ABSTRACT

Uniform grape maturity can be sought by producers to minimise underripe and/or overripe proportions of fruit and limit any undesirable effects on wine quality. Considering that grape heterogeneity is a multifaceted phenomenon, a composite index summarising overall grape heterogeneity was developed to benefit vineyard management and harvest date decisions. A grape heterogeneity index (GHI) was constructed by aggregating the sum of absolute residuals multiplied by the range of values from measurements of total soluble solids, pH, fresh weight, total tannins, absorbance at 520 nm (red colour), 3-isobutyl-2-methoxypyrazine, and malic acid. Management of grape heterogeneity was also studied, using Cabernet Sauvignon grapes grown under four viticultural regimes (normal/low crop load, full/deficit irrigation) during the 2019/2020 and 2020/2021 seasons. Comparisons of GHI scores showed grape variability decreased throughout ripening in both vintages, then significantly increased at the harvest time point in 2020, but plateaued on sample dates nearing the harvest date in 2021. Irrigation and crop load had no effect on grape heterogeneity by the time of harvest in both vintages. Larger vine yield, leaf area index, and pruning weight significantly increased GHI score early in ripening, but no significant relationship was found at the time of harvest. Differences in the Ravaz index, normalised difference vegetation index, and soil electrical conductivity did not significantly change the GHI score.

ABSTRACT

Grapevine virus-associated disease such as grapevine leafroll disease (GLD) affects grapevine health worldwide. Current diagnostic methods are either highly costly (laboratory-based diagnostics) or can be unreliable (visual assessments). Hyperspectral sensing technology is capable of measuring leaf reflectance spectra that can be used for the non-destructive and rapid detection of plant diseases. The present study used proximal hyperspectral sensing to detect virus infection in Pinot Noir (red-berried winegrape cultivar) and Chardonnay (white-berried winegrape cultivar) grapevines. Spectral data were collected throughout the grape growing season at six timepoints per cultivar. Partial least squares-discriminant analysis (PLS-DA) was used to build a predictive model of the presence or absence of GLD. The temporal change of canopy spectral reflectance showed that the harvest timepoint had the best prediction result. Prediction accuracies of 96% and 76% were achieved for Pinot Noir and Chardonnay, respectively. Our results provide valuable information on the optimal time for GLD detection. This hyperspectral method can also be deployed on mobile platforms including ground-based vehicles and unmanned aerial vehicles (UAV) for large-scale disease surveillance in vineyards.

ABSTRACT

Generations of sensors have been developed for predicting food sensory profiles to circumvent the use of a human sensory panel, but a technology that can rapidly predict a suite of sensory attributes from one spectral measurement remains unavailable. Using spectra from grape extracts, this novel study aimed to address this challenge by exploring the use of a machine learning algorithm, extreme gradient boosting (XGBoost), to predict twenty-two wine sensory attribute scores from five sensory stimuli: aroma, colour, taste, flavour, and mouthfeel. Two datasets were obtained from absorbance-transmission and fluorescence excitation-emission matrix (A-TEEM) spectroscopy with different fusion methods: variable-level data fusion of absorbance and fluorescence spectral fingerprints, and feature-level data fusion of A-TEEM and CIELAB datasets. The results for externally validated models showed slightly better performance using only A-TEEM data, predicting five out of twenty-two wine sensory attributes with R2 values above 0.7 and fifteen with R2 values above 0.5. Considering the complex biotransformation involved in processing grapes to wine, the ability to predict sensory properties based on underlying chemical composition in this way suggests that the approach could be more broadly applicable to the agri-food sector and other transformed foodstuffs to predict a product's sensory characteristics from raw material spectral attributes.

