ABSTRACT

Yeast responds to alterations in plasma membrane lipid asymmetry and external alkalization via the sensor protein Rim21 in the Rim101 pathway. However, the sensing mechanism used by Rim21 remains unclear. Here, we found that the C-terminal cytosolic domain of Rim21 (Rim21C) fused with GFP was associated with the plasma membrane under normal conditions but dissociated upon alterations in lipid asymmetry or external alkalization. This indicates that Rim21C contains a sensor motif. Rim21C contains multiple clusters of charged residues. Among them, three consecutive Glu residues (EEE motif) were essential for Rim21 function and dissociation of Rim21C from the plasma membrane in response to changes in lipid asymmetry. In contrast, positively charged residues adjacent to the EEE motif were required for Rim21C to associate with the membrane. We therefore propose an "antenna hypothesis," in which Rim21C moves to or from the plasma membrane and functions as the sensing mechanism of Rim21.

ABSTRACT

INTRODUCTION: This study aims to explore the efficacy of the topical application of 10% benzocaine for treating pruritus and pain as compared to vehicle ointment. METHODS: Twenty male subjects were treated in a randomized double-blind fashion with the investigational medicinal product (IMPD) and vehicle. Immediately after the injection of 100 µg histamine on both arms, subjects received topical treatment and pruritus was subsequently assessed with visual analogue scale (VASpruritus) and Eppendorfer questionnaire. Ultraviolet B radiation (UVB) was administered on the back to induce slight sunburn. Twelve hours after UVB application again the IMPD was applied on the right or left upper back and vehicle on the other side and pain related to sunburn was measured with VASpain and pressure algometry. RESULTS: A trend towards better reduction of pruritus was shown for benzocaine in VASpruritus. For the VASpain significant differences in group comparison (p = 0.02) were observed. Algometer measurements showed onset of pain reduction in the verum group after 20 min whereas in the vehicle-treated area pain relief occurred only after 60 min after application. CONCLUSIONS: The topically administered ointment containing 10% benzocaine was found superior over vehicle for treating pain, but not pruritus.

ABSTRACT

OBJECTIVE: This study aimed to determine the influence of an object's z-axis location and location on the axial plane on the voxel values in cone beam computed tomography. STUDY DESIGN: The CTP401 and CTP486 of the Catphan 500 phantom were scanned with an Alphard-3030 at 6 locations in the z-axis. RESULTS: In CTP401, the voxel values of 3 inserts showed the lowest values when the center of the radiation field corresponded to under 2 or 4 cm of the z-axis center of CTP401. In CTP486, the voxel values showed the lowest values when the center of the radiation field corresponded to under 2 or 4 cm of the z-axis center of CTP486. On the same axial plane, the voxel values at the center showed significantly lower values than the off-center locations (P = .002). CONCLUSIONS: The voxel values in the Alphard-3030 changed based on the z-axis location of the radiation field and were nonuniform on the same axial plane.
