ABSTRACT

This review assessed the effectiveness of fluconazole as antifungal prophylaxis on the incidence of oral fungal diseases in patients undergoing cancer treatment. The secondary outcomes evaluated were the adverse effects, discontinuation of cancer therapy due to oral fungal infection, mortality by a fungal infection, and the mean duration of antifungal prophylaxis. Twelve databases and records were searched. The RoB 2 and ROBINS I tools were used to assess the risk of bias. The relative risk (RR), risk difference, and standard mean difference (SMD) were applied with 95% confidence intervals (CI). The certainty of the evidence was determined by GRADE. Twenty-four studies were included in this systematic review. In randomized controlled trials pooling, fluconazole was a protective factor for the primary outcome (RR = 0.30; CI: 0.16, 0.55; p < 0.01, vs placebo). Compared to other antifungals, fluconazole was only more effective than the subgroup of amphotericin B and nystatin (alone or in combination) (RR = 0.19; CI: 0.09, 0.43; p < 0.01). Fluconazole was also a protective factor in non-randomized trials pooling (RR = 0.19; CI: 0.05, 0.78; p = 0.02, vs untreated). The results showed no significant differences for the secondary outcomes. The certainty of the evidence was low and very low. In conclusion, prophylactic antifungals are necessary during cancer treatment, and fluconazole was shown to be more effective in reducing oral fungal diseases only compared with the subgroup assessing amphotericin B and nystatin, administered alone or in combination.

ABSTRACT

Giant anteater (Myrmecophaga tridactyla) is an endangered species that resides in much of Latin America, but it has been losing its habitat, especially in the Cerrado biome, where it constantly suffers traumas resulting from fires and roadkill. The anatomical knowledge of structures of the respiratory system is important for a better morphophysiological understanding of the species. Thus, this study aimed to perform the macroscopic and histomorphological description of the pharynx and larynx of the giant anteater. Twelve adult giant anteaters were used, three of them fixed in buffered formalin for further dissection and pharynx and larynx macroscopic analysis of structures. From the other animals, samples of the pharynx and larynx were collected and prepared for histological evaluation under optical microscope. Macroscopically, their pharynx and soft palate are extensive, and the anatomical location of these structures and the larynx differs greatly from that described in other species. The larynx, although more caudal, was similar to that of other animals. Histologically, the epithelium of these regions varied between the pseudostratified ciliated columnar and the non-keratinized stratified squamous epithelium. Laryngeal cartilages were composed of elastic (epiglotti) and hyaline cartilages (arytenoid, cricoid and thyroid cartilage), with an ossification process and glandular clusters around the hyaline cartilage. The distinct anatomical location of the pharynx and larynx of Myrmecophaga tridactyla is the main macroscopic finding of this study, besides the length of the pharynx and soft palate of these animals.

ABSTRACT

Pure human and canine mammary invasive micropapillary carcinoma is a rare malignant epithelial tumor accounting for 0.9 to 2% of all invasive mammary carcinomas and present a high rate of lymphatic invasion and metastasis, with unfavorable prognosis. Surgery and chemotherapy are standard treatments for almost all mammary cancer in both species, as well as hormonal and target therapies available for human patients. However, depending on the patient's clinical staging, satisfactory therapeutic results for invasive micropapillary carcinoma are a challenge due to its high capacity of invasion and metastasis. Cyclooxygenase-2 (COX-2) isoform is an important enzyme stimulated by cytokines, growth factors and oncogenes activation to synthetizes prostaglandins in inflammatory process. COX-2 overexpression is associated with angiogenesis and invasion and contributes to cancer development, disease progression, tumor recurrence and regional lymph node metastasis in human and canine mammary carcinomas. This enzyme can be targeted by non-steroidal anti-inflammatory drugs and its inhibition can reduce tumor growth and metastasis in several cancer types. Given the similarity between both species, the present study aims to elucidate the involvement of COX-2 mRNA and protein expression in canine (cIMPC) and human (hIMPC) pure invasive mammary micropapillary carcinoma, with clinicopathological and survival data. Twenty-nine cases of cIMPC and 17 cases of hIMPC were analyzed regarding histologic type, grade, age, tumor size, lymph node condition, extracapsular extension, inflammatory infiltrate and immunophenotype. When available, information on adjuvant treatment, recurrence, metastasis and overall survival were collected. The present study demonstrated COX-2 protein expression in 65.5% of cIMPC and 92.3% of hIMPC, and an association with more advanced histological grades in bitches and higher Ki67 in women. COX-2 mRNA expression was significantly higher in cIMPC than in hIMPC, and its expression was not associated with COX-2 protein expression in both species. COX-2 mRNA expression was associated with negative-ER hIMPC as well as higher Ki67. cIMPC demonstrated proportional early development, more regional metastasis, and a prevalence of negative estrogen receptor, than hIMPC. This is the first time COX-2 expression is associated with negative prognostic factors in both cIMPC and hIMPC, besides the overexpression of COX-2 protein in such unfavorable histological type, which suggests that COX-2 can act as a potential target in IMPC.

