ABSTRACT

BACKGROUND: Flowering time has an important effect on regional adaptation and yields for crops. The tyrosine kinase-like (TKL) gene family is widely existed and participates in many biological processes in plants. Furthermore, only few TKLs have been characterized functions in controlling flowering time in wheat. RESULTS: Here, we report that TaCTR1, a tyrosine kinase-like (TKL) gene, regulates flowering time in wheat. Based on identification and evolutionary analysis of TKL_CTR1-DRK-2 subfamily in 15 plants, we proposed an evolutionary model for TaCTR1, suggesting that occurrence of some exon fusion events during evolution. The overexpression of TaCTR1 caused early flowering time in transgenic lines. Transcriptomics analysis enabled identification of mass differential expression genes including plant hormone (ET, ABA, IAA, BR) signaling, flavonoid biosynthesis, phenolamides and antioxidant, and flowering-related genes in TaCTR1 overexpression transgenic lines compared with WT plants. qRT-PCR results showed that the expression levels of ethylene (ET) signal-related genes (ETR, EIN, ERF) and flowering-related genes (FT, PPD1, CO, PRR, PHY) were altered in TaCTR1-overexpressing wheat compared with WT plants. Metabonomics analysis showed that flavonoid contents were altered. CONCLUSIONS: Thus, the results show that TaCTR1 plays a positive role in controlling flowering time by activating various signaling pathways and regulating flowering-related genes, and will provide new insights on the mechanisms of wheat flowering regulation.

ABSTRACT

Objectives: In recent years, the free triiodothyronine/free thyroxine (FT3/FT4) ratio, a new comprehensive index for evaluating thyroid function, which could reflect thyroid function more stably and truly than serum thyroid hormone level, has been demonstrated to correlate with the risks of diabetes and cardiovascular disease in euthyroid adults. However, the correlation between thyroid hormone sensitivity and long-term prognosis in euthyroid patients with acute coronary syndrome (ACS) and diabetes after percutaneous coronary intervention (PCI) remains unclear. Methods: A total of 1,786 euthyroid patients with ACS who successfully underwent PCI at Beijing Anzhen Hospital from August 2021 to April 2022 were included in our study, which was divided into three groups according to tertiles of thyroid hormone sensitivity index. Cox regression, Kaplan-Meier, and receiver operating characteristic analyses were applied to analyze the associations between the FT3/FT4 ratio with ACS and diabetes after PCI. Results: Our analysis indicated that a lower level of FT3/FT4 ratio in euthyroid patients with acute coronary syndrome (ACS) and diabetes after PCI showed significantly higher incidences of major adverse cardiac and cerebrovascular events (MACCE) when compared with a higher level of FT3/FT4 ratio. After adjusting for other covariates, patients with a lower level of FT3/FT4 ratio were negatively associated with the risk of MACCE than those with a higher level of FT3/FT4 ratio (adjusted OR =1.61, 95% CI 1.05-2.47, P = 0.028). In subgroup analyses, individuals were stratified by age, sex, BMI, ACS type, hypertension, and dyslipidemia, showing that there were no significant interactions between the FT3/FT4 ratio and all subgroups for MACCE. In addition, the FT3/FT4 ratio performed better on ROC analyses for cardiac death prediction [area under the curve (AUC), 0.738]. Conclusion: A reduced level of FT3/FT4 ratio was a potential marker of poor prognosis in euthyroid patients with ACS and diabetes after PCI.

ABSTRACT

This study proposes a novel multi-stage multi-attribute group decision making method under a probabilistic linguistic environment considering the development state and trend of alternatives. First, the probabilistic linguistic term set (PLTS) is used by decision makers (DMs) to describe qualitative evaluation information. Subsequently, the weights of DMs for different attributes in different periods are determined by the credibility degree, which is combined with the hesitancy degree and the similarity degree. The evaluations of different DMs for alternatives and the evaluations of DMs' intentions to reward or punish are then aggregated. Later, the trend change level and the trend change stability of alternatives are measured through the means of reward and punishment incentives. Additionally, the probabilistic linguistic time-ordered incentive operator is proposed to aggregate the development state evaluation information and development trend evaluation information in different periods, and alternatives are prioritized by the extended TOPSIS method in the probabilistic linguistic environment. Finally, the practical use of the proposed decision framework is validated by using a sustainable supplier selection problem, and the effectiveness and the applicability of the framework are discussed through comparative analysis. The results show that the proposed approach can select suitable sustainable suppliers by considering their development state and trend in multiple stages.

