ABSTRACT

The botanical insecticide market is growing because of limitations placed on the use of certain synthetic chemical insecticides. In this sense, the lesser mealworm Alphitobius diaperius (Coleoptera: Tenebrionidae) is the main poultry pest. The insect causes weight loss and damage to the digestive system of poultry, and it is a vector and reservoir of pathogens. Consequently, this study explored the following hypotheses: (i) essential oils (EOs) derived from Mentha spp. are toxic to A. diaperius; (ii) these EOs are compatible with Beauveria bassiana, the natural enemy of the poultry pest, that parasite A. diaperinus; (iii) these EOs also exhibit activity against bacteria that are pathogenic to poultry. In topical applications and ingestion tests, EOs from Mentha arvensis, Mentha spicata, and Mentha piperita were toxic to A. diaperinus. Chromatographic analyses revealed that menthol is the predominant compound in M. arvensis and M. piperita, whereas carvone is the major compound in M. spicata. Both (-)- and (+)-menthol, along with (-)- and (+)-carvone, underwent testing with A. diaperinus. Nevertheless, their activity was not as potent as those of the EOs, suggesting a possible synergistic and/or additive effect. The EOs did not have any adverse effects on the conidial germination, vegetative growth, or conidia production per colony of the entomopathogenic fungus B. bassiana. Consequently, these EOs are compatible with this natural enemy. The EO extracted from M. spicata exhibited significant toxicity against Staphylococcus aureus (ATCC 25923), whereas the remaining EOs displayed moderate toxicity against this bacterium. The EOs derived from Mentha spp., as assessed in this study, hold promise for the development of botanical insecticides tailored for the control of A. diaperinus. These insecticides are selective in favor of the natural enemy B. bassiana and can also serve as effective sanitizers, thanks to their antibacterial properties.

ABSTRACT

To contribute to the development of products capable of complexing with the SARS-CoV-2 spike protein, and thus preventing the virus from entering the host cell, this work aimed at discovering binding sites in the whole protein structure, as well as selecting substances capable of binding efficiently to such sites. Initially, the three-dimensional structure of the protein, with all receptor binding domains in the closed state, underwent blind docking with 38 substances potentially capable of binding to this protein according to the literature. This allowed the identification of five binding sites. Then, those substances with more affinities for these sites underwent pharmacophoric search in the ZINC15 database. The 14,329 substances selected from ZINC15 were subjected to docking to the five selected sites of the spike protein. The ligands with more affinities for the protein sites, as well as the selected sites themselves, were used in the de novo design of new ligands that were also docked to the binding sites of the protein. The best ligands, regardless of their origins, were used to form complexes with the spike protein, which were subsequently used in molecular dynamics simulations and calculations of ligands affinities to the protein through the molecular mechanics/Poisson-Boltzmann surface area method (MMPBSA). Seven substances with good affinities to the spike protein (-12.9 to -20.6 kcal/mol), satisfactory druggability (Bioavailability score: 0.17 to 0.55), and low acute toxicity to mice (LD50: 751 to 1421 mg/kg) were selected as potentially useful for the future development of new products to manage COVID-19 infections.Communicated by Ramaswamy H. ABSTRACT

Although new nematicides have appeared, the demand for new products less toxic and more efficient for the control of plant-parasitic nematodes are still high. Consequently, studies on natural secondary metabolites from plants, to develop new nematicides, have increased. In this work, nineteen extracts from eleven Brazilian plant species were screened for activity against Meloidogyne incognita. Among them, the extracts of Piterogyne nitens showed a potent nematostatic activity. The alkaloid fraction obtained from the ethanol extract of leaves of P. nitens was more active than the coming extract. Due to the promising activity from the alkaloid fraction, three isoprenylated guanidine alkaloids isolated from this fraction, galegine (1), pterogynidine (2), and pterogynine (3) were tested, showing similar activity to the alkaloid fraction, which was comparable to that of the positive control Temik at 250 µg/mL. At lower concentrations (125-50 µg/mL), compound 2 showed to be the most active one. As several nematicides act through inhibition of acetylcholinesterase (AChE), the guanidine alkaloids were also employed in two in vitro AChE assays. In both cases, compound 2 was more active than compounds 1 and 3. Its activity was considered moderated compared to the control (physostigmine). Compound 2 was selected for an in silico study with the electric eel (Electrophorus electricus) AChE, showing to bind mostly to the same site of physostigmine in the AChEs, pointing out that this could be the mechanism of action for this compound. These results suggested that the guanidine alkaloids 1,2 and 3 from P. nitens are promising for the development of new products to control M. incognita, especially guanidine 2, and encourage new investigations to confirm the mechanism of action, as well as to determine the structure-activity relationship of the guanidine alkaloids.

