ABSTRACT

Here, we evaluate the relationships between the infestation rates of five monogenean parasites species with the dry and wet seasons, with the organic and inorganic parameters of the water of two rivers: the Jacaré-Pepira and Jacaré-Guaçú, and with the condition factors of its fish hosts: Serrasalmus maculatus and Astronotus crassipinnis, in the state of São Paulo, southeastern Brazil. Fish were collected between January and December 2017. Anacanthorus serrasalmi, Amphithecium speirocamarotum and Gussevia asota had higher abundance rates (Student's t test, p ≤ 0.05) in the wet season. Gussevia asota had its abundance negatively correlated to nitrate in the Jacaré-Pepira River and with total nitrogen and potassium in the Jacaré-Guaçú River. Regarding the fish hosts condition factors, was observed a positive correlation with the abundances of G. asota in the Jacaré-Guaçú River, and with A. serrasalmi in the Jacaré-Pepira River. In general, wet season favored an increasing in the infestation rates of the monogeneans parasites in their host species, mainly in the river considered as the most polluted, the Jacaré-Guaçú River. Of the five parasites species analyzed in this study, only Gussevia astronoti and Rhinoxenus piranhus had no interaction with seasonality, river water variables, or fish host condition factors. On the other hand, G. asota had interactions both with water parameters (nitrate and total nitrogen) and with the hosts condition factors, which reflected in the abundance and intensity rates, showing itself as a species sensitive to changes in the environment and, therefore, that can be considered as a bioindicator organism.

ABSTRACT

MLH1 and MSH2 are important genes for DNA mismatch repair and crossing over during meiosis and are implicated in male infertility. Therefore, the methylation patterns of the DNA mismatch repair genes MLH1 and MSH2 in oligozoospermic males were investigated. Ten oligozoospermic patients and 29 normozoospermic donors were analysed. Methylation profiles of the MLH1 and MSH2 promotors were analysed. In addition, sperm motility and seminal reactive oxygen species (ROS) were recorded. Receiver operating characteristic (ROC) analysis was conducted to determine the accuracy of the DNA methylation status of MLH1 and MSH2 to distinguish between oligozoospermic and normozoospermic men. In oligozoospermic men, MLH1 was significantly (p = .0013) more methylated compared to normozoospermic men. Additionally, there was a significant positive association (r = .384; p = .0159) between seminal ROS levels and MLH1 methylation. Contrary, no association between MSH2 methylation and oligozoospermia was found. ROC curve analysis for methylation status of MLH1 was significant (p = .0275) with an area under the curve of 61.1%, a sensitivity of 22.2% and a specificity of 100.0%. This pilot study indicates oligozoospermic patients have more methylation of MLH1 than normozoospermic patients. Whether hypermethylation of the MLH1 promoter plays a role in repairing relevant mismatches of sperm DNA strands in idiopathic oligozoospermia warrants further investigation.

Abstract This paper describes the occurrence of digenetic Rhipidocotyle santanaensis Lunaschi, 2004 in Brazil. This parasite was found in the liver, heart, gonads, intestine, pyloric caeca, stomach, swim bladder and cavity of Acestrorhynchus lacustris (Lütken, 1875) from the Batalha River, São Paulo State, with higher incidence in gonads and digestive tract. The parasite specimens found in this study showed morphological characteristics very similar to specimens from Argentina, with differences only in relation to the ejaculatory duct and seminal vesicle. However, Brazilian specimens presented great morphometrical differences between specimens described in Argentina, with much higher measures. This is the first record of this metazoan parasite species in Brazil and in this host fish.


Resumo O presente trabalho descreve a ocorrência do digenético Rhipidocotyle santanaensis Lunaschi, 2004 no Brasil. Estes parasitos foram encontrados no fígado, coração, gônadas, intestino, cecos pilóricos, estômago, bexiga natatória e cavidade do peixe Acestrorhynchus lacustris (Lütken, 1875) provenientes do rio Batalha, Estado de São Paulo, com maior abundância nas gônadas e trato digestório. Os espécimes dos parasitos encontrados neste trabalho apresentaram características morfológicas muito semelhantes aos espécimes provenientes da Argentina, com diferenças apenas em relação ao ducto ejaculatório e vesícula seminal. Porém, os exemplares brasileiros apresentaram grandes diferenças morfométricas entre os espécimes descritos na Argentina, com medidas muito superiores. Este é o primeiro registro desta espécie de parasito no Brasil e neste hospedeiro.

