ABSTRACT

OBJECTIVE: The current study aimed to investigate the functional roles and clinical significance of microRNA-148a (miR-148a) in the progression of oral squamous cell carcinoma (OSCC). METHODS: Relative expression of miR-148a in OSCC cells and tissues were detected using quantitative real-time polymerase chain reaction (qRT-PCR). Chi-square test was performed to estimate the relationship between miR-148a expression and clinical characteristics of OSCC patients. Cell transfection was carried out using Lipofectamine® 2000. Biological behaviors of tumor cells were detected using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) and transwell assays. Bioinformatics analysis and luciferase reporter assay were used to identify the target genes of miR-148a. Protein expression was detected through Western blot analysis. RESULTS: MiR-148a expression was obviously decreased in OSCC tissues and cells, and such down-regulation was closely correlated with lymph node metastasis (P=0.027) and tumor node metastasis (TNM) stage (P=0.001) of OSCC patients. miR-148a overexpression could significantly impair OSCC cell proliferation, migration and invasion in vitro (P<0.05 for all). Insulin-like growth factor-I receptor (IGF-IR) was a potential target of miR-148a. MiR-148a could inhibit ERK/MAPK signaling pathway through targeting IGF-IR. CONCLUSION: MiR-148a plays an anti-tumor role in OSCC and inhibits OSCC progression through suppressing ERK/MAPK pathway via targeting IGF-IR.

ABSTRACT

With the deepening population aging process in China, the medical expenses of older adults has become a widespread concerned. Medical insurance is a major source of Chinese medical financing and payment. The study aims to understand the current status of medical expenses for older adults and explore the effect of different types of health insurance on medical expenses in China.The data came from the Chinese Longitudinal Health Longevity Survey (CLHLS) in 2014. The Kruskal-Wallis test and general multivariate linear regression model were applied to analyze the current situation and to explore how medical insurance as the main payment impacts medical expenses.A total of 4376 older participants were included in this study. The median of medical expenses of a total was 1500 Yuan per year. The proportions of participants who had the urban employee-based basic medical insurance (UE-BMI), the urban residents basic medical insurance (UR-BMI), the new rural cooperative medical insurance scheme (NCMS), and the commercial medical insurance were 10.8%, 8.4%, 72.7%, and 0.9%, respectively. 34.8% of older adults paid the health care service via the NCMS and 11.9% paid via the UE-BMI. Participating in the NCMS and UR-BMI are significantly related to the level of the medical fees of older adults. UE-BMI, UR-BMI, and NCMS as main payment eased the pressure of medical expenses.The influence of different types of medical insurances as main payments on the medical expenses of older adults is varied. Implementation of medical insurance should be taken to further relieve the medical expenses of older adults.

ABSTRACT

We demonstrate a master oscillator power amplifier (MOPA) architecture based on Yb:YAG amplifiers and adaptive optics (AO) systems with a high power and high beam quality laser output. With two conduction cooled, dual-end-pumped Yb:YAG zigzag-slab amplifiers at room temperature, the fiber laser of 300 W was scaled to 11.9 kW. Moreover, AO system positioned downstream was utilized to correct wavefront of amplified laser. The beam quality ß at maximum output power was 2.8 times diffraction limited with closed-loop AO system.

ABSTRACT

PURPOSE: To observe the changes of endotoxin levels after different teeth with periodontitis were treated with different methods. METHODS: Six healthy premolars extracted for orthodontic reasons and 36 posterior teeth extracted due to severe periodontitis were selected. Each tooth was processed from two 4 mm×4 mm×1 mm cementum pieces 2 mm under the cementum-enamel junction, each tooth with periodontitis was numbered. Healthy teeth served as negative control group, one of the two tablets from each tooth with periodontitis was selected in the periodontitis group, which was not treated with root surface treatment. The remaining 36 teeth with periodontitis were randomly divided into 6 groups: SRP group, SRP + antimicrobial peptide A group , SRP + antimicrobial peptide B group, SRP + EDTA group, SRP + Er:YAG laser group and SRP + Nd:YAG laser group. Endotoxin concentration in each tooth was determined by chromogenic substrate limulus reagent. The endotoxin concentration in each tooth was recorded according to the serial number, and the changes of endotoxin concentration were calculated before and after treatment. SPSS17.0 software package was used to analyze the data. RESULTS: Compared with the teeth with periodontitis, endotoxin concentration decreased to varying degrees, there were significant differences in each treatment group(P<0.01). Compared with SRP group, endotoxin levels in SRP + antimicrobial peptide A group, SRP + antimicrobial peptide B group, SRP + Er:YAG laser group were significantly decreased(P<0.01). No significant difference decreased was from between SRP + EDTA group and SRP + Nd:YAG laser group(P>0.05). CONCLUSIONS: Treatment of teeth with periodontitis using different methods can decrease the level of endotoxin, and the treatment of periodontitis root surface with antimicrobial peptide A + SRP may be more effective.