ABSTRACT

The poultry red mite Dermanyssus gallinae (De Geer) is the most important haematophagous ectoparasite in the poultry industry. The use of synthetic acaricides for this control is presenting risks related to human food. In this sense, plant secondary metabolites are promising for controlling this pest. Thus, this study aimed to evaluate the acaricidal activity of Duguetia lanceolata A.St.-Hil. (stem bark), Xylopia emarginata Mart. (stem bark), and Xylopia sericea A.St.-Hil. (stem bark and fruits) against D. gallinae. Additionally, the secondary metabolite profile of the X. emarginata was analysed by UFLC-DAD-ESI(+)-MS/MS (micrOTOF-QII) and data analysis was performed using the Molecular Networking. In a topical application test, all plant species tested showed bioactivity, in that order of toxicity with the respective probability survival: X. emarginata (stem bark) (0.28) > X. sericea (stem barks) (0.35) > X. sericea (fruits) and D. lanceolata (stem bark) (0.47). The most promising results were found for X. emarginata (LC50 = 331.769 µg/cm2). It is noteworthy that the LC50 of the insecticide cypermethrin was 1234.4 µg/cm2, which was 73% higher than that of X. emarginata. The metabolomic profile of X. emarginata revealed the presence of alkaloids, amides, terpenoids, and phenolic compounds. This is the first report of X. emarginata acaricidal activity against D. gallinae and exploratory chemical analysis by untargeted metabolomics and the molecular network of this plant.

ABSTRACT

Alternaria brown spot (ABS), caused by the fungus Alternaria alternata pathotype tangerine, is one of the main phytosanitary problems for mandarin growers. About 15 applications per year of harmful fungicides are required for controlling ABS disease in citrus orchards. As chalcones seem to be less toxic to humans and environment than the commercial fungicides in use, this study initially aimed at synthesizing 137 chalcones through aldolic condensations between benzaldehydes and acetophenones. The resulting chalcones were screened for activity against A. alternata through a fungal growth assay that was carried out in 96-cell polypropylene plates, using the same concentration to all studied substances. The four active chalcones underwent conidia germination and mycelial growth, which confirmed the antifungal activity of the compounds. These chalcones were then poured onto Murcott tangor fruit that had been inoculated with conidia of the fungus. All four chalcones reduced the ABS progress to values significantly smaller (P0.05) than that observed for the control. Statistical calculations showed that the best results were afforded by two compounds, bearing a 2,4,5-trimethoxyphenyl group at position 3 of prop-2-enal and a 3-nitro- or 3-hydroxyphenyl group at position 1 of the aldehyde. Such compounds reduced the incidence of the disease in Murcott tangor fruit to values that did not differ statistically from those obtained with a commercial fungicide.


