ABSTRACT

Ethnoveterinary medicine (EVM) has been used by local South African communities for centuries. However, the knowledge of EVM is under threat due to the influence of Western medicine and acculturation. This study aimed to document the knowledge of ethnoveterinary medicinal plants in four villages within the Mahikeng Local Municipality to help preserve this valuable knowledge. The study used a qualitative research approach and targeted practitioners and holders of ethnoveterinary knowledge in four villages. Participants were recruited using key informants and snowball techniques, and in-depth interviews were conducted using semi-structured interview schedules. The data collected was analyzed thematically, and herbarium specimens were prepared from medicinal plants during field walks and sent to the South African National Biodiversity Institute (SANBI) for identification. The study documented the use of thirty-one medicinal plant species, with Senna italica (sebetebete) emerging as the most frequently used species for multiple animal health indications. The most common diseases treated using EVM were gala (general malaise due to bile reflux) and retained placenta. Participants strongly agreed on the cultural significance of plants for treating five animal diseases, including coughs and diarrhea. Although the study revealed a high level of trust in ethnoveterinary medicine among participants, they expressed concern regarding the loss of this knowledge due to the influence of Western medicine and acculturation. The study concluded that the indigenous knowledge and use of ethnoveterinary medicine was prevalent among livestock owners in the communities of the North West Province, and more studies need to be conducted in other areas to preserve this valuable knowledge.

ABSTRACT

Childhood diseases remain an increasing health problem in many developing countries and also associated with an enormous financial burden. In South Africa, many people still rely on traditional medicine for their primary healthcare. However, inadequate documentation of medicinal plants used to manage childhood diseases remain a prominent gap. Hence, the current study explored the importance of medicinal plants to treat and manage childhood diseases in the North West Province, South Africa. An ethnobotanical survey was conducted with 101 participants using semi-structured interviews (face-to-face). Ethnobotanical indices such as Frequency of citation (FC), Use-value (UV) and Informed Consensus Factor (ICF) were used for data analysis. A total of 61 plants from 34 families were recorded as medicine used for managing seven (7) categories of diseases resulting from 29 sub-categories. Skin-related and gastro-intestinal diseases were the most prevalent childhood health conditions encountered by the study participants. Based on their FC values that ranged from approximately 0.9-75%, the most popular medicinal plants used by the participants were Aptosinum elongatum (75.2%), Commelina diffusa (45.5%), Euphorbia prostrata (31.6%) and Bulbine frutescens (31.7%). In terms of the UV, A. elongatum (0.75), C. diffusa (0.45), E. prostrata (0.31), H. hemerocallidea (0.19) and E. elephantina (0.19) were the dominant plants used for treating and managing childhood diseases. Based on ICF, skin-related diseases dominated with the highest ICF value of 0.99. This category had 381 use-reports, comprising 34 plants (55.7% of total plants) used for childhood-related diseases. Particularly, B. frutescens and E. elephantina were the most-cited plants for the aforementioned category. Leaves (23%) and roots (23%) were the most frequently used plant parts. Decoctions and maceration were the main preparation methods, and the plant remedies were mainly administered orally (60%) and topically (39%). The current study revealed the continuous dependence on the plant for primary health care relating to childhood diseases in the study area. We generated a valuable inventory of medicinal plants and associated indigenous knowledge for child healthcare needs. However, investigating the biological efficacies, phytochemical profiles and the safety of these identified plants in relevant test systems remain essential in future research.

ABSTRACT

Annually, significant crop losses are reported due to diseases caused by phytopathogens. Most subsistence farmers cannot afford the high cost of chemical treatments thereby resulting in the increasing dependence on plant extracts to manage crop diseases. In this study, we documented plants used for the management of cabbage and spinach diseases in OR Tambo Municipality, Eastern Cape Province. An ethnobotanical survey using semi-structured questionnaires was used to document plants and plant parts used by the subsistence farmers in managing cabbage and spinach diseases. Semi-structured questionnaires were administered to 41 consenting subsistence farmers from November to December in 2021, using snowball sampling. The collected data were subjected to descriptive statistical and ethnobotanical analyses. A total of 17 plants belonging to 10 families were identified by the participants as being used in mitigating cabbage and spinach diseases. Tulbaghia violacea, Aloe ferox, and Capsicum annuum had the highest use value of 0.32 each, whereas Tulbaghia violacea had the highest relative frequency of citation of 0.39. This current study revealed the importance of plants in managing crop diseases in local communities. It provides baseline data for future pharmacological evaluations in authenticating the efficacies of the identified plants in managing crop diseases.