ABSTRACT

Candidiasis may affect the central nervous system (CNS), and although Candida albicans is predominant, non-albicans Candida species can also be associated with CNS infections. Some studies have suggested that Candida infections could increase the odds of multiple sclerosis (MS) development. In this context, we investigated whether systemic infection by non-albicans Candida species would affect, clinically or immunologically, the severity of experimental autoimmune encephalomyelitis (EAE), which is an animal model used to study MS. For this, a strain of C. glabrata, C. krusei, and C. parapsilosis was selected and characterized using different in vitro and in vivo models. In these analysis, all the strains exhibited the ability to form biofilms, produce proteolytic enzymes, and cause systemic infections in Galleria mellonella, with C. glabrata being the most virulent species. Next, C57BL/6 mice were infected with strains of C. glabrata, C. krusei, or C. parapsilosis, and 3 days later were immunized with myelin oligodendrocyte glycoprotein to develop EAE. Mice from EAE groups previously infected with C. glabrata and C. krusei developed more severe and more prevalent paralysis, while mice from the EAE group infected with C. parapsilosis developed a disease comparable to non-infected EAE mice. Disease aggravation by C. glabrata and C. krusei strains was concomitant to increased IL-17 and IFN-γ production by splenic cells stimulated with fungi-derived antigens and with increased percentage of T lymphocytes and myeloid cells in the CNS. Analysis of interaction with BV-2 microglial cell line also revealed differences among these strains, in which C. krusei was the strongest activator of microglia concerning the expression of MHC II and CD40 and pro-inflammatory cytokine production. Altogether, these results indicated that the three non-albicans Candida strains were similarly able to reach the CNS but distinct in terms of their effect over EAE development. Whereas C. glabrata and C. Krusei aggravated the development of EAE, C. parapsilosis did not affect its severity. Disease worsening was partially associated to virulence factors in C. glabrata and to a strong activation of microglia in C. krusei infection. In conclusion, systemic infections by non-albicans Candida strains exerted influence on the experimental autoimmune encephalomyelitis in both immunological and clinical aspects, emphasizing their possible relevance in MS development.

ABSTRACT

PURPOSE: The present study aims to record the occurrence of a haemogregarine of the genus Dactylosoma in anurans in the eastern Brazilian Amazon. METHODS: Specimens of Leptodactylus macrosternum, Rhinella major, and Rhinella marina were collected in the municipality of Santarém, west of the State of Pará, in Brazil. Parasitic forms identified as Dactylosoma sp. were recorded by light microscopy, with magnification of 400 and 1000 X. RESULTS: Aspects about the morphology and morphometry of Dactylosoma sp. are presented. The mean intensity of infection was 200,000 parasites/mL in R. major and 50,000 parasites/mL in R. marina. CONCLUSION: The present study expands the occurrence of parasites of the genus Dactylosoma in Brazil, presenting a new anuran host for this parasite in the Brazilian Amazon region.

ABSTRACT

Giant anteater (Myrmecophaga tridactyla) is a wild mammal distributed in Central and South America; nowadays, it is classified as an endangered species. Research about the macroscopic and histomorphological aspects of its respiratory tract is scarce, and, sometimes, it limits the treatment provided to sick animals and impairs species preservation. Thus, the present study aims to describe the macroscopic and microscopic morphology of its lower respiratory tract, including trachea and lungs. To do so, 12 adult giant anteaters from "Centro de Triagem de Animais Silvestres de Goiânia" (CETAS-GO), Goiás State, Brazil, were used in the research after natural death or euthanasia. Three of these animals were used for macroscopic assessments; they were fixed in 10% buffered formalin and dissected. Trachea and lung tissue samples were collected from nine animals right after death and fixed in 10% buffered formalin for histomorphological analysis; they were processed, embedded in paraffin, and inked with hematoxylin-eosin (HE), periodic acid-Schiff (PAS), and Masson's trichrome. The macroscopic analysis showed that the trachea in this species is proportionally short and presents from 19 to 27 tracheal cartilages. The right lung presents four lobes and the left one, two. The microscopic analysis evidenced respiratory epithelium of the ciliated cylindrical pseudostratified type, without evident goblet cells in the mucosa layer of the trachea and bronchi. The pulmonary visceral pleura is thick, similar to other large domestic mammals - complete septa extend from the pulmonary visceral pleura. In conclusion, the macroscopy and histomorphology of giant anteater's lower respiratory tract, represented by trachea and lungs, are similar to that of other domestic and wild mammals. Pulmonary histomorphology is mainly similar to that of pigs and ruminants: it has thick visceral pleura that emits complete septa of conjunctive tissue, which enable lobular parenchymal architecture.(AU)