A mancha marron de Alternaria (MMA), causada pelo fungo Alternaria alternata patótipo tangerina, é um dos maiores problemas fitossanitários dos produtores de tangerina. Aproximadamente 15 aplicações de fungicidas de elevada periculosidade, por ano, são necessárias para o controle de MMA em várias plantações de citros. Como as chalconas parecem menos tóxicas para os seres humanos e para o ambiente que os fungicidas comerciais atualmente em uso, este estudo teve como objetivo inicial sintetizar 137 chalconas através da condensação aldólica de benzaldeídos com acetofenonas. As atividades das chalconas resultantes contra A. alternata foram avaliadas através do emprego de teste de crescimento fúngico em placas de polipropileno com 96 cavidades, empregando a mesma concentração para todas as substâncias estudadas. As quatro chalconas mais ativas foram submetidas a testes de germinação de conídios e de crescimento micelial, que confirmaram as atividades antifúngicas dos compostos selecionados. Estes foram então, aplicados em frutos de tangor Murcote que tinham sido inoculados com conídios do fungo. Todas as quatro chalconas reduziram o progresso de MMA a valores significativamente inferiores (P0.05) ao observado para o controle. Cálculos estatísticos mostraram que os melhores resultados foram obtidos para dois compostos, que tinham um grupo 2,4,5-trimetoxifenil na posição 3 do prop-2-enal e um grupo 3-nitro- ou 3-hidroxifenil na posição 1 do aldeído. Tais compostos reduziram a incidência da doença em frutos de tangor Murcote a valores que não diferiam estatisticamente do obtido com um fungicida comercial.

ABSTRACT

Meloidogyne exigua is a parasitic nematode of plants that causes great losses to coffee farmers. In an effort to develop parasitic controls, 154 chalcones were synthesized and screened for activity against this nematode. The best results were obtained with (2E)-1-(4'-nitrophenyl)-3-(2,4,5-trimethoxyphenyl)prop-2-en-1-one (6) with a 50% lethal concentration (LC50) of 171 µg/ml against M. exigua second-stage juveniles, in comparison to the commercially-available nematicide carbofuran which had an LC50 of 260 µg/ml under the same conditions. When coffee plants were used, 6 reduced the nematode population to ~50% of that observed in control plants. To investigate the mechanism of action of 6, an in silico study was carried out, which indicated that 6 may act against M. exigua through inhibition of a putative caffeic acid 3-O-methyltransferase homodimer, the amino acid sequence of which was determined by examining the genome of Meloidogyne incognita.

ABSTRACT

Objetivou-se, com o presente trabalho, verificar o efeito do extrato metanólico de Coffea racemosa Lour., espécie silvestre de cafeeiro portadora de resistência a uma das principais pragas da cultura cafeeira, o bicho-mineiro. Inicialmente, o extrato metanólico das folhas de C. racemosa foi pulverizado em mudas de cafeeiro Coffea arabica cv. Catuaí, em casa-de-vegetação, as quais foram expostas a adultos de L. coffeella para o teste de oviposição com chance de escolha. Observou-se que as plantas tratadas foram menos preferidas para oviposição. A seguir, realizou-se o teste de efeito ovicida, no qual mudas de C. arabica contendo ovos do bicho-mineiro foram pulverizadas com o mesmo extrato. Os experimentos foram conduzidos em delineamento inteiramente casualizado com cinco repetições. Verificou-se redução significativa na eclosão de larvas, e segundo análise por microscopia eletrônica de varredura, os ovos não viáveis apresentaram anomalias nas superfícies externas. Entretanto, os adultos provenientes de ovos tratados não foram afetados na percentagem de emergência, razão sexual e oviposição. Os resultados obtidos demonstram que C. racemosa apresenta-se como uma alternativa promissora ao controle do bicho-mineiro, necessitando, no entanto, de novos estudos para isolamento e identificação das substâncias bioativas.


The objective of this study was to evaluate the effect of Coffea racemosa Lour. methanolic extract , a wild coffee specie that presents resistance against the coffee leaf-miner, a major pest of coffee crop. The methanol extract of C. racemosa leaves was sprayed on coffee seedlings, Coffea arabica cv. Catuaí, in greenhouse, which were exposed to adults of L. coffeella for oviposition in free choice test. Plants treated were less preferred for oviposition. An ovicidal assay was carried out by spraying the methanol extract on C. arabica seedlings containing L. coffeela eggs on their leaves. The experiments were conducted in a completely randomized design with five replicates. Larvae hatching out from treated eggs were significantly reduced by the extract and non viable eggs presented some anomalies on their external surfaces according to analyses by scanning electron microscopy. However, the adults derived from treated eggs did not have the percentage of emergence, sex ratio and oviposition affected by the treatment. The results show that C. racemosa presents as a promising alternative to control the coffee leaf-miner, being necessary new studies to isolate and caracterize the bioactive substances against this pest.