ABSTRACT

The use of medicinal plants for contraception remains a common practice among South African ethnic groups. The present study assessed the phytochemical profile, cytotoxicity, acute oral toxicity and efficacy of a herbal mixture used for contraception by the Batswana of South Africa. An aqueous extract was prepared from equal quantities (in terms of weight) of Bulbine frutescens (roots), Helichrysum caespititium (leaves) and Teucrium trifidum (leaves) based on a recipe used by traditional health practitioners. The phytochemical profiles of the freeze-dried herbal mixture were analyzed using gas chromatography-mass spectrometry (GC-MS). In addition, cytotoxicity was determined using an MTT assay on Vero cells and in vivo contraceptive efficacy was evaluated using seven Sprague Dawley rats per control and treatment groups. The control group received distilled water while test groups received 5, 50 and 300 mg/kg of the herbal mixture, which was administered orally once a day for three consecutive days. Subsequently, female rats were paired 1:1 with males for 3 days. Their weights were measured weekly and incidence of pregnancy was recorded. The GC-MS chromatogram revealed the presence of 12 identified and 9 unidentified compounds. In terms of safety, the herbal mixture had an IC50 value of 755.2 µg/mL and 2000 mg/kg, which was the highest tested dose that caused no mortality or morbidity in the rats. A contraceptive efficacy of 14.5% was exerted with 50 mg/kg herbal mixture extract while other doses had no effects given that all the rats were pregnant. Based on a chi-square test (p < 0.05), there was no correlation between the tested herbal mixture doses and contraception, nor on the weight of the rats. Overall, the herbal mixture extract was found to be safe but had limited contraceptive efficacy at the tested doses. In future studies, exploring increased dose range, solvent extract types and hormonal analysis will be pertinent.

ABSTRACT

Cancer occurrence is rapidly increasing all over the world, including in developing countries. The current trend in cancer management requires the use of herbal remedies since the majority of anticancer drugs are known to be costly, with unwanted side effects. In the Eastern Cape province, the use of medicinal plants for cancer management has been climbing steadily over the past two decades due to their cultural belief, low cost, efficacy, and safety claims. With the aim of identifying some potential anticancer plants for probable drug development, this study was undertaken to review plants reported by ethnobotanical surveys in the Eastern Cape province of South Africa for the traditional management of cancer. Information regarding plants used for cancer management in the Eastern Cape province was obtained from multidisciplinary databases and ethnobotanical books. About 24 plant species belonging to twenty families have been reported to be used for the traditional management of cancer in the Eastern Cape province. Among the anticancer plant species, only 16 species have been explored scientifically for their anticancer activities. This review authenticated the use of anticancer plant species in the Eastern Cape province and, therefore, identified several promising unexplored species for further scientific evaluation.

ABSTRACT

Bulbine asphodeloides (L.) Spreng (Xanthorrhoeaceae family), popularly known in South Africa as "ibhucu" or "Balsamkopieva," is a perennial plant traditionally used to treat skin diseases, including sunburns, rough skin, dressing burns, itches, and aging. The present study reports the cytotoxic, cellular antioxidant, and antiglucuronidase properties of the ethanol leaf extract from B. asphodeloides. The cytotoxic effect of the plant extract on human dermal fibroblast (MRHF) cells was evaluated by the bis-Benzamide H 33342 trihydrochloride/propidium iodide (Hoechst 33342/PI) dual-staining method. A validated biological cell-based assay was used to determine the cellular antioxidant activity of the extract. The antiglucuronidase and metal chelating activities were evaluated using standard in vitro methods. Lipopolysaccharide- (LPS-) induced RAW 264.7 cell model was used to determine the anti-inflammatory effect of the plant extract, and the immune-modulatory activity was performed using RAW 264.7 cells. The extract demonstrated no cytotoxic effect towards the MRHF cells at all the tested concentrations. Furthermore, the extract also possessed significant cellular antioxidant and antiglucuronidase activities, but a weak effect of metal chelating activity in a dose-dependent manner. However, the extract showed no significant anti-inflammatory and immune-stimulatory activities. Overall, the results showed that B. asphodeloides may be a useful therapeutic agent for the treatment of skin diseases, therefore supporting its ethnomedicinal usage.