O tamanduá-bandeira (Myrmecophaga tridactyla) é um mamífero silvestre com distribuição na América Central e do Sul e, atualmente, encontra-se classificado como ameaçado de extinção. Pesquisas acerca dos aspectos macroscópicos e histomorfológicos do seu sistema respiratório são escassas, o que, por vezes, limita o tratamento e o manejo de eventuais animais doentes, bem como a preservação da espécie. Desse modo, o objetivo deste estudo foi descrever a morfologia macroscópica e microscópica do aparelho respiratório inferior do tamanduá-bandeira, incluindo traqueia e pulmões. Para tanto, foram utilizados 12 tamanduás-bandeiras adultos, provenientes do Centro de Triagem de Animais Silvestres de Goiânia (CETAS-GO), Goiás, Brasil, após morte natural ou eutanásia. Destes, três foram utilizados para o estudo macroscópico, sendo fixados em formalina tamponada a 10% e dissecados. Para a análise histomorfológica, amostras teciduais da traqueia e do pulmão foram colhidas de nove animais logo após o óbito, fixadas em formalina tamponada a 10%, processadas, incluídas em parafina e coradas com hematoxilina e eosina (HE), ácido periódico-Schiff (PAS) e tricrômico de Masson. À análise macroscópica notou-se que a traqueia é proporcionalmente curta, apresentando 19 a 27 cartilagens traqueais. O pulmão direito apresenta quatro lobos e o esquerdo dois. À análise microscópica foi constatado epitélio respiratório do tipo pseudoestratificado cilíndrico ciliado, sem células caliciformes evidentes na camada mucosa da traqueia e dos brônquios. A pleura visceral pulmonar é espessa, assim como nos grandes mamíferos domésticos, e, a partir desta, estendem-se septos completos. Conclui-se que a macroscopia e a histomorfologia do sistema respiratório inferior do tamanduá-bandeira, representado pela traqueia e pulmões, são semelhantes àquelas de outros mamíferos domésticos e silvestres. A histomorfologia pulmonar é especialmente semelhante à de suínos e ruminantes, com a pleura visceral espessa e emitindo septos completos de tecido conjuntivo, que conferem arquitetura parenquimal lobular.(AU)

ABSTRACT

Multiple sclerosis (MS) is an inflammatory and demyelinating disease of the central nervous system (CNS). The persistent inflammation is being mainly attributed to local oxidative stress and inflammasome activation implicated in the ensuing demyelination and axonal damage. Since new control measures remain necessary, we evaluated the preventive and therapeutic potential of a beta-selenium-lactic acid derivative (LAD-ßSe), which is a source of organic selenium under development, to control experimental autoimmune encephalomyelitis (EAE) that is an animal model for MS. Two EAE murine models: C57BL/6 and SJL/J immunized with myelin oligodendrocyte glycoprotein and proteolipid protein, respectively, and a model of neurodegeneration induced by LPS in male C57BL/6 mice were used. The preventive potential of LAD-ßSe was initially tested in C57BL/6 mice, the chronic MS model, by three different protocols that were started 14 days before or 1 or 7 days after EAE induction and were extended until the acute disease phase. These three procedures were denominated preventive therapy -14 days, 1 day, and 7 days, respectively. LAD-ßSe administration significantly controlled clinical EAE development without triggering overt hepatic and renal dysfunction. In addition of a tolerogenic profile in dendritic cells from the mesenteric lymph nodes, LAD-ßSe also downregulated cell amount, activation status of macrophages and microglia, NLRP3 (NOD-like receptors) inflammasome activation and other pro-inflammatory parameters in the CNS. The high Se levels found in the CNS suggested that the product crossed the blood-brain barrier having a possible local effect. The hypothesis that LAD-ßSe was acting locally was then confirmed by using the LPS-induced neurodegeneration model that also displayed Se accumulation and downmodulation of pro-inflammatory parameters in the CNS. Remarkably, therapy with LAD-ßSe soon after the first remitting episode in SJL/J mice, also significantly downmodulated local inflammation and clinical disease severity. This study indicates that LAD-ßSe, and possibly other derivatives containing Se, are able to reach the CNS and have the potential to be used as preventive and therapeutic measures in distinct clinical forms of MS.

