ABSTRACT

OBJECTIVE: To understand Primary Health Care nurses' role in treating Lower Urinary Tract Dysfunction. METHOD: Cross-sectional multi-methodological research, composed of quantitative and qualitative steps, independently and sequentially. Data collected remotely, through a questionnaire and focus group, analyzed using descriptive statistics and thematic analysis by Braun and Clarke, respectively. The project was approved under Opinion 22691119.0.0000.0030. RESULTS: A total of 145 nurses participated in the study in the quantitative step and 20 in the qualitative step, working in Primary Health Care in Brazil. Of the 93.1% nurses who reported having already cared for people with Urinary Tract Dysfunction, only 54.4% provided guidance, mainly for training the pelvic floor muscles. CONCLUSION: Even though they have legal support and access to demand, nurses do not have the knowledge to offer conservative treatment for Lower Urinary Tract Dysfunction. Despite this, they were motivated to do so as long as they received specific training.

ABSTRACT

In this study, we investigated the effects of the Payment for Ecosystem Services (PES) in an agrisystem located in the Brazilian Midwest. This PES benefits the owners of rural properties that encompass springs that feed the Abóbora River microbasin which supplies water for consumption in the city of Rio Verde, Goiás. We evaluated the percentage of native vegetation cover around the springs of the watercourses and also estimated its change over time (2005, 2011 and 2017). The vegetation cover of the Areas of Permanent Preservation (APP) increased by 2.24% on average, seven years after the implantation of the PES. There was little difference in the change of vegetation cover maintained over the study years (2005, 2011, and 2017), however, the vegetation cover increased for 17 springs, decreased for 11 springs, and was completely degraded for other two. To improve the performance of this PES, we recommend (1) expanding the program to include the APPs surrounding the springs, together with the legal reserves of each property, (2) implementing measures to ensure that properties are environmentally adequate, (3) including the properties in the Brazilian Rural Environment Register (CAR), and (4) obtaining environmental licensing for the activities undertaken within the Abóbora River basin.

ABSTRACT

Triple-negative breast cancer (TNBC) is an aggressive, fast-growing tumor that is more likely to spread to distant organs. Among women diagnosed with breast cancer, the prevalence of TNBC is 20%, and treatment is currently limited to chemotherapy. Selenium (Se), an essential micronutrient, has been explored as an antiproliferative agent. Therefore, this study aimed to evaluate the effects of exposure to organic (selenomethionine, ebselen, and diphenyl diselenide) and inorganic (sodium selenate and sodium selenite) Se molecules in different breast cell lines. The compounds were tested at 1, 10, 50, and 100 µM for 48 h in the non-tumor breast cell line (MCF-10A) and TNBC derivatives cell lines (BT-549 and MDA-MB-231). The effects of Se on cell viability, apoptotic and necrotic processes, colony formation, and cell migration were analyzed. Exposure to selenomethionine and selenate did not alter the evaluated parameters. However, selenomethionine had the highest selectivity index (SI). The exposure to the highest doses of selenite, ebselen, and diphenyl diselenide resulted in antiproliferative and antimetastatic effects. Selenite had a high SI to the BT cell line; however, the SI of ebselen and diphenyl diselenide was low in both tumoral cell lines. In conclusion, the Se compounds had different effects on the breast cell lines, and additional tests are needed to reveal the antiproliferative effects of Se compounds.

ABSTRACT

Regarding free genistein small delivery to the central nervous system, physico-chemical parameters of dimiristoylphosphatidylcholine liposome-loaded genistein were investigated, as well as its in vitro activity against the DPPH radical and glioma cells. Data obtained by UV-vis spectroscopy, Fourier Transform Infrared Spectroscopy, Nuclear Magnetic Resonance, Differential Scanning Calorimetry and Dynamic Light Scattering were used to characterize the liposomal system with respect to motion restriction, hydration degree, trans-gauche isomerization and phase state. In vitro antitumoral effects were monitored through conting and viability assays. Genistein hydroxyl group and lipid hydrogen bonds may have important role in dimiristoylphosphatidylcholine phosphate and choline motion restriction. Genistein-induced choline restriction may be also related to a decrease in the group rotation rate. Genistein: dimiristoylphosphatidylcholine system showed higher molecular package at the acyl chains region compaired to empty liposomes, and it may be related to a decrease in gauche bonds quantity and system size. Lipid acyl chain length seems to influence different genistein effects on membranes, due to the presence of gauche conformers. Genistein: dimiristoylphosphatidylcholine liposome was more efficient as DPPH reducting system than the free-Gen. Liposomal system, at genistein 100 µM, was so efficient as the correspondent free-form genistein, probably showing higher stability to cross the blood-brain barrier. Genistein and the lipid did not show an additive activity against glioma cells. Antioxidant and anti-glioma genistein-loaded liposome potential may be related to the isoflavone location and its restriction effect in the lipid molecular motion. Anti-glioma activity may also be related to a decrease of system size and trans-gauche isomerization.