BACKGROUND: Skin-related diseases affect every individual irrespective of age, gender or social status. Since time immemorial, humans have explored natural resources from their environment for the maintenance of the skin. This explorative survey was conducted to document the natural resources (plant and non-plant materials) used for folk cosmeceuticals by rural communities in Vhembe district municipality, Limpopo province, South Africa. METHODS: The research was conducted in six communities namely: Tshakuma, Shigalo, Tshamutilikwa, Luvhimbi (Masikhwa), Khakhanwa, and Folovhodwe in Vhembe district. Random and convenient sampling was used to access the target population. Semi-structured questionnaires were used to interview 71 participants that comprised traditional practitioners, herbalists and laypeople from the study area. Collected data were analysed using both quantitative (for e.g. frequency, use-value and relative frequency of citation) and qualitative (thematic) analytical methods. RESULTS: A total of 52 plants from 27 families and 22 non-plant materials were used as folk cosmeceuticals in the study area. The most cited plants included Dicerocaryum zanguebarium (Pedaliaceae), Ricinus communis (Euphorbiaceae) and Helinus integrifolius (Rhamnaceae). Trees and shrubs were the most common plant-life form while leaves were the most popular plant part. Pig fats, red ochre (Luvhundi soil) and ashes were the most cited non-plant materials. These documented natural resources are frequently prepared by crushing and mostly used to heal wounds. CONCLUSION: Traditional knowledge concerning folk cosmeceuticals is mostly held by elders. The high number of natural resources documented is an indication that Vhembe district is rich in ethnopharmacological knowledge. Scientific investigation of the efficacies and safety of these natural resources is highly recommended as a drive aimed at innovations with benefits to the rural communities who are the custodians of this valuable knowledge.

ABSTRACT

In Africa, indigenous methods of contraception continue to play a significant role in preventing unwanted pregnancies despite the introduction and popularity of modern contraceptives. The current review identified the common techniques and practices of African indigenous contraception, and examined their mechanisms and reasons for use. We searched data bases such as Google Scholar, Scopus, Web of Science, EBSCohost, African Journals, Science Direct, textbooks, thesis and dissertations for research articles on African indigenous contraception. The six common techniques of African indigenous contraception included periodic abstinence, withdrawal, breastfeeding, use of herbs, postpartum abstinence and waist bands, whilst practices relate to child (birth) spacing, postponement of first birth (virginity), stopping of reproduction and indigenous emergency contraception. Mother and infant health was stated as one of the reasons for using African indigenous contraception. African indigenous contraception continues to play a critical reproductive role in preventing unwanted pregnancies. However, there is lack of clarity regarding mechanisms, the safety, and efficacy of some techniques.

ABSTRACT

Although orthodox medications are available for skin diseases, expensive dermatological services have necessitated the use of medicinal plants as a cheaper alternative. This study evaluated the pharmacological and phytochemical profiles of four medicinal plants (Drimia sanguinea, Elephantorrhiza elephantina, Helichrysum paronychioides, and Senecio longiflorus) used for treating skin diseases. Petroleum ether and 50% methanol extracts of the plants were screened for antimicrobial activity against six microbes: Bacillus cereus, Shigella flexneri, Candida glabrata, Candida krusei, Trichophyton rubrum and Trichophyton tonsurans using the micro-dilution technique. Antioxidant activity was conducted using 2,2-diphenyl-1-picryhydrazyl (DPPH) free radical scavenging and ß-carotene linoleic acid models. Cytotoxicity was determined against African green monkey Vero kidney cells based on the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) colorimetric assay. Spectrophotometric and Gas Chromatography-Mass Spectrometry (GC-MS) methods were used to evaluate the phytochemical constituents. All the extracts demonstrated varying degrees of antimicrobial potencies. Shigella flexneri, Candida glabrata, Trichophyton rubrum and Trichophyton tonsurans were most susceptible at 0.10 mg/mL. In the DPPH test, EC50 values ranged from approximately 6-93 µg/mL and 65%-85% antioxidant activity in the ß-carotene linoleic acid antioxidant activity model. The phenolic and flavonoid contents ranged from 3.5-64 mg GAE/g and 1.25-28 mg CE/g DW, respectively. The LC50 values of the cytotoxicity assay ranged from 0.015-5622 µg/mL. GC-MS analysis revealed a rich pool (94-198) of bioactive compounds including dotriacontane, benzothiazole, heptacosane, bumetrizole, phthalic acid, stigmasterol, hexanoic acid and eicosanoic acid, which were common to the four plants. The current findings provide some degree of scientific evidence supporting the use of these four plants in folk medicine. However, the plants with high cytotoxicity need to be used with caution.