ABSTRACT

Induced pluripotent stem cells (iPSCs) have great potential as seed cells for tissue engineering applications. Previous studies have shown that iPSCs could be induced to differentiate into bone forming cells. However, in a tissue engineering approach, seeding cells in biomaterials is required, and the effect of biomaterials on cell growth and differentiation is critical for the success of the formation of engineered tissues. In this study, we investigated the effect of akermanite, a bioactive ceramic, on the osteogenic differentiation of embryoid body (EB) cells derived from human iPSCs. The results showed that, in the presence of osteogenic factors (ascorbic acid, dexamethasone, and ß-glycerophosphate), ionic extracts of akermanite enhanced the osteogenic differentiation of EB cells as compared with normal osteogenic medium. Alkaline phosphatase (ALP) activity and the expression of osteogenic marker genes such as osteocalcin (OCN), collagen (COL-1), RUNX2, and BMP2 are significantly increased by the stimulation of akermanite ceramic extracts at certain concentration ranges. More interesting is that the medium containing extracts of akermanite but without osteogenic factors also showed stimulatory effects on the osteogenic differentiation of EB cells as compared to normal growth medium without osteogenic factors, such as ascorbic acid, dexamethasone, and ß-glycerophosphate, not at the early stage of culture, but only at the later stage of the culture period (21 days). These results suggest that akermanite as a bioactive material together with human iPSCs might be used for bone tissue engineering applications.

BACKGROUND: Periodontal disease is closely related to type 2 diabetes and is an important complication of diabetes. This study aimed to investigate the effect of periodontal treatment on levels of blood glucose (Glu) and glycosylated hemoglobin (HbA1c) among elderly patients with type 2 diabetes and periodontal disease. METHODS: A total of 107 elderly patients with type 2 diabetes and periodontal disease were selected and divided into two groups according to their HbA1c levels. Group A was a well-controlled diabetic group and group B was uncontrolled. Their probing depth (PD), attachment loss (AL), the value of Glu and HbA1c were analyzed before periodontal treatment and 4 months later. RESULTS: There was a significant difference in periodontal condition between groups A and B (P < 0.01). The periodontal condition for both groups was significantly (P < 0.01) improved after periodontal therapy. The effect of treatment in group A was more pronounced than group B, and the difference was significant (P < 0.01). After the periodontal treatment, Glu and HbA1c were reduced significantly in both groups (P < 0.05). CONCLUSIONS: Periodontal condition is related to the control of Glu level among elderly patients with type 2 diabetes and periodontal disease. Periodontal treatment can effectively reduce the level of Glu and HbA1c as well as improve the periodontal condition in elderly type-2 diabetes patients with periodontal disease.

ABSTRACT

OBJECTIVE: To evaluate the effect of calcification of autogenous bone marrow stem cell transplantation in periodontal tissue regeneration. METHODS: Bone marrow stem cells derived from the same dog were cultured with alpha-MEM. 1 x 10(7) cells of first passage were allowed to attach to the collagen membrane for 24 hours. The membrane-cells were transplanted into periodontal defect in the same dog. Then the defects were covered with e-pTFE membranes. The defects covered only with e-pTFE without membrane-cells were served as control. Eighteen teeth of 6 dogs for every group were studied. The dogs were sacrificed after 6 weeks. RESULTS: The results showed that new bone formation in test group was significantly higher than that of control group. The calcification of new bone in test group was better than control group. CONCLUSIONS: The results suggested that autogenous bone marrow stem cell transplantation with guided tissue regeneration technique could enhance periodontal tissue regeneration and could form new bone tissue fast and could shorten times of periodontal tissue regeneration in dogs.