ABSTRACT

INTRODUCTION: Physical training has been recommended to improve overall well-being in patients with fibromyalgia. Body relaxation exercises also seem to have some beneficial effect, however there is no consensus regarding this modality. OBJECTIVE: Comparing the effectiveness of sophrology and resistance training in improving the pain of women with fibromyalgia. METHOD: A randomized controlled clinical trial with a blind evaluator. Sixty (60) women with a medical diagnosis of fibromyalgia were randomized and included in two groups: sophrology group (SG) who participated in a relaxation program based on sophrology (n = 30), and resistance group (RG) (n = 30) who participated in a resistance training program for biceps, pectoral, triceps, knee extensors, trapezius, knee flexors, hip abductors. Both groups were treated twice a week for 12 weeks and reevaluated every 4 weeks. The assessment instruments used were the Visual Analog Scale for Pain (VAS), the one-repetition maximum test (1 RM), the overall quality of life (SF-36), the 6-min walk test (6MWT), the Timed Up and Go test (TUG) and the Fibromyalgia Impact Questionnaire (FIQ). RESULTS: We found that the RG presented statistically significant decreases in pain (VAS) during the evaluations (p < 0.05) and increased strength of the evaluated muscles (p < 0.05). A statistically significant decrease in pain (p < 0.05) was observed in the SG compared to T0, with no significant differences in muscle strength. Differences between groups were observed, with better indices only for 6MWT and functional capacity domain of the SF36 for the RG (p < 0.05). CONCLUSION: No differences in pain were found between the groups. Resistance training was more effective than sophrology in improving strength and functional capacity of women with fibromyalgia.

ABSTRACT

Auxin modulates a range of plant developmental processes including embryogenesis, organogenesis, and shoot and root development. Recent studies have shown that plant hormones also strongly influence metabolic networks, which results in altered growth phenotypes. Modulating auxin signalling pathways may therefore provide an opportunity to alter crop performance. Here, we performed a detailed physiological and metabolic characterization of tomato (Solanum lycopersicum) mutants with either increased (entire) or reduced (diageotropica-dgt) auxin signalling to investigate the consequences of altered auxin signalling on photosynthesis, water use, and primary metabolism. We show that reduced auxin sensitivity in dgt led to anatomical and physiological modifications, including altered stomatal distribution along the leaf blade and reduced stomatal conductance, resulting in clear reductions in both photosynthesis and water loss in detached leaves. By contrast, plants with higher auxin sensitivity (entire) increased the photosynthetic capacity, as deduced by higher Vcmax and Jmax coupled with reduced stomatal limitation. Remarkably, our results demonstrate that auxin-sensitive mutants (dgt) are characterized by impairments in the usage of starch that led to lower growth, most likely associated with decreased respiration. Collectively, our findings suggest that mutations in different components of the auxin signalling pathway specifically modulate photosynthetic and respiratory processes.

ABSTRACT

Abstract Objective : To evaluate the functional capacity and mobility of older adults treated in primary health care and the association between adverse outcomes (functional dependence and reduced mobility) and sociodemographic and health conditions. Method : A cross-sectional and analytical epidemiological study was carried out in the municipal region of Caicó, Rio Grande do Norte, Brazil. Sociodemographic variables, presence of comorbidities, practice of physical activity, functional capacity (Lawton Scale) and mobility (Timed Up and Go Test) were investigated. Data analysis was performed using descriptive statistics, followed by bivariate analysis to investigate association variables and multivariate analysis (logistic regression). Results : Among the 109 elderly people evaluated, 29.4% were dependent in instrumental activities of daily living (IADL) and 67.9% had reduced mobility. The present study found a significant association between dependence in IADL and age equal to or greater than 75 years; while not practicing physical activity remained a factor of association with reduced mobility, regardless of sex, age and presence of co-morbidities. Conclusion : The findings emphasize the importance of the practice of physical activity, which was associated with better mobility, the construction of spaces of health promotion and disease prevention to encourage active aging for older adults, as well as a multi and interprofessional approach to comprehensive health care for older adults, with the use of functionality assessment tools.

Foram investigadas variáveis sociodemográficas, presença de comorbidades, prática de atividade física, capacidade funcional (Escala de Lawton) e mobilidade (Time Up and Go Test).