ABSTRACT

This study aimed to evaluate and compare the effects of ozonized solutions on tissue wound repair in rats. Treatments consisted of ozonized water (GA), 0.9% sodium chloride (GCL), ozonized oil (GO), and 0.2% allantoin cream (GAL). The morphometric evaluation showed that wounds of the GA group presented a higher degree of retraction (p<0.05) at three and eight days of treatment (37.96 and 84.81%, respectively). Picrosirius red staining showed that groups GA and GO presented higher deposition (p<0.05) of type I collagen at 15 and 22 days of treatment, respectively. The neovascularization was higher in wounds of group GO on days 3, 8, and 15 (p<0.05), with higher VEGF immunostaining. (p<0.05). Thus, ozonized water enhances wound retraction and assists in the maturation and remodeling phase, while ozonized oil promotes higher neovascularization during tissue repair and higher deposition of type I collagen from the third week of treatment.(AU)


O objetivo deste estudo foi avaliar e comparar os efeitos de soluções ozonizadas sobre o reparo tecidual de feridas em ratos. Foram realizados os tratamentos: água ozonizada (GA), cloreto de sódio 0,9% (GCL), óleo ozonizado (GO) e creme de alantoína 0,2% (GAL). À avaliação morfométrica, as feridas do grupo GA apresentaram maior grau de retração (p<0,05) aos três e oito dias de tratamento (37,96% e 84,81%, respectivamente). A coloração de picrosirius red mostrou que os grupos GA e GO apresentaram maior deposição (p<0,05) de colágeno do tipo I aos 15 e aos 22 dias de tratamento, respectivamente. Já a variável neovascularização foi maior (p<0,05) nas feridas do grupo GO nos dias três, oito e 15, o que fora ratificado à imunoistoquímica, com maior imunomarcação de VEGF nas feridas do grupo GO (p<0,05). Conclui-se que a água ozonizada potencializa a retração da ferida e auxilia na fase de maturação e remodelamento, enquanto o óleo ozonizado promove maior neovascularização durante o reparo tecidual e maior deposição de colágeno do tipo I a partir da terceira semana de tratamento.(AU)

ABSTRACT

O desenho é uma forma de expressão que vai muito além do lápis e papel. Muitos artefatos e materiais foram utilizados pelo homem ao longo do tempo para registrar sua história. Por meio de pesquisa bibliográfica, objetivou-se, com o presente trabalho, apresentar uma nova teoria sobre o desenho infantil, denominada como Perspectiva Dinâmica e Estética. Essa perspectiva divide o desenvolvimento do grafismo infantil em estágios, como as teorias clássicas, mas se diferencia delas quando propõe que a expressão de estados emocionais, por meio do rabisco da criança, já é possível por volta dos dois-três anos de idade, na fase chamada rabisco expressivo. O rabisco é mais que a consequência do gesto, não é motivado somente pelo prazer motor e parece ser capaz de expressar estados emocionais. A Perspectiva Dinâmica e Estética integra funcionalmente aspectos biológicos, psicológicos e sociais na descrição de um processo fluído na evolução do desenho. AU


Drawing is a form of expression that goes far beyond pencil and paper. Many artifacts and materials have been used by man over time to record our history. By means of a bibliographical study, the aim of this work was to present a new theory about childish drawing called the Dynamic and Aesthetic Perspective. This perspective divides the development of childish drawing into stages, in the same way as the classical theories, however, differs from them when it proposes that the expression of emotional states through the child's scribble is already possible at around 2-3 years of age, in the phase called expressive scribbling. The scribble is more than the gesture's consequence; it is not motivated solely by motor pleasure and seems to be able to express emotional states. The Dynamic and Aesthetic Perspective functionally integrates biological, psychological, and social aspects into the description of a fluid process in the evolution of the drawing. AU


El dibujo es una forma de expresión que va mucho más allá del lápiz y el papel. Muchos artefactos y materiales fueron utilizados por el hombre a lo largo del tiempo para registrar su historia. Son el presente trabajo se objetivó, por medio de investigación bibliográfica, presentar una nueva teoría sobre el dibujo infantil denominada Perspectiva Dinámica y Estética. Esta perspectiva divide el desarrollo de la expresión gráfica infantil en etapas, como las teorías clásicas, pero se diferencia de ellas cuando propone que la expresión de estados emocionales, a través del garabato de los niños, ya es posible alrededor de los dos-tres años, en la fase denominada como garabato expresivo. El garabato es más que la consecuencia del gesto, no es motivado solamente por el placer motor y parece ser capaz de expresar estados emocionales. La Perspectiva Dinámica y Estética integra funcionalmente aspectos biológicos, psicológicos y sociales en la descripción de un proceso fluido en la evolución del dibujo. AU