ABSTRACT

Drought stress results in significant yield losses in wheat production. Although studies have reported a number of wheat drought tolerance genes, a deeper understanding of the tolerance mechanisms is required for improving wheat tolerance against drought stress. In this study, we found that "Deguo 2" exhibited higher tolerance to drought than "Truman". Transcriptomics analysis enabled identification of 6084 and 7146 differentially expressed genes (DEGs), mainly mapping flavonoid biosynthesis, plant hormone, phenolamides and antioxidant pathways and revealed altered expression levels of about 700 genes. Exogenous melatonin application enhanced wheat tolerance against drought stress. Co-expression analysis showed that bHLH and bZIP transcription factors may be involved in the regulation of various pathway genes. Take together, these results provide new insights for us on exploring the crosstalk between phytohormones and secondary metabolites, and will deepen the understanding of the complex tolerance mechanisms against drought stress in wheat.

ABSTRACT

Drought is a major abiotic stress that reduces crop yields and quality worldwide. Although some genes involved in the response to drought stress have been identified, a more in-depth understanding of the mechanisms underlying wheat tolerance to drought is needed for the control of drought tolerance. Here, we evaluated the drought tolerance of 15 wheat cultivars and measured their physiological-biochemical parameters. Our data showed that the drought tolerance of the resistant wheat cultivars was significantly higher than that of drought-sensitive cultivars, which was associated with a greater antioxidant capacity of the former. Transcriptomic analysis revealed that different mechanisms of drought tolerance exist between the wheat cultivars Ziyou 5 and Liangxing 66. Transcriptomic analysis also revealed a large number of DEGs, including those involved in flavonoid biosynthesis, phytohormone signalling, phenolamides and antioxidants. qRT-PCR was performed, and the results showed that the expression levels of TaPRX-2A were significantly different among the various wheat cultivars under drought stress. Further study revealed that overexpression of TaPRX-2A enhanced tolerance to drought stress through the maintenance of increased antioxidase activities and reductions in ROS contents. Overexpression of TaPRX-2A also increased the expression levels of stress-related genes and ABA-related genes. Taken together, our findings show that flavonoids, phytohormones, phenolamides and antioxidants are involved in the plant response to drought stress and that TaPRX-2A is a positive regulator of this response. Our study provides insights into tolerance mechanisms and highlights the potential of TaPRX-2A overexpression in enhancing drought tolerance in crop improvement programmes.

ABSTRACT

Objective: In minimally invasive spinal surgery, the treatment of lumbar spinal stenosis with microendoscopic discectomy (MED) or unilateral biportal endoscopic discectomy (UBED) shows effective results, but which is more effective is controversial. Our study aimed to evaluate the efficacy and safety of UBED versus MED in the treatment of lumbar spinal stenosis by a systematic review and meta-analysis, so as to provide reference for the promotion of UBED in clinical practice. Methods: The multiple databases like PubMed, EMBASE, Web of Science, Cochrane Library, Chinese National Knowledge Databases, Chinese BioMedical Database, and Wanfang Database were used to search for the relevant studies. Review Manager 5.4 was adopted to estimate the effects of the results among selected articles. Odds ratio (OR) and mean difference (MD) with 95% confidence intervals (CIs) were used to estimate the overall pooled effect. Subgroup analysis, forest plots, funnel plots and Egger's test for the articles included were also conducted. Results: Three randomized clinical trials and seven cohort studies were finally retrieved, these studies included 685 and 829 patients in the UBED and MED groups, respectively. There were no differences in terms of operation time (MD = -0.92, P =0.72), estimated blood loss (MD = -26.31, P =0.08), complications (MD =0.81, P =0.38) and Oswestry Disability Index (ODI) score (P >0.05 in four subgroup) between the two groups. The visual analog scale (VAS) score of back pain in the UBED group was better than MED group only at 6 months (MD = -0.23, P =0.006) after operation, the VAS score of leg pain in the UBED group was better than that of MED group at 3 mouths (MD = -0.22, P =0.002) and 6 months (MD = -0.24, P =0.006) after operation, the UBED group had a less postoperative length of stay than the MED group (MD = -1.85, P <0.001). The bias analysis showed that there was no potential publication bias in the included literature. Conclusion: This study showed that compared with MED, UBED has the advantages of short hospital stay and good short-term curative effect, but there is no significant difference in long-term efficacy and safety, they can be replaced by each other in clinical application.

ABSTRACT

Alkaline pretreatment was applied to enhance hydrogenolysis conversion of cellulose to C2-C3 polyols. The alkali cellulose was obtained by treating cellulose with different concentration of NaOH solution. The scanning electron microscopy (SEM) and X-ray diffraction (XRD) results indicate that the cleavage of cellulose chains occurs and the amorphous part is increased after alkaline treatment, which means the alkali cellulose has more accessible structure. Moreover, the absorbed NaOH crystal in alkali cellulose could make the further reaction perform in weak basic condition. When hydrogenolysis of alkali cellulose over Ru/C was conducted at 433 K, 59.23% of the substrate was converted with 1,2-propanediol and ethylene glycol as main products, whereas the corresponding conversion rate of untreated cellulose was 25.05% and no C2-C3 polyols were detected. These preliminary results suggested the advantages of activating the cellulose by alkaline pretreatment and potentials for efficient conversion of cellulose. Finally the plausible mechanism was also discussed.