ABSTRACT

O objetivo deste trabalho foi avaliar o efeito alelopático de extratos de Coleus barbatus sobre a germinação e desenvolvimento de plântulas de alface. Os experimentos foram conduzidos em laboratório, em diferentes concentrações de extratos obtidos de folhas de falso-boldo (0; 7,5; 15; 22,5; 30%), coletadas nas quatro estações do ano, obtidos por trituração, maceração, infusão e decocção. As sementes de alface foram distribuídas sobre placas de petri umidecidas com 10 mL de solução. Os resultados mostraram que os extratos de folhas afetaram a germinabilidade de sementes de alface apenas na concentração de 30% do extrato preparado por decocção de folhas coletadas no inverno. O índice de velocidade de germinação apresentou redução significativa nos extratos triturados preparados no outono e inverno; na maceração de folhas coletadas no verão e outono e no extrato de decocção preparado no inverno. Os extratos estimularam o crescimento da parte aérea das plântulas de alface, ao passo que, o comprimento das raízes foi afetado pelos extratos, ora inibindo, ora estimulando o crescimento das mesmas. Os extratos de folhas de falso-boldo coletadas nas quatro estações do ano apresentaram efeito alelopático sobre a germinação e o desenvolvimento da alface, sendo que estes variaram em função da sazonalidade da coleta, da forma de obtenção dos extratos e das concentrações testadas.


The aim of this study was to evaluate the allelopathic effects of extracts from Coleus barbatus on germination and seedling growth of lettuce. The experiments were conducted in the laboratory using different concentrations (0, 7.5, 15, 22.5, 30%) of aqueous extracts from "falso-boldo" leaves harvested in all four seasons; extracts were obtained by grinding, maceration, infusion and decoction. Seeds were distributed on Petri dishes moistened with 10 mL solution. Results showed that the leaf extracts affected the germination of lettuce seeds only at the concentration of 30% extract prepared by decoction of leaves harvested in the winter. Germination speed index showed significant reduction for ground extracts prepared in the fall and winter, as well as for macerated leaves harvested in the summer and fall and for extracts prepared through decoction in the winter. The extracts stimulated the growth of shoots of lettuce seedlings, whereas root length was affected by the extracts, which either stimulated or inhibited its growth. Extracts of "falso-boldo" leaves harvested in all four seasons had allelopathic effect on germination and development of lettuce, and the latter varied depending on the harvest season, the form of preparing the extracts and the tested concentrations.

OBJECTIVE: To evaluate the effect of pars plana vitrectomy in the management of patients with pars planitis. METHODS: A retrospective analysis of the clinical course, post-operative complications and recurrent uveal inflammation following pars plana vitrectomy in patients with pars planitis. RESULTS: The study included 22 eyes of 19 patients. The mean follow-up was 55.7 (± 39.6) months (range 7 - 144 months). The surgical indications were, persistent vitreous opacities in 10 eyes, vitreous haemorrhage in 9 eyes, and epiretinal membrane in 3 eyes. There was an improvement in the clinical course of the uveitis in 19 of the 22 eyes (86.4%), allowing the suspension of the systemic treatment in 16 patients. An improvement of the visual acuity was observed in 20 eyes (90.9%). The most common post-operative complications were, lens opacities in 9 eyes (40.9%), and glaucoma in 4 eyes (18.2%). CONCLUSIONS: The results of this study suggest that pars plana vitrectomy has a beneficial effect on the course and visual function of patients with vitreo-retinal complications associated with pars planitis.

ObjetivoEvaluar el efecto de la vitrectomía pars plana en pacientes diagnosticados de pars planitis.MétodosSe realizó un análisis retrospectivo del curso clínico, las complicaciones postoperatorias y recurrencia de la uveítis tras la realización de vitrectomía pars plana en pacientes con pars planitis.ResultadosSe incluyeron en el estudio 22 ojos de 19 pacientes con pars planitis. El tiempo medio de seguimiento postoperatorio fue de 55,7 meses (rango 7 – 144 meses). Las indicaciones quirúrgicas fueron opacidades vítreas persistentes en 10 ojos, hemorragia vítrea en 9 ojos, y membrana epirretiniana en 3 ojos. En 19 de los 22 ojos (86,4%) hubo una mejoría en el curso de la uveítis que permitió la suspensión del tratamiento sistémico en 16 pacientes. Veinte ojos (90,9%) mejoraron la agudeza visual postoperatoriamente. Las complicaciones postoperatorias más frecuentes fueron opacidad de cristalino en 9 ojos (40,9%) y glaucoma en 4 ojos (18,2%).ConclusionesLos resultados de este estudio sugieren que la vitrectomía vía pars plana tiene un efecto beneficioso en el curso y en la función visual de los pacientes con complicaciones vitreo-retinianas asociadas a pars planitis.