ABSTRACT

Por meio de revisão integrativa de literatura, objetivou-se discutir a utilização do desenho na prática clínica em Gestalt-terapia. Consta do final dos anos de 1800 o primeiro estudo sobre os desenhos infantis. As teorias clássicas descrevem o desenvolvimento do grafismo por meio de estágios que são superados à medida que a criança se desenvolve. Na ciência psicológica, inicialmente, o desenho foi utilizado em testes psicológicos para medir aspectos cognitivos e, posteriormente, como técnica projetiva. Atualmente, ele também é utilizado como um meio para a investigação diagnóstica em diversas abordagens teóricas. Na Gestalt-terapia, o desenho pode ser utilizado como técnica, experimento e instrumento para o estabelecimento de vínculo, fundamentado no método fenomenológico. O desenho é uma importante ferramenta terapêutica que ao ser utilizada dentro de um processo psicoterapêutico consistente, fundamentado no vínculo de confiança e pautado no arcabouço teórico da Gestalt-terapia, é capaz de contribuir para o processo de atualização e de crescimento do cliente. Through an integrative literature review, the goal is to discuss the use of drawings on the clinical practice of Gestalt-therapy. The first study regarding children's drawings refers to the ending of the 1800's. The classical theories describe the development of the graphism through stages that are overcome as the child develops. In psychological science, initially, the drawing was used in psychological tests to measure cognitive aspects, and, further on, as a projective technique. Currently, it is also used as a manner of investigations for diagnosis in various theories. In Gestalt-therapy the drawing can be used as a technique, an experiment and an instrument to establish bonding, based on the phenomenological method. The drawing is an important therapeutic tool that, when used within a consistent psychotherapeutic process, founded on a tie of trust and based on the theoretical background of Gestalt-therapy, is capable of contributing to the update and growth process of the client. Por medio de una revisión integradora de literatura, se objetivó discutir la utilización del dibujo en la práctica en clínica en terapia Gestalt. Se pudo encontrar el primer estudio cerca los dibujos infantiles al final de los 1800. Las teorías clásicas describen el desarrollo del grafismo a través de etapas que son rebasadas con el desarrollo de los niños. En la ciencia psicológica, inicialmente, el dibujo fue utilizado en pruebas psicológicas, para medir aspectos cognitivos y, después, como una técnica de proyección. Actualmente, también se pone como una forma de investigación diagnóstica en las teorías diversas. En la terapia Gestalt, el dibujo puede ser usado como técnica, para experimentación y instrumento de formación de vínculo, fundamentado por el método fenomenológico. El dibujo es una herramienta significativa que, cuando utilizada en un proceso psicoterápico consistente, fundamentado en un vínculo de confianza y basado en la teoría de la terapia Gestalt, puede contribuir con el proceso de actualización y crecimiento del cliente. ABSTRACT: The objective of this study was to evaluate the effect of digestible methionine and cystine (Met + Cys) levels on the hematological and serum biochemical parameters of broiler chickens during the initial and growth stages. For this, 1,800 male chicks of the Coob 500 strain were used, with 900 chicks in the initial phase (1 to 21 days old) and 900 chicks in the growth phase (22 to 42 days old), distributed in a completely randomized design of five treatments with six replicates of 30 birds. The treatments consisted of 0.545, 0.616, 0.711, 0.782, and 0.853%; and 0.514, 0.571, 0.647, 0.704, and 0.761% digestible Met + Cys for "1 to 21" and "22 to 42" days of breeding, respectively. Results showed that digestible Met + Cys levels in broiler feed altered some hematological parameters (erythrocyte, hematocrit hemoglobin, total leukocytes, heterophile: lymphocyte) and serum biochemistry (uric acid, PST, total LDL, and TG). The digestible Met + Cys levels in the diet of broilers affected the hematological parameters and serum biochemistry, especially at higher levels. From the inclusion level 0.761 of Met + Cist in the broiler diet, red blood cells, hemoglobin and hematocrit changes begin to appear.


RESUMO: Teve-se como objetivos neste estudo avaliar o efeito dos níveis de metionina + cistina (Met+Cis) digestível na dieta sobre os parâmetros hematológicos e bioquímicos séricos de frangos de corte na fase inicial e de crescimento. Para isso, utilizou-se 1.800 pintos, machos, da linhagem Coob 500, sendo 900 pintos para a fase inicial (1 a 21 dias de idade) e mais 900 pintos para a fase de crescimento (22 a 42 dias de idade), distribuídos em delineamento inteiramente casualizado composto de cinco tratamentos, com seis repetições de 30 aves. Os tratamentos consistiram em 0,545; 0,616; 0,711; 0,782 e 0,853% e 0,514; 0,571; 0,647; 0,704 e 0,761% de Met+Cis digestíveis para a fase inicial e de crescimento, respectivamente. Os resultados demonstraram que níveis de Met+Cis digestível na alimentação de frango de corte, acarretam alterações em parâmetros hematológicos como: hemácia, hematócrito, hemoglobina, leucócitos totais, relação heterófilo: linfócito; e bioquímico sérico como: ácido úrico, albumina, colesterol total, proteínas séricas totais (PST), lipoproteína de baixa densidade (LDL) e triglicerídeos (TG). Os níveis digestíveis de Met + Cys na dieta de frangos de corte afetam os parâmetros hematológicos e a bioquímica sérica, principalmente em níveis mais elevados. A partir do nível de inclusão 0.761 de Met + Cist na dieta de frangos de corte, começam a aparecer alterações nos glóbulos vermelhos, hemoglobina e hematócrito.