ABSTRACT

Gas exchange mechanisms play crucial roles in maintaining fruit post-harvest quality in perishable fruit such as strawberry (Fragaria×ananassa Duch.) and blueberry (Vaccinium corymbosum L.). The internal oxygen concentration ([O2 ]) of strawberry and blueberry were measured using Clark-type oxygen sensing electrodes. The volume of intercellular voids in strawberry was obtained by micro-computed tomography (micro-CT). In both berries, internal [O2 ] was consistent and relatively high across measured tissues. The overall [O2 ] was well above the Michaelis constant (K m ) for cytochrome c oxidase in both fruit and different from previously examined grape (Vitis vinifera L.) berry mesocarp with near zero minimum [O2 ]. In strawberry and blueberry, cell vitality was also maintained at full maturity in the mesocarp. Higher storage temperature (i.e. 20 vs 4°C) reduced internal [O2 ] of strawberry. Pedicel detachment in blueberry was associated with greater fruit dehydration and lower internal [O2 ] after short-term storage of 12h. The results suggest that the intercellular voids of the fruit's mesocarp provide an efficient gas exchange route for maintaining high fruit internal [O2 ] post-harvest.

ABSTRACT

Absorbance-transmission and fluorescence excitation-emission matrix (A-TEEM) spectroscopy was investigated as a rapid method for predicting maturity indices using Cabernet Sauvignon grapes produced under four viticulture treatments during two growing seasons. Machine learning models were developed with fused spectral data to predict 3-isobutyl-2-methoxypyrazine (IBMP), pH, total tannins (Tannin), total soluble solids (TSS), and malic and tartaric acids based on the results from traditional analysis methods. Extreme gradient boosting (XGB) regression yielded R2 values of 0.92-0.96 for IBMP, malic acid, pH, and TSS for externally validated (Test) models, with partial least squares regression being superior for TSS prediction (R2 = 0.97). R2 values of 0.64-0.81 were achieved with either approach for tartaric acid and Tannin predictions. Classification of grape maturity, defined by quantile ranges for red colour, IBMP, malic acid, and TSS, was investigated using XGB discriminant analysis, providing an average of 78 % correctly classified samples for the Test model.

ABSTRACT

The sessile plant has developed mechanisms to survive the "rough and tumble" of its natural surroundings, aided by its evolved innate immune system. Precise perception and rapid response to stress stimuli confer a fitness edge to the plant against its competitors, guaranteeing greater chances of survival and productivity. Plants can "eavesdrop" on volatile chemical cues from their stressed neighbours and have adapted to use these airborne signals to prepare for impending danger without having to experience the actual stress themselves. The role of volatile organic compounds (VOCs) in plant-plant communication has gained significant attention over the past decade, particularly with regard to the potential of VOCs to prime non-stressed plants for more robust defence responses to future stress challenges. The ecological relevance of such interactions under various environmental stresses has been much debated, and there is a nascent understanding of the mechanisms involved. This review discusses the significance of VOC-mediated inter-plant interactions under both biotic and abiotic stresses and highlights the potential to manipulate outcomes in agricultural systems for sustainable crop protection via enhanced defence. The need to integrate physiological, biochemical, and molecular approaches in understanding the underlying mechanisms and signalling pathways involved in volatile signalling is emphasised.

ABSTRACT

Vascular bundles in the grape pedicel and berry contain the conduits, phloem and xylem, for transport of water, sugar, nutrients and signals into and through the grape berry and play a critical role in berry growth and composition. Here, we assess the vascular anatomy within the proximal region of the berry. Guided using a 3D berry model generated by micro-CT, differential staining of transverse sections of berries and receptacles was followed by fluorescent microscopy. Morphometric and vascular characteristics were analyzed within the central proximal region (brush zone, a fibrous extension from the pedicel vascular system into the berry) of the seeded cultivars Shiraz and Sauvignon Blanc, as well as the stenospermocarpic cultivars Ruby Seedless and Flame Seedless. Observations revealed a change in vascular arrangement from the receptacle into the berry brush zone and differences in xylem element size as well as xylem and phloem area relationships. Xylem anatomical and derived hydraulic parameters, as well as total tissue area of xylem and phloem varied between cultivars and in receptacle and berry components. Variation in vascular growth between grape pedicels and berries was independent of seededness. Differences in receptacle xylem vessel size and distribution could contribute to cultivar-dependent xylem backflow constraint.

