ABSTRACT

Somatogen, a human somatotropic hormone (HSH) prepared with the use of gene engineering (genHSH) does not differ from the highly purified substance of hypophysial hormone (hypHSH) in affinity to the antibodies produced on hypHSH and to the membrane receptor of the rabbit liver. 50 per cent inhibition of 125I hypHSH binding to the antibodies was observed at somatogen concentration of 8.2 ng/ml and hypHSH concentration of 7.9 ng/ml. The affinity constants of somatogen and hypHSH to the receptor amounted to 3.8 X 10(9) and 3.6 X 10(9) M-1, respectively. Somatogen for injections as a medicine contains 94.6 per cent of the hormone monomer and by the level of its homogeneity exceeds hypHSH as a medicine manufactured by Serano (Italy) and genHSH as a medicine manufactured by Kabi Vitrum (Sweden).

ABSTRACT

The human growth hormone gene, containing mouse metallothionein gene promotor, was injected into the male pronuclei of fertilized mouse ova. The progeny of transgenic mice included animals with both accelerated and inhibited growth. Radioimmunochemical analysis has revealed human growth hormone synthesis in both groups of transgenic mice. The molecular weight of the hormone synthesized in liver cells was 25,000 daltons. A possible mechanisms of foreign hormone effect on the growth of transgenic mice is discussed.

ABSTRACT

Main characteristics of 45Ca accumulation by myometrum plasma membrane were obtained. It was shown that bradykinin and D 600 significantly altered calcium accumulation by the membrane vesicles. Bradykinin increases the rate of calcium accumulation and D 600 suppresses general accumulation and abolishes bradykinin stimulation of this process. The obtained evidence indicates that bradykinin interaction with the membrane receptor results in activation of calcium transport across the myometrium plasma membrane.