This product is traditionally used in the baking industry for the manufacture of biscuits and Brazilian cheese breads. However, the end of fermentation is evaluated empirically, and the process occurs without standardization, which results in products of inconsistent quality. Predominant microbiota from a cassava flour manufacturer was isolated in order to select starter cultures for the production of sour cassava starch in a pilot-scale fermentation process. Lactic acid bacteria and yeasts were isolated, enumerated and grouped by Restriction Fragment Length Polymorphism, and PCR fingerprinting, respectively. One isolate of each molecular profile was identified by sequencing of the rRNA gene. LAB were prevalent throughout the entire process. Lactobacillus brevis (21.5%), which produced the highest values of acidity, and Lactobacillus plantarum (13.9%) were among the most frequent species. Pichia scutulata (52.2%) was the prevalent yeast and showed amylolytic activity. The aforementioned species were tested as single and mixed starter cultures in a pilot-scale fermentation process for 28 days. L. plantarum exhibited better performance as a starter culture, which suggests its potential for the production of sour cassava starch.

The giant anteater (Myrmecophaga tridactyla) is classified as a vulnerable species on Brazil's list of species at risk of extinction mainly due to deforestation and forest fires. This has contributed to a considerable increase in detailed clinical case records of the treatment of wild species at veterinary institutions. However, the paucity of serum biochemical profiles of healthy giant anteaters has made it difficult to evaluate these animals, preventing diagnosis, treatment and prognosis. The objective of this work was to collect data about the biochemical profile of healthy giant anteaters from the Brazilian Cerrado raised in captivity, in order to better understand the physiological characteristics inherent to this species. Eighteen analytes from 12 healthy giant anteaters were measured. The following means and standard deviations were found in the biochemical analyses: albumin 3.29±0.33g/dL, ALT 15.49±7.98 IU/L, amylase 1037.92±149.04 IU/L, AST 21, 12±7.50 IU/L, total cholesterol 62.79±20.08mg/dL, HDL cholesterol 14.73±4.98mg/dL, LDL cholesterol 26.60±11.05mg/dL, VLDL cholesterol 2.14±1.06mg/dL, CK 111.61±70.16 IU/L, creatinine 1.05±0.37mg/dL, iron 194.64±81.17µg/dL, GGT 65.18±54.57 IU/L, glucose 103.71±29.63mg/dL, globulins 2.76±0.36g/dL, lipase 28.80±5.11 IU/L,TSP 6.05±0.56g/dL, triglycerides 10.71±5.29mg/dL, and urea 53.46±18.28mg/dL. The values found in this study can be used as references for the laboratory evaluation of giant anteaters living in conditions similar to those of this study. This is one of the first reports of biochemical examinations on giant anteaters of the Cerrado biome.(AU)


O tamanduá-bandeira (Myrmecophaga tridactyla) está classificado como espécie vulnerável na lista brasileira de espécies ameaçadas de extinção devido principalmente ao desmatamento e aos incêndios florestais. Tal fato contribuiu com o aumento da casuística de atendimento de espécies silvestres em instituições veterinárias. Porém, a escassez de valores bioquímicos séricos em tamanduás-bandeiras hígidos tem dificultado a avaliação destes animais, impedindo o diagnóstico, tratamento e prognóstico. O objetivo deste trabalho foi fornecer dados sobre o perfil bioquímico de tamanduás-bandeiras saudáveis do cerrado brasileiro, criados em cativeiro, a fim de compreender melhor as características fisiológicas inerentes a esta espécie. Foram mensurados 18 analitos de 12 tamanduás-bandeiras hígidos. As médias e o desvio padrão correspondentes às análises bioquímicas foram: albumina 3,29±0,33g/dL; ALT 15,49±7,98 UI/L; amilase 1037,92±149,04 UI/L; AST 21, 12±7,50 UI/L; colesterol total 62,79±20,08mg/dL; colesterol HDL 14,73±4,98mg/dL; colesterol LDL 26,60±11,05mg/dL; colesterol VLDL 2,14±1,06mg/dL; CK 111,61±70,16 UI/L; creatinina 1,05±0,37mg/dL; ferro 194,64±81,17µg/dL; GGT 65,18±54,57 UI/L; glicose 103,71±29,63mg/dL; globulinas 2,76±0,36g/dL; lipase 28,80±5,11 UI/L; PST 6,05±0,56g/dL; triglicerídeos 10,71±5,29mg/dL; ureia 53,46±18,28mg/dL. ABSTRACT