ABSTRACT

O fracionamento do extrato metanólico das folhas de Gochnatia barrosii Cabrera (Asteraceae) resultou no isolamento do flavonol glicosídico trans-tilirosídeo [kaempferol 3-O- -D-(6''-O-E-p-cumaroil)-glicopiranosídeo], que nunca tinha sido identificado na referida espécie vegetal. Em teste realizado in vitro, observou-se que tal substância a 500 μg/mL, não tem efeito sobre a mortalidade de juvenis do segundo estágio do nematóide Meloidogyne exigua Goeldi.


Fractionation of the methanolic extract from Gochnatia barrosii Cabrera (Asteraceae) leaves resulted in the isolation of the flavonol glycoside trans-tiliroside [kaempferol 3-O- -D-(6''-O-E-p-coumaroyl)-glucopyranoside], which had never been found in such plant species. Such substance at 500 μg/mL caused no in vitro effect on the mortality of second-stage juveniles of the nematode Meloidogyne exigua Goeldi.

Native and exotic Brazilian plants collected in the State of Minas Gerais were evaluated for their anticancer potential. Methanol extracts from leaves of 51 plant species were tested for cytotoxicity against four tumor cell lines: B16 (murine skin), HL-60 (human leukemia), MCF-7 (human breast), and HCT-8 (human colon). Plant extracts that exhibited IC(50) values less than 30 microg/ml against any tumor cell line were tested on sea urchin egg development and mouse erythrocytes. In addition, all extracts were evaluated for their general toxicity using the brine shrimp lethality assay. The most active extracts against the tumor cells were those obtained from Lantana fucata, Copaifera langsdorffii, and Momordica charantia. These three extracts inhibited sea urchin development from the first cleavage, but those from C. langsdorffii and M. charantia were very active against mouse erythrocytes. Only the L. fucata extract presented no hemolytic activity. Consequently, although the extracts of L. fucata, M. charantia, and C. langsdorffii could be useful in the development of new anticancer products, the first of these extracts is the most promising since it did not present unspecific toxicity, as suggested by negative results obtained with brine shrimp lethality and mouse erythrocytes assays.

As nematodes cause great damage to Brazilian coffee production, effective methods to control these parasites are necessary. In a previous work Allium cepa L., Cajanus cajan (L.) Mill., Crotalaria juncea L., Ficus elastica Roxb., Ruta graveolens L., Stylosanthes guianensis Aubl., Leucaena leucocephala (Lam.) Dewit., Brachiaria decumbens Stapf., Catharanthus roseus G. Don, Tagetes minuta L., Ricinus communis L. and Coffea arabica L. produced active substances against Meloidogyne exigua Goeldi, a nematode widely disseminated through Brazilian coffee fields. Thus, aqueous extracts of such plants, collected in a different season from that of the previous work, as well as crude metabolites produced in liquid medium by Fusarium moniliforme Shelden and Cylindrocarpon magnusianum (Sacc.) Woll., were submitted to in vitro assays with M. exigua second-stage juveniles (J2). All plants and fungi produced active substances against J2. Therefore, their metabolites were applied to six-month-old coffee plants inoculated with M. exigua. After 90 days in a greenhouse, those samples obtained from A. cepa, L. leucocephala, R. graveolens and F. moniliforme inhibited the production of galls and eggs by M. exigua, demonstrating potential to control such parasite.