ABSTRACT

Monastrol is a small cell-permeable heterocyclic molecule that is recognized as an inhibitor of mitotic kinesin Eg5. Heterocyclic-fatty acid derivatives are a new class of compounds with a broad range of biological activities. This work describes a comparative study of the in vitro antitumoral activity of a series of new long-chain monastrol analogues against rat glioblastoma cells. The novel analogues C6-substituted monastrol and oxo-monastrol were synthesized via Biginelli multicomponent condensation of fatty ß-ketoester in good yields using a simple approach catalyzed by nontoxic and free-metal sulfamic acid. Following synthesis, their in vitro antitumoral activities were investigated. Notably, all analogues tested were active against rat glioblastoma cells. Superior activity was observed by analogues derived from palmitic and stearic fatty acid chains; these compounds were the most potent molecules, showing 13-fold higher potency than monastrol with IC50 values of 5.11 and 6.85 µM, respectively. These compounds could provide promising new lead derivatives for more potent antitumor drugs.

ABSTRACT

Chitosan has been indicated as a safe and promising polycation vector for gene delivery. However its low transfection efficiency has been a challenging obstacle for its application. To address this limitation, we synthesized chitosan derivatives which had increasing amounts of diethylethylamine groups (DEAE) attached to the chitosan main chain. The plasmid DNA VR1412 (pDNA), encoding the ß-galactosidase (ß-gal) reporter gene was used to prepare nanoparticles with the chitosan derivatives, and the transfection studies were performed with HeLa cells. By means of dynamic light scattering and zeta potential measurements, it was shown that diethylethylamine-chitosan derivatives (DEAE(x)-CH) were able to condense DNA into small particles having a surface charge depending on the polymer/DNA ratio (N/P ratio). Nanoparticles prepared with derivatives containing 15 and 25% of DEAE groups (DEAE(15)-CH and DEAE(25)-CH) exhibited transfection efficiencies ten times higher than that observed with deacetylated chitosan (CH). For derivatives with higher degrees of substitution (DS), transfection efficiency decreased. The most effective carriers showed low cytotoxicity and good transfection activities at low charge ratios (N/P). Vectors with low DS were easily degraded in the presence of lysozyme at physiological conditions in vitro and the nontoxicity displayed by these vectors opens up new opportunities in the design of DEAE-chitosan-based nanoparticles for gene delivery.

ABSTRACT

This article aims to present an experience report from the practice developed by the extension project "Weaving the Web: a proposal for training in the field of mental health from the relationship between university and community service" into a Psychosocial Care Center for children and teenagers (CAPSi). Through this report, we discuss about the role and consequences of contemporary psychiatric discourse in the treatment of children in the network of mental health services, further more we are going to show how the methodology of construction of the case clinical opens possibilities for appearance of the subject, often hidden by the nosology. In the meetings for the construction of a clinical case of David we can see a change in some of the professional service team as it began to treat the symptoms of children in different ways advocated by the diagnostic and statistical manual of mental disorders (DSMIV). Thus, the symptoms fail to be understood only as a sign of dysfunction, opening space for reflection on the subjective construction of David to hold a symbolic network.(AU)

ABSTRACT

Allometric growth of bone, muscle and fat, as well as of the carcass cuts of feedlot Texel × Ile de France crossbred lambs was evaluated. Twenty non-castrated male lambs weaned at 42 days of age (15.9 ± 2.1kg live weight) were housed in individual stalls and fed ad libitum. Five lambs were slaughtered ten days after weaning, and the remaining were randomly assigned to one of three slaughter weights: 25, 30 and 35kg live weight. To determine allometric growth of individual cuts and tissues, exponential equation Y = aXb was logarithmically transformed and used. Shoulder presented early maturity (b = 0.89), whereas allometric growth of leg, rib and neck were isogonics (b = 1.00, 1.03 e 1.11, respectively), i.e., similar to the carcass. Increased carcass weight resulted in decreased rate of bone and muscle growth (b = 0.60 and 0.92, respectively), and increased rate of fat growth (b = 1.78). Muscle growth was different among carcass cuts, being isogonics in shoulder (b = 0.98) and leg (b = 0.99), and early in rib (b = 0.90) and neck (b = 0.79). Maximum slaughter weight of Texel × Ile de France crossbred lambs is recommended to be 30kg live weight.

ABSTRACT

Carcass traits of twenty Texel x Ile de France crossbred lambs slaughtered after weaning (52 days of age), or at 25, 30 and 35kg live weight were evaluated. Lambs were placed in individual stalls (1.5m²) and fed ad libitum with a diet composed of sorghum silage (Sorghum bicolor (L.) Moench) and a concentrate mix (ground corn, soybean meal, calcium carbonate and salt). Commercial dressing percentage was related quadratically with slaughter weight, being highest in lambs slaughtered after weaning. Real dressing percentage increased linearly. Shoulder proportion decreased with slaughter weight, whereas leg, rib and neck proportions remained constant. Carcass fat was positively correlated to the slaughter weight. Slaughter of feedlot Texel x Ile de France crossbred lambs can be recommended at 28kg live weight.