Giant Anteater (Myrmecophaga tridactyla) is a vulnerable species because of progressive habitat destruction, mostly affected by wildfires and car accidents. The increasing number of animals that are attended by wildlife rescue centres reinforces the need of knowledge about haematological parameters, especially for those that inhabit Brazilian cerrado biome. For this purpose and in order to establish reference values for this species and also to compare them with previous studies, haematological analysis of captive giant anteaters from Brazilian cerrado were performed. Moreover, the alterations of blood samples after 24 and 48 hours of storage at refrigeration temperatures (4oC) and preserved with two different EDTA concentrations (5% and 10%) were studied. Means and standard deviations of haematological parameters analysed immediately after collection were: RBC: 2,07 x106/µL ± 0,40; hematocrit: 38,08%± 5,93; haemoglobin: 11.33g/dL±2.15; MCV:186.52 fL±21.72; MCHC: 29.68g/dL±2.56; MCH: 55.08pcg±5,94; total leucocytes: 8.142/µL±2.441; neutrophils: 5.913/µL±2.168; lymphocytes: 1.460/µL±740; eosinophil: 522/µL±385; monocytes: 247/µL±176; thrombocytes: 123.458/µL±31.362 and total plasma protein: 6.23g/dL±0.49. This data shows evidence of the existence of important differences between these values and others from other areas, either from Brazil or from other South American countries. Those variations might be connected to environment, genetic, nutritional and/or management factors. Regarding the storage effect analysis, it can be concluded that in giant anteaters, haematological analysis can be performed until 24h after collection without any significant alterations on the haematological parameters, except for thrombocytes. Concerning the different EDTA concentrations, it can be concluded that there are no quantitative differences in haematological variables. Nevertheless, relevant morphologic alterations in blood cells can be observed after a 24h storage period, being most noticeable in the leucocytes. Those alterations can lead to misinterpretation of the results, interfering diagnosis, prognosis and treatment.(AU)


O tamanduá-bandeira (Myrmecophaga tridactyla) é uma espécie vulnerável devido à destruição progressiva do seu habitat natural, sendo afetado por queimadas e atropelamentos. O aumento na casuística de atendimentos de animais silvestres reforça a necessidade de se ter conhecimento dos parâmetros hematológicos, em especial para os que vivem no bioma do cerrado. Por isso, este trabalho teve por objetivos realizar o hemograma de tamanduás-bandeiras de cativeiro localizados no cerrado brasileiro, a fim de estabelecer valores de referência para essa espécie e compará-los a estudos prévios. Além disso, verificar quais alterações podem ser encontradas em amostras armazenadas por 24 e 48 horas após a colheita, em temperatura de refrigeração (4oC) e tratadas com duas concentrações distintas de EDTA (5% e 10%). A média e o desvio padrão das variáveis hematológicas encontradas nas amostras processadas logo após a colheita foram: hemácias (2,07x106/µL±0,40); volume globular (38,08%± 5,93); hemoglobina (11,33g/dL±2,15); VCM (186,52 fL±21,72); CHCM (29,68g/dL±2,56); HCM (55,08pcg±5,94); leucócitos totais (8.142/µL±2.441); neutrófilos (5.913/µL±2.168); linfócitos (1.460/µL±740); eosinófilos (522/µL±385); monócitos (247/µL±176); plaquetas (123.458/µL±31.362) e proteínas plasmáticas totais (6,23g/dL±0,49). Tais dados permitem afirmar que existem importantes diferenças entre os valores hematológicos destes em relação aos animais provenientes de outras regiões, tanto do Brasil quanto de outros países da América do Sul. Provavelmente, tais divergências estão associadas a fatores ambientais, genéticos, nutricionais e/ou de manejo. Quanto à análise das amostras estocadas, conclui-se que, em tamanduás-bandeiras, as amostras para a realização de hemograma podem ser processadas até 24 horas após a colheita, sem alteração significativa das variáveis hematológicas, com exceção das plaquetas. Com relação às duas concentrações de EDTA, pode-se inferir que não há diferença quantitativa entre ambas para as variáveis hematológicas. Contudo, é possível constatar que o EDTA promove alterações morfológicas relevantes nas células sanguíneas após 24 horas de armazenamento, sendo os leucócitos os mais afetados. Tais alterações, quando relatadas, podem induzir interpretações equivocadas, interferindo no diagnóstico, prognóstico e tratamento.(AU)

The wild grass species Brachypodium distachyon (L.) has been proposed as a new model for temperate grasses. Among the biotechnological tools already developed for the species, an efficient induction protocol of somatic embryogenesis (SE) using immature zygotic embryos has provided the basis for genetic transformation studies. However, a systematic work to better understanding the basic cellular and molecular mechanisms that underlie the SE process of this grass species is still missing. Here, we present new insights at the morpho-histological, histochemical, and molecular aspects of B. distachyon SE pathway. Somatic embryos arose from embryogenic callus formed by cells derived from the protodermal-dividing cells of the scutellum. These protodermal cells showed typical meristematic features and high protein accumulation which were interpreted as the first observable steps towards the acquisition of a competent state. Starch content decreased along embryogenic callus differentiation supporting the idea that carbohydrate reserves are essential to morphogenetic processes. Interestingly, starch accumulation was also observed at late stages of SE process. Searches in databanks revealed three sequences available annotated as BdSERK, being two copies corresponding to SERK1 and one showing greater identity to SERK2. In silico analysis confirmed the presence of characteristic domains in a B. distachyon Somatic Embryogenesis Receptor Kinase genes candidates (BdSERKs), which suggests SERK functions are conserved in B. distachyon. In situ hybridization demonstrated the presence of transcripts of BdSERK1 in all development since globular until scutellar stages. The results reported in this study convey important information about the morphogenetic events in the embryogenic pathway which has been lacking in B. distachyon. This study also demonstrates that B. distachyon provides a useful model system for investigating the genetic regulation of SE in grass species.