Os nematóides acarretam grandes perdas aos produtores brasileiros de café, sendo necessário o desenvolvimento de métodos eficientes para o seu controle. Em trabalho anterior, Allium cepa L., Cajanus cajan (L.) Mill., Crotalaria juncea L., Ficus elastica Roxb., Ruta graveolens L., Stylosanthes guianensis Aubl., Leucaena leucocephala (Lam.) Dewit., Brachiaria decumbens Stapf., Catharanthus roseus G. Don, Tagetes minuta L., Ricinus communis L. e Coffea arabica L. produziram substâncias ativas contra o nematóide Meloidogyne exigua Goeldi, que é amplamente disseminado pelos cafezais brasileiros. Dando continuidade a esse trabalho, extratos aquosos das plantas mencionadas, coletadas em época diferente daquela mencionada no trabalho anterior, bem como metabólitos na forma bruta de Fusarium moniliforme Shelden e Cylindrocarpon magnusianum (Sacc.) Woll., produzidos em meio de cultura líquido, foram submetidos a testes in vitro com juvenis do segundo estádio (J2) de M. exigua. Todas as plantas e fungos produziram substâncias ativas contra J2. Portanto, seus metabólitos foram aplicados em mudas de café de seis meses de idade, inoculadas com M. exigua. Após 90 dias em casa de vegetação, verificou-se que os materiais oriundos de A. cepa, L. leucocephala, R. graveolens e F. moniliforme inibiram a produção de ovos e de galhas por M. exigua, demonstrando potencial para o controle de tal parasita.

ABSTRACT

Aiming to develop more efficient and environmental friendly methods than those available to control Colletotrichum gloeosporioides Penz, which causes blister spot in coffee trees, a search for plants able to produce substances active against such pathogen was carried out. Thus, extracts of 48 plant species, collected at Alto Rio Grande region, in Minas Gerais, were prepared and submitted to in vitro assays with that fungus. The best results were obtained with the extracts prepared from Digitalis lanata Ehrh, Origanum manjorona L., Plantago lanceolata Hook. and Stevia rebaudiana (Bertoni) Bertoni, which inhibited C. gloeosporioides spores germination. After dilution of some active extracts with aqueous 1 percent Tween 80 solution in a 1:2 or 1:3 ratio (extract:aqueous solution), their antifungal activity vanished. Some of the active extracts were also submitted to freeze drying and none of them presented any alteration in their antifungal activity. Concluding, several plants presented potential to be used in the search for new bioactive substances to control C. gloeosporioides, especially O. manjorona L., which inhibited 96 percent of the fungus spores germination.


Com vistas a contribuir para o desenvolvimento de métodos mais eficientes e menos agressivos ao ambiente que aqueles disponíveis para o controle de Colletotrichum gloeosporioides Penz, causador da mancha manteigosa em cafeeiros, buscou-se identificar espécies vegetais produtoras de substâncias ativas contra o referido patógeno. Para tanto, prepararam-se extratos de 48 espécies vegetais, coletadas na região do Alto Rio Grande, em Minas Gerais, para serem submetidos a testes in vitro com o mencionado fungo. Constatou-se que os melhores resultados foram obtidos com os extratos oriundos de Digitalis lanata Ehrh, Origanum manjorona L., Plantago lanceolata Hook. e Stevia rebaudiana (Bertoni) Bertoni que inibiram a germinação dos esporos de C. gloeosporioides. Alguns dos extratos ativos também foram submetidos a diluições em solução aquosa de Tween 80 a 1 por cento, o que permitiu observar que, na proporção de 1:2 ou 1:3 (extrato: solução aquosa), os extratos perdiam a atividade antifúngica. Verificou-se também que alguns dos extratos ativos não alteraram suas atividades quando submetidos ao processo de liofilização. Desse modo, identificaram-se várias plantas promissoras na pesquisa por novas substâncias bioativas para o controle de C. gloeosporioides, com destaque para O. manjorona L., que inibiu 96 por cento da germinação dos esporos do referido fungo.

