ABSTRACT

Apart from its role in plant taxonomy, the understanding of pollen morphology is considered an essential interdisciplinary tool in diverse applied fields, including plant systematics, melissopalynology, aeropalynology, forensic palynology, paleopalynology, and copropalynology. In palynotaxonomy, it is frequently employed to classify and validate natural groups across a range of hierarchical levels, from higher categories down to the infrageneric level. The subfamily Acanthoideae, within the Acanthaceae family, consists of a diverse group of flowering plants that are distributed globally. The present study attempted a comprehensive analysis of the pollen morphology, employing both light microscopy (LM) and scanning electron microscopy (SEM), for a total of 13 Acanthoideae species from the Shivalik Foothills in Rajaji National Park (located in the northern Indian state of Uttarakhand, Western Himalaya). The findings indicated that the Acanthoideae is characterized by eurypalynous features, and the studied species exhibited pollen grains that were monads, radially symmetrical, and varied in size from small to large. The pollen grains were predominantly tricolporate or heteroaperturate, with porate occurrences being rare. The significant variation in exine sculpturing, including reticulate, coarsely reticulate, and bireticulate patterns, holds substantial taxonomic significance. The detailed presentation encompasses pollen morphological characters described with LM and SEM micrographs, along with a species-level identification key. RESEARCH HIGHLIGHTS: This article provides a thorough analysis of the pollen morphology of Acanthoideae taxa using both light microscopy and scanning electron microscopy, covering 13 species across 10 genera and revealing a broad spectrum of pollen characteristics, including size, shape, aperture type, and exine sculpturing. The microscopic investigation of these Acanthoideae species not only enhances our understanding of their pollen morphology but also aids in species identification through the development of a pollen-based key.

ABSTRACT

A ligninolytic peroxidase called versatile peroxidase, VP, (EC 1.11.1.16) is an iron-containing metalloenzyme. The most distinctive feature of this enzyme is its composite molecular framework, which combines lignin peroxidase's capacity to oxidize compounds with high-redox potential with manganese peroxidase's capacity to oxidize Mn2+ to Mn3+. In this study, we have extracted amino acid sequences from the Citrus sinensis source and subjected them to various computation tools to visualize the insight secondary and 3D structure, physicochemical properties, and validation of the structure which have not been studied so far to further investigate the catalytic efficiency and effectiveness of VP. The binding energies of HEME and HEME C (HEC) ligands with produced PDB (6rqf.1. A) have been also assessed, analyzed, and confirmed utilizing AutoDock. Binding energies were calculated using the AutoDock and validated by MD simulation using SCHRODINGER DESMOND. Most stable confirmation was achieved through a protein-ligand interaction study. Bio-technological use of VP in the biotransformation of ß-naphthol has also been studied. The findings in the current study will have a substantial impact on proteomics, biochemistry, biotechnology, and possible uses of versatile peroxidase in the bio-remediation of different toxic organic compounds. ABSTRACT

BACKGROUND & OBJECTIVES: Breast cancer is the second most common malignancy in Indian women. Among the members of the steroid receptor superfamily the role of estrogen and progesterone receptors (ER and PR) is well established in breast cancer in predicting the prognosis and management of therapy, however, little is known about the clinical significance of androgen receptor (AR) in breast carcinogenesis. The present study was aimed to evaluate the expression of AR in breast cancer and to elucidate its clinical significance by correlating it with clinicopathological parameters, other steroid receptors (ER and PR) and growth factors receptors (EGFR and CD105). METHODS: Expression of AR, ER, PR, epidermal growth factor receptor (EGFR) and endoglin (CD105) was studied in 100 cases of breast cancer by immunohistochemistry (IHC). Risk ratio (RR) along with 95% confidence interval (CI) was estimated to assess the strength of association between the markers and clinicopathological characteristics. Categorical principal component analysis (CATPCA) was applied to obtain new sets of linearly combined expression, for their further evaluation with clinicopathological characteristics (n=100). RESULTS: In 31 cases presenting with locally advanced breast cancer (LABC), the expression of AR, ER, PR, EGFR and CD105 was associated with response to neoadjuvant chemotherapy (NACT). The results indicated the association of AR+ (P=0.001) and AR+/EGFR- (P=0.001) with the therapeutic response to NACT in LABC patients. The AR expression exhibited maximum sensitivity, specificity and likelihood ratio of positive and negative test. The present results showed the benefit of adding AR, EGFR and CD105 to the existing panel of markers to be able to predict response to therapy. INTERPRETATION & CONCLUSIONS: More studies on the expression profiles of AR+, AR+/CD105+ and AR+/EGFR- in larger set of breast cancer patients may possibly help in confirming their predictive role for therapeutic response in LABC patients.
