ABSTRACT

Reliable information on population size is fundamental to the management of threatened species. For wild species, mark-recapture methods are a cornerstone of abundance estimation. Here, we show the first application of the close-kin mark-recapture (CKMR) method to a terrestrial species of high conservation value; the Christmas Island flying-fox (CIFF). The CIFF is the island's last remaining native terrestrial mammal and was recently listed as critically endangered. CKMR is a powerful tool for estimating the demographic parameters central to CIFF management and circumvents the complications arising from the species' cryptic nature, mobility, and difficult-to-survey habitat. To this end, we used genetic data from 450 CIFFs captured between 2015 and 2019 to detect kin pairs. We implemented a novel CKMR model that estimates sex-specific abundance, trend, and mortality and accommodates observations from the kin-pair distribution of male reproductive skew and mate persistence. CKMR estimated CIFF total adult female abundance to be approximately 2050 individuals (95% CI (950, 4300)). We showed that on average only 23% of the adult male population contributed to annual reproduction and strong evidence for between-year mate fidelity, an observation not previously quantified for a Pteropus species in the wild. Critically, our population estimates provide the most robust understanding of the status of this critically endangered population, informing immediate and future conservation initiatives.

OBJECTIVE: Repeated autobiographical narratives have recently received increased attention as measures of the stability of narrative identity. We propose that one way to map change in life narratives is to rate the degree to which the autobiographical meaning of renarrated events changes. We aimed to test the influence of age, traits (openness, extraversion), and event characteristics on how much autobiographical meaning changes. METHOD: In waves 3 and 4 of the MainLife study, 123 participants (15-72 years) narrated their lives twice, 4 years apart. Life events that were told both times were rated for change in autobiographical meaning (n = 531). Multilevel models tested individual and event characteristics as predictors. RESULTS: Autobiographical meaning changed more the more individuals were open to experience, the more recently the events had happened, and the more negative emotions the event narratives contained. It was unrelated to extraversion and to the use of autobiographical arguments. A decrease in change with age was due to older individuals narrating older events. CONCLUSION: Our findings add to understanding how traits and life story are related and underscore the need to further study the role of event characteristics for stability and change in narrative identity.

ABSTRACT

The alarmin interleukin-33 (IL-33) is released upon cell stress and damage in peripheral tissues. The receptor for IL-33 is the Toll/Interleukin-1 receptor (TIR)-family member T1/ST2 (the IL-33R), which is highly and constitutively expressed on MCs. The sensing of IL-33 by MCs induces the MyD88-TAK1-IKK2-dependent activation of p65/RelA and MAP-kinases, which mediate the production of pro-inflammatory cytokines and amplify FcεRI-mediated MC-effector functions and the resulting allergic reactions. Therefore, the investigation of IL-33-induced signaling is of interest for developing therapeutic interventions effective against allergic reactions. Importantly, beside the release of IL-33, heat shock proteins (HSPs) are upregulated during allergic reactions. This maintains the biological functions of signaling molecules and/or cytokines but unfortunately also strengthens the severity of inflammatory reactions. Here, we demonstrate that HSP90 does not support the IL-33-induced and MyD88-TAK1-IKK2-dependent activation of p65/RelA and of mitogen-activated protein (MAP)-kinases. We found that HSP90 acts downstream of these signaling pathways, mediates the stability of produced cytokine mRNAs, and therefore facilitates the resulting cytokine production. These data show that IL-33 enables MCs to perform an effective cytokine production by the upregulation of HSP90. Consequently, HSP90 might be an attractive therapeutic target for blocking IL-33-mediated inflammatory reactions.

ABSTRACT

Reflective functioning (RF) is defined as the ability to infer mental states of others and oneself. While RF has been predominantly studied in attachment research, it might also occur in other autobiographical narratives because of its strong connection to self-organization and self-understanding. Therefore, this study took a first step combining research on RF with developmental narrative research. In a longitudinal lifespan study covering up to three measurements across 8 years and six age groups (N = 172), we aimed to detect RF in entire life narratives to explore its development with age and its contribution to causal-motivational coherence of life narratives. Although scores were initially low, RF could be identified in life narratives, and was found to develop throughout adolescence and to predict life narrative coherence above and beyond age. Results confirm RF as significantly contributing to narrative self-organization, indicating promising new paths in research on autobiographical narratives and self.
