genomics studies, and dairy production studies. The present review would be helpful in understanding the metabolomics methodologies and use of livestock metabolomics in various areas in a brief way.

ABSTRACT

AIM: The present study aims to evaluate the impact strength of PMMA incorporated with TiO2 nanoparticles by two different processing techniques i.e. water bath and microwave processing techniques. MATERIALS AND METHODS: A total of 80 samples made of PMMA were divided into four groups. Each group includes 20 samples with group I and II comprising of samples made of normal acrylic resin and acrylic resin reinforced with 1 wt% TiO2 nanoparticles processed with conventional water bath technique respectively, groups III and IV include normal acrylic resin and acrylic resin reinforced with 1 wt% TiO2 processed using microwave technique respectively. The specimens were tested for impact strength using IZOD pendulum impact tester. The impact energy values obtained were analyzed statistically. RESULTS: The results through one-way ANOVA showed a high mean impact strength with group IV samples (23.13) and lowest with respect to group I (19.42) with highest statistical significance (p < 0.001). A post hoc Tukey test intergroup analysis showed a statistically significant difference between group I and other groups, whereas there was no statistical significance associated with other intergroup comparisons. CONCLUSION: PMMA with its current drawbacks in the physical and mechanical properties requires modifications to make it an ideal denture base material. The current investigation evaluates that a high mean impact strength with samples made of PMMA incorporated with TiO2 nanoparticles processed by microwave technique was obtained when compared with normal acrylic resins processed by water bath technique. CLINICAL SIGNIFICANCE: The methodology of current study can be used while processing of denture bases for patients to evaluate the effect of oral environment on inclusion of TiO2 nanoparticles with microwave processing in a clinical setup. This could help in reducing the amount of fractures associated with heavy load masticatory stresses and improving the mechanical properties in denture bases. ABSTRACT

Peristaltic pumps were normally used to pump liquids in several chemical and biological applications. In the present study, a peristaltic pump was used to pressurize the chamber (positive as well negative pressures) using atmospheric air. In the present paper, we discuss the development and performance study of an automatic pressurization system to calibrate low range (millibar) pressure sensors. The system includes a peristaltic pump, calibrated pressure sensor (master sensor), pressure chamber, and the control electronics. An in-house developed peristaltic pump was used to pressurize the chamber. A closed loop control system has been developed to detect and adjust the pressure leaks in the chamber. The complete system has been integrated into a portable product. The system performance has been studied for a step response and steady state errors. The system is portable, free from oil contaminants, and consumes less power compared to existing pressure calibration systems. The veracity of the system was verified by calibrating an unknown diaphragm based pressure sensor and the results obtained were satisfactory.
