ABSTRACT

The experimental research was carried out on the juvenile carp (Cyprinus carpio L.). The impact from supplemental feeds consisting of variable concentrations of chelate compounds, biogenic trace elements and probiotic lactobacillus-based product Bacillus subtilis VKPM B-2335 was evaluated. Optical and qualitative parameters of the lactobacillus base were studied in order to identify the major group of substances potentially able to influence the end result. The purpose of this research was to identify changes in the structure of the zymogen granules and their dimensions at which supplemental feeds produce a stimulating effect on the synthesis of zymogens in exogenous cells of the secretory part of pancreas. At the outcome of the study, for the first time, it was possible to prove that the integrated action of chelates and lactobacillus-based probiotics complemented each other. Metal chelate compounds contributed to enlargement of the zymogen granules, if compared to the control values. The bacterial products accelerated production of the zymogen granules in acinar cells diffusely located in carp hepatopancreas.


A pesquisa experimental foi realizada em carpas juvenis (Cyprinus carpio L.). O impacto de suplementos alimentares consistindo em concentrações variáveis de compostos quelatos, oligoelementos biogênicos e produto probiótico baseado em lactobacilos Bacillus subtilis VKPM B-2335 foi avaliado. Parâmetros ópticos e qualitativos da base de lactobacilos foram estudados a fim de identificar o maior grupo de substâncias potencialmente capazes de influenciar o resultado final. O objetivo dessa pesquisa foi identificar mudanças na estrutura dos grânulos de zimogênio e suas dimensões nas quais alimentos suplementares produzem um efeito estimulante na síntese de zimogênios em células exógenas da parte secretora do pâncreas. No resultado do estudo, pela primeira vez, foi possível comprovar que a ação integrada dos quelatos e dos probióticos à base de lactobacilos se complementava. Compostos quelatos metálicos contribuíram para o aumento dos grânulos de zimogênio, se comparados aos valores de controle. Os produtos bacterianos aceleraram a produção dos grânulos de zimogênio nas células acinares localizadas difusamente no hepatopâncreas da carpa.

ABSTRACT

Macronutrient and micronutrient deficiencies in the fish diet can affect fish growth rate, ability to resist disease, and fillet quality. Determination of effective dose of developed feed additives consisting of chelate compounds of biogenic elements and probiotic preparation based on Bacillus subtilis. Young rainbow trout were divided into four groups consisting of 100 fish each. The control group received standard food; the experimental groups received food with chelates and probiotics in different concentrations. At the end of the experiment (30 days), the middle intestine histology and morphometric parameters were studied. The use of chelated compounds alone affected the state of the intestinal villi and the infiltration of the epithelium by lymphocytes. The addition of a probiotic had a positive effect on the migration of immune cells in the intestinal villi; however, the probiotic in high concentrations contributed to a decrease in the number and area of goblet cells. Synergistic effects of chelated compounds and probiotic preparations on the morphometric parameters of the medium intestine were observed. In the proper doses, the studied feed additives might be recommended for rainbow trout farming under industrial aquaculture conditions.

ABSTRACT

Growing fish in a recirculating aquaculture system (RAS) involves various stress factors that directly affect their physiological condition. This study aimed to investigate the effect of a chelated organic-mineral additive including Fe, Mn, Cu, Zn, Co, Se and I; on the rearing performance of juvenilecatfish (Clariasgariepinus) under RAS conditions. Four groups of fish (n=50) were formed: a control group (receiving standard feed) and three experimental groups (receiving standard feed with biogenic elements chelate compounds in different concentrations). At the end of the experiment (30 days), the physiological condition of the fish was evaluated by the growth rate, internal organs condition, blood serum biochemical indices and histological examination of the middle intestine. The survival rate of fish in the experimental groups was 96-98%. According to the results, the absolute increase was 14,30% in group III, 11,13% in group II and 6,71% in group I, compared to the control. However, the use of chelated compounds in high concentrations can cause necrosis and erosion of the apical part of the villi (groups II and III). Blood biochemical analysis of fish (group II and III) receiving medium and high concentrations of chelated compounds showed high ALT activity, which was 23,02% (p<0,05) and 45,19% (p<0,05) higher compared to control, respectively. Mineral-chelate compounds, of the studied composition, at a concentration of 0,5 g/kg positively affect the histological structure of the midgut of Clarias gariepinus. This dosage of the investigated feed additive can be recommended for the practical application of rearing Clariasgariepinus in closed water installations.