ABSTRACT

Wine made from grapes exposed to bushfire smoke can exhibit unpleasant smoky, ashy characters, which have been attributed to the presence of smoke-derived volatile phenols, in free or glycosylated forms. Here we report the uptake and glycosylation of volatile phenols by grapes following exposure of Cabernet Sauvignon vines to smoke, and their fate during winemaking. A significant delay was observed in the conversion of volatile phenols to their corresponding glycoconjugates, which suggests sequestration, the presence of intermediates within the glycosylation pathway and/or other volatile phenol storage forms. This finding has implications for industry in terms of detecting smoke-affected grapes following vineyard smoke exposure. The potential for an in-canopy sprinkler system to mitigate the uptake of smoke-derived volatile phenols by grapes, by spraying grapevines with water during smoke exposure, was also evaluated. While "misting" appeared to partially mitigate the uptake of volatile phenols by grapes during grapevine exposure to smoke, it did not readily influence the concentration of volatile phenols or the sensory perception of smoke taint in wine. Commercial sensors were used to monitor the concentration of smoke particulate matter (PM) during grapevine exposure to low and high density smoke. Similar PM profiles were observed, irrespective of smoke density, such that PM concentrations did not reflect the extent of smoke exposure by grapes or risk of taint in wine. The sensors could nevertheless be used to monitor the presence of smoke in vineyards during bushfires, and hence, the need for compositional analysis of grapes to quantify smoke taint marker compounds.

ABSTRACT

Grapevine viruses are found throughout the viticultural world and have detrimental effects on vine productivity and grape and wine quality. This report provides a comprehensive and up-to-date review on grapevine viruses in Australia with a focus on "Shiraz Disease" (SD) and its two major associated viruses, grapevine virus A (GVA) and grapevine leafroll-associated virus 3 (GLRaV-3). Sensitive grapevine cultivars like Shiraz infected with GVA alone or with a co-infection of a leafroll virus, primarily GLRaV-3, show symptoms of SD leading to significant yield and quality reductions in Australia and in South Africa. Symptom descriptors for SD will be outlined and a phylogenetic tree will be presented indicating the SD-associated isolates of GVA in both countries belong to the same clade. Virus transmission, which occurs through infected propagation material, grafting, and naturally vectored by mealybugs and scale insects, will be discussed. Laboratory and field-based indexing will also be discussed along with management strategies including rogueing and replanting certified stock that decrease the incidence and spread of SD. Finally, we present several cases of SD incidence in South Australian vineyards and their effects on vine productivity. We conclude by offering strategies for virus detection and management that can be adopted by viticulturists. Novel technologies such as high throughput sequencing and remote sensing for virus detection will be outlined.

ABSTRACT

Hydraulics of plants that have different strategies of stomatal regulation under water stress are relatively poorly understood. We explore how root and shoot hydraulics, stomatal conductance (g s), leaf and root aquaporin (AQP) expression, and abscisic acid (ABA) concentration in leaf xylem sap ([ABA]xylemsap) may be coordinated under mild water stress and exogenous ABA applications in two Vitis vinifera L. cultivars traditionally classified as near-isohydric (Grenache) and near-anisohydric (Syrah). Under water stress, Grenache exhibited stronger adjustments of plant and root hydraulic conductances and greater stomatal sensitivity to [ABA]xylemsap than Syrah resulting in greater conservation of soil moisture but not necessarily more isohydric behavior. Correlations between leaf (Ψleaf) and predawn (ΨPD) water potentials between cultivars suggested a "hydrodynamic" behavior rather than a particular iso-anisohydric classification. A significant decrease of Ψleaf in well-watered ABA-fed vines supported a role of ABA in the soil-leaf hydraulic pathway to regulate g s. Correlations between leaf and root AQPs expression levels under water deficit could explain the response of leaf (K leaf) and root (Lp r) hydraulic conductances in both cultivars. Additional studies under a wider range of soil water deficits are required to explore the possible differential regulation of g s and plant hydraulics in different cultivars and experimental conditions.