This study assessed the efficiency of Scheffersomyces amazonensis UFMG-CM-Y493T, cultured in xylose-supplemented medium (YPX) and rice hull hydrolysate (RHH), to convert xylose to xylitol under moderate and severe oxygen limitation. The highest xylitol yields of 0.75 and 1.04 g g-1 in YPX and RHH, respectively, were obtained under severe oxygen limitation. However, volumetric productivity in RHH was ninefold decrease than that in YPX medium. The xylose reductase (XR) and xylitol dehydrogenase (XDH) activities in the YPX cultures were strictly dependent on NADPH and NAD+ respectively, and were approximately 10% higher under severe oxygen limitation than under moderate oxygen limitation. This higher xylitol production observed under severe oxygen limitation can be attributed to the higher XR activity and shortage of the NAD+ needed by XDH. These results suggest that Sc. amazonensis UFMG-CM-Y493T is one of the greatest xylitol producers described to date and reveal its potential use in the biotechnological production of xylitol.

ABSTRACT

Three strains of a new xylanase-producing yeast species were isolated from rotting wood samples collected in the Atlantic Rain Forest of Brazil. The sequences of the internal transcribed spacer region and D1/D2 domains of the large subunit of the rRNA gene showed that this novel yeast species belongs to the genus Spencermartinsiella, and its closest relatives among recognized species are Spencermartinsiella europaea and Spencermartinsiella ligniputridi. A novel species, named Spencermartinsiella silvicola sp. nov., is proposed to accommodate these isolates. The type strain is UFMG-CM-Y274T ( = CBS 13490T). The MycoBank number is MB 813053. In addition, Candida cellulosicola is reassigned to the genus Spencermartinsiella as a new combination.

BACKGROUND/AIM: Soft tissue injuries (STI) are common in children and adolescents. This retrospective study evaluated the frequency of STI, gender, type of injury, and its location in pediatric patients, comparing the different age groups and how the presence of these lesions influences the pursuit for immediate care. MATERIAL AND METHODS: Medical records of patients 0-15 years old who had been treated between 2005 and 2013 at the Dental Trauma Surveillance Center, School of Dentistry, Universidade Federal do Rio de Janeiro were included in this study. Records with incomplete data and those in which the patient took more than 2 weeks to seek attendance were excluded. The data collected included the age and gender of the patient, the frequency, type and location of the injury, and immediate treatment (<24 h). Data were analyzed descriptively and with the chi-square test (P ≤ 0.05). RESULTS: Of the 543 patients who met the eligibility criteria, 56.2% had STI, and males (65.6%), and the age group of 0-3 years (39.7%) were the most affected. The concomitant presence of lesions in the intra- and extra-oral region was the most frequent in all age groups. In the extra-oral region, contusion (20.7%) and abrasion (19.3%) were more common to the upper lip and laceration to the lower lip (18.4%). In the intra-oral region, the injury was more frequent to the gums (41%), while the lips were the most affected by abrasion (12.2%) and laceration (24.9%). Regardless of the age groups, the pursuit for immediate care was statistically influenced by the presence of soft tissue lesions (P < 0.05). CONCLUSION: We conclude that there is a high prevalence of STI in the study sample; the lesions were more common in boys and in patients aged 0-3 years. The presence of STI proved to be a determining factor for the pursuit for immediate care.

Objetivo: caracterizar as alterações citopatológicas cervicais e o perfil epidemiológico de mulheres com vida sexual ativa. Método: estudo quantitativo, descritivo, analítico, de corte transversal e base documental retrospectiva, realizado no município de Campo Grande/MS. Foram analisados os resultados dos exames citopatológicos referentes ao período de janeiro de 2010 a 2011. Os dados foram coletados com instrumento próprio. O projeto de pesquisa teve a aprovação pelo Comitê de Ética em Pesquisa, protocolo nº 063/11. Resultados: obteve-se a amostra de 527 exames. As alterações citopatológicas foram encontradas nas pacientes mais jovens, com menor nível de escolaridade. A afecção mais encontrada foi a Inflamação (22,5%) e os agentes mais comuns foram Cocos, Bacilos e Lactobacilos. Conclusão: os profissionais de saúde devem ser preparados para atender o público feminino em suas especificidades, bem como estimular a realização do exame Papanicolaou.(AU)
