ABSTRACT
Oligonucleotides (ON) 4 to 60 nucleotides in length were isolated by ion-exchange chromatography on a column with Fractogel TSK DEAE-650 (M) from human milk which was hydrolyzed with proteinase K. ON from 60 to 16 nucleotides were degraded by RNase A but were resistant to DNase I, and, thus, they were ribooligonucleotides. In the presence of [gamma-32P]ATP, ON and heparin inhibited the phosphorylation of 38- and 20-kD milk proteins and failed to affect the phosphorylation of a 76-kD protein. Human milk is believed to contain polyanion-dependent and polyanion-independent protein kinases. The influence of the ON on the activity of the cytotoxic fraction of human milk alpha-lactalbumin towards human mammary gland carcinoma MCF-7 cells was studied. The ON inhibited the cytostatic and cytotoxic effects of alpha-lactalbumin. Synthetic oligonucleotides and heparin had similar effects. The endogenous ON are suggested to be involved in the regulation of cytotoxic activity of human milk.

Monosaccharide composition of glycopolymers of five mollicutes (Mycoplasma pneumoniae FH, M. hominis PG 21, M. fermentans PG 18, Acholeplasma laidlawii PG 8 and A. laidlawii var. granulum 118) integrated with their membrane has been studied by means of nine plant lectins with certain carbohydrate specificity labelled with colloid gold. Monosaccharide composition of glycopolymers depends both on the mollicute species and on its capacity to evoke diseases in different microorganisms. Thus genetically relative A. laidlawii PG 8 (saprophytic microorganism) and A. laidlawii var. granulum 118 (an agent of pale-green dwarfness of cereals) have the ratio between N-acetyl-D. galactosamine and N-acetyl-D-glucosamine 2:1 and 1:2, respectively. Large amounts of such exotic sugar as L-fucose have been found in M. hominis PG 21, M. fermentans PG 18 and A. laidlawii PG 8. L-fucose plays the basic role in disguising cells of these mollicutes in human organism (as O(H)-group blood erythrocytes), which permits these mollicutes disseminating through the whole organism. The composition of extracellular glycopolymers of mollicutes is discussed to reveal their role in the processes of adhesion and specialization (organotropism) of these microorganisms in populating the corresponding organs and tissues of macroorganisms.

The nucleotides sequence has been determined for the viral RNA and some of its cDNAs coding for the major antigenic protein of the epidemic stomatitis O1-194 and O1-1618 VP1 viruses. The expressed microheterogeneity has been registered for the population of the strain O1-194.

The nucleotide sequence of the cDNA for the viral RNA region coding for the main antigenic protein of the epidemic stomatitis virus of Asia 1 serotype has been identified. The amino acid sequences in the regions of VP1 protein antigenic determinants of the serotype Asia 1 virus and other serotypes viruses have been compared.

The DNA-copy of the major antigen (VP1) coding region of the FMDV A22 sero-type has been cloned and sequenced. A comparison of the respective amino acid sequence with those of other VP1 of A-serotype revealed considerable differences in the structure of antigenic determinants.

The use of MUV-5 as a recording microspectrophotometer for enzymatic and chromatographic analysis on miscroscale is proposed. The tuning of MUV-5 for working with a thermostated microcuvette is described. A microspectrophotometric method of the assay of lactate dehydrogenase activity in a single ovum of the mouse according to the kinetics of enzyme reaction is described. The proposed method allows to analyse the enzyme quantity 470 times as small as that which can be measured with SPh-16. A method for chromatographic separation of RNA with poly-U-sepharose column is described. The use of MUV-5 for the procedure of chromatographic separation of poly-RNA permits to reduce the amount of RNA in the assayed sample to 50 ng.