ABSTRACT

Recent climatic trends of higher average temperatures and erratic precipitation patterns are resulting in decreased soil moisture availability and, consequently, periods of water stress. We studied the effects of seasonal water stress on grapevine (Vitis vinifera L. cv. Riesling grafted onto 101-14 (Vitis riparia Michx.×Vitis rupestris Scheele) rootstock) shoot growth, leaf gas exchange, xylem morphology and hydraulic performance in the cool-climate Finger Lakes region of New York. A plastic rain exclusion tarp was installed on the vineyard floor to create a soil moisture deficit and consequently induce vine water stress. Weekly measurements of predawn leaf and midday stem water potentials (Ψmd) were made, and two contrasting shoot length classes, long (length >2.0m) and short (length <1.0m), were monitored. Growth of both long and short shoots was positively correlated with Ψmd but no difference in water status was found between the two. Compared with rain-fed vines, water-stressed vines had lower photosynthesis and stomatal conductance later in the season when Ψmd dropped below -1.2MPa. Long shoots had three-fold higher xylem-specific hydraulic conductivity values than short shoots. Long shoots experiencing water stress were less vulnerable to xylem cavitation than shorter shoots even though they had more large-diameter vessels. The lower vulnerability to cavitation of long shoots may be attributed to less xylem intervessel pitting being found in long shoots, consistent with the air-seeding hypothesis, and suggests that a hydraulic advantage enables them to maintain superior growth and productivity under water stress.

ABSTRACT

Tensiometers sense the chemical potential of water (or water potential, Ψw) in an external phase of interest by measuring the pressure in an internal volume of liquid water in equilibrium with that phase. For sub-saturated phases, the internal pressure is below atmospheric and frequently negative; the liquid is under tension. Here, we present the initial characterization of a new tensiometer based on a microelectromechanical pressure sensor and a nanoporous membrane. We explain the mechanism of operation, fabrication, and calibration of this device. We show that these microtensiometers operate stably out to water potentials below -10 MPa, a tenfold extension of the range of current tensiometers. Finally, we present use of the device to perform an accurate measurement of the equation of state of liquid water at pressures down to -14 MPa. We conclude with a discussion of outstanding design considerations, and of the opportunities opened by the extended range of stability and the small form factor in sensing applications, and in fundamental studies of the thermodynamic properties of water.

ABSTRACT

In perfusion experiments, the hydraulic conductance of stem segments ( ) responds to changes in the properties of the perfusate, such as the ionic strength ( ), pH, and cationic identity. We review the experimental and theoretical work on this phenomenon. We then proceed to explore the hypothesis that electrokinetic effects in the bordered pit membrane (BPM) contribute to this response. In particular, we develop a model based on electroviscosity in which hydraulic conductance of an electrically charged porous membrane varies with the properties of the electrolyte. We use standard electrokinetic theory, coupled with measurements of electrokinetic properties of plant materials from the literature, to determine how the conductance of BPMs, and therefore , may change due to electroviscosity. We predict a nonmonotonic variation of with with a maximum reduction of 18%. We explore how this reduction depends on the characteristics of the sap and features of the BPM, such as pore size, density of chargeable sites, and their dissociation constant. Our predictions are consistent with changes in observed for physiological values of sap and pH. We conclude that electroviscosity is likely responsible, at least partially, for the electrolyte dependence of conductance through pits and that electroviscosity may be strong enough to play an important role in other transport processes in xylem. We conclude by proposing experiments to differentiate the impact of electroviscosity on from that of other proposed mechanisms.
