ABSTRACT

Prosopis juliflora is an invasive plant distributed throughout the world and presents metabolites of interest for cosmetology. The aim of this work was to develop a new polysaccharide-based ingredient from P. juliflora and analyze its application in a solid core formulation that upon contact with water instantly forms a gel to improve moisturizing and anti-aging skin properties. Purified extracts by gel chromatography were characterized by NMR and LC-DAD-MS-MS. The in vitro and in vivo safety, antioxidant activity, formulation development and clinical evaluation were performed. The extract was characterized as containing an α-glucan and phenolics. It was non-cytotoxic, non-phototoxic and no skin reactions were observed in vivo. Antioxidant activity were present through different mechanisms. Clinical evaluation reinforced the potential of P. juliflora in skin hydration and microrelief improvement. This innovative form proved to be a prototype of a new product and the first study of an α-glucan as a cosmetic ingredient.

ABSTRACT

It is challenging to disperse lipophilic substances in a validated cytotoxicity assay, especially for compounds with log Kow greater than or equal to 5 that may show false negative results. The purpose of this study was to explain the challenges in conducting a cytotoxicity validated test of lipophilic substances: Minthostachys setosa, Pimenta pseudocaryophyllus, and Drimysbrasiliensis essential oils. Additionally, we compared the equivalence of Neutral Red (NR) and 3- (4,5-dimethylthiazol-2-yl) -5- (3- carboxymethoxyphenyl) -2- (4-sulfophenyl) -2H -tetrazolium, inner salt (MTS) in detecting cell viability. The Hydrophile-Lipophile Balance (HLB) technique was used to evaluate the dispersion of essential oils and cytotoxicity in accordance to the guidelines of the OECD / GD 129 validated cytotoxicity assay. We compared the equivalence of vital dyes by TOST equivalence test. According to the results, we demonstrated the possibility of using other ways to disperse the lipophilic substances. Based on the HLB theory, we selected polysorbate 20 as the best solubilizing agent of the essential oils studied in D10 culture medium.

ABSTRACT

Várias pesquisas vêm sendo desenvolvidas e direcionadas no descobrimento de novos agentes antimicrobianos provenientes de extratos de plantas e outros produtos naturais, para serem aplicados em produtos farmacêuticos e cosméticos. Atualmente, existem vários métodos para avaliar a atividade antibacteriana e antifúngica dos extratos vegetais. Os mais conhecidos incluem método de difusão em ágar, método de macrodiluição e microdiluição. A proposta dessa revisão é apresentar diferentes métodos comumente utilizados na pesquisa de novos agentes antimicrobianos, provenientes de extratos vegetais, e elucidar os principais fatores interferentes. Dessa maneira, contribuir como fonte de pesquisa para o desenvolvimento de futuros trabalhos relacionado ao estudo de atividade antimicrobiana de produtos naturais.


Several researches have been developed to search for new antimicrobial agents from extracts of plants and other natural products to be used in pharmaceutical and cosmetic products. Nowadays there are many methods to evaluate the antibacterial and antifungal activity of the plant extracts. The most known assays have been based on diffusion in agar; and micro and macrodilution methods. The purpose of this review is to describe the different methods commonly used for the determination of new antimicrobial agents from the plants extracts and elucidate the main interference factors. Moreover, this contributes as research source for future development of investigations related to the study of antimicrobial activity from natural products.