Estudou-se a influência de extratos vegetais no crescimento, in vitro, de plântulas de Rosa x hybrida, objetivando o desenvolvimento de novos reguladores vegetais para a propagação dessa planta ornamental. Extratos metanólicos de Saintpaulia ionantha Wendl. (folhas), Hibiscus rosa-sinensis L. (folhas) e Bougainvillea spectabilis Wild. (folhas e flores) foram obtidos nas concentrações de 150 e 300 mg.L-1. Na ocasião de preparo do extrato todas as plantas se encontravam no período de floração. O experimento foi realizado em delineamento inteiramente casualizado com dez tratamentos e quatro repetições. Dois desses tratamentos foram utilizados como controle: meio de cultura com e sem BAP (6-benzilaminopurina). As variáveis analisadas foram: número e comprimento de brotos, número e comprimento de raízes, e número de folhas por broto. Quanto à variável número de brotos, todos os extratos testados se comportaram como o controle sem o BAP. No que diz respeito ao número e comprimento de raízes, apenas o extrato das folhas de H. rosa-sinensis na concentração de 300 mg.L-1 se mostrou diferente do controle sem o BAP, com valores estatisticamente iguais ao do controle com BAP. Excetuando-se o extrato de H. rosa-sinensis, todos reduziram o número de folhas por broto. Embora os efeitos observados não tenham sido tão pronunciados quanto os causados pelo BAP, os resultados indicam a presença de fitormônios nos extratos estudados.


This work aimed to study the influence of plant extracts on the in vitro growth of Rosa x hybrida plantlets, to contribute to the development of new plant growth regulators to propagate such ornamental plant. Methanolic extracts of Saintpaulia ionantha Wendl. (leaves), Hibiscus rosa-sinensis L. (leaves) and Bougainvillea spectabilis Wild. (leaves and flowers) were employed at 150 and 300 mg.L-1. Flowering plants were used to obtain the extracts. The experiment was carried out in a randomized design, with ten treatments and four repetitions. Culture medium with and without BAP (6-benzylaminopurine) were used as controls. The analyzed variables were shoot number and length, root number and length, and leaves per shoot. Regarding shoot number, all extracts were statistically identical to the control without BAP. When root number and length were taken into account, only the extract of H. rosa-sinensis leaves at 300 mg.L-1 differed from the control without BAP, with values statistically identical to the control with BAP. Except for the H. rosa-sinensis extract, all the others reduced the number of leaves per shoot. Although the observed effects were not as pronounced as those obtained with BAP, these results indicate the presence of plant growth regulators in the studied extracts.

ABSTRACT

1300 ppm (1.3 g / L), water and ethanol extracts prepared from stems or roots of Picrolemma sprucei Hook. f. were lethal (85-90 percent mortality) in vitro to Haemonchus contortus (Barber Pole Worm) larvae, a gastrointestinal nematode parasite found in domestic and wild ruminants. Neosergeolide and isobrucein B were isolated in 0.0083 and 0.0070 percent yield from dry, ground P. sprucei stems (0.89 kg). Neosergeolide, isobrucein B and the anthelmintic drug standard levamisole all caused comparable mortality rates (68-77 percent) in vitro to H. contortus at similar concentrations (81-86 ppm). The anthelmintic activity of P. sprucei infusions (teas), alcohol extracts, and neosergeolide and isobrucein B, has therefore been demonstrated for the first time.


Na concentração de 1300 ppm (1.3 g / L), extratos aquosos e etanólicos preparados a partir dos caules ou raízes de Picrolemma sprucei Hook. f. apresentaram letalidade (85-90 por cento de mortalidade) in vitro para Haemonchus contortus, um nematóide parasítico do aparelho gastrointestinal de ruminantes domesticos e silvestres. Neosergeolida e isobruceina B foram isoladas dos caules em rendimentos de 0.0083 and 0.0070 por cento, respectivamente. Essas últimas e a droga anti-helmíntica levamisole provocaram mortalidade semelhante in vitro (68-77 por cento) em H. contortus em concentrações semelhantes (81-86 ppm). A atividade anti-helmíntica in vitro de infusões e extratos alcoólicos dos caules, bem como da neosergeolida e isobruceina B isoladas de P. sprucei, foi demonstrada pela primeira vez.