ABSTRACT

ETHNOPHARMACOLOGICAL RELEVANCE: Hypoxis hemerocallidea (commonly known as African Potato) is popular in African traditional medicine. It is used in the management of diverse ailments including burns, wounds and skin-related diseases. AIM OF THE STUDY: The current study investigated the antimicrobial effects of Hypoxis hemerocallidea against six microorganisms associated with skin diseases. In addition, the antioxidant activity, phytochemical profiles and cytotoxicity of the bulb extracts were evaluated. MATERIALS AND METHODS: The antimicrobial activity of 50% methanol (MeOH) and petroleum ether (PE) extracts of Hypoxis hemerocallidea bulbs was tested against two bacterial and four fungal strains implicated in causing opportunistic skin-related diseases. Antioxidant potential of the extract was investigated via the 2,2-diphenyl-1-picryhydrazyl (DPPH) free radical scavenging assay and ß-carotene linoleic acid model. Phytochemical profiling of the 50% MeOH extract of Hypoxis hemerocallidea was done using spectrophotometric assay and Gas Chromatography-Mass Spectrometry (GC-MS). The extracts were also evaluated for cytotoxicity against African green monkey Vero kidney cell lines based on the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) colorimetric assay. RESULTS: Both 50% MeOH and PE extracts showed considerable inhibitory effects against all six microorganisms. The extracts were potent against Shigella flexneri and Trichophyton tonsurans with minimum inhibitory concentration (MIC) values less than 1 mg/ml. However, there was relatively poor antifungal activity against the other fungal strains. In the DPPH assay, the MeOH extract of the bulb had an EC50 of 29.8 µg/ml while 76.91% antioxidant activity was observed in the ß-carotene-linoleic acid model. The extract contained total phenolics (41 mg GAE/g) and flavonoids (10 mg CE/g). The GC-MS analysis of Hypoxis hemerocallidea bulb revealed 29 and 160 bioactive compounds for 50% MeOH and PE extracts, respectively. Based on the cytotoxicity, Hypoxis hemerocallidea had LC50 value of 210.9 ±â€¯18.4 and 95.5 ±â€¯13.3 µg/ml for PE and MeOH extracts, respectively. CONCLUSIONS: The bulb extracts of Hypoxis hemerocallidea exhibited good antimicrobial and antioxidant activities, which could be attributed to the presence of phenolics, flavonoids and the other bioactive compounds identified through GC-MS, making it a potentially effective cosmetic plant. These findings also account for the multi-pharmacological use of Hypoxis hemerocallidea in traditional medicine, especially related to skin diseases. The plant extracts can be considered as safe based on their LC50 values (< 20 µg/ml). However, other form of cytotoxicity studies need to be carried out on Hypoxis hemerocallidea, as well as in vivo tests, to confirm its safety and efficacy as a treatment for skin-related diseases.

BACKGROUND: Obesity is a global epidemic that affects 500 million people worldwide and is predicted to increase to one billion people by 2030. The prevalence of obesity is increasing across populations in South Africa. However, questions still remain surrounding the predisposing factors and obesity-related health problems especially in the rural areas. This study evaluated several lifestyle factors such as dietary habits, physical activity, smoking, alcohol intake, co-morbidities and their association with the prevalence of obesity and overweight in Nkonkobe Municipality of the Eastern Cape. METHODS: A cross-sectional, population-based survey was conducted among 118 residents in four rural/sub-urban townships of the study area. Measurements including weight, height, body mass index (BMI), physical activity and dietary habits were determined using a validated questionnaire. RESULTS: The overall prevalence of obesity and overweight was 38 and 19%, respectively. The highest prevalence of obesity (70%) was observed among those who do not undertake any physical activity. Close to half (48.48%) of the respondents who eat fast foods always were obese, and 30.30% were overweight; when combined, the prevalence for obesity is 78.78%. A negative association with obesity was observed among regular smokers (26.92%) and consumers of alcohol (4.00%). Arthritis, hypertension and tuberculosis were co-morbidities significantly (P < 0.05) associated with obesity in the study area. CONCLUSIONS: The findings of this study reveal that lack of physical activity, overindulgence on fast and fried foods, low fruit and vegetable consumption as well as arthritis, hypertension and tuberculosis were significant risk factors of obesity in Nkonkobe Municipality.
